Objective:Establish eukaryotic expression vector of glucocorticoid receptor GR,to explore the relation of GR and tradition Chinese medicine.Methods:Recombinant were designed and established Supported by the National Natural Science Foundation of China(3047119)by targeting gene GR and plasmid pBS/U6-Neo based on GR mRNA sequence.Two pairs of oligonucleotides were synthesized and inserted into plasmid pBS/U6-Neo to generate siRNA eukaryotic expression vectors.DH5a strains were transformed.plasmids were extracted,and the recombinant sequences were identified by PCR and sequencing.Results:The result of recombinant sequence was the same as aim sequence.The recombinant vectors were established successfully.Conclusion:siRNA recombinant can be established successfully by RNAi technique,to use in further gene stability transfer and cultivate transgenic mouse.

Objective To explore the influence of Xingnaojing injection on the duration of fever,convulsions and rate of relapse in the treatment of children with febrile convulsion.Methods Reviewed the medical records of 157 patients with febrile convulsion.Patients were assigned to Xingnaojin treatment (n =68) and conventional therapy group according to whether Xingnaojing injection was added in the conventional treatment(n =89).Collected the duration of fever since they accepted treatment in hospital,whether occurred convulsion again and untoward effect,had a follow -up of 6 months,in order to record whether they had relapse.Results In Xingnaojing group,the duration of fever was (2.43 ±1.38)d,which was shorter than (3.75 ±2.24)d of the control group(t =4.28,P <0.05).The occurrence rate of convulsion again in Xingnaojing group was 11.76%,which was lower than 25.83% of the control group(χ2 =4.821,P <0.05).Following -up for 6 months,Xingnaojing group had better rate of relapse (7.35%) than the control group(20.22%) (χ2 =5.108,P <0.05).No untoward effect was observed.Conclusion Xingnao-jing injection can shorter the duration of fever and prevent convulsion,while without obvious untoward effect,could apply to the treatment of children febrile convulsion.

Objective To explore the relationships between life events,automatic thoughts,coping style and internet addiction disorder(IAD) of adolescent . Methods 54 cases of the IAD were assessed with the Adolescent Self-rating Life Events,Automatic Thought questionnaire and Coping Style questionnaire. Results Life events, automatic thoughts,and coping style were significantly correlated with IAD (r =0.312,r =0.325,r =0.295) ( P <0.01).There were significant correlations between life events and automatic thought, life events and coping style(r =0.554,r =0.392) ( P <0.01).The multiple linear stepwise regression revealed that only life events entered on the regression equation when the score of IAD server as dependent variable. Conclusion There may be three ways that life events influence in IAD:life events influence in IAD directly,life events influence in IAD by the mediators,automatic thought and coping style indirectly.Automatic thoughts and life events is not a necessary condition that led to internet addiction and life events may be influence in IAD by other ways.

The cases of cardiac arrest are increasing because of the increased incidence of cardiovascular diseases. Cardiopulmonary resuscitation (CPR) is the necessary measure to rescue these patients. But the success rate of recovery is still low. Acid-base disturbance is one of the important factors affecting the recovery effect. Overseas and domestic studies in recent years focused on the acid-base changes and mechanism in CPR have founded that it exits extraordinary hypercapnia and CO2 spike, which have important guidance to the clinical work. The research for the arterial blood gas analysis during CPR, changes in the exact time, and treatment of hypercapnia and acid-basic change in different etiology and different populations were very few, therefore the a lot of clinical data is needed to accumulate for further study.

OBJECTIVE To evaluate the effect of AG490 on the invasion of Hep2 laryngeal carcinoma cells, and to investigate the role of STAT3 signaling pathway in human laryngeal carcinoma invasion.METHODS Janus kinase inhibitor AG490 was used to inhibit the activation of STAT3 signaling pathway of Hep2 cells.Hep2 cell invasion was detected by quantitative Matrigel invasion assay.The expression of p-STAT3 was measured by flow cytometry. RESULTS The activation of STAT3 signaling pathway and the invasion of laryngeal carcinomas Hep2 cell line were suppressed by AG490.The expression of p-STAT3 and the invasion cell number of Hep2 cell line in the group treated by AG490 for 24h decreased compared with that in control group(P

Recurrent cerebral infarction (RCI) is very common in clinical practice. Intracranial and extracranial vessels diseases closely associate with the recurrence of cerebral infarction. Hypertension, diabetes, high homocysteine, etc., result in vascular stenosis and more in-farction recurrence by atherosclerosis and arterial plaque formation. Study of the etiology of RCI is important for the secondary prevention of cerebral infarction.

Health problems caused by age-related hearing loss can not be ignored, and elderly hearing screening that aimed to detect early and intervene early is of importance. As one of the main screening tools, elderly hearing screening scales are rarely used in China, so the paper mainly reviewed the development and application of hearing screening scales for elderly and provided a reference for future research and clinical application.

OBJECTIVE To discuss the risk factors of hosocomial infection and nursing care in order to prevent and control its occurrence. METHODS The risk factors of nosocomial infections were investigated. RESULTS The passing rate of nosocomial infection monitoring has been significantly improved. Nosocomial infection rate decreased from 5.23% in 2001 dropped to 2.12% in 2007. CONCLUSIONS Only increaseing nosocomial infection understanding among the nursing staff,strengthening accountability,strict aseptic technique rules and self-protection,can avoid and reduce the incidence of nosocomial infections.

Objective To explore the application of comfort care in loop electrosurgical excision procedure (LEEP) in treatment of chronic cervicitis.Methods 130 patients who used LEEP to treat chronic cervicitis were collected from June to December 2011 in our Hospital,then they were randomly divided into the observation group (68 cases) and the control group (62 cases).The observation group used comfort care and the control group used general nursing.The effects of care,operation time,bleeding volume,anxiety and pain degree of the two groups were compared.Results The effect of care in the observation group was superior to the control group.Compared with the control group,the operation time was shortened,the amount of bleeding was decreased,the degree of anxiety and pain was obviously relieved.Conclusions Comfort care can improve the degree of comfort of the patients who used LEEP to treat chronic cervicitis,and is conducive to the rehabilitation of the disease.

OBJECTIVE To investigate the distribution and drug resistance status of urinary tract infections in diabetics and offer scientific basis for reasonable usage of antibiotics.METHODS Pathogenic bacteria isolated from specimens of 236 cases with urinary tract infections were identified and drug susceptibility tests were performed.RESULTS Among 236 strains of pathogenic bacteria,Escherichia coli rated the top one,59.7%.Then Enterococcus rated the second and accounted for 10.6%.The isolates were susceptible to imipenem and amikacin and showed higher drug-resistance to the other common antibiotics.CONCLUSIONS Diabetics easily suffer from urinary tract infections.Drug-resistance of pathogenic bacteria to antibiotics is higher.Therefore,we should take precautions to control the urinary tract infections in diabetics.
