Objective To examine whether the prenatal hypoxia adaptation has any protective effect on the brain of the newborn rat Methods 12 22 day pregnant Wistar rats were randomly divided into 2 groups: the control group and the treated group In the treated group the pregnant rats were placed in a tightly closed hypoxia adaptation chamber When the O 2% in the chamber dropped to 15%, the rat was taken out to breathe fresh air for 5 min then put back in the chamber This process was repeated until its natural delivery In control the chamber was not tightly closed (O 2%=21%) 40 newborn rats weighing 6 8g were selected and subjected to brain ischemia and hypoxia, Left common carotid artrey was ligated under ether anesthesia 2h after recovery from surgery the newborn rats were placed in hypoxia chamber (T=36℃?1℃,O 2%=9%)for 1 5h 24h later they were sacrificed and brain was removed for microscopic examination (optical and electron) and flow cytometry measurement Results In the treated group most newborns were normal There were a few apoptosis cells in early stage The rate of apoptosis was 2 9%, necrosis cells could hardly be seen In the control group, although most neurons were also normal but there were apoptosis cells in early, middle and late stage and even necrosis cells The rate of apoptosis was 9 51%, which was significantly different from that in the treated group Conclusions Prenatal hypoxia adaptation has protective effect on the brain of newborn rat

AIM: This study is designed to demonstrate the drug resistance breast cancer cell line MCF-7/ADM has a higher proportion of cancer stem cells than its original parent cell line MCF-7 in vitro.METHODS: Firstly,the drug resistance of MCF-7/ADM was estimated by MTT method,and then higher proportion of cancer stem cells in MCF-7/ADM than that in MCF-7 was demonstrated by three aspects: side population analysis,sphere culture and cell surface markers of breast cancer stem cells.RESULTS: The drug resistance index of MCF-7/ADM compared to MCF-7 was 37.1.The proportion of side population in MCF-7/ADM and MCF-7 was 9.60%?0.66% versus 0.39%?0.11%;The proportion of sphere-initiating cells in MCF-7/ADM and MCF-7 was 10.27%?0.64% versus 1.03%?0.15%,and the proportion of CD+44CD-24 cells in these two cell lines was 64.87%?3.87% versus 3.70%?0.53%,all are statistically significant.CONCLUSION: ADM resistance breast cancer cell line MCF-7/ADM has a higher proportion of cancer stem cells than that in its original cell line MCF-7.

OBJECTIVE To investigate the role of XIAP in cisplatin-induced apoptosis and its possible mechanism. METHODS Hep-2 cells were treated with different concentrations of cisplatin for different times. Using crystal violet assay, RT-PCR and flow cytometry (FCM), we investingated the rate of the cell apoptosis and the expression of XIAP protein and mRNA at different times after treating with different concentrations of cisplatin. RESULTS Hep-2 cells were adherent and in normal survival condition with very low apoptosis rate. After treating with cisplatin, the rate of inhibition, the expression of XIAP protein and the rate of apoptosis were remarkably different between different concentrations and times. The expression of XIAP protein was down regulated accompanied by the augmentation of the rate of Hep-2 cell apoptosis. The products of RT-PCR were analyzed, demonstrating that the XIAP mRNA was down regulated with the increased concentrations of cisplatin overtime. Correlation analysis showed the rate of inhibition and the rate of apoptosis were positively correlated with increased concentrations of cisplatin for different times, however, the expression of XIAP protein was negatively correlated. The expression of XIAP protein and the rate of apoptosis were conversely correlated. CONCLUSION The most important pathway that cisplatin induces cell death is apoptosis. The levels of expression of XIAP proteinand mRNA in Hep2 cells are time-and concentration-dependent after treating with cisplatin. Down-regulation of the XIAP protein expression to augment the rate of apoptosis is one of the mechanisms for the cisplatin tokill the carcinoma cells.

Objective To evaluate the effect of rocuronium on entropy to endotracheal intubation during anesthesia induction with propofol. Methods Forty patients anesthetized induction with propofol using a target-controlled infusion were randomly divided into two groups: rocuronium group (R group, 20 cases) received 0.6 mg/kg rocuronium or saline group (S group, 20 cases) received saline. 2-3 min later, endotracheal intubation was performed. Response entropy(RE) and state entropy(SE) were recorded during baseline(Ta), at steady state(Tb), 2 min after rocuro nium or saline administration (Tc) and 0, 1, 2 and 3 min after endotracheal intubation (T0, T1, T2, T3). Results At T2, the RE-SE was higher in S group than that in R group. Endotracheal intubation induced increasing in RE and SE. Comparing T2 and T0 values in R group and S group, SE increased from 42 ± 7 to 50 ± 8 and 43 ± 13 to 55 ± 12, and RE increased from 45 ± 6 to 54 ± 9 and 48 ± 16 to 66 ± 15, respectively. At T0, RE and RE-SE were higher in S group. Conclusion Rocuronium affects RE-SE and RE and RE-SE responses to endotracheal intubation and may confound interpretation of entropy monitoring.

BACKGROUND:Alveolar bone remodeling and sustained absorption due to tooth extraction seriously affect the implanting conditions and morphology of hard and soft tissue in implant zone. OBJECTIVE:To evaluate the effect of nano-hydroxyapatite/polycaprolactone electrospinning scaffolds to improve the osteogenic effect of bone defects around immediate implants. METHODS:Tissue-engineered bone was prepared by combining canine bone marrow mesenchymal stem cels with nano-hydroxyapatite/polycaprolactone electrospinning scaffold. Bilateral mandibular second premolars from six dogs were extracted mandibular second premolar, and an immediate implant was placed in the mesial fossa of the mandibular second premolar. Three-wal bone defects was made buccaly using titanium nails, then tissue-engineered bone and Bio-Oss bone powders were implanted bilateraly covered by colagen membranes (Bio-Gide). Imageology examination was performed to measure bone gray levels immediately, 4, 8, 12 weeks after surgery. After 12 weeks, the mandible was removed completely, toluidine blue staining was used for observation of microstructure, new bone formation, bone morphology and implant osseointegration. RESULTS AND CONCLUSION: Between the two groups, there was no difference in bone mineral density at each time point after surgery, indicating that the effects of the two materials to promote bone regeneration process are basicaly the same. After implantation, the dense lamelar bone formed in the bone defect region of tissue-engineered bone group, mature bone cels, Haversian canal, and implant osseointegration were visible. While, in the Bio-Oss group, the lamelar bone was dense, a smal amount of Bio-Oss particles distributed within new bone tissues, fewer bone cels were found, a part of Haversian canal was shown to have blood capilaries, and new bone was in close conjunction with the implant. These findings indicate that the nano-hydroxyapatite/polycaprolactone electrospinning scaffold combined with bone marrow mesenchymal stem cels and Bio-Gide colagen membrane can promote the regeneration of alveolar bone around the implant.

Objective To investigate the distribution and amount of human immunodeficiency virus (HIV) infection in gastric mucosa from untreated acquired immune deficiency syndrome (AIDS) patients and highly active antiretroviral therapy (HAART) treated patients.Methods Thirty-five AIDS patients (14 untreated patients and 21 patients receiving HAART) and 10 HIV-1 seronegative patients with gastrointestinal symptoms were enrolled and examined by upper endoscopy.The labeled HIV-1 double-stranded cDNA probe was a PCR product corresponding to the LTR and gag gene of the HIV-1 genome.HIV in gastric mucosal tissues from AIDS patients was detected using in situ hybridization (ISH) and compared with that in peripheral blood mononuclear cells (PBMC).Results ① No obvious character was found in gastrointestinal symptoms,endoscopy examination and pathology results of AIDS patients.② The expression of HIV gene was mainly detected in the gastric mucosal mononuclear cell (MMC).Other cells were also observed with HIV expression including mucosal epithelial cells,gland epithelial cells and interstitial cells.③There was no difference in HIV expression between sinus ventriculi and gastric body.④ HIV gene expression from AIDS patients was (1.97±3.25)% in gastric mucosa,no difference in HIV gene expression between two groups (P＞0.05).⑤ HIV gene expression in PBMC smear from AIDS patients was (12.38 ± 9.17)%.HIV expreesion in PBMC from patients who had received HAART for 1-4 years were markedly lower than that from patients who had not received HAART (P＜0.05).Conclusions The gastric mueosa is one of HIV infected sites.The potential effect of HAART on the decrease of HIV infected cells in gastric mucosa was unsatisfactory.

Objective To compare and analyze different factors that influence hypertension between subjects with and without gout, and to recognize and understand them further. Methods A total of 7395 patients ( 6935 males and 460 females) from the gout clinic of Affiliated Hospital of Qingdao University between May 2009 and January 2016 were chosen as gout group, while 8379 people without gout (7858 males and 521 females) were served as control group. The height, weight, waist circumference, hip circumference, blood pressure, fasting plasma glucose ( FPG) , triglyceride(TG), cholesterol(TC), blood urea nitrogen (BUN), creatinine(Cr), and uric acid (UA) of both groups were monitored. Clinical and biochemical differences of the two groups were analyzed. The morbidity rate of hypertension in the two groups was also compared. According to different criteria, the subjects were divided into several subgroups. The data were analyzed mainly by Empower statistical software. Results The risk ratio of hypertension in gout group was 63. 25%, and it was higher than that in control group(49. 19%,x2=316. 25,P<0. 01). The risk ratio of hypertension in gout group was 1. 173 times higher than that in control group. After adjusting UA, it would drop to 1. 065 times, but the difference still remained significant. In groups with diabetes, hyperlipidemia, hypercholesterolemia, obesity, the risk ratio of hypertension increased by 13. 7%, 15. 3%, 21. 8%, and 23. 6% respectively, in gout group. FPG, TG, TC, BMI, and WHR were all associated with HP in both gout and normal groups, but Cr was associated with HP in gout group only(OR=1. 396, 95%CI 1. 197-1. 629). Age had different saturation effects in two groups. Conclusion The factors influence hypertension differently in patients with and without gout, especially those of gout itself and creatinine. The precision medicine should be applied.

Objective To investigate the inducing effect of acitrotin on the growth and apoptosis of human cutaneous squamous cell carcinoma cell line SCC13 and on caspases expression.Methods Human cutaneous squa-mous cell carcinoma cell line SCC13 was treated with five different concentrations of acitrefin [5×10-7,1×10-6,5×10-6,1×10-5,5×10-5mol/L].Cell proliferation was evaluated by MTT assay.Apoptosis was assessed by en-zyme-linked immunosorbent assay.The cell cycle was assessed by flow cytometry.The protein expressions of caspase-8 and caspase-9 were examined with Western blot.Results Acitretin inhibited the growth ( F = 83.64,96.34 and 123.17, respectively on the first, third and fifth day)and induced the apoptosis of SCC13 cells(F=74.45,107.37,and 64.28, respectively on the first, third and fifth day) in a dose- and time-dependent manner(P<0.05).Acitretin altered cell cycle distribution of SCC13 cells as compared with controls, the G1-phase population increased by 77.66% 72 hours after acitretin treatment, while the control increased only by 63.55%.An active fragment of caspase-8 occurred following 12 hours treatment of acitretin on SCC13 cells, whereas the caspase-9 active fragment occurred 24 hours after acitretin treatment, which increased time-dependently (P<0.01).Conclusion Acitretin plays an important role on the growth inhibition and apoptosis of cutaneous squamous cell carcinoma cells, which may be affected through both Fas receptor way and mitochondria way.

Objective To investigate the compartmentalization of human immunodeficiency virus (HIV)infection and CD4~+ T lymphocytes between gastric mueosa and peripheral blood from patients with acquired immune deficiency syndrome(AIDS).Methods Thirty-five AIDS patients and 10 HIV seronegative subjects were enrolled in this study.The gastric mucosal biopsies were done by gastroscope and peripheral blood samples were collected.The digoxin labeled HIV-1 double-stranded cDNA probes for the long terminal repeat(LTR)and gag gene of HIV-1 genome were prepared by polymerase chain reaction(PCR).In situ hybridization was used to detect HIV infection in gastric mucosal tissues and peripheral blood mononuclear cells(PBMC)of AIDS patients.CD4~+ T lymphocytes were determined by immunohistochemistry method.The data were analyzed using t test.Results HIV positive rates were(1.67±1.48)% in gastric mucosa and(19.37±9.23)% in PBMC of AIDS patients who had not received highly active antiretroviral therapy(HAART).The HIV positive rates in gastric mucosa were not significantly different between AIDS patients without treatment and those treated with HAART(t=-0.996,t=-0.794,t=-0.461;P＞0.05).HIV positive rate on PBMC film preparation in patients who had received HAART for 1-4 years was (4.25±3.47)%,which was significantly lower than untreated group[(19.37±9.23)%](t=3.000,P＜0.05).The positive rate of CD4~+ T lymphocytes was(12.53±8.14)% in gastric mucosal mononuclear cells(MMC)and(19.00±9.55)% in PBMC of AIDS patient who had not received HAART.Even after received HAART for 1-4 years,the positive rate of CD4 ~+ T lymphocytes in MMC were(37.44±18.00)%,which was still lower than control group[(50.35±3.41)%](t=-4.620,P＜0.01);while the positive rate of CD4~+ T lymphocyte in PBMC was similar with that of control group(t=-2.094,P＞0.05).Conclusion The compartmentalization of HIV infection and CD4~+ T lymphocyte counts exists between gastric mucosa and peripheral blood from AIDS patients.
