1.Interleukin-17 regulates the proliferation of fibroblast-like synoviocytes of rheumatoid arthritis through signal transducer and activator of transcription 3 signaling

Objective To determine the effect of interleukin (IL)-17 on the proliferation of fibroblastlike synoviocytes (FLSs) from rheumatoid arthritis (RA)patients and to investigate whether signal transducer and activator of transcription 3 (STAT3) was implicated in this process.Methods FLSs were acquired by primary culturing from RA patients.STAT3,p-STAT3,Bcl-2 and Bax expression in FLSs were determined using western blotting analysis.The proliferation of FLSs was determined by MTT assay.Analysis of variance was used to determine the differences between two groups.Results IL-17 promoted the proliferation of FLSs from RA patients and inhibited their apoptosis.The pro-apoptotic Bax was decreased and anti-apoptotic Bcl-2 was increased in FLSs from RA (t=2.612,P<0.05;t=2.723,P<0.05).However,targeted silencing of STAT3 could inhibit the effect of IL-17 on FLSs (t=2.745,P<0.05).[the control group:Bax (3.814±1.623),Bcl-2 (1.429± 0.311);IL-17 group:Bax (0.972±0.017),Bcl-2 (3.175±1.356);STAT3 silent group:Bax (5.729±1.236),Bcl-2 (0.637±0.006)].Conclusion STAT3 is critical in IL-17-induced proliferation of FLS from RA patients.IL-17/STAT3 pathway might be a good candidate for RA treatment modalities.
2.Clinical and Pathological Analysis of Acute Renal Failure in Children

Objective To study the etiology, clinical and pathological characteristics, therapy and prognosis of acute renal failure (ARF). Methods The literature of 80 treated children with ARF from 1988. 1 to 2003.10 was reviewed. Results Sixty three cases (78.8%)suffered from mtrarenal ARF;47 glomerulonephritls (58. 8%); 15(18. 8%)prerenal ARF;2(2 5%)postrenal obstructive ARF. Twenty three cases with renal biopsy,9(39. 1%)MsPGN;4 (17. 4% ) MPGN; 4 (17. 4%) crescentic glomerulonephntis; 3 (13% ) endocapillary glomerulonephritls;2(8 7% )proliferative and sclerosing glomerulonephritis; 1(1. 3% )minimal changes. According to the different etiologic and pathological characteristics,different therapies were applied,infusion,oral prednisone ,and intravenous pulses of methylprednisone and cyclosphosphamide ,dialysis, or diuretic therapy and lowering blood. Thirty one (38. 8% )cases recov-ered;27(33 8% )cases improved; 10( 12.5% ) cases gave up treatments;21 (26 3% )died. 4.GC Fingerprint analysis for quality control of volatile oil in Radix Bupleuri

Objective To establish the GC fingerprint analysis for the quality control of volatile oil in Radix Bupleuri. Methods The GC with capillary column DB-1 (30 m?0.25 mm?0.25 ?m) was used. The column was maintained at 50 ℃ for 5 min after injection then programmed at 3 ℃/min to 170 ℃ and at 5 ℃/min to 230 ℃ which was maintained for 5 min. Gasification temperature: 250 ℃, carrier gas: N_2, flow rate: 1.03 mL/min, inlet volumn: 0.5 ?L, spliting ratio: 10∶1, FID Injector temperature: 250 ℃. The internal standard was [WTBX]n-nonane used to determine 25 batches of Radix Bupleuri from different habitats by GC fingerprint. Results The 25 batches of Radix Bupleuri are classified to be the qualified and unqualified based on the results of cluster and similarity analyses. Conclusion The method is simple and reliable and it is capable of effectively controlling the quality of Radix Bupleuri.
5.The effect of mental abacus calculation training on form recognition of children: an ERP study

ObjectiveTo explore the influence of mental abacus calculation (MAC) training on children's form recognition.Methods 28 children ( 14 children with MAC training and 14 children with non-MAC traning)were investigated by using event-related potential(ERP) technology.The event-related potentials were recorded when subjects were in the different form shape recognition.Results ( 1 ) The amplitudes of the posterior P1 evoked by the children with MAC training ( (9.59 ± 3.58) μV) were significantly greater than that of the children without MAC training ( (7.06 ± 2.84) μV) ( P < 0.05 ).(2) The amplitudes of right temporal-posterior N170 of children with MAC( ( -9.83 ±2.97) μV) were markedly less than that of children with non-MAC( ( - 12.45 ±2.51 ) μV) ( P < 0.05 ).( 3 ) The amplitudes of prefrontal P3 of children with MAC ( (7.65 ± 2.25 ) μV) were remarkably greater than that of children with non-MAC ( (4.89 ± 3.35 ) μV) ( P < 0.05 ).ConclusionThe brain function of form recognition in children is influenced by mental abacus calculation training.
6.Effects of permanent magnetic fields on superoxide dismutase and malondialdehyde in human microvascular endothelial brain cells under normal and hypoxic conditions

Objective To observe the effects of permanent magnetic fields with different magnetic inductions on oxidative damage to human brain microvascular endothelial cells (HBMECs) under normal and hypoxic conditions. Methods HBMECs were cultured in vitro under normal and hypoxic conditions,then divided into a control group and groups receiving magnetic induction at 8.1,16.5,20.3,26.0,27.3,62.5,110.7 and 215.6 mT.Changes in superoxide dismutase (SOD) activity and malondialdehyde (MDA) content in each group were measured 72 h after exposure to the magnetic fields. Results Under normal conditions the SOD activities of the magnetic groups were not significantly different from that of the control group. Only the MDA content of the 215.6 mT group was statistically different (slightly higher) than that of the control group (P < 0.05 ).Under hypoxic conditions,there was again no statistically significant difference in SOD activity,but the MDA content of the 110.7 mT group was significantly lower. Conclusions Under normal conditions,a 215.6 mT permanent magnetic field has an effect on MDA in HBMECs; under hypoxic conditions a 110.7 mT permanent magnetic field has such an effect.
7.Clinical Value of Serum Cystatin C and suPAR Assay in Patients with Malignant Tumor

Objective To explore the relationship of serum level of Cystatin C and suPAR with tumor infiltration, metastasis, and treatment of patients with malignant tumor. Methods: The serum levels of Cystatin C was detected by particle enhanced nephelometic immunoassay (PENIA) by 7600-010 full-automatic biochemical analyzer made in Japan. The level of suPAR was detected by ELISA. The serum levels of Cystatin C and suPARof 82 normal adults and 172 patients with malignant tumor were measured and compared. Results: The serum level of Cystatin C and suPAR in patients with malignant tumor was significantly increased compared with that of normal adults (P<0.01 and P<0.01). The level of Cystatin C and suPAR in terminally ill patients or patients with metastasis was significantly higher than that in the control group. The levels of the two indices in postoperative patients were lower than those in preoperative patients. No significant difference was found in the levels of the two indicies before chemotherapy or radiotherapy and after therapy. Conclusion: The serum levels of Cystatin C and suPARin patients with malignant tumor are correlated with tumor invasion, metastasis and surgical intervention. Detection of Cystatin C and suPAR levels in patients with malignant tumor is valuable for disease monitoring and treatment evaluation.
8.Value of procalcitonin in the etiological diagnosis of bloodstream infections

Objective:To evaluate the accuracy of serum concentration of procalcitonin (PCT) in differential diagnosis of the etiology of bloodstream infections (BSI).Methods:Patients hospitalized in ICU of China-Japan Friendship Hospital from January 2015 to June 2020 with BSI and with PCT test simultaneously when blood drawing for blood culture were enrolled. Sequential Organ Failure Assessment (SOFA) were calculated based on parameters on the day of blood culture. Difference of various indicators among different pathogen infections were compared. Receiver Operating Characteristic (ROC) Curve was used to analyze the value of PCT in differential diagnosis of BSI by different pathogens.Results:Among 1 456 patients with BSI,1 261 (86.6%) patients with monobacterial infection, 80 (5.5%) patients with candidiasis and 115 (7.9%) patients with mixed infection. The 28-day mortality was 24.5% (356/1 456) and the 60-day mortality was 30.6% (446/1456). Mortality of both 28-day and 60-day in the mixed group was significantly higher than that in the bacteriacemia group and candidemia group. PCT levels was significantly higher in patients with bacteremia caused by gram-negative bacteria (GNB) than that in gram-positive bacteria (GPB) infected bacteremia and candidemia {3.4 μg/L[95% confidence interval (95% CI) 0.7-17.0 μg/L] vs 1.3 μg/L (95% CI 0.4-7.3 μg/L); 3.4μg/L (95% CI was 0.7-17.0 μg/L) vs 1.1 μg/L (95% CI was 0.4-3.4 μg/L); P<0.01} . ROC curve analysis showed that: ① the optimal cut-off value of PCT in differential diagnosis of monobacterial bacteremia and candidemia was 7.25 μg/L, with specificity of 90.0% and the area under the ROC curve (AUROC) was 0.612 (95% CI 0.533-0.691). When PCT value was greater than 0.51 μg/L, the sensitivity of diagnostic of bacteremia could reach 73.3%. ② the optimal cut-off value of PCT in differential diagnosis of bacteremia caused by GNB infection and candidemia was 7.32 μg/L, with specificity of 90.0% and AUROC was 0.695 (95% CI 0.614-0.776). When PCT value was greater than 0.51 μg/L, the sensitivity of diagnostic of bacteremia caused by GNB infection was 84.9%.③ the optimal cut-off value of PCT in differential diagnosis of bacteremia caused by GNB and GPB infection was 0.52 μg/L, with sensitivity of 84.9% and AUROC was 0.713 (95% CI 0.672-0.755). When PCT value was greater than 7.36 μg/L, the specificity of diagnostic of bacteremia caused by GNB infection could reach 80.1%. Conclusions:PCT can provide additional information about the possible etiology of patients with BSI, especially as high levels often indicate the possibility of GNB bacteremia.
9.Study on hepatocyte-like cells differentiated from human peripheral blood mononuclear cells labeled by the Fluorescent Dye PKH26 in vivo

ObjectiveTo induce human peripheral blood mononuclear cells differentiate into hepatocyte-like cells by hepatocyte growth factor (HGF) and fibroblast growth factor-4 (FGF-4) in vitro and determine whether PKH26 could be used to serve as an effective tracer for the cells,and observe the ability of transplanted hepatocyte-like cells differentiate into hepatic cells in nude mice.MethodsGroup A and B were set up respectively.In Group A,mononuclear cells were cultivated without hepatocyte growth factor (HGF) and fibroblast growth factor-4 (FGF-4) in cell culture.They were used as negative control group.In Group B,mononuclear cells were cultured with the administration of both HGF and FGF-4 to induce the differentiation into liver hepatocyte-like cells.The changes in cell morphology were observed and the expressions of AFP and CK 19 were detected by immunocytochemical staining in two groups at different times after induction.The hepatocyte-like cells differentiated from human peripheral blood mononuclear cells labeled by the fluorescent dye PKH26 injected into caudal vein in nude mice is experimental group.The nude mice injected with equal amount of normal saline in control group.The migration of the labeled cells into the liver are observed by the fluorescence microscope in the hepatic tissue sections of nude mice and the expressions of ALB were detected by immunocytochemical staining two weeks after the cells transplantation.ResultsCells in group B have a strong proliferative activity.It becomes large and oval,grows in colonies following induction.Cells in group A that showed spherical shape when peripheral blood mononuclear cells were just isolated are gradually becoming inconformity in morphology,spindle or fibroid,and a few cells are round:cells developed apoptosis and cracked following incubation.The expressions of AFP and CK19 were positive after induction in group B as detected by immunocytochemicat staining.Inversely,the expressions of AFP and CK19 were negative in group A after incubation.The experimental group showed numerous PKH26 labeled cells in the hepatic tissue sections of nude mice.But the control group did not show PKH26 labeled cells.The expressions of ALB were positive in the experimental group as detected by immunocytochemical staining after two weeks of the cells transplantation.ConclusionHuman peripheral blood mononuclear cells have the potential to differentiate into hepatocyte-like cells under the induction of HGF and FGF-4.Additionally,PKH26 is an effective tracer in hepatocyte-like cell transplantation.The hepatocyte-like cells settled in hepatic tissue begin to differentiate into mature hepatocyte after two weeks of the cells transplantation.It plays hepatic cells function and expresses alhnmin.
10.Observation on the Effect of Lidocaine Treated in Three Different Ways on the Prevention of General Anesthesia in the Elderly Patients

Objective:To compare the three effects of lidocaine in the prevention of general anesthesia in elderly patients.Methods:A total of 120 elderly patients (65-85 years old) underwent anesthesia with general anesthesia (ASA) Ⅰ ~ Ⅱ were randomly divided into thyrocricocentesis group (group H),throat surface anesthesia group (Group Y),intravenous injection group (group J) and control group (group D).Group H was injected with lidocaine for surface anesthesia;group Y used laryngeal spray for laryngeal sprayed lidocaine for surface anesthesia;group J was anesthetized induction of intravenous lidocaine to prevent intubation reaction;group D the control group was not treated with lidocaine.(SBP) and heart rate (HR) were measured before and after induction (T0),tracheal intubation (T1) and 1 (T2),3 (T3) and 5 min (T4),the changes of hemodynamics related indexes were compared.Results:Compared with the same group of T0,the SBP and HR of four groups of T1 moments were significantly decreased (P <0.05);In the other three groups,SBP was significantly increased at T2 and T3 (P <0.05),HR ofT2 was significantly increased (P <0.05);Compared with group D,SBP and HR in group H,Y and J were significantly different at T2,T3 and T4,he difference was statistically significant (P <0.05);but there was no statistically significant difference between the three groups (P> 0.05).Conclusion:The three ways of lidocaine can be used to prevent the general anesthesia reaction in elderly patients,the effect of three ways is parallel.However,cricothyroid membrane puncture increased the patient's pain,throat spray method increased the cumbersome operation and enhanced the cost of the patient,and intravenous injection method is simple and worthy of popularization and application in clinic.
