1.Effect of tissue factor pathway inhibitor-1 on no-reflow phenomenon in rabbit

Objective To observe the effects of different doses of human recombinant tissue factor pathway inhibitor-1 (TFPI-1) on no-reflow (NR) phenomenon in rabbit.Methods Fifty-two New Zealand white rabbits were subjected to coronary artery occlusion for 120 min and followed by reperfusion for 60 min,and then were randomly (random number) assigned into four groups:control group,large,moderate and low doses TFPI-1 groups ( 1000 ng/kg,100 ng/kg,10 ng/kg bolus and thenl0 ng/kg,1 ng/kg and 0.1 ng/kg per minute infusion for maintenance,each group n =13).The no-reflow area (NA) and ischemic area (IA) was measured by thioflavin S and Evan's blue.The NR severity was expressed by NA/IA.The difference in NR severity was compared between groups.The thrombi and myocardial injury were observed under light microscope.The infarction and NR severity in different groups were compared by using one-way ANOVA followed by LSD procedure.Results There were no significant differences in IA and body weight among four groups (P>0.05).NR severity in the large,moderate,low doses TFPI-1 groups and control group were (0.210 ±0.061 ),(0.389 +0.110),(0.478 ±0.077) and (0.536 ±0.061 ),respectively.NR severity in the large dose TFPI-1 group was slightest among the four groups (P <0.01 ).NR severity in the moderate dose TFPI-1 group was significantly decreased than that in control group ( P < 0.01 ) and in low dose TFPI-1 group (P <0.05 ).There was no significant difference in NR severity between the low dose TFPI-1 group and control group ( P > 0.05 ).There was less thrombus formation and lower grade myocardial injury found in the large dose TFPI-1 group. Conclusion Human rTFPI-1 might lessen NR severity in rabbit in dose-dependent,suggesting the option on human rTFPI-1 for treatment of NR phenomenon.
2.Transcriptomic Analysis and Systematic Mining of Genes Involved in Biosynthetic Pathway of Triterpenoid Saponins in Ilex Asprella

This study was aimed to screen candidate genes involved in the triterpenoid saponins biosynthetic pathway of the Ilex asprella root. The Illumina platform was applied to perform transcriptomic sequencing of I. asprella root, followed by a series of bioinformatics analysis. The results showed that a total of 272 candidate unigenes were anno-tated to be involved in the biosynthetic pathway of terpenoid in the transcriptome of I. asprella root, including 72 u-nigenes for the upstream pathway and 26 unigenes for cyclization, oxidation and glycosylation in the downstream pathway. Phylogenetic analysis was carried out to further analyze the evolution relationship of some candidate uni-genes and their homologous genes. Two genes IaA S1 and IaA S2 were proved to be mixed amyrin synthases in yeast expression system. Moreover, IaA S1 was identified to one of the rare ASs with α-amyrin as the major product. It was concluded that a series of candidate genes, which might be involved in the biosynthetic pathway of triterpenoid saponins, were screened out from the transcriptome of I. asprella root. Further investigation of these candidate genes will provide insight into their actual functions in the triterpenoid saponins biosynthetic pathway in I. asprella.
3.Analysis of the cognition of medical staff on scientific research ethics and its influencing factors: A case study of a 3A-grade general hospital in Fujian Province

Objective:To explore the status quo and influencing factors of scientific research ethics cognition of medical staff in 3A-grade hospitals in Fuzhou, and to put forward suggestions for strengthening the construction of scientific research ethics in hospitals.Methods:A questionnaire survey was conducted using a self-designed questionnaire, and the data were statistically analyzed using SPSS 25.0 software.Results:Among the 478 respondents, 184 (38.49%) did not participate in ethics training, 123 (25.73%) were not aware of the organization of their hospital ethics committee, and 47 (9.83%) thought that carrying out ethics review was beneficial to patients but not to scientific research. Respondents with higher education, previous experience with ethics application review, who would consider relevant research ethics issues during clinical study protocol design and development, and who believed that ethical review of medical research would benefit both patients and research had a better overall cognition of research ethics ( P<0.05). Conclusions:Medical staff's cognition of various aspects of scientific research ethics is relatively weak. Hospitals should take multiple initiatives to improve the efficiency of ethical training, optimize the ethical review mechanism, and strengthen the construction of scientific research ethics.
4.Research progress of visual art therapy in patients with dementia

This article summarizes the concept, implementers, intervention subjects, intervention methods, intervention mechanisms, theoretical basis and application effects of visual art therapy (VAT) . This article also proposes training professional implementation personnel, screening patients through the community, promoting rehabilitation through multidisciplinary cooperation, establishing interest groups with peer support, setting up relevant courses in colleges, and cooperation between nursing home and universities to promote the promotion and application of VAT in patients with dementia in my country.
5.Evaluation of subjective scale in allergen nasal provocation test.

Objective:To compare the clinical value of visual analogue scale (VAS), Lebel scale and total nasal symptom scores (TNSS) in evaluating nasal allergen provocation test (NAPT). Methods:A total of 151 patients suspected of allergic rhinitis admitted to the Department of Otolaryngology-Head and Neck Surgery of our hospital from April 2020 to September 2020 were included, of which 76 were positive for house dust mites and 75 were negative for allergens. Nasal airway resistance(NAR) was measured by active anterior nasal manometry. Nasal symptoms were evaluated by VAS, Lebel and TNSS. House dust mite allergen was used for NAPT by spray method. An increase≥40% in NAR was used as the gold standard for objective evaluation of NAPT. ROC curves of VAS, Lebel and TNSS were drawn to compare the evaluation effectiveness of different subjective evaluation methods, and the optimal critical point of each ROC curve was obtained. Results:With NAR increased by ≥40% as the gold standard, the area under ROC curve of VAS was 0.884, and the sensitivity and specificity were 97.75% and 80.65%, respectively. The area under ROC curve of Lebel was 0.773, and the sensitivity and specificity were 68.54% and 75.81%, respectively. The area under ROC curve of TNSS was 0.792, and the sensitivity and specificity were 68.54% and 79.03%, respectively. There was no significant difference between Lebel and TNSS(P>0.05). The VAS differed significantly from Lebel and TNSS(P<0.05). The Kappa values of VAS, Lebel, TNSS and NAR were 0.803, 0.432 and 0.459, respectively. Conclusion:The VAS, Lebel, TNSS subjective scale and NAR are consistent in evaluating the efficacy of NAPT, with the VAS assessment showing highest consistency with NAR. As objective assessment instruments are not widely used in China, subjective assessment method could be adopted to evaluate the efficacy of NAPT in clinical practice, and VAS scale is recommended as a priority.
