1.Effects of road transport on hematological and biochemical parameters in new zealand rabbits

Objective To observe the effects of road transport on hematological and biochemical parameters in New Zealand rabbits.Methods A total of 12 healthy New Zealand rabbits were selected for 2 h road transport.Blood samples were collected at 0, 24, 48, 72 and 96 h after transport, respectively.White blood cells (WBC), red blood cells (RBC), hemoglobin (HGB), hematocrit (MCV), mean erythrocyte hemoglobin content (MCH), mean erythrocyte hemoglobin concentration (MCHC) and platelets (PLT) were measured using a blood analyzer.Blood alanine aminotransferase (ALT), aspartate aminotransferase (AST), albumin (ALB), total protein (TP), urea nitrogen (UREA), creatinine, uric acid (UA), triglycerides (TG), total cholesterol (COHL), glucose (GLU), hypersensitive C-reactive protein (CRP), α-amylase (AMYL), and creatine kinase (CK) were detected by an automatic biochemical analyzer.Results Compared the parameters before and after transport, The WBC count was increased first (P< 0.05 or P < 0.01) and then decreased after transport, the levels of RBC, HGB, HCT and PLT were decreased first (P< 0.05 or P < 0.01) and then increased after transport, and MCV was significantly high at 96 h after transport (P< 0.05).Among the clinical biochemical parameters, ALT, AST and BUN were firstly elevated (P< 0.05 or P < 0.01) and then decreased.TP, ALB as well as CREA and TG were firstly decreased (P< 0.05 or P < 0.01) and then increased.GLU was significantly low at 24 h after transport (P< 0.05).All parameters except MCV at 96 h after transport were not significantly different from those before transport.Conclusions Changes of blood routine, liver and kidney function indexes, lipid metabolism indexes, glucose metabolism index and creatine kinase index are observed in the New Zealand rabbits after 2-hour road transportation, and all the indicators except MCV return to pre-transport levels within 96 h.
2.Hemodynamic analysis in the fetuses with ductus arteriosus constriction or closure by conventional fetal echocardiography combined with fetal heart quantification technology

Objective:To evaluate the heart hemodynamics in fetuses with premature ductus arteriosus constriction or closure using fetal heart quantification (FHQ).Methods:The clinical data of 50 singleton fetuses with ductus arteriosus constriction ( n=35) or ductus arteriosus closure ( n=15) who underwent echocardiography in Department of Ultrasound, Beijing Anzhen Hospital were retrospectively analyzed, from May 2013 to January 2020. Fifty healthy singleton fetuses were randomly selected as the control group. The ductus arteriosus diameter (DA), pulsatility index (PI), diameter of the left atrium(LA) and right atrium(RA), diameter of the left ventricle (LV) and right ventricle (RV), tricuspid regurgitation/right atrium area ratio (TR/RA Ratio), pressure gradient of tricuspid regurgitation (PG of TR), and heart/chest ratio were measured using conventional fetal echocardiography; the correlations among the parameters were analyzed. Speckle-tracking analysis was used to analysis and compute the LV and RV global spherical index (GSI), fractional area change (FAC) and global strain (GS), the LV ejection fraction(EF) and stroke volume (SV). These variables and their correlations were compared and analyzed. Results:Compared with the control group, the GS and FAC of the LV and RV in the ductus arteriosus constriction or closure groups were lower ( P<0.05) while the LV-SV was higher ( P<0.05). The FAC, GS, and EF values of the LV were higher in the premature ductus arteriosus closure group than in the ductus arteriosus constriction group ( P<0.05), while the RV-FAC was lower ( P<0.05), the RV-GS and LV-SV showed no significant changes ( P>0.05). Correlation analyse showed that the PI was positively correlated with DA( r=0.364, P<0.05); the PG of TR was negatively correlated with DA( r=-0.414, P<0.05); the TR/RA Ratio was negatively linearly correlated with PI( r=-0.388, P<0.05), and positively correlated with RV/LV Ratio ( r=0.369, P<0.05); the other parameters were not significantly correlated with the DA or PI ( P>0.05). Conclusions:Fetal heart hemodynamics in the premature ductus arteriosus constriction or closure groups change significantly, FHQ can provide valuable information for the evaluation of the fetal heart with ductus arteriosus constriction or closure.
3.Study on the correlation between the acceptance of disability and the social support in patients with permanent colostomy

Objective To explore the correlation between the acceptance of disability and the social support in patients with permanent colostomy .Methods One hundred and twenty-four patients with colostomy were surveyed by the acceptance of disability scale ( AODS ) and social support rating scale ( SSRS ) . Results The total score of AODS in patients with permanent colostomy was (82.10 ±8.86), and was overall in the middle level; the total score of SSRS was (41.80 ±2.87), and was higher than (34.56 ±3.73) in the national norm, and the difference was statistically significant (t=28.11, P<0.01).The total score of AODS was positively correlated with the total score of SSRS (r=0.332, P<0.01), and the score of tolerance dimension in the AODS was positively correlated with subjective support (r=0.327, P<0.01).Conclusions The acceptance of disability in patients with permanent colostomy is needed to improve , and the nurses should explore the effective psychological intervention mode according to its related factors in order to improve the level of psychological health in patients with colostomy .
4.A correlative study on self-efficacy and positive aspects among caregivers of disabled elderly

ObjectiveTo investigate the level of positive aspects and current status of their general self-efficacy among the family caregivers of disabled elderly,and to explore the correlation between them. MethodsBy convenience sampling, a cross-sectional survey was conducted among 120 family caregivers of disabled elderly in Fangzhuang Community, Heping Li Community, Tiancun Community and Peking Union Medical College Hospital from April to June 2018 by Self-designed General Questionnaire,Activities of Daily Living Scale(ADL), General Self-Efficacy Scale(GSES) and Positive Aspects of Caregiver(PAC) to investigate the current status of the caregivers' self-efficacy and positive aspects and analyze the correlation between them using statistical software. ResultsThe 120 caregivers of disabled elderly got (27.98±7.82)points from GSES, which is at a moderate level; the caregivers achieved (35.60±8.16)points from PAC among which the score of"self-affirmation"dimension was (19.93±4.51) points, and "life prospects" dimension was (15.68±4.24)points;the total score of GSES and PAC as well as all dimensions of the family caregivers of disabled elderly showed a moderate level of positive correlation (r=0.524, 0.435, 0.541;P<0.01). ConclusionsThe family caregivers of the disabled elderly had positive aspects which correlated positively with their general self-efficacy. Medical staff can take corresponding measures to improve the family caregivers' self-efficacy and positive aspect so as to improve the quality of their care and as well as the life quality of the elderly.
5.Microbiological culture of skin lesion exudates and drug sensitivity test in 176 inpatients with pemphigus

Objective:To analyze results of microbiological culture of lesion exudates and drug sensitivity test in patients with pemphigus, and to optimize the management protocol of pemphigus.Methods:Inpatients with pemphigus were collected from Department of Dermatology and Venereology, the First Affiliated Hospital of Anhui Medical University from January to December in 2008, and the results of microbiological culture of skin lesion exudates and drug sensitivity test were retrospectively analyzed.Results:A total of 176 patients were collected, and skin lesion exudate culture yielded 25 kinds of bacteria and 11 kinds of fungi in 156 (88.64%) patients. Of the 156 patients, 1 kind of pathogen was yielded in 104 (66.67%), and 2 or more kinds of pathogens were yielded in 52 (33.33%). Gram-positive cocci were detected in 134 (76.14%) cases, including Staphylococcus aureus in 80, Staphylococcus hemolyticus in 21, and Staphylococcus epidermidis in 16; Gram-negative bacilli were detected in 66 (37.50%) cases; fungi were detected in 24 (13.64%) cases. In addition, 89 (93.68%) of 95 relapsed patients showed positive culture results, 67 (82.72%) of 81 patients with newly developed pemphigus had positive culture results, and the positive rate significantly differed between the two groups ( χ2 = 5.22, P <0.05). Drug sensitivity test for Staphylococcus aureus isolates showed that 71 (97.26%) of 73 were sensitive to vancomycin, 69 (97.18%) of 71 to linezolid, and 61 (87.14%) of 70 to rifampin. Conclusion:Cutaneous infection, especially Gram-positive cocci infection, is very common in patients with pemphigus, and Staphylococcus aureus clinical isolates were highly sensitive to vancomycin, linezolid and rifampicin.

OBJECTIVE To study the unknown impurity in potassium aspartate and magnesium aspartate injection by heart-cutting two-dimensional liquid chromatography coupled with Quadrupole time-of-flight mass spectrometry (2D-LC-Q-TOF-MS). METHODS The Waters ACQUITY UPLC 2D system and Xevo G2-XS QTof-MS system were used. One- dimensional chromatographic conditions were as listed. An Agilent ZORBAX-NH2 column was used, and potassium dihydrogen thophosphate(0.05 mol·L-1)-acetonitrile(37∶63) as the mobile phase, at a flow rate of 1.3 mL·min-1. The detection wavelength was set at 200 nm. Two-dimensional chromatographic conditions were as listed. A Waters ACQUITY UPLC HSS T3 column was used, and 0.1% formic acid solution(ESI+)/5 mmol·L-1 ammonium formate(ESI-) as the mobile phase A, and acetonitrile as the mobile phase B, in gradient mode. The mass spectrometry used an electro spray ionization source(ESI) for positive and negative ion mode detection. RESULTS Combined the high resolution mass, MS2 fragment ions and the UNIFI software, the possible structure of the unknown impurity in potassium aspartate and magnesium aspartate injection was infered. CONCLUSION The 2D-LC-Q-TOF-MS method can be used to identify impurities in potassium aspartate and magnesium aspartate injection, which provides reference for the improvement of quality control and process optimization of potassium aspartate and magnesium aspartate injection.
8.Determination of Fourteen Fluorescent Whitening Agents in Cosmetics by HPLC-MS/MS

OBJECTIVE To establish a method for the determination of fourteen fluorescent whitening agents in cosmetics by HPLC-MS/MS. METHODS Samples were extracted on a Waters ACQUITY UPLC ®BEH C18(2.1 mm×50 mm, 1.7 μm) column after ultrasonic extracted by DMF with the mobile phase consisted of methanol-0.1% ammonia water solution by gradient elution. The flow rate was 0.3 mL·min-1. The column temperature was 40 ℃. MS was performed using triple quadrupole mass spectrometry. Electrospray ionization source was operated in the positive/negative mode using multiple reaction monitoring scanning mode. RESULTS The results showed that there were good linear relationships for the fluorescent whitening agents in a certain concentration range with correlation coefficients(r) greater than 0.99. The limits of quantification were 0.01-20 μg·g-1 and the limits of detection were 0.004-8 μg·g-1. The average recoveries at three spiked levels were in the range of 85.4%-108.9%, and the relative standard deviation were in the range of 0.3%-7.2%. CONCLUSION The method has high sensitivity, strong specificity, simple and convenient operation, and is suitable for the detection of fourteen fluorescent whitening agents in cosmetics.

AIM: To investigate the pharmacokinetic properties of the main active components of Dalitong extract in SD rats after oral administration using UPLC-MS / MS. METHODS: An UPLC-MS / MS method was established to simultaneously detect tetrahydropalmatine, nobiletin and costunolide in the plasma and tissues of SD rats. The method was applied to investigate the pharmacokinetic characteristics and tissue distribution. RESULTS: After a single oral administration, the three active components were rapidly absorbed into the body, with a peak concentration (Cmax) of (13.73 ± 7.50), (27.01 ± 17.69) and (6.73 ± 29.94) ng / mL for tetrahydropalmatine, nobiletin, and costunolide, respectively. The time to reach the peak concentration (Tmax) was (1.40 ± 0.93), (0.63 ± 0.28) and (2.38 ± 8.81) h, respectively. Objective:
The urine concentrations of phthalate metabolites were used to estimate the cumulative risk assessment in preschool children in Ma’anshan of Anhui province.
Methods:
Based on the China-Anhui Birth Cohort, the demographic information and urine samples of 3 743 children were collected in Ma’anshan from April 2014 to April 2015. The concentrations of 7 metabolites’ [monomethyl phthalate (MMP), monoethyl phthalate (MEP), monobutyl phthalate (MBP), monobenzyl phthalate (MBzP), mono (2-ethylhexyl) phthalate (MEHP), mono (2-ethyl- 5-oxohexyl) phthalate (MEOHP) and mono (2-ethyl-5-hydroxyhexyl) phthalate (MEHHP)] of 5 phthalates [dimethyl phthalate (DMP), diethyl phthalate (DEP), dibutyl phthalate (DBP), butyl benzyl phthalate (BBzP), and di (2-ethylhexyl) phthalate (DEHP)] in the urine samples of the children were measured by solid-phase extration-triple quadrupole high performance liquid chromatography-tandem mass spectrometry-isotope method. In addition, the estimated daily intakes (EDIs) of 5 phthalates were calculated according to the metabolites’ concentrations. Cumulative risk assessment was performed using hazard quotient (HQ) and hazard index (HI) methods.
