1.Clinical significance of the defermination of serum leptin and soluble leptin receptor levels in patients with lupus erythematosus

Objective To investigate the possible association of serum leptin(Lp)and soluble leptin receptor(sLR)levels in patients with different clinical types of lupus erythematosus(LE)and the disease activity of system lupus erythematosus(SLE).Methods 39 patients with LE were randomly enrolled from Department of dermatology of Sheng Jing Hospital of China Medical University and No.1 Hospital of China Medical University from 2004 to 2005,and 31 normal controls(age-,sex-and body mass index(BMI)-matched)were enrolled in this study.Fasting blood samples were obtained,and serum Lp levels were measured by using radioimmunoassay,and sLR levels were measured by using enzyme-linked immunosorbent assay(ELISA).Results(1)The patients had significantly higher serum Lp levels,but lower sLR levels than that of the control group(P0.05).(2)Lp was in positive relation with BMI,but in negative relation with sLR.The disease activity of SLE showed positive correlation with the serum Lp levels,but negative correlation with sLR levels by Kendall's correlation analysis(P
2.Fabrication of IgG Functionalized Mn Doped ZnS Quantum Dots for Low Background Phosphorescent Sensing of IgG in Human Serum

IgG functionalized Mn doped ZnS quantum dots ( IgG-QDs ) were fabricated via the amide condensation between the carboxy groups on the surface of Mn doped ZnS quantum dots and the amino groups on the surface of IgG. The IgG antibody functionalized Au nanoparticals were synthesized via the electrostatic interaction between the Au nanoparticals and IgG antibody. Based on the fluorescence resonance energy transfer effect between the IgG-QDs and the IgG antibody functionalized Au nanoparticals, the IgG-QDs could be applied for low background phosphorescent sensing of IgG with the linear range of 1-10 μg/mL and the limit of detection of 0. 45 μg/mL. The proposed method was successfully applied to detect IgG in human serum without the disturbance of the background fluorescence and scattered light.

Objective To study the anti-proliferation and apoptosis induction effect of cisplatin combined with Astragalus on human laryngeal cancer Hep-2 cells.Methods The proliferation inhibition of cisplatin and Astragalus alone or in combination on Laryngeal cancer Hep-2cells was measured by MTT assay.Effects of cisplatin and Astragalus alone or in combination on apoptosis of Hep-2 cells were detected by flow cytometry (FCM).Western blot was used to analyze the expressions of Bcl-2 and Bax proteins.Results The inhibition ratio of proliferation of Hep-2 cells was (53.83 ± 17.12) % in the astragalus group,(69.12 ± 27.12)% in the cisplatin group,and (84.55 ± 27.84)% in the cisplatin combined with Astragalus group,and was significantly greater than the control group (0%) (t =16.87,16.67,40.90,P <0.01),respectively.The apoptotic ratio of Hep-2 cells was (38.2 ± 13.6)% in the astragalus group,(67.2 ± 17.8)% in the cisplatin group,and (86.4 ± 25.1)%] in the cisplatin combined with Astragalus group,and was significantly greater than control group (17.1 ± 1.3) % (t =8.11,12.77,24.92,P <0.05),respectively.The effect in the combination group is better than the other group (t =11.33,9.37,P < 0.01).The expressions of Bcl-2 and Bax proteins were changed.Conclusions The tumor-killing effect of cisplatin on laryngeal cancer Hep-2 cells could be enhanced significantly by the combination application of astragalus by the way of regulating the expression of Bcl-2/Bax.
4.Fetal adrenal area space-occupying lesions:prenatal diagnosis on ultrasound and prognosis

Objective To explore the prenatal ultrasonic types ,sonographic features of malformation , ultrasonic diagnosis and prognosis of fetuses with adrenal area space‐occupying lesions . Methods Forty‐four fetuses with adrenal area space‐occupying lesions were retrospectively analyzed . Compared with postpartum neonatal special inspection results or the anatomical and pathological examination results of the corpses after induced labor ,the ultrasound types ,sonographic feature ,the key points of differential diagnosis were summarized . The newborns were followed up to 6 months after birth ,tracking the prognosis . Results Totally 44 fetuses among 166 471 fetuses(166 471 pregnant women) were detected with adrenal area space‐occupying lesions , 41 cases after delivery , including 24 cases adrenal hematoma ( 2 cases prenataly misdiagnosed as neuroblastoma) ,14 cases adrenal cyst and 3 cases neuroblastoma ,were accurately diagnosed through color Doppler or MRI or CT or pathological examination results . Three cases after induced labor , including 1 case adrenal neuroblastoma ,1 case mullerian duct cyst and 1 case adrenal hematoma( prenataly misdiagnosed as neuroblastoma ) were confirmed through the anatomical and pathological examination results of the corpses . So the conformity rate of adrenal area space‐occupying lesions was 90 .9% (40/44) . The postnatal ultrasound showed that the tumor disappeared in twenty‐four cases of adrenal hematoma without surgery ,and the longest time of the tumor disappearance was 1 year . The size of 14 cases adrenal cyst had no obvious change . One case of adrenal neuroblastoma was operated at 2 months ,2 cases were operated at 2 years of age . Conclusions Ultrasound can be used as qualitative diagnosis and differential diagnosis for fetal adrenal area space‐occupying lesions . The majority of the children with adrenal area space‐occupying lesions have a good prognosis .

AIM: To investigate the role of the nucleus tractus solitarius (NTS) in the regulation of paraventricular nucleus (PVN) AVP-ergic neurons on gastric ischemia- reperfusion injury (GI-RI). METHODS: Male SD rats were used in experiments. The celiac artery were clamped for 30 min and reperfused 1 h by removal of the clamp to obtain the ischemia-reperfusion state. Objective:To study the bioactivity of thymosin ?16 (T?16) in vitro and in vivo.Methods:Recombinant His-SUMO-T?16 was constructed and transformed into E.coli BL21(DE3) for induced expression.The product was treated by ultrasonication,ion-exchange chromatography and metal chelation chromatography respectively for purification.The fusion protein was cut by His-SUMO protease and then further purified by metal chelation chromatography and Superdex 30 gel chromatography.Results:Recombinant fusion protein His-SUMO-T?16 was soluble,whose specific activity was 5.3?105 U/mg.It could promote the proliferation of BALB/c 3T3 cells,rabbit corneal cells,and chicken embryo chorion vessels in vitro,and both the proliferation and migration of vascular endothelial cells in vitro were enhanced,and rabbit skin healing of alkali burns in vivo was accelerated.Conclusion:E.coli expressing vector of recombinant His-SUMO-T?16 fusion protein is constructed successfully,and recombinant protein T?16 has significant repairing effects.The study established a good foundation for further industrialization of T?16.
7.Study on the application of big cup membranate stent on restenosis after stenting for carcinoma of esophago cardia

Objective To evaluate the clinical value of self-designed big cup membranate stent on restenosis after stenting for carcinoma of esophago-cardia. Methods 12 cases of restenosis after stenting for carcinoma of esophago-cardia were involved in the study. Self-designed big cup membranate stent made by Nanjing Weichuang Company(the length of the cup was 3.5 cm)was placed into the constricted stent under guidance of fluoroscopy. Clinical effect, restenosis, and complications were observed during followed up. Results All the 12 cases of big cup membranat stent placement went along smoothly without indigitation of the cup of the stent. Follow-up of 1.5-8 months showed that 2 cases developed severe restenosis on the big cup of the stent, resulting in third grade difficult deglutition. Among them, one occurred 1 month after stenting, caused by hyperplasia of large amount of granulation tissue; another occurred 6 months after stenting, caused by growth of tumor tissue. 3 cases developed mild to moderate restenosis, 2.3-7 months(mean: 4.6 months)after stenting, with result of first grade difficult deglutition. The remaining 7 cases(mean5.6 months follow-up) did not have difficulty during deglutition. Conclusions Application of big cup membranate stent on restenosis after stenting for carcinoma of esophago-cardia can effectively prevent the stent from moving downwards and thus lower down the rate of restenosis, and postpone the occurrence of restenosis.
8.The neuroregulatory mechanism of lateral hypothalamic area on gastric ischemia-reperfusion injury in rats

AIM: To determine the effect of electrical stimulation of lateral hypothalamic area(LHA) on gastric ischemia-reperfusion injury(GI-RI) in rats and to analyse its possible neuroregulatory mechanisms. METHODS: The methods of electrical stimulation(ES), chemical stimulation, electrolytic lesion(EL) and denervation were used to investigate the effect of LHA ES on gastric mucosal injury in rats subjected to 30 min gastric ischemia followed by 60 min reperfusion and to analyse the role of dorsal vagal complex(DVC), vagus and sympathetic nerve in this effect. RESULTS: ①Electrical stimulation of LHA and microinjection of L-glutamic acid into LHA obviously aggravated GI-RI; ②Electrolytic lesion of the LHA attenuated the GI-RI;③DVC lesion eliminated the effect of electrical stimulation of LHA on GI-RI; ④Vagotomy or sympathectomy eliminated the effect of electrical stimulation of LHA on GI-RI too.CONCLUSION: These results indicate that the LHA is a specific area in the CNS for exerting aggravative effects on the GI-RI; the DVC, vagus and sympathetic nerve may be involved in regulatory effects of LHA on GI-RI.
9.Clinical study of treatment of peri-chemotherapy nausea and vomiting of cancer patients using thalidomide

Objective To explore the therapeutic effects and adverse reactions of thalidomide in the treatment of peri-chemotherapy nausea and vomiting of cancer patients.Methods Total of 70 patients were randomly divided into two groups:the treatment group (38 cases) and the control group (32 cases).The treatment group was treated with thalidomide (oral administration at a dose of 100 mg per night,then dose can be added by 50 mg until the top dose of 200 mg per day).The original will be maintained if they cannot be tolerate of extensive dose.The treatment group was also injected 2mg tropisetron in 30 minutes before chemotherapy.The control group was only injected same dose tropisetron.All cases were examined antiemetic effects and evaluated adverse reactions.Results Nausea and vomiting control rates were 89.5 % (34/38) and 68.8 % (22/32) respectively in the treatment group and control group respectively with significant difference.The adverse reactions were similar between the two groups.Conclusion Thalidomide joint tropisetron can effectively control the peri-chemotherapy nausea and vomiting,and the adverse reactions can be acceptable.It could improve further indications of the drug.
10.Multislice spiral CT and digital subtraction angiography in interventional therapy of hepatocellular carcinoma

Objective To investigate the value of multislice spiral CT(MSCT,)in interventional therapy of the hepatocellular carcinoma(HCC)emphasising on transcatheter hepanc arterial chemoembolization(TACE).Methods MSCT were performed in 60 cases of HCC before interventional procedure,CT findings of hepatic artery phase,portal venous phase and hepatic venous phase were observed respectively,among which CTA were done in 15 cases,and the anatomy of celiacartery and its branches were observed in 45 cases.The schemes of interventional therapy were worked out according to the findings of MSCT.Results MSCT showed 250 lesions,10 cases of tumor thrombosis in portal vein and 19 cases of hepatic arterioportal shunt.There was no significant difference between MSCT and digital subtraction angiography(DSA)in positive rate of in showing number of tumor or tumor thrombosis in portal vein(P>0.05),but the 3D construction of celiac artery branches in CTA was better than that in DSA,while angles between celiac artery and abdominal aorta in MSCT were more convenient than that in DSA.MSCT showed 5 eases of hepatic artery original abnormality,according to that in DSA.Conclusion MSCT is of importance for guidance of interventional therapy of hepatocellular carcinoma.
