3.Clinical Observation on Treatment of 27 Cases of Blepharospasm with Acupuncture

Objective To observe clinical effect of treating blepharospasm with acupuncture. Methods 27 patients with blepharospasm were randomly recruited into a control group and a treatment group. The control group was treated with western medicine, and the treatment group was treated with acupuncture. The clinical effects of both groups were observed. Results Total effective rate in the treatment group and the control group was 85.7% and 69.2%, respectively. There was a significant dieffference between the two groups (P<0.05) . Conclusion The treatment of acupuncture on blepharospasm is better than western medicine.

Objective To study the changes and roles of serum anticardiolipin antibodies (ACA) and plasma TXB_2, 6-K-PGF_ 1? in acute cerebral infarction (ACI).Methods Serum ACA and plasma TXB_2, 6-K-PGF_ 1? levels were measured by enzyme linked immunosorbent assay (ELISA) in 143 patients with ACI and were compared with 59 healthy controls. The patients with ACA positive in ACI group were further divided into subgroupⅠwith stroke risk factors and subgroup Ⅱ without stroke risk factors. Objective To observe clinical effect of treating ophthalmoplegia with acupuncture combined with point injection. Methods 25 cases patients with ophthalmoplegia were randomly recurited into a control group and a treatment group. The control group was treated with western medicine and the treatment group was treated with acupuncture combined with point injection. The clinical effect of both groups was observed after treatment. Results The clinical effect in the treatment group was significantly higher than that of the control group (P<0.05) . Conclusion The treatment of ophthalmoplegia with acupuncture combined with point injection has a good effect.
6.Experimental Study on the Anti-aging Effect of Yinao Capsule

Objective To observe the anti-aging effect of Yinao capsule. Methods The rat model of acute incomplete cerebral ischemia was established, and determined the content of superoxide dismutase (SOD), lipid peroxidation (LPO) and Evans blue in brain tissue, determined the serum SOD content of normal rats. Formed blood platelet thrombus by common carotid artery-external jugular vein blood flow bypass operation of rats, and weighed wet weight of thrombus. Results The Yinao capsule can increase SOD and Evans blue content of brain tissue from rat model of acute incomplete cerebral ischemia, at the same time, decrease the LPO content. It can enhance serum SOD content of normal rats, and inhibit the formation of experimental thrombus. Conclusion The Yinao capsule can prevent the occurrence of senile diseases, and defer organism aging.
7.The proliferative effect on silencing of endogenetic mdr1 gene expression by RNA interference in ovarian cancer resistant strain SKOV3/TAXOL

Objective To construct the short hairpin RNA recombinant plasmids targeting mdr1 gene which expresses highly in ovarian cancer resistance strain SKOV3/TAXOL to silence endogenefic mdr1 gene expression and investigate the role of mdr1 gene in the development of resistant ovarian cancer. Methods The pGPU6/GFP/Neo-mdr1 were constructed by gene clone technology. The influence on proliferation and apoptosis were investigated by CCK-8 in SKOV3/TAXOL after transfected pGPU6/GFP/Neo-mdr1. Results The expression against mdr1 proteins were inhibited by pGPU6/GFP/Neo-mdr1. The cell proliferation were inhibited after transfected pGPU6/GFP/Neo-mdr1 by CCK-8. The apoptosis were observed in DAB experiments and the apoptosis rate increased. Conclusion mdr1 plays an important role in proliferation of resistant ovarian cancer and the short hairpin RNA of mdr1 can efficiently suppress mdr1 expression and enhance the apoptosis in SKOV3/ATAXOL.
8.Study on the expression,purification and bioactivity of recombinant T?16

Objective:To study the bioactivity of thymosin ?16 (T?16) in vitro and in vivo.Methods:Recombinant His-SUMO-T?16 was constructed and transformed into E.coli BL21(DE3) for induced expression.The product was treated by ultrasonication,ion-exchange chromatography and metal chelation chromatography respectively for purification.The fusion protein was cut by His-SUMO protease and then further purified by metal chelation chromatography and Superdex 30 gel chromatography.Results:Recombinant fusion protein His-SUMO-T?16 was soluble,whose specific activity was 5.3?105 U/mg.It could promote the proliferation of BALB/c 3T3 cells,rabbit corneal cells,and chicken embryo chorion vessels in vitro,and both the proliferation and migration of vascular endothelial cells in vitro were enhanced,and rabbit skin healing of alkali burns in vivo was accelerated.Conclusion:E.coli expressing vector of recombinant His-SUMO-T?16 fusion protein is constructed successfully,and recombinant protein T?16 has significant repairing effects.The study established a good foundation for further industrialization of T?16.
10.Toxicologic Study on Ke'erxing Capsules

Objective: To observe the acute toxicity of Ke'erxing Capsules (KC) in mice and its long-period toxicity in rats. Methods: The maximal tolerance dose was tested by gastric infusion of single oral dose of KC. Three groups of rats (in high, moderate and low dosage of KC) were administered with repeated gastric infusion of KC for 8 weeks. Body weight, hematological and hematobiochemical parameters, coefficient and histomorphological figure of organs (the heart, liver, spleen, lungs, kidneys, adrenals, testes, prostate, pancreas, uterus, brain, thymus) were examined in 8 weeks and 2 weeks after the cessation of KC. Results: The oral maximal tolerance dose of KC in mice was more than 4000 mg/kg (equal to 600 times of clinical dose). General conditions were changed and the body weight was increased slowly in high-dosage group. The coefficient of prostate was increased in high-dosage group and moderate-dosage group and recovered to normal 2 weeks after stopping the drug. Conclusion: KC has a certain effect on mice and rats but the effect was reversible.
