BACKGROUND: Recently, miR-378 has been shown to modulate the anti-hypoxia capacity of bone marrowmesenchymal stem cells (BMSCs) and reduce cell apoptosis under hypoxic conditions.OBJECTIVE: To investigate the benefits of miR-378-upregulated BMSC transplantation in a rat model of acutemyocardial infarction.METHODS: Primary rat BMSCs were cultured in vitro. Until passage 3, the cells were infected with the lentiviruscarrying synthetic miR-378 gene fragments. A rat model for acute myocardial infarction was constructed by ligatingthe left anterior descending artery. Thereafter, the animals were randomly assigned to three groups: control group(n=10), BMSCnull group (n=16) and BMSCmiR-378 group (n=16). In the latter two groups, 50 μL of normal salinecontaining 1×107 empty virus-transfected or miR-378-transfected BMSCs was injected into the region of myocardialinfarction, respectively. Only 50 μL of normal saline was injected in the control group. Twenty-four hours later, theapoptosis of transplanted BMSCs was evaluated with TUNEL, and expression level of vascular endothelial growthfactor and transforming growth factor-β was detected using western blot assay. Four weeks after treatment, the leftventricular function of rats was assessed by echocardiography, and then histological and molecular biology analyseswere performed.RESULTS AND CONCLUSION: At 24 hours postoperatively, there were less apoptotic BMSCs and higher expressionlevels of vascular endothelial growth factor and transforming growth factor-β in the BMSCmiR-378 group than in theBMSCnull group (n=6, P < 0.001). Four weeks later, there were more transplanted BMSCs and BMSCs-derivedcardiomyocytes in the BMSCmiR-378 group than the BMSCnull group (n=10, P < 0.001). Moreover, increased new vesseldensity (P < 0.001), decreased infarcted area (P < 0.001), preserved left ventricular ejection fraction (P < 0.05), reducedleft ventricular end-diastolic volume (P < 0.05) were found in the BMSCmiR-378 group, compared with the other two groups. The above parameters were better in the BMSCnull group than the control group (P < 0.05). Overall, the upregulation ofmiR-378 could enhance the capability of BMSCs against hypoxia, and consequently promote myocardial repair afterimplantation, providing a new strategy for cell therapy of myocardial infarction.

Chinese Journal of Tissue Engineering Research 2006;10(40):151-153
BACKGROUND: At present, international classification of diseases by WHO, cerebrovascular disease classification used in China and TOAST classification used abroad are all based on patho-physiology. There is no report about human cerebral infarction by magnetic imaging types in Beijing and other areas.OBJECTIVE: To analyze retrospectively the subtype distribution of acute cerebral infarction of patients in Beijing by magnetic resonance imaging in order to make Beijing patients with acute ischemic stroke receive more exact and individualized treatment.DESIGN: Retrospective case study.SETTING: Department of Neurology, General Hospital of Beijing Military Area Command of Chinese PLA.PARTICIPANTS: The study was carried out in General Hospital of Beijing Military Area Command of Chinese PLA, Beijing Sino-Japan Friendship Hospital, Beijing Chaoyang Hospital, Beijing Naval General Hospital and Beijing Jishuitan Hospital between January and December 2004. Totally 602 inpatients with ischemic stroke who received treatment in each hospital were recruited. They were 352 male and 250 female with average age of (65.6±10.2) years.METHODS: The 602 patients with acute ischemic cerebral stroke who received the treatment in the 5 hospitals from Dongcheng, Xicheng,Chaoyang and Haidian districts in Beijing were recruited in the study. According to results of magnetic resonance imaging of brain, the maximal infarction area were determined. The patients were classified as: large infarction: over 1 lobe of brain, > 5.0 cm in diameter; moderate infarction: less than 1 lobe of brain, 3.1-5.0 cm; small infarction: 1.6-3.0 cm; lacunar infarction, < 1.5 cm.MAIN OUTCOME MEASURES: Constitution ratio of MRI classification of patients in each groupRESULTS: Totally 602 patients with ischemic stroke participanted in final result analysis. Among the patients with acute cerebral infarction of 5 hospitals in Beijing, 280 (46.5%) suffered from lacunar infarction, 299(38.1%) from small infarction, 57 (9.4%) from moderate infarction, and 36 (6%) from large infarction. There were significant differences between lacunar infarction and other types (P < 0.01).CONCLUSION: Acute cerebral infarction of patients of Beijing mostly occurs in the type of lacunar infarction. It suggests that small arterial diseases are the major disease type of ischemic stroke in Beijing. This MRI classfication can be used for early classification, treatment guidance, evaluation and prognosis of acute ischemic cerebral stroke.

Objective To investigate the follicle development and pregnancy in clomiphene citrate-resistant patients with polycystic ovary syndrome (PCOS) after transvaginal follicular aspiration and gonadotropin administration. Methods Seventeen clomiphene citrate-resistant patients with PCOS failed to gonadotropin treatment associated with ovarian hyperstimulation syndrone(OHSS). They are anovulatory infertility with normal tubal and husbands'sperm condition. In this treatment regimen, all patients was administered with gonadotropin from day 5 of menstrual cycle(or withdrawal bleeding) and were asked to undergo ultrasound-guided transvaginal follicular aspirations for 5 days later. Only one or two follicles for every ovary were remained and gonadotropin administration was continued. A follicle development, ovulation rate, pregnancy rate and sex hormone were monitored. Results Fifteen of the 17 patients experienced dominant follicles development and ovulation. 9 of them with one(52.9%) ,4 with two(23.5%) and 2 with three(11.8%) dominant follicles ovulated and pregnancy occurred in 7 patiends(41.2%). All of pregnancy are singleton. Conclusion A single follicle development and singleton pregnancy can be realized in clomiphene citrate-resistant PCOS patients by combined ultrasound-guided transvaginal follicular asperation with gonadotropin administration.

Objective To explore the effects of narrow-band ultraviolet B (NB-UVB)on the serum levels of thymic stromal lymphopoietin (TSLP)and interleukin-25 (IL-25), as well as on the expressions of TSLP receptor (TSLPR)and IL-25 receptor (IL-25R)mRNAs in peripheral blood mononuclear cells (PBMCs)from patients with atopic dermatitis(AD). Methods A total of 40 patients with AD and 30 healthy volunteers were enrolled in this study. All the patients were treated with NB-UVB at 0.3 - 2.5 J/cm2 thrice a week for 12 consecutive weeks. Venous blood samples were obtained from these patients before and after the treatment as well as from these healthy controls. Double-antibody sandwich enzyme-linked immunosorbent assay (ELISA)was performed to detect serum levels of TSLP and IL-25, and reverse transcription PCR(RT-PCR)to determine the mRNA expression levels of TSLPR and IL-25R in PBMCs from these subjects. The scoring atopic dermatitis (SCORAD)system developed by the European Task Force on Atopic Dermatitis was used to estimate the severity of AD, and visual analogue scale (VAS)to evaluate the degree of itch. Statistical analysis was carried out by the two-independent-sample t-test for intergroup comparisons and paired t-test for comparisons between pre- and post-treatment samples from these patients. Results After the treatment with NB-UVB, the total response rate reached 75%(30/40)in these patients, with a significant decrease in SCORAD score from 55.26 ± 10.88 before the treatment to 20.36 ± 5.12 after the treatment (t = 10.29, P < 0.05)and in VAS score from 8.20 ± 1.37 to 3.05 ± 1.02(t = 8.16, P < 0.05). Before the treatment, the patients showed a significant increase in the serum levels of TSLP(198.24 ± 29.47 ng/L vs. 120.13 ± 19.65 ng/L, t = 29.70, P < 0.05)and IL-25(160.54 ± 34.89 ng/L vs. 120.41 ± 43.07 ng/L, t = 14.65, P < 0.05), as well as in the mRNA expression levels of TSLPR (8.57 ± 1.34 vs. 1.94 ± 0.39, t =7.07, P < 0.05)and IL-25R(6.81 ± 0.50 vs. 1.48 ± 0.47, t = 18.89, P < 0.05)compared with the healthy controls. With the improvement of conditions after the treatment, a significant decrease was observed in the serum levels of TSLP (151.87 ± 14.78 ng/L, t = 18.56, P < 0.05)and IL-25(130.52 ± 29.65 ng/L, t = 9.07, P < 0.05), as well as in the mRNA expression levels of TSLPR (2.89 ± 0.53, t = 5.21, P < 0.05)and IL-25R (3.90 ± 0.37, t = 7.35, P < 0.05)in PBMCs from these patients compared with those before the treatment, and differences disappeared in all of these parameters between the patients and controls (all P > 0.05). Conclusions TSLP and IL-25 may play important roles in the development of AD, and NB-UVB may treat AD by downregulating the expressions of them and their receptors.
