1.Expression of PPAR? in gastric carcinma tissue and effect of its ligand on human gastric carcinoma cell growth

Objective To investigate the expression of peroxisome proliferator-activated receptor?(PPAR?) in human gastric carcinoma and the effect of its ligand 15-deoxy-△~(12,14)-Prostagliandxin J_(2)(15d-PGJ_(2)) on the growth of human gastric carcinoma MGC803 cells.Methods PPAR? and survivin mRNA were examined by RT-PCR;PPAR?,survivin,Skp2 and p27 protein were examined in cells by Western blot;proliferation was examined by MTT assay,apoptosis and cell cycle distribution were examined by flow cytometry;PPAR? protein in tissue was examined by immunohistochemistry;morphological change of apoptosis was examined by Hoechst33342 staining.Results The positive rate of PPAR? protein was significantly higher in gastric carcinomas 75.0%(40 cases) than in their paired adjacent mucosa(25.0%)((40 cases))(and)(normal)(mucosa)(20.0%(40cases)),(and)(there)(was)(no) difference between the latterones. The positive rate of PPAR?mRNA and protein were significantly higher in intestinal gastric carcinoma 95.0%(20 cases)than that in diffuse gastric carcinoma 55.0%(20 cases).Proliferation inhibition rate and apoptosis rate of MGC803 cells after_()24h were all significantly higher following 5,10,20,30 and (40 mol/L) 15d-PGJ_(2)treatment than(0 mol/L)(P

Epithelial-mesenchymal transition (EMT) is the basis of biological process of embryonic development,and is also closely related with tumor cell in situ invasion and distant metastasis.Recent studies find that EMT and cancer stem cells (CSCs) have a close relationship.CSCs have a strong ability to selfrenewal,tumorigenicity and cell differentiation potential.Identifing the markers,in-depth study of CSCs resistance,may provide a new path for cancer treatment.

4.The relationship between histological activity index and serum of tumor necrosis factor-α,peripheral blood T cell subsets in patients with chronic hepatitis B

Objective To study the relationship between histological activity index and serum of tumor nec-rosis factor-α( TNF-α) ,peripheral blood T cell subsets in patients with chronic hepatitis B.Methods Histopatholog-ical examinations were performed in 237 patients with chronic hepatitis B and 12 controls.The histological activity index( HAI) were analyzed by knodels method.These patients were divided into the four groups according to histologi-cal activity index classification:the control group(n=12),mild group(n=67),moderate group(n=89),and severe group(n=81).The serum levels of TNF-αwere determined by ELISA,and peripheral blood T cell subsets were ana-lyzed by flow cytometry.Results The serum level of TNF-αin the mild group[(29.65 ±10.15)μg/L],moderate group[(38.96 ±7.32)μg/L]and severe group[(47.73 ±6.99)μg/L]were higher than those in the control group [(13.78 ±6.40)μg/L](q=14.38,19.97,24.83,all P<0.05),significant positive correlation lied between the histological activity index classification and the serum level of TNF-α(r=0.708,P<0.05).The rate of CD8+cells in the moderate group[(27.66 ±6.63)μg/L]and the severe group[(28.98 ±5.92)μg/L]were higher than those in the control group[(22.32 ±1.84)μg/L](q=3.84,4.76,all P<0.05),and the ratio of CD4+/CD8+in the moderate group(1.32 ±0.37) and the severe group(1.19 ±0.30) were lower than those in the control group(1.67 ±0.14) (q=4.20,5.72,all P<0.05),but the rate of CD8+cells and the ratio of CD4+/CD8+showed no significance among the mild group,moderate group and severe group.Conclusion TNF-αand disorder of cellular immunity may play an important role in the development and progression of intrahepatic vascular lesion.
5.Study on the relationship between vascular obstruction and inflammation and the pseudolobuli formation of chronic hepatitis B

Objective To study the relationship between vascular obstruction,inflammation and the pseudolobuli formation in liver tissues with chronic hepatitis B.Methods The liver biopsies from 1256 patients with chronic hepatitis B was observated by HE,histochemical,immunohistochemical and the electricity mirror techniqe.Results (1)The intrahepatic vascular inflammation and obstruction was a histopathological characteristics of liver cirrhosis of chronic hepatitis B;(2)The lesions of vascular inflammation caused the vascular obstruction and induced the proliferations of venous and fiber;(3)The blood vessel of hyperplasia revolved round the liver cell into the irregular cell circular(tiny node).The tiny node settled the composition base in the interest of pseudolobuli formation;(4)The blood vessel of hyperplasia once more inflammation,blocking,fiber deposit constituted the fiber brings,pseudolobuli formation.Conclusion The vascular obstruction,inflammation has a closely relationship with the pseudolobuli formation in liver tissues with chronic hepatitis.
6.An ERP Study on Visual Illusory Motion

Objective To explore the neural basis of the visual illusory motion using the event-related potential(ERP).Methods One hundred trials(each duration 2 000 ms)of a visual illusory motion figure,rippling wheat pattern made by Akiyoshi Kitaoka,and a visual static figure made of modified rippling wheat pattern were randomly presented with equal probability.Ten healthy right handed undergraduate students as subjects were asked to judge the stimulus whether motional or not.The EEG was recorded from 128 scalp sites using with electrodes mounted in HydroCel GSN cap(Electrical Geodesics Incorporated,Oregon,USA).Results 1)The C1 component of ERP could be evoked by both visual illusory motion pattern and static pattern in POz.The peak of C1 component was presented about 75 ms after each stimulus.C1 component evoked with visual illusory motion was negative,but it's static pattern to be positive.2)P100 and P200 components could be obviously evoked by both kinds of stimuli,but in O2 the amplitudes of P100 evoked by different stimuli were significant different and in T3 the amplitudes of P200 were significant different too.Conclusion The visual illusory motion is formed at the primary visual cortex.It may be related to the organized mode of perception.
7.Effect on radiosensitivity of human lung adenocarcinoma cells transfected with wild-type p16 gene

0.05); The result of FCM indicates that the cells transfected with p16 changed in cell cycle distribution ; proportion of G 2 M phase was increased, while that of S phase was decreased.Conclusions:Introduction of p16 gene into low expression human lung adenocarcinoma cell line Anip973 alters the radiation effect in a way which perhaps was correlated with the inhibition of sublethal damage repair and the cell cycle distribution change, but not in the high expression cell line AGZY83a.
8.Long-term results of postoperative electronic irradiation for 53 patients with keloids.

OBJECTIVETo analyze the results of postoperative radiotherapy with electronic beam for patients with keloids in our hospital.
METHODSFrom September 2006 to May 2009, radiotherapy was given within 24 hours after operation in 53 keloid patients. With single vertical field irradiation, 6-12 Mev electronic beams of Linear Accelerator were selected for different incision depth in different sites. The field size was 1.0 cm (range: 0.5-2.0 cm) away from both incision ends and 1.25 cm (range: 0.75-2.50 cm) away from incision laterally. The radiation was given daily with median treatment course of 4 days (range: 3-21 days) at 3.5 Gy/Fx to a median total dose of 14 Gy (range: 8-20 Gy). SPSS 21. 0 was used for analysis.
RESULTSAll postoperative incisions healed in one stage, the median follow-up was 34 months (range: 18-63 months). The overall local control rate was 79.7%. For patients who received the dose of more than 14 Gy versus less than 14 Gy, the local control rate was 81.6%, 75.2%, respectively (P > 0.05). For male and female, the 3 year local recurrence rate were 45.3%, 9.9% respectively (P = 0.008). Multivariate analysis showed that the sex (male versus female) was an independent prognostic factor (P = 0.036).
CONCLUSIONSurgery combined with electronic beam irradiation is a rather effective way to treat keloids. The local control rate would have a better trend if the total dose was higher than 14 Gy. Sex is an independent prognostic factor.

9.A modified equivalent uniform dose with the dosimetric parameters of perfusion imaging correlates with radiation pneumonitis in radiation therapy planning

Background and purpose: The literature on dose-volume parameters and pneumonitis is extensive. The results are inconsistent, both for the best predictive metrics and significant comorbid factors. This study aimed to investigate a prospective functional equivalent uniform dose (fEUD) with perfusion single photon emission computed tomography (SPECT) images as predictors of radiation pneumonitis (RP) in patients undergoing curative radiotherapy (RT). Methods: Functional lung imaging was performed using SPECT for perfusion imaging. Perfusion factors were defined as the mean percentile perfusion levels of the 4 areas, top to 75%, 75% to 50%, 50% to 25%, 25% to 0%, re-spectively. fEUD was calculated from perfusion factors and standard dose-volume parameters extracted from treatment planning computed tomography (CT) scans. Total lung (TL), ipsilateral (IL) and contralateral lung (CL) volumes minus gross tumor volume (GTV), whole-lung V5, V20, whole lung fEUD, IL and CL fEUD, and general equivalent uniform dose (gEUD) were analyzed to evaluate correlations between RP using Common Terminology Criteria for Adverse Events (CTCAE) version 4.03. Statistical significance was defined as P<0.05. Results: A total of 15 patients treated with intensity modulated RT or 3D conformal RT were analyzed, grades≥3 RP were observed in 6 patients. There was only a trend toward significance for unilateral (UL) fEUD of higher dose side (P=0.007). Whole-lung V5, V20 were almost identical between patients who developed pneumonitis and patients who did not, as the values were below the recommended thresholds from published papers. Unilateral fEUDs were linear with unilateral gEUDs (t=0.815, P=0.000). Conclusion: SPECT-based equivalent uniform dose appears to be a better predictor of RP compared to stan-dard dose-volume parameters. Planning constraints should aim to keep unilateral fEUD below 21 Gy.
10.Study on relationship between expression of survivin, galectin-3 and TPO and BRAFV600E mutation in patients with papillary thyroid microcarcinoma and its clinical significance

Objective:To detect the expressions of survivin, galectin-3 and thyroid peroxidase in papillary thyroid microcarcinoma ( PTMC) and cancer adjacent normal tissue to explore the clinical significant.The correlations between the expressions of survivin,Gal-3 and TPO with BRAFV600E gene mutation in PTMC were analyzed.Methods: The expressions of survivin,Gal-3 and TPO were measured by immunohistochemical ( IHC ) method in 56 cases of PTMC tissue and adjacent normal tissue; BRAFV600E mutation was detected by PCR amplification and subsequently sequencing.Chi square test was used to analyse the relation in the expression rates of survivin,Gal-3 and TPO and BRAFV600E gene mutation.Results:The survivin and Gal-3 were strongly expressed but TPO was negatively expressed.The survivin and Gal-3 were negative in adjacent normal tissues but TPO was expressed.There were sig-nificant differences in the expression rates of survivin, Gal-3 and TPO between PTMC tissue and adjacent normal tissue (all P<0.001).The BRAFV600E mutation rate (32.1%) and the expression rates of survivin,Gal-3 and TPO in PTMC tissue were found to be positively related to lymph node metastases (P=0.009,P=0.025,P=0.007,P=0.008),and negatively related to gender and age (all P>0.05).There were no correlation was found between the expressions of survivin,Gal-3,TPO and the BRAFV600E gene mutation in PTMC(all P>0.05).Conclusion: The strong expressions of survivin and Gal-3,the mild expression of TPO and BRAF mutation may be important in the development of PTMC,and the detection of BRAFV600E gene mutation and the expressions of survivin, Gal-3 and TPO could be used to the judgment of pathogenetic condition and prognostic outcomes.
