1.The relationship between serum CC10 protein and ventilator-associated lung injury in rats

Objective To investigate the relationship between serum CC10 protein and lung injury induced by mechanical ventilation.Methods Forty healthy Wistar rats of both sexes weighing 200-250 g were randomly divided into 5 groups(n=8 each):group Ⅰ control;group Ⅱ mild lung injury[VT=7 ml/kg,duration of mechanical ventilation(t):2 h];group Ⅲ moderate lung injury(VT=7 ml/kg,t=4 h);group Ⅳ severe lung injury(VT=40 ml/kg,t=2 h);group Ⅴ extremely severe lung injury(VT=40 ml/kg,t=4 h).The animals were anesthetized with intraperitoneal 10% chloral hyrdrate 3.5 ml/kg and tracheostomized.Group Ⅰ received no mechanical ventilation.The animals in group Ⅱ-Ⅴ were mechanically ventilated with air(FiO2=21%,RR=40 bpm,I:E=1:2).The animals were sacrificed at the end of mechanical ventilation.The lungs were immediately removed for microscopic examination and determination of W/D lung weight ratio.The left lung was lavaged.The CC10 protein level in broncho-alveolar lavage fluid(BALF)and serum were determined by Western blotting.The Clara cells in the bronchiolar epithelium were examined by immno-histochemistry.Results The level of CC10 protein in BALF was significantly lower in group Ⅳ and Ⅴ while the serum CC10 protein level was significantly higher in group Ⅱ-Ⅴ than in group Ⅰ.The serum CC10 protein level was positively correlated while the CC10 protein level in BALF was negatively correlated with the severity of lung injury and W/D ratio.Conclusion The serum CC10 protein is closely related to the severity of lung injury induced by mechanical ventilation.
2.Effect and mechanisms of tetrahydroxystilbene glucoside on neuron proliferation in the hippocampus of epileptic rats

Objective To explore the influence of tetrahydroxystilbene glucoside ( TSG ) on the neuron proliferation in epileptic rats and the related mechanisms.Methods 54 healthy SD male rats were randomly divided into three groups: sham-operated group, epilepsy group and epilepsy with TSG treatment group.The epilepsy group was established by stereotactic brain trace injection with kainic acid ( KA ) . TSG solution ( 3 mg/kg ) was injected intraperitoneally at 6 hours after the epilepsy group established, and then q.d.for consecutive 42 days.The sham-operated group and epilepsy group were injected with normal saline.The influence of TSG on cell proliferation of rat hippocampal dentate gyrus BrdU-positive granular cells was observed by immunohistochemistry.Results Compare with the epilepsy group, the amount of glial fibrillary acid protein ( GFAP)-positive astrocytes was significantly reduced and dentate gyrus BrdU-positive cells were significantly increased in the TSG group ( P <0.01 ) .Conclusions Tetrahydroxystilbene glucoside ( TSG) promotes neurogenesis of neural stem cells and neurons, and inhibits the growth of hippocampal dentate gyrus astrocytes in epilepsy rats.
3.Expression of Runx3 protein and its cliniopathologic significance in gastric carcinoma

Purpose To investigate the relationship between the expression of Runx3 protein and pathogenesis, development,clinicopathological features and its prognostic significance in gastric adenocarcinoma.Methods Immunohistochemical SP method was used to detect the expression of Runx3 protein in 63 cases of gastric cancer,19 cases of atypical hyperplasia and 10 cases of normal gastric mucosa obtained from patients whose partial gastrotomy was performed for benign diseases.Results Compared with gastric atypical hyperplasia tissue (68.4%) and normal gastric mucosa (90%), the positive rate of Runx3 protein in gastric cancer (39.7%) was significantly lower (P<0.05); the expression of Runx3 protein was related to tumor differentiation, invasive depth and lymphnode metastasis, but not to age, sex, tumor size and TNM stages; the survival rate of the patients with Runx3 protein expression was higher than that without Runx3 expression (P<0.05).Conclusion Runx3 protein may play an important role in the occurrence and development of gastric cancer, and it is an important marker in evaluating the prognosis of the patients.
4.Effects of Yushen Heji on Expressions of Heme Oxygenase-1 and Heme Oxygenase-2 in Kidney of Diabetic Rats

Objective To observe the effects of Yushen Heji on expressions of Heme Oxygenase (HO-1) and HO-2 in kidney of diabetic rats;To explore its mechanism in diabetic nephropathy. Methods SD rats were randomly divided into normal group, model group, control group and treatment group. Models were duplicated by subcutaneous injection STZ. Control group and treatment group were given gavage with related medicine, while normal group and model group were given gavage with the same amount of distilled water. Eight weeks later, the urine of rats was collected to detect the 24 h Upro and Ucr;the rats were sacrificed to detect the levels of CO, BUN and Scr;the kidneys were harvested for observing the expressions of HO-1 and HO-2 by immunohistochemistry.Results Compared with normal group, the contents of BUN, 24 h Upro and Scr all increased in model group;Ccr and plasma CO decreased;the expressions of HO-1 and HO-2 in kidney tissues were obviously reduced (P<0.01). Compared with model group, the contents of BUN, 24 h Upro and Scr all obviously decreased in treatment group and control group;Ccr and plasma CO increased;the expressions of HO-1 and HO-2 in kidney tissues obviously increased (P<0.05,P<0.01). Compared with control group, the contents of 24 h Upro decreased in treatment group;plasma CO increased;the expressions of HO-1 and HO-2 in kidney tissues obviously increased (P<0.05,P<0.01).Conclusion Yushen Heji has therapeutic effect on diabetic nephropathy rats, which may related with HO/CO system.

6.Accuracy of stroke volume variation in monitoring blood volume in patients undergoing off-pump coronary artery bypass grafting

Objective To evalute the accuracy of stroke volume variation (SVV) in monitoring blood volume in patients undergoing off-pump coronary artery bypass grafting.Methods Twenty-one ASA Ⅱ or Ⅲ patients of both sexes aged 44-77 yr undergoing off-pump coronary artery bypass grafting were enrolled in this study.Anesthesia was induced with midazolam,etomidate,fentanyl,rocuronium and dolicaine and maintained with target-controlled infusion of propofol,infusion of remifentanil,intermittent iv injetion of atracurium and inhalation of sevoflurane.The patients were mechanically ventilated (VT 8 ml/kg,RR 12 bpm,I:E 1:2,PEEP 0,FiO2 80% ).PEr CO2 was maintained at 35-44 mm Hg.Radial artery was cannulated and connected to FloTrac pressure transducer and Vigileo monitor.6% hydroxyethyl starch 130/0.4 sodium chloride solution 7 ml/kg was infused at a rate of 0.25 ml· kg- 1 1· min- 1 at 5 min of haemodynamics stabilization after pericardiotomy (T1).HR,MAP,CVP,systemic vascular resistance (SVR),systemic vascular nesistance index (SVRI),SVV,stroke volume index (SVI)and CI were recorded at T1 and at 10 min after loading dose (T2).The change rate of HR(△HR),MAP(△MAP),CVP(△CVP),SVR(△SVR),SVV(△SVV),SVI(△SVI) and CI(△CI) were calculated.△SVI≥25% was considered effective volume expansion.The ROC curves for HR,MAP,CVP,SVR and SVV in determining the volume expansion efficacy were plotted.The area under the curves and 95 % confidence interval were calculated.Results Compared with T1,CVP,SVI,CO and CI were significantly increased,SVRI and SVV decreased at T2 (P < 0.01).There was no significant difference in MAP and HR between T1 and T2(P >0.05).△SVI was negatively correlated with △HR and △SVR ( r =- 0.737,r =- 0.480,P < 0.05).△SVI was not correlated with △CVP,△MAP and( P > 0.05).The change in SVI was determined by SVV 8.8% (sensitivity =52.6%,specificity =100.0% ).The area under the curve for SVV and 95% confidence interval were 0.579(0.346-0.812).Conclusion SVV can not be used to accuratelymonitor the changes in blood volume in patients undergoing off-pump coronary artery bypass grafting.
7.The analysis of correlation between the occurrence of tourette syndrome with one-child family environment

Objective The analysis of correlation between the occurrence of tourette syndrome with one-child family environment. Methods The authors investigated home environment of two groups. One group included 200 patients with tourette syndrome and is the other group included healthy children of the same age in 3 different primary and secondary schools. By comparing with the home environmental factors, parenting styles and parent emotional factors between two groups. Results The instabilities of home environment included bad family relationship, separation between parents, divorce, scolding and even beating children. It showed that anxiety of the direct supervisors among the sickness group were higher than healthy control group as well. Among the ten factors of the Family Environments Scale, the significant differences showed in six factors of affection,cohesion,expressiveness, conflict, intellectual-culture orientation, active-recreational orientation, organization between the two groups. Conclusion So we concluded that ①There is a correlation between the parents relations,parenting styles and the incidence of tourette syndrome; ②An anxiety of the direct supervisor may initiate, even aggravate the incidence of tourette syndrome; ③It is important to convert the education mode and take family overall treatment in order to reduce the incidence and improve the effectiveness of tourette syndrome.
8.Transarterial chemoembolization as an adjuvant therapy in patients with hepatocellular carcinoma treated with hepatectomy

Objective To evaluate the role of transarterial chemoembolization (TACE) as an adjuvant therapy in patients with hepatocellular carcinoma (HCC) treated with hepatectomy. Methods Clinical data of 386 consecutive patients who underwent hepatectomy for HCC were analyzed retrospectively.Of the 386 patients, 156 patients did not undergo TACE served as controls (non-TACE group), the remaining 230 patients underwent TACE (TACE group) preoperatively (n=71), postoperatively (n=86), or both (n =73). For the purpose of comparison, patients who did not undergo preoperative TACE were assigned to group A (n=242), and those patients who underwent preoperative TACE were assigned to group B (n =144). Patients cumulative survival rates were calculated by survival table and analyzed using Kaplan-Meier survival curves. Results There were significantly higher complete necrosis rates in group B (18/144) than those in group A (0/242) ( P < 0.01). The difference between the survival rate of patients with complete necrosis and those with incomplete necrosis was statistically significant (P<0.01).The 1-,3-,5-and 10-year survival rates were 90.4%(66/73),72.9%(42/73),51.9%(22/73) and 25.4%(2/73) in combined TACE group, 74.0% (50/71) ,46.2%(28/71) ,27.3%(5/71) and 0(0/71) in preoperative TACE group, 88. 0% (73/86) ,59. 6% (39/86) ,36. 7% (11/86) and 0(0/86) in postoperative TACE group, and 75. 8% (110/156), 63.4% (48/156), 31.0% (13/156) and 23.9% (10/156) in non-TACE group,respectively. Combined TACE group got a significantly higher survival rate compared with non-TACE group or preoperative TACE group or postoperative TACE group (P<0.05). The survival rates in either preoperative or postoperative TACE group were not significantly better than those in non-TACE group (P>0.05).Conclusions As an adjuvant treatment, combined pre-and post-operative TACE can increase survival rate in patients with surgically resectable HCC. No significant benefit for patient's long term survival when either preoperative or postoperative TACE was performed in addition to surgery.

Objective To investigate the mechanisms of decreasing insulin degrading enzyme (IDE) level by mutation V97L in the gene presenilin 1 (PS-1).Methods Transcription factor GATA binding protein 3 (GATA-3) activity was assessed by protein/DNA array and verified by Western blot in SH-SY5Y cells transfected by PS-1 mutation V97L.Results Protein/DNA array and Western blot revealed that there was increased transcript factor activity (5.5 times high) and protein level of GATA-3 in V97L-PS-1 transfected SH-SY5Y cells.Transcription factor GATA-3 can bind to the IDE promoter and negatively control the IDE transcription level.Conclusion PS-1 mutation V97L may regulate the transcription of IDE via GATA-3, and subsequently involve in deposition of Aβ42 and development of Alzheimer's disease.

Objective To discuss real time three-dimensional reconstruction techniques of spiral CT in the detection of tracheoesophageal groove lymph node metastasis and its diagnostic value.Methods Spiral CT RT3D reconstruction of the supraclaviculrar region were applied to 45 patients with scanning and post-processing on the workstation.Mediastinum,color in chest,clip planes 3D images were obtained by adjusting the CT value threshold and encoding pseudo color techniques.Results RT3D images can correctly reveal the macroscopic morphology of lymphatic metastasis and it's pressure to the trachea and carotid artery.There were significant differennces of visualization capabilities of radiological signs among different 3D techniques.Conclusion With the utilization of multiple methods,RT3D reconstruction can play an important role in detecting tracheoesophageal groove lymph node metastasis of esophageal cancer.It is an important complement to the axial CT images and the basis of clinical treatment.
