Objective To investigate the changes of nuclear factor?B activity in peripheral blood mononuclear cell(PMNC)in newly diagnosed type 2 diabetic patients.Methods Thirty normal individuals and 30 newly diagnosed type 2 diabetics were selected.Phosphorylation status of NF-?B P65 of PMNC was determined by immunoblotting with phosphor-specific antibodies to NF-?B P65(Ser 536 ).Serum hs-CRP was assayed by enzyme linked immunosorbent assay(ELISA).Results The level of NF-?B P65 phosphorylation was increased in newly diagnosed type 2 diabetic patients compared with control subjects[0.85(0.49,1.02)vs 0.47(0.26,0.67),P
2.Environmental safety assessment of elderly patients admitted in different grade hospitals

Objective To provide evidences for fall prevention through environmental safety assessment of elderly patients admitted in different grade hospitals. Methods 24 different levels hospitals located in 10 cities in southwest of China were surveyed with assessment form of fall-related factors of hospital environment for elderly patients. Results Hospital public environment and ward public environment were assessed well by elderly patients which were 4.10 points and 4.06 points, but the scores for ground channel environment and toilet environment were lower which were 3.92 points and 3.65 points;Scores of tertiary-class hospitals were better than that in second-class hospitals, including ward facilities (t=-2.756, P<0.01), bathroom facilities (t=-3.752, P<0.01), public ward environment (t=-2.438, P<0.05) and public hospital environment (t=-2.263, P<0.05). As the ground-channel was concerned, there was no statistically significant different between them (P>0.05). Conclusions Public hospital environment and public ward environment got higher scores, and the scores of ground-channel environment and the bathroom environment were relatively lower. Environment in second-class hospitals need to improve in order to prevent fall of elderly patients. Assessment form of fall-related factors of hospital environment for elderly patients is helpful to disclose the unsafety factors of the hospital environment, and should be windely used in futher clinical practice.
3.Test-retest Reliability of Distal Upper Extremity Muscle Strength by Muscle Evaluation and Treatment System on Healthy Adults

Objective To investigate the test-retest reliability of Muscle Evaluation and Treatment System on healthy adults. Methods 30 healthy adults participated in this study and were tested by Muscle Evaluation and Treatment System, designed by University of Shang-hai for Science and Thechnology and Rehabilitation Department of Huashan Hospital, on muscle strength of both sides' flexor carpi mus-cles, musculus flexor digitorum sublimis, flexor digitorum profundus, extensor digitorum muscle, extensor carpi muscles. Peak torque, peak torque per weight, largest single work, largest single work per weight, average torque, average power were the main parameters. Each partici-pant was tested 2 times, and the interval was 7 days. Results Almost perfect test-retest reliability of peak torque was performed on flexor car-pi muscles, musculus flexor digitorum sublimis, flexor digitorum profundus, and extensor carpi muscles. Moderate to excellent test-retest re-liability of average power were performed on extensor digitorum muscle. Conclusion Good test-retest reliability of Muscle Evaluation and Treatment System is proved in the study.
4.Expression and clinical significance of CAK complex in estrogen receptor-positive breast cancer

Objective To evaluate the expression and clinical significance of cyclin dependent kinase (CDK)-activating kinase (CAK) complex including CDK7, cyclin H and accessory protein menage a trios 1 (MAT1) in estrogen receptor-positive breast cancer.Methods A total of 40 patients with estrogen receptor-positive breast cancer from Department of Galactophore, Baoji Maternal and Child Care Service Centre of Shaanxi Province were investigated in this study.Breast cancer tissues and adjacent normal tissues were obtained from patients undergoing surgery.The mRNA expressions of CDK7, cyclin H and MAT1 in two types of tissues were measured by real-time fluorescent quantitative (qRT)-PCR, and their correlations with clinicopathologic features of patients were analyzed.Results The expressions of CDK7, cyclin H and MAT1 in cancer tissues were 2.54±0.78, 2.21±0.56 and 2.46±0.58, while those in adjacent normal tissues were 1.26±0.30, 1.16±0.42 and 1.17±0.39, and there were significantly differences between different types of tissues (t=9.654, P<0.001;t=9.433, P<0.001;t=11.741, P<0.001).The higher expressions of cyclin H and MAT1 in patients′ cancer tissues had the lower clinical stage, with significant correlations (U=3.17, P=0.01;U=2.53, P=0.01).In addition, the tumors were smaller in patients with higher expression levels of MAT1 (χ2=14.16, P=0.01).Conclusion The expressions of CAK complex CDK7, cyclin H and MAT1 are elevated in estrogen receptor-positive breast cancer patients.Cyclin H and MAT1 are closely associated with clinical grade, and MAT1 is also significantly associated with tumor size.
5.Determination and analysis of free amino acids in snake venom

Objective To establish a method for simulataneous determination of 18 free amino acids in snake venom. Methods Preparation of free amino acid samples by membrane. HPLC analysis was performed after derivatization by using phenyl isothiocyanate (PITC) as a derivative reagent, samples were analyzed on Ultimate LP-C18 column with gradient elution column of 0.05 mol/L sodium acetate buffer and methanol-acetonitrile- water (40:40:20), and current speed was1.0 mL/min, and the column temperature was set at 35℃, and detection wavelength was 254nm. Results The 18 kinds of amino acids showed a good linearity with the correlation coefficients ≥0.99. The recovery rate was 74.59%~110.62%. Snake venom contained 17 kinds of amino acids, the total content of amino acids was 0.2%. Conclusion The method was accurate, reproducible and reliable, and can be used for the determination of amino acids in snake venom and related products.
6.Active control of the trajectory of the center of gravity in balance training

Objective To study the impact of active control of the trajectory of the center of gravity on the effectiveness of balance training and balance assessment. Methods Two groups of subjects ( group 1 30-45 years old, group 2 45-60 years old ) were evaluated and trained using both active center of gravity trajectory control and static balance methods. Results The percentages of success in controlling the center of gravity were the same when both groups were trained using static balance. Group 1's success percentage was higher than that of group 2 after active center of gravity trajectory training. Their affected lower limbs performed better in wave trace training than after static balance training, and performance improved with increased wave trace amplitude. Conclusions Wave tracing can stimulate the lower limbs of patients with active control force and improve their balance. The wave assessment is superior to static assessment, as it can objectively reflect ability in active center of gravity control and adjust the lower limbs of subjects whose static balance ability is at the same level.

Objective To verify the main index of Sysmex XN-1000 hematology analyzer and check whether performance of XN-1000 hematology analyzer meet the quality requirement.Methods The background value,carryover rate,precision,line-arity,reportable range,accuracy and comparability of different modes of sample absorbing of Sysmex XN-1000 hematology analyzer were verified and WS/T 406-2012 Analytical Quality Specifications for Routine Tests in Clinical Hematology was referred.Results The background counts,carryover rate,inter-batch precision and inter-day precision all met the require-ment.Linearity verification of WBC,RBC,HGB and PLT all met the requirement of the value of a within 1±0.05 and the correlation r≥0.975.The reportable range surpassed the range stated by the manufacture,which was accepted as the clinic reportable range.The accuracy test showed the mean and bias all met the requirement.The difference between the values de-tected by different absorbing modes all met the comparability requirement.Conclusion Performance of Sysmex XN-1000 he-matology analyzer was verified to meet the requirement.
8.Correlation Study between hMOF Expression and Prognosis in Patients with Non-small Cell Lung Cancer after Radiotherapy

Objective To investigate the hMOF protein expression in non?small cell lung cancer and explore the relationship between its expres?sion and radiotherapy prognosis. Methods Immunohistochemical staining was used to detect the hMOF expression in 59 cases of non?small cell lung cancer after radiotherapy. The relationship between hMOF expression with clinicopathological and radiation prognosis was analyzed. Results Among the 59 cases of non?small cell lung cancer tissues,there were 30 cases found to be high expression with hMOF. The rate of positive expression of hMOF in non?small cell lung cancer were 50.85%. Clinical stage and hMOF expression were independent predictors for non?small cell lung can?cer. Conclusion The expression of hMOF had a positive correlation with the radiation prognosis in non?small cell lung cancer,which could be used as a prognostic indicator of radiotherapy.
9.The Application and Effectiveness Evaluation of Continuing Care Based on Humanistic Care for Out Patients with Facial Plastic Surgery

Objective:To evaluate the effect of continuing care in facial plastic operation .Method:Select 120 patients who had aesthetic plastic surgery in our hospital during the period from January to July in 2014 and ran-domize them into the observation group (60 cases)and the control group (60 cases).The control group merely re-ceive conventional treatment .The observation group is employed continuing care based on conventional treatment . Results:The satisfaction of two groups has no significant difference a week after operation (P>0.05).Six months after operation , the satisfaction of the observation group is obviously higher than that of the control group .The difference is statistically different ( P<0 .0001 ) .The anxiety level of the observation group is obvious lower than that of the control group and the difference is Statisticallydifferent ( P<0 .0001 ) .Conclusion: Based on the hu-manistic care, carry on the continuing care could considerately improve the satisfaction of patients in clinic facial plastic surgery and is one of the most important solution of preventing medical disputes ;mean while , it is also one of the most effective stress reduction measures which makes the patients feel humanized nursing .
10.Construction and Identification for Over-expressing Lentiviral Vector of mSema3A

Objective To construct and identify over?expressing lentiviral vector of mSema3A. Methods Sema3A gene of mice was amplified by PCR,then the gene was inserted into plasmids pDown?mSema3A?IRES/EGFPby Gateway technology. The plasmids pLV/EXPNZ?puro?mSema3A?IRES/EGFP were produced by recombination. After sequencing identification,the vector pLV(Exp)?Puro?CMV?mSema3A?IRES/EGFP was packed and condensed. Finally the recombinant vectors were used to transfect 293T cells to obtain virus pools. Results The recombinant lentiviral vectors were 11 538 bp with EGFP marker,and Sema3Agenes were inserted into the lentiviral vector correctly,indicating the over?expressing vector of Sema3A gene in mice was successfully constructed. Conclusion The over?expressing lentiviral vector of mSema3A was constructed correctly, which lay a foundation of screening of over?expressing strains of such gene in specific cells.
