Aim To explore the role of KATP in the pa-raventricular nucleus in inflammatory pain. MethodsMale Sprague-Dawley rats,250~280 g, were randomly assigned into 5 groups ( each, n =6 ): Normal group, Saline group ( for control, subcutaneous injection of 100 μl of saline into the plantar surface of the left hindpaw) , CFA group ( subcutaneous injection of 100μl of complete freund's adjuvant into the plantar sur-face of the left hindpaw) , Vehicle group ( treated with dimethylsulfoxide), KATP selective agonist group(trea-ted with diaoxide) . Rats in each group were tested for TWL with radiant heat apparatus. Immunofluorescent technique was used to label KATP in PVN and c-Fos in lumber spinal cord. Three days after injected with CFA, a selective KATP agonist, diaoxide, was injected into one side of PVN to test its effect on inflammatory pain and c-Fos expression in lumber spinal cord. Re-sults ① Compared with pre-operation and saline group, rats showed significantly lower TWL on day 1, 3, 7 after injection of CFA;the numbers of KATP posi-tive cells were significantly lower; the numbers of c-Fos positive cells were significantly higher. ② Com-pared with those of vehicle group, TWL and the num-ber of c-Fos in lumber spinal cord were both signifi-cantly lower after injection of diaoxide into one side of PVN. Conclusion KATP in PVN is related to inflam-matory pain.

Objective To investigate the value ofsomatosensory evoked potentials (SEP) in early diagnosis of cervical spondylotic radiculopathy (CSR). Methods The median nerve SEP (SEPm) was examined in 65 cases of CSR (experimental group), and was compared with 30 controls(control group). All the patients were examined by MRI of the cervical spine which were compared with SEPm. All cases were followed up 12 months, at the same time MRI and SEPm were examined repeatedly. Results The experimental group with CSR had significantly prolonged peak latency (PL) of N11 and N13 wave and interpeak latency(IPL) of wave N9-NI3 within 3 months [( 12.92 ± 0.97), (14.81 ± 1.16), (4.45 ±0.65)ms] and at the 12th month [( 14.20 ± 1.06), ( 16.35 ± 1.75), (5.62 ± 0.88) ms] compared with those of control group[(11.65 ± 0.69), (13.26 ± 0.78), (3.56 ± 0.31 ) ms](P< 0.05). The PL of Nll and N13 wave and IPL of wave N9-N13 significantly prolonged at the 12th month compared with those within 3 months.SEPm provided more sensitive modality for the diagnosis[87.69%(57/65 )] of CSR within 3 months than that of MRI [70.77% (46/65)] ( X2 =4.35,P <0.05). While there was no significant difference in sensitive between SEPm [90.77%(59/65)] and MRI [81.54%(53/65)] at the 12th month (X2= 1.56,P>0.05).Conclusion SEP is valuable for the early diagnosis of CSR and may be clinically utilized as a sensitive indicator of CSR.

BACKGROUND:Thrombus formation and neointimal hyperplasia still limit the use of small-caliber expanded poly(tetrafluoroethylene) (ePTFE) vascular prosthesis with a diameter less than 6 mm for revascularization in the coronary or peripheral circulation. Bioactive surface heparin coating is one conceivable path for above-mentioned problems.OBJECTIVE: To elevate the anticoagulant property of ePTFE, this study promoted the patency of a novel small-caliber ePTFE vascular graft by modifying its luminal surface with covalently crosslinked poly(vinyl alcohol)/ p-diazonium diphenyl amine polymer/heparin gel (PVA/PA/Hep gel) and examined the hemocompatibility of the graft.DESIGN, TIME AND SETTING: Observational experiments were performed at the Nanomedicine and Biosensor Lab,Biomedical Engineering Center, Harbin Institute of Technology from May 2006 to June 2007.MATERIALS: The ePTFE vascular grafts (diameter of 4 mm), Nafion (Naf) and Poly(vinyl alcohol) (Aldrich, USA), heparin (Mw 12 000- 14 000) (Calbiochem, USA), p-diazonium diphenyl amine polymer (PA) (this lab, China) were used in this study.METHODS: ①The vascular graft surface was firstly modified with Nafion. ②Following the impregnation of the mixture of PVA/PA/Hep, covalent crosslinking between polyvinyl alcohol and heparin was performed using crosslinker PA under ultraviolet radiation.MAIN OUTCOME MEASURES: ①Contact angles, ②Attenuated total reflection-fourier transform infrared spectroscopy (ATR-FTIR), ③Activated partial thromboplastin time (APTT) and prothromhin time (PT), ④hemolysis test, ⑤platelet adhesion test and ⑥thrombosin inactivation test.RESULTS: ①The water contact angle of the vascular graft surface was greatly decreased after modifying. ATR-FTIR revealed the disappearance of diazonium groups at 2 172 cm-1 and 2 224 cm-1. Vascular prosthesis after modifying had prolonged APTT and PT, low percent hemolysis and low amount of platelet adhesion. Modified vascular prosthesis had inhibitory effect on thrombosin activity and good coating stability.CONCLUSION: Converage of PVA/PA/Hep has good antithrombotic function and low percent hemolysis, resulting in improving hemocompatibility of vascular prosthesis.

Objective To investigate the effects of berberine on cholesterol efflux in THP-1 macrophage derived foam cells, and explore the possible mechanism. Methods HP-1 cells were induced into macrophages by phorbol myristate acetate (PMA), and were treated with acetylated low-density lipoprotein (Ac-LDL) to establish the THP-1 macrophage derived foam cell models. Foam cells were divided into blank control group and berberine (5 to 20 μmol/L) treatment groups according to the way of treatment and berberine concentrations. After treatment for 24 h, flow cytometry was employed to detect AcLDL aggregation, enzymic method was adopted to detect contents of cholesterol and triglyceride, scintillation counting technique was used to detect cholesterol efflux, and effects of peroxisome proliferator-activated receptor-γ (PPARγ) antagonist GW9662 pretreatment on cholesterol efflux (pioglitazone as positive control) were analysed. Besides, RT-PCR was applied to detect expression of liver X receptor α (LXRα) and ATP binding cassette transporter A1 (ABCA1) mRNA. ResultsCompared with blank control group, AcLDL aggregation and contents of cholesterol and triglyceride of foam cells in various berberine treatment groups decreased significantly (P<0.01), while cholesterol efflux increased (P<0.01) in a dose-dependent manner. After GW9662 pretreatment, there was no significant difference in cholesterol efflux between various berberine treatment groups and control group (P>0.05). Furthermore, expression of LXRα and ABCA1 mRNA of foam cells in various berberine treatment groups was higher than that in blank control group. Conclusion Berberine may increase cholesterol efflux in THP-1 macrophage derived foam cells, the mechanism of which may be associated with activation of PPARγ pathway and increase of expression of LXRα and ABCA1 mRNA.

Objective To compare the image quality, radiation dose and diagnostic accuracy on film-screen, digital radiography (DR) and computed radiography (CR) mammography. Methods Three different kinds of mammography machines (film-screen, DR and CR) were tested to make sure they were qualified and comparable. Radiographies were taken on Mammo-152 phantom and ACR phantom using filmscreen, DR and CR systems with the same radiation dose. Next, radiographies were taken on two phantoms using DR and CR systems with classic mode or AEC and the radiation dose was recorded. The images were numbered and scored by eight independent experienced radiologists under the same reading condition according to ACR method. Statistics was performed with the randomized complete-block design variance analysis. The diagnostic accuracy of three kinds of mammography methods were compared with Kappa test in clinical cases who had pathologic results. Results The spacial resolution of film-screen system was the highest (7.0-8.0 Lp/mm) and that of CR was the lowest (2.5 Lp/mm). When the radiation dose was increased (27. 0%-30. 0% ), the resolution was improved with DR ( +11. 0% ) but almost no change with CR ( +1.5%). The radiation dose of DR was the lowest on ACR phantom. The delineation of mass was better on film-screen than on CR, but the delineation of fibers and speck was better on CR. When the radiation dose of CR was increased (25. 0% ), the delineation of fibers and speck on CR was similar to that on DR and the delineation of mass was similar to that on film-screen. There was a good correlation between the mammography diagnosis and pathological results (film-screen is 50/56, DR is 83/90 and CR is 61/69,P < 0. 01 and Kappa value > 0. 75) . The diagnostic accuracy of three mammography systems had no statistical difference [film-screen 89. 3% (50/56), DR 92. 2% (83/90) and CR 88. 4% (61/69),Pearson X 2=0. 722 ,P =0. 697]. Conclusion Film/screen system has the highest space resolution and DR has the lowest radiation dose. Three mammography systems have the same diagnostic accuracy for breast disease.
