1.Mechanism of delaying aging process with optimal stress

Objective To study the mechanism of delaying aging process under optimal stress . Methods Kunming mice swam at the 18℃ water temperature for 15min everyday based upon the model of aging mice induced by D-galactose .After six weeks continuously , the ability of memory and learning , the contents of SOD and MDA were measured, at the same time ,the mitochondria membranous fluidity ,MDA's contents in mitochondria and the activity of ATPase in hippocampus were also determined.Results In the aging mice, a significant reduction of learning and memory ability was observed, the contents of SOD decreased , but MDA contents increased markedly, we also found a obvious declining at the ATPase activity and the fluidity of mitochondria membrane . But the optimal swimming stress could effectually antagonize the above-mentioned changes . Conclusions The optimal swimming stress could delay the aging process by regulating the balance between the oxidant system and the antioxidant system.
2.The guidance of TCM theory manifestation and root cause to the prevention and treatment of tumor

TCM theory of manifestation and root cause which originates in Nei Jing is valuable to guide the prevention and treatment of malignant tumor.It is a specific method for research and analysis of manifestation and essence of diseases.This theory focuses on the diagnosis of tumor,proposes that etiological diagnosis of tumor is the most important.The carcinogenic factors,yin-yang disharmony and visceral dysfunction are the root causes and the formation of tumor and exterior pathogens are the manifestations.The theory emphasize on the diagnosis and treatment before the formation of tumor,dialectic relationship between the tumor and the system,strengthening the body resistance and driving away evil(treating both manifestation and root cause of disease).On the basis of human,improvement of survival period and quality of life is more important than tumor size;prevention is more important than treatment.
3.Influence of Acupuncture on Cephalic Acupuncture Points Region Over the Brain-stem Evoked Potential of Patients Suffering Insufficient Blood-Supply of Vertebral Basilar Artery

Objective From the angle of electrophysiology, go further into the mechanism of action of acupuncture treatment to the insufficient blood-supply of vertebral basilar artery and at the same time, make a comparison of the effects between the acupuncture on cephalic meridian points and zonation cephalic acupuncture. Methods Through the contrast before and after adopting the self-acupuncture on the testees of 28 cases (including healthy persons of seven cases and TIA patients of 21 cases), observe the change of immediate effect of all waves latent period values of BAEP. Results The BAEP wave latent period of healthy persons (exclusive of I wave of auditory fainting region) assumes a relative delay within the physiological range and by comparison between points and region, the delay of V-wave Baihui point is evidently higher than that of auditory fainting region. However, the latent period of all waves of TIA patients is relatively moved up, especially there are evident differences before and after the I-wave acupuncture is carried out. Viewing from the comparison between points and region, there is no significant difference. Conclusion The function of acupuncture lies in the adjustment of antagonistic body state and it is most effective under the pathological state. Acupuncture will play the main role of excitation to the auditory nerve conduction of BAEP of TIA patients (from cochlea to midbrain) and the influence over the peripheral nerves is more sensitive than over the center ones. Although points and region have their own relative specificities, there is no significant difference between them.
4.Desensitization of nicotinic receptors

Nicotinic acetylcholine receptors, a ligand-gated ion c hannels,mediates many important physiological processes. Chronic nicotine admini stration could desensitize nicotinic receptors. Many factors which modulate dese nsitization have been described, including different subunits, agonist, signal t ransduction and cellular microenivornment. The important physiological functions of desensitized receptors still remain controversial, may be modulate synaptic transmission,synapse plasticity and mediate self-protection.
5.A Preliminary Study on the Role of Mitochondrial Protein, Bcl-2 and Bax in Tumorigenesis

Objective: To study the expressions of mitochondrial protein (MAB1273), Bcl-2, and Bax in renal cell carcinoma and to investigate their role in the tumorigenesis. Methods: The expressions of mitochondrial protein, Bcl-2 and Bax were detected by immunohistochemistry SP method in 9 tissue samples of renal on-cocytoma, 6 samples of chromophobe carcinoma, 23 samples of clear cell carcinoma and 12 samples of normal renal tissue. Results: The expression of MAB1273 in renal oncocytoma, chromophobe carcinoma and clear cell carcinoma was much higher than that in normal renal tissue (P=0.006). No significant difference was found in MAB1273 expression among the three types of renal carcinoma. The expression of Bcl-2 was higher in the renal carcinoma groups than in the normal renal tissue group, with a significant difference (P=0.008). The expression of Bax was not different among all of the groups (P=0.057). Rank correlation analysis showed a positive correlation between MAB1273 and Bcl-2 expression (r=0.341, P=0.015). The expression of Bcl-2 was negatively correlated with Bax expression (r= -0.287, P=0.043). Conclusion: Overexpression of mitochondrial protein and Bcl-2 and underexpression of Bax may play a part in the genesis of renal oncocytoma, chromophobe carcinoma and the clear cell carcinoma, indicating that overexpression of MAB1273 may be correlated with apoptosis of renal cell carcinoma cells.
6.Immunosensor Based on Immobilizing Antibody of Aflatoxin B_1 Using Silica Sol-Gel Technology

In the presence of hydrochloric acid, tetraethoxysilicane was hydrolyzed and formed silica sol. Non-labeled immunosensor was fabricated by droping the mixture solution of the silica sol and antibody of aflatoxin B_1 on the surface of glassy carbon electrode. In this work, a Fe(CN)_6~(3-/4-) phosphate buffer solution) was employed as base solution for investigating cyclic voltammetry(CV) and electrochemical impedance spectroscopic(EIS) performances of the sensor, respectively. The experimental results t indicated that because of the complex formed by the immunoreaction hindered the diffusion of Fe(CN)_6~(3-/4-) on the electrode surface, the redox peak current of the immunosensor in CV obviously decreased, and its electron transfer impedance linearly) increased with increasing the concentration of aflantoxin B_1(AFB). When the medium acidit and incubation) time were pH 6.5 and 20 min, respectively, the biggest electron transfer impedance changed value before and after the immunoreaction was obtained. Under the optimal conditions, a linear range to concentration of aflatoxin B_1 was 1-10 μg/L with a detection limit of 0.1 μg/L(S/N=3). Proposed method is of high sensitivity and stability, it has been successfully applied to determine AFB_1 in maize, rice and peanut.
7.On mitochondrial quality control regulated by Bcl-2 family

Mitochondrial quality control is the important mecha-nism that regulates the morphology,quantity and quality of mito-chondrial in cell to maintain cellular homeostasis and thus,cell survival and health.It has been revealed that members of Bcl-2 family are linked to mitochondrial function and integrity.Bcl-2 family proteins are the key regulators of mitochondrial quality control,participating in the signaling pathways regulating the
crosstalk between mitophagy and apoptosis,as well as mitochon-drial fission and fusion.This paper mainly reviews their impact on mitochondrial quality and the major signaling pathways regula-ted by Bcl-2 family proteins.
8.Global views on clinical trials and data quality.

The quality and integrity of clinical trials and associated data are not only derived from accuracy of trial data analyses, but also closely embodied to the authenticity and integrity of those data and data documents as well as the compliant procedures obtaining those data and relevant files in the life cycle of clinical trials. The compliances of good clinical practices and standards suggest the reliability, complete and accuracy of data and data documents, which is constructing the convincible foundation of drug efficacy and safety validated via clinical trials. Therefore, the monitoring and auditing on clinical trials and associated data quality keep eyes on not only verifications of reliability and correctness on the data analytic outcomes, but also validation of science and compliance of the trial management procedure and documentations in the process of data collections.
9.Effects and mechanism of interferon-gamma on vasculogenic mimicry of melanoma cells

Objective To investigate the effects of interferon-gamma (IFN-γ) on migration,invasion and vasculogenic mimicry (VM) formation of human melanoma cell line MUM-2B. Methods MUM-2B cells were divided into three groups, control group (10%FBS in DMEM), treatment group1 (10μg/L IFN-γ) and treatment group2 (100μg/L IFN-γ). Different concentrations of IFN-γ were added in the culture medium of MUM-2B cells. Wound-healing assay and matrigel invasion assay were performed to examine the migration and invasion ability of MUM-2B cells. Three-D culture was used to observe the VM formation. The expression of vascular endothelial growth factor (VEGF) of MUM-2B cells was detected by Western blot assay. Results The result of wound-healing assay showed that the migration distance of cells was decreased in treatment groups compared with that of control group. The migration distance of cells was decreased in treatment group 2 compared with that of treatment group 1(P<0.05). The result of matrigel invasion assay showed that the number of invaded cells was decreased in treatment groups compared with that of control group, and which was significantly decreased in treatment group2 than that of treatment group1 (P<0.05). The result of 3-D culture showed that cells in control group can form typical VM tube-like structures, whereas cells in treatment groups cannot. Western blot assay showed that the expression of VEGF protein was significantly decreased in treatment groups compared with that of control group, and the expression of VEGF protein was significantly decreased in treatment group2 than that of treatment group 1(P<0.05). Conclusion These data suggest that IFN-γinhibits migration and invasion of MUM-2B cells, and inhibits VM formation by down regulating VEGF expression in vitro.
10.The Influence of SYT-SSX Fusion Gene, E-cadherin and β-Catenin on the Metastasis of Synovial Sarcoma

Objective: To study the prognostic significance of the subtype of SYT-SSX fusion gene, E-cadherin, β-Catenin and clinicopathologicel parameters for the metastasis of synovial sarcomas. Methods: A total of 98 synovial sar-coma patients with complete clinical and follow-up data were reviewed. RT-PCR was used to detect the subtype of SYT-SSX fusion geneo The expression of E-cadherin and β-catenin was detected by immunohistochemistry. Univariate and multivariate analyses were performed to analyze the influence of the above factors and clinicopathological parameters on the metastasis free survival to explore the factors affecting the metastasis of synovial sarcoma. Results: Of all the pa-tients, 69.4% (68/98) had metastasis during follow-up. The median metastasis free survival was 48 months. The metastasis free 1-, 2-, 3-, 4-, and 5-year survival rate after surgery was 97.5%, 75.5%, 63.5%, 54.0%, and 48.5%, respectively; 31.6% (31/98) patients were found with SYT-SSX1 and 68.4% (67/98) patients with SYI-SSX2. The positive rate of E-cadherin ex-pression was 38.8% (38/98), the positive rate of β-catenin expression was 39.8% (39198) on cellular membrane and 53.1% (52/98) in cellular nucleus/cytoplasm. Univariate analysis showed that age (P=0.003), mitotic figure (P=0.002), histological grade (P=0.001), the subtype fusion gene of SYT-SSX (P=0.014), E-cadherin expression (P=0.015) and β-catenin expres-sion on cellular membrane (P=0.020) were significantly correlated with metastasis free survival of synovial sarcoma pa-tients. Sex (P=0.190), tumor location (P=0.105), tumor size (P=0.180), histological type (P=0.354), necrosis (P=0.451), β-catenin expression in cell nucleus/cytoplasm (P=0.911), radiotherapy (P=0.193), and chemotherapy (P=0.249) had no sig-nificant correlation with metastasis free survival of synovial sarcoma patients. Multivariate analysis revealed that the sub-type of SYT-SSX1 fusion gene (RR=2.505, P=0.003), negative expression of E-cadherin (RR=3.282, P=0.000), patient age (RR=2.157, P=0.004), and grade Ⅲ (RR=1.784, P=0.030) were independent risk factors for metastasis of synovial sarco-ma. Conclusion: The subtype of SYT-SSX, expression of E-cadherin, histological grade and the age of patients are impor-tant factors for evaluating the metastasis and prognosis of synovial sarcoma.
