1.Preliminary analysis of miRNA expression profile of gastric cancer tissues by microarray

Objective To analyze the expression profile of specific miRNA of gastric cancer tissues.Methods miRNA were isolated from five paired gastric cancer tissues and corresponding paracancerous normal tissues.To identify the profile of gastric cancer,miRNA hybridization and cluster analysis were processed by significance analysis of microarrays (SAM,version 2.1) and cluster software.The relative expression of hsa-miR-30c,hsa-miR-196a and hsa-miR-193b were validated by fluorescence Real-Time PCR using taqman probe.Further hsa-miR-30c taget gene was predictived by online software.Results Revealed by miRNA microarray detection,there were 16 miRNA with differential expression in gastric cancer tissues and para-cancerous normal tissues,among which HS_100,hsa-miR-27a,hsa-miR-214*,solexa-555-1991 and hsa-miR-196 were significantly up-regulated,and hsa-miR-502-3p,hsa-miR-29c*,hsa-miR-64,hsa-miR-193b,hsa-miR-551b,hsa-miR-30c,hsa-miR-582-5p,hsa-miR-625*,hsa-miR-660,hsa-miR-363 and hsa-miR-486-5p were down-regulation.The relative expression of hsa-miR-30c,hsa-miR-196a and hsa-miR-193b was in high concordance with microarray.Conclusion There is specific miRNA expression profile in gastric cancer tissues,among which differentially-expressed miRNAs may be involved in the regulation of gastric cancer progress and serve as new potential biomarkers.

3.Trend in disease burden of stroke in Linping District from 2014 to 2023

Objective:
To analyze the trend in disease burden of stroke in Linping District, Hangzhou City from 2014 to 2023, so as to provide the basis for prevention and control of stroke.
Methods:
Data on incidence and mortality of stroke in Linping District from 2014 to 2023 were collected through the Chronic Disease and Death Cause Monitoring System of Hangzhou Center for Disease Control and Prevention. The disease burden was evaluated by calculating years of life lost (YLL), years lived with disability (YLD) and disability-adjusted life years (DALY) with reference to the methodology of the Global Burden of Disease Study, and standardized using the data of the seventh national population census in 2020. The trend was analyzed using average annual percent change (AAPC). The disease burden of stroke attributed to risk factors was calculated.
Results:
The crude incidence of stroke was 298.13/105 and the crude mortality was 62.72/105 in Linping District from 2014 to 2023. The YYL, YLD and DALY caused by stroke were 49 413.60, 8 197.68, and 57 611.28 person-years, respectively, with decreasing trends in YLL and DALY, and an increasing trend in YLD (AAPC=-12.663%, -10.427% and 3.035%, all P<0.05). The standardized YLL rate, standardized YLD rate and standardized DALY rate were 9.54‰, 1.53‰ and 11.07‰, respectively, showing decreasing trends (AAPC=-16.011%,-0.975% and -13.791%, all P<0.05). Higher standardized DALY rate (P<0.05) and slower decline were seen in men than in women (AAPC=-13.025% vs. -14.550%, both P<0.05). The highest DALY of stroke was attributed to combined risk factors (men, 18 754.05 person-years; women, 13 256.60 person-years), followed by hypertension (men, 12 231.23 person-years; women, 9 373.77 person-years). The DALY of stroke attributed to smoking was 15.67 times higher in men than in women, and the DALY of stroke attributed to a lack of exercise was 1.27 times higher in women than in men.
Conclusion
There was an overall decreasing trend in the disease burden of stroke in Lingping District from 2014 to 2023, with a higher disease burden in men. The coexistence of multiple risk factors had a greater impact on stroke.
4.The curative effect of music-regulated laser therapy and the quality of life of patients with primary hyper-tension

Objective To assess the therapeutic effect of music- regulated laser therapy on mild and moderate primary hypertension (PH) so as to provide clinical evidence for the development of an effective non-drug therapy for hypertension. Methods One hundred mild and moderate PH patients were divided into obser-vation ( n = 50 ) and control groups ( n = 50 ) according to their stage of hypertension. There were two phases of treatment with 6 weeks in each phase for a total of 12 weeks. In the first phase, patients in the observation group received music-regulated laser therapy while the control group received drug therapy. In the second phase, the treatments were reversed, with the observation group receiving the drugs. The blood pressure and quality of life (QOL) of all the patients was evaluated after each phase. Results Before therapy there were no significant differences in average blood pressure or QOL between the groups. After the first phase, systolic blood pressure (SBP) and diastolic blood pressure (DBP) had been reduced significantly in both groups, but the decrease was significantly greater in the control group. The score on each item of the QOL evaluation was not significantly different between the two groups. After the second phase, blood pressure had decreased further in the observation group, but no significant change was observed in the control group, and there was no significant difference in blood pressure between two groups. The scores on each item of the QOL evaluation were not significantly different between the two groups, but average somatic symptoms, healthy and pleasant feelings, task perform-ance and life satisfaction were significantly ameliorated in both groups. Conclusions Music-regulated laser therapy can lower the blood pressure of mild and moderate PH patients effectively. Compared with antihyperten-sion drugs, music-regulated laser therapy provides a weaker effect in lowering blood pressure, but plays an e-quivalent role in improving the QOL of patients.
5.Global views on clinical trials and data quality.

The quality and integrity of clinical trials and associated data are not only derived from accuracy of trial data analyses, but also closely embodied to the authenticity and integrity of those data and data documents as well as the compliant procedures obtaining those data and relevant files in the life cycle of clinical trials. The compliances of good clinical practices and standards suggest the reliability, complete and accuracy of data and data documents, which is constructing the convincible foundation of drug efficacy and safety validated via clinical trials. Therefore, the monitoring and auditing on clinical trials and associated data quality keep eyes on not only verifications of reliability and correctness on the data analytic outcomes, but also validation of science and compliance of the trial management procedure and documentations in the process of data collections.
6.Mediating effect of cognitive emotion regulation strategies between emotional intelligence and loneliness

Objective To explore the mediating effect of cognitive emotion regulation strategies between emotional intelligence and loneliness. Methods 542 university students were surveyed by the Chinese version of Cognitive Emotion Regulation Questionnaire ( CERQ) ,the Emotional Intelligence Questionnaire and UCLA Loneli?ness Scale,and the correlation analysis was conducted by using SPSS17.0. The path model was set up and its path analysis was conducted by using AMOS17. 0. Results Positive cognitive emotion regulation strategies ( 28. 64 ± 5.90) were positively correlated with emotional intelligence(2.38±0.27, r=0.346, P<0.05) and negatively correla?ted with loneliness(42.29±7.88, r=0.301, P<0.05);Path analysis showed that the estimates of indirect and direct effects support the hypothesis respectively(P<0.05). Conclusion Positive cognitive emotion regulation strategies serve as partial mediators between emotional intelligence and loneliness.
7.THE PARANODAL REGION AND AXO-GLIAL JUNCTION OF MYELINATED FIBERS

By using electron microscopy, the paranodal region and axo-glial junction were examined in optic nerves of rats aged 14 days. The paranodal region was characterized in longitudinal sections by the sequential termination of the myelin lamellae, beginning proximally with the innermost and ending, at the Ranvier node, with the outermost lamella. The termination of each lamella was accom- panied by a separation of the major dense line of the compact myelin and the consequent formation of a "loop" of glial cytoplasm. Each paranodal loop inde- nted the axonal surface as it became junctionally apposed to the axolemma. The periaxonal extracellular space, 10-20nm in width in the internodal region and reduced at the paranodal junction to approximately 3nm, forming an axo-glial junction, which was thought to be held together by dense structure. The parano- dal junction seems to serve strong adhesion between the apposed axonal and glial membranes. Conduction of the nerve impulses in myelinated axons was saltatory. Axons and sheath cells probably maintain vital communication with one another, presumably at the paranodal junctional complex. This communication was viewed as vital to the stability and maintenance of myelin. We found some clear vesi- cles in axoplasm near the Ranvier node and speculated that there were endocyto- sis and exocytosis in paranodal region. This was a direct morphological evidence supporting metabolic coupling between axons and sheeth cells.
8.Effects and mechanism of interferon-gamma on vasculogenic mimicry of melanoma cells

Objective To investigate the effects of interferon-gamma (IFN-γ) on migration,invasion and vasculogenic mimicry (VM) formation of human melanoma cell line MUM-2B. Methods MUM-2B cells were divided into three groups, control group (10%FBS in DMEM), treatment group1 (10μg/L IFN-γ) and treatment group2 (100μg/L IFN-γ). Different concentrations of IFN-γ were added in the culture medium of MUM-2B cells. Wound-healing assay and matrigel invasion assay were performed to examine the migration and invasion ability of MUM-2B cells. Three-D culture was used to observe the VM formation. The expression of vascular endothelial growth factor (VEGF) of MUM-2B cells was detected by Western blot assay. Results The result of wound-healing assay showed that the migration distance of cells was decreased in treatment groups compared with that of control group. The migration distance of cells was decreased in treatment group 2 compared with that of treatment group 1(P<0.05). The result of matrigel invasion assay showed that the number of invaded cells was decreased in treatment groups compared with that of control group, and which was significantly decreased in treatment group2 than that of treatment group1 (P<0.05). The result of 3-D culture showed that cells in control group can form typical VM tube-like structures, whereas cells in treatment groups cannot. Western blot assay showed that the expression of VEGF protein was significantly decreased in treatment groups compared with that of control group, and the expression of VEGF protein was significantly decreased in treatment group2 than that of treatment group 1(P<0.05). Conclusion These data suggest that IFN-γinhibits migration and invasion of MUM-2B cells, and inhibits VM formation by down regulating VEGF expression in vitro.
9.Activation and significance of the PI3K/Akt pathway in endometrium with polycystic ovary syndrome patients

Objective To investigate activation of the phosphatidylinositol 3-kinase(PI3 K)/protein kinase B(Akt)pathway in the endometrium of women with polycystic ovary syndrome(PCOS)and its role in endometrium hyperplasia and carcinogenesis,and the factors affecting the activation of the PI3K/Akt pathway.Methods From Jan 2007 to Jun 2008,52 patients with PCOS who underwent dilatation and curettage were selected as experimental group matched with 32 non-PCOS patients as control group.Serous hormonal parameters,fasting blood glucose and insulin,body mass index(BMI),and endometrium pathology were measured and evaluated in all patients.The PCOS patients were divided into insulin resistance and non-insulin resistance group according to homeostasis model assessment-insulin resistance index (HOMA-IR).Meanwhile,the PCOS patients were grouped as normal,endometrial hyperplasia and carcinoma depending on outcome of pathology.The expression of Akt and phosphorylated Akt(p-Akt)were determined by western blot.Results(1)The expression of p-Akt was significantly higher in PCOS group [(46±18)%]than that in control[(33 ±9)%,P <0.01)].(2)The expression of p-Akt was significantly higher in group of endometrial hyperplasia and carcinoma[(56 ± 19)%]when compared with those in normal endometria group[(31 ± 12)%,P < 0.05]; the expression of p-Akt was significantly higher in group of insulin resistance[(50 ± 19)%]compared with that in non-insulin resistance group [(34 ± 10)%,P <0.01].(3)There was a positive correlation between the expression level of p-Akt in endometrium with PCOS and HOMA-IR and BMI respectively(r =0.400,0.326,both P < 0.05).Conclusions The PI3K/Akt pathway was over activated in endometrium with PCOS which may be associated with the formation of endometrial hyperplasia and carcinoma in PCOS patients.Insulin resistance and obesity may be high risk factors for over-activation of the PI3K/Akt pathway in endometrium with PCOS.
10.Applying Six Sigma management to shorten the waiting time of outpatients accepting radiography

Objective To evaluate the efficacy of Six Sigma management for shortening the waiting time of outpatients accepting radiography and improving patient satisfaction. Methods Six Sigma management was applied to identify the key points critical to quality in the process of radiography,analyze the factors influencing the waiting time,and take targeted measures to modify the process. Results The waiting time of patients accepting radiography was reduced by an average of 25.2min and the rate of dissatisfaction was decreased by 18%. Conclusion The application of Six Sigma management can effectively shorten the waiting time of outpatients accepting radiography and increase patient satisfaction.
