OBJECTIVE To discuss risk factors of hospital infection in patients who suffered from primary cancer of the liver in the medium and advanced stage and give the countermeasures.METHODS The liver cancer cases in medium and advanced stage in 2001-2006 were retrospectively investigated and analyzed.RESULTS The hospital infection rate was 33.92% for the liver cancer patients who were in the medium and advanced period;the more common sites of infection were lungs,abdominal cavity,intestinal tract,blood.The motality was 20.83% in the infection group and 10.16% in non-infection group and there were significant differences between two groups(P
2.Establishment and biological characteristics of nude mice xenograft tumor models of human endometrial carcinoma

Objective To establish nude mice xenograft tumor models of human endometrial carcinoma for basic and clinical study. Methods Fourteen samples of human endometrial carcinoma were subcutaneously heterotransplanted to nude mice (BALB/C, nu/nu),3~4 mice were transplanted for every sample. Results The initial take rate was 42.8%,then it increased to 100.0% after the 5th passage. Five of 14 samples were transplanted for 48~63 passages.The characteristics of histology,ultrastructure and chromosome were identical to those of human donour tumors. The DNA ploidy was consistent for SL-1,SL-2 and SL-3,but changed for SL-4 and SL-5 by cytometric analysis. The expression of estrogen receptor and progesterone receptor for 5 models was all negative. The expression of p53 was positive for SL-1,SL-2 and SL-4,but negative for SL-3 and SL-5.The expression of c-erbB-2 was positive for SL-1,SL-2 and SL-4,negative for SL-3 and SL-5.The expression of p16 was positive for SL-2 and SL-3,negative for SL-1,SL-4 and SL-5. Conclusions Five nude mice xenograft tumor models of human endometrial carcinoma were established successfully. It will be helpful for the basic and clinical study of human endometrial carcinoma.
3.Expression of long non-coding RNA 1010001N08Rik in bronchopulmonary dysplasia and its bioinformatics analysis

Objective To explore the expression feature of long non-coding RNA (lncRNA) 1010001N08Rik in hyperoxia-induced bronchopulmonary dysplasia (BPD) and predict the mechanism that 1010001N08Rik might be involved in the occurrence and development of BPD by a series of bioinformatics analysis.Method The sequence,genomic position and structure characteristics of 1010001N08Rik were acquired from UCSC genome browser,and its target gene was predicted by Ensemble database.We successfully established the animal model of BPD by making newborn C57BL/6J mice exposed to 95% concentrations of ambient oxygen for seven days.The expression of 1010001N08Rik and Gata 6 were detected using real-time quantitative polymerase chain reaction (PCR).Student's t test was used to compare their expression levels during the BPD process.Result The relative expression of 1010001N08Rik in BPD process at d1,d3,d5,d7 was 1.21 ± 0.33,2.02 ± 0.41,2.95 ± 0.45,4.20-± 0.48 respectively,and there were significant difference between adjacent time points (P < 0.05).The relative expression of Gata 6 mRNA was 0.92 ±0.30,1.10 ± 0.31,0.86 ± 0.24,0.45-± 0.08 respectively,and there was significant difference between d5 and d7 (P <0.05).1010001N08Rik had highly conserved property among different species.The chromosomal regions of 1010001N08Rik existed transcriptional factors binding locations and epigenetic regulation clues,and its possible candidate target gene was Gata 6.Conclusion The expression of 1010001N08Rik increased during the formation process of BPD.Bioinformatics analysis and preliminary experiment results suggested that 1010001N08Rik might participate in the process of BPD by down-regulating Gata 6 expression.
4.Angiogenesis related gene expression profiles of human endometrial endothelial cells isolated from tissue of endometrial cancer

Objective To identify and verify the different genes expression pattern between human endometrial endothelial cells (HEEC) isolated from endometrial cancer and normal endometrium.Methods Endothelial cells were isolated from 5 patients with endometrial cancer (endometrial cancer group 1) and 5 patients with normal endometria tissue (control group 1) admitted from June to November 2007 in Shandong Cancer Hospital.Global expression patterns of endothelial cells were examined using oligonucleotide microarrays.Tissues from 36 patients with endometrial cancer(endometrial cancer group 2) and 10 normal endometrial tissues (control group 2) admitted from January 2007 to April 2008 were selected to verify the expression of different genes,in which up-regulated genes including ESM1,MMP-10,SPP1 and HMGB1 were tested by quantitative real-time PCR and immunohistochemistry.Results Microarray analyses revealed 317 genes that exhibited > 2-fold or < 0.5 differences were identified (including 191 genes up-regulated and 126 down-regulated).Pathway analysis showed that these genes involved cell cycle,cell adhesion molecules,and extracellular matrix-receptor interaction were obviously predominant.Of them,97 up-regulated genes and 44 down-regulated genes were related to angiogenesis.The mRNA expression of ESM1,MMP-10,SPP1 and HMGB1 in endometrial cancer group 2 were 0.898、3.890、1.433 and 1.881,respectively.Positive expression of SPP1,MMP-10,ESM1 and HMGB1 was observed in endometrial cancer group 2.However,the SPP1,ESM1 and HMGB1 was negative expressed in control group 2.Conclusion It shows that there are the different angiogenesis related genes between endometrial cancer and normal endothelium,which will provide insights into the anti-angiogenesis therapy for endometrial cancers.
5.Effects of optimization of experimental specimens on practical teaching of systematic anatomy

We optimized the randomly combined specimens of limb bones into paired specimens of limb bones and used optimized (paired) specimens in systemic anatomy experimental teaching to strengthen the practical ability of medical students.Results showed that the teaching effect was better by using optimized (paired) specimens than by using randomly combined specimens.Applying optimized (paired) specimens was conducive to training students' practical ability and increasing the using efficiency of specimens.
6.Clinicopathological characteristics of hereditary ovarian cancer syndrome

Objective To explore the clinicopathological characteristics of hereditary ovarian cancer syndrome(HOCS). Methods From Jan. 2000 to Jan. 2007, among 580 cases of primary ovarian cancer, 42 cases(herediatary group),who had a positive family history of ovarian cancer and met the diagnostic criteria of HOCS, were analyzed retrospectively. One hundred cases without a family history of ovarian cancer were enrolled randomizely as control group (sporadic group). Results The incidence of HOCS was 7.2% (42/580). Forty-two cases associated tumors affected at least 2 successive generations in 31 families and affected 1 generation in 8 families. Eighty-seven percent (27/31)was from maternal lineage, while 13% (4/31)from paternal lineage. Earlier age of onset was significantly difference between two groups[(49±10) years vs. (55±10) years, P<0.05]. There were 90% belong to serous adenocarcinoma in the herediatary group, while 84% in the sporadic group. There was statistical difference in the proportion of mucinous adenocarcinoma (0 vs. 11%, P<0.05). The most common clinical manifestations were abdominal distention and anorexia (64% vs. 70%, P>0.05), International Federational of Gynecology Obstetrics(FIGO)stage Ⅲ (62% vs. 63%, P>0.05) between two groups. Fourteen cases (33%,14/42) were previously untreated in the herediatary group, while 40 cases (40%, 40/100) in the sporadic group. There were 15 cases (36%, 15/42) underwent secondary surgery and 15 cases (36%, 15/42) underwent third surgery or more in berediatary group, while 50 cases (50%, 50/100) and 27 cases(27%, 27/100) in the sporadic group. The mean number of ehemotberapy cycles received in two groups was 13.3 and 11.8 (P>0.05). The 3-year and 5-year survival rate in herediatary group were 73.6% and 54.9% respectively, compared with 47.4% and 21.2% (P<0.05) in sporadic group. Conclusion Hereditary ovarian cancer mostly from maternal lineage are featuring in early age of onset, serous adenocarcinoma, advanced stage (stage Ⅲ), and better prognosis after the comprehensive treated by cytoreductive surgery plus with chemotherapy.
7.Analysis of molecular features of clinical Vibrio parahaemolyticus strains in China.

OBJECTIVETo explore the phenetic and genetic features of clinical Vibrio parahaemolyticus strains from 2007-2009 in China.
METHODSA total of 135 clinical Vibrio parahaemolyticus strains, isolated from Zhejiang, Jiangsu, Sichuan, Guangxi, Liaoning Provinces during 2007 to 2009, were selected for the research. The occurrence of virulence genes thermostable direct hemolysin (tdh) and TDH-related hemolysin (trh), species-specific genes thermolabile hemolysin (tlh), toxR, VPM and gyrB, the pandemic clone gene markers(GS-PCR, PGS-PCR, orf8 and HU-α) in 135 Vibrio parahaemolyticus strains was detected by PCR. The antimicrobial susceptibilities to eight antimicrobial agents of the experimental strains were determined by the broth microdilution method. All strains were serotyped and underwent the cluster analysis with pulsed-field gel electrophoreses.
RESULTSThe results of PCR methods claim that all experiment strains carry species-specific genes such as tlh, toxR, gyrB, VPM. Among clinical strains, 85.9% (116/135) carry tdh and/or trh. 85.2% (115/135) were positive for tdh, and 3.0% (4/135) were positive for trh; while 3 strains carried both.66.7% (90/135) , 80.7% (109/135) , 65.2% (88/135) , 66.7% (90/135) clinical strains carried the genes of GS-PCR, PGS-PCR, orf8, HU-α, respectively. The results of antibiotics susceptibility test showed that 8.1% (11/135) strains were resistant to at least one agent, including 9 strains were resistant to ampicillin, 2 strains were resistant to trimethoprim and sulphamethoxazole, and 1 strain were resistant to tetracycline. All clinical strains were sensitive to cefotaxime, ceftazidime, gentamicin, ciprofloxacin and chloromycetin.Serological analysis of the O and K antigens claimed that a total of 29 serotypes were identified for clinical strains, predominantly O3, O4 and O1 groups, accounting for 89.6% (121/135). O3: K6 was dominant serotype, accounting for 56.3% (76/135). The pandemic flora in China included O3: K6, O4: K68, O1: K36, O1: K25, O1: K5 and O3: K29 serotypes.Genomic DNAs of 135 clinical strains were digested with SfiI and NotI, the molecular size of PFGE restriction fragments used for analysis mainly ranged from 30-700 kb.When subjected to UPGMA clustering, 6 and 9 clusters were grouped by SfiI and NotI, and the minimal similarity was 52.6% and 58.7%, and pandemic flora were located in C groups and D group, respectively.
CONCLUSIONMost of Vibrio parahaemolyticus strains isolated from clinical sources in China were pathogenic. The pandemic clone, especially O3: K6 was prevalent. The GS-PCR and HU-α genes were reliable markers to identify the pandemic flora. The serotype by PFGE was reliable to distinguish the pandemic flora and the sporadic strains.

8.Study on the Improvement Effect and Mechanism of Ilicifoliosids Alkaloid A on Non-alcoholic Fatty Liver Di- sease in Rats

OBJECTIVE:To s tudy improvement effect and mech anism of ilicifoliosids alkaloid A (HBOA)on non-alcoholic fatty liver disease in rats. METHODS :SD rats were randomly divided into blank control group ,model group ,Silybin capsule group(positive control ,26.25 mg/kg),HBOA high-dose ,medium-dose and low-dose groups (100,50,25 mg/kg),with 10 rats in each group. Except that blank control group fed normal feed ,the other groups were continuously fed with high-fat diet for 8 weeks to induce non-alcoholic fatty liver disease model. Form the 9th week ,blank control group and model group were given constant volume of 0.6% CMC-Na solution ,and administration groups were given corresponding drugs by intragastric admini- stration,once a day ,for consecutive 4 weeks. The general information of rats were observed and the body weight increase ,organ (liver,kidney and spleen )indexes were calculated ;the contents of AST ,ALT,TC,TG and NEFA in liver tissue were detected , and SOD,GSH-Px activities and MDA content in the serum were also determined. The protein expression of PPARα in liver tissue was detected by immunohistochemistry. RESULTS :Compared with blank control group ,the body mass increase and liver index of rats in model group were increased significantly (P<0.01);fat deposition could be observed in the liver ;the activities of SOD and GSH-Px in serum were reduced significantly ,and the contents of MDA ,the contents of AST ,ALT,TC,TG and NEFA in liver tissue were significantly increased ,and the protein expression of PPARα was decreased significantly(P<0.01). Compared with model group ,the body mass increase and liver index of the rats were decreased significantly in administration groups (P<0.05 or P<0.01),liver fat deposition was improved ,the activity of SOD and GSH-Px in serum (except for HBOA low-dose group )were increased significantly while MDA content ,the contents of AST ,ALT,TC(except for HBOA low-dose group ),TG(except for HBOA low-dose group ) and NEFA in liver tissue were decreased significantl y,while protein expression of PPAR α 15177460685@163.com was increased significantly (P<0.05 or P<0.01). 9.Effects of 4-hydroxy-2(3H)-benzoxazolone on Inflammatory and Apoptosis Signaling Pathways in Non-alcoholic Fatty Liver Disease Model Rats

OBJECTIVE:To inv estigate the effects of 4-hydroxy-2(3H)-benzoxazolone on inflammatory and apoptosis signaling pathways in non-alcoholic fatty liver disease (NAFLD)model rats. METHODS :SD rats were divided into normal control group(10 rats)and modeling group (50 rats). Normal control group was given basic diet ,and modeling group were given high-fat diet to induce NAFLD model. After modeling ,the rats were divided into normal control group ,model group ,silibinin group (26.25 mg/kg),and 4-hydroxy-2(3H)-benzoxazolone high-dose ,medium-dose and low-dose groups (100,50,25 mg/kg),with 8 rats in each group. Normal control group and modeling group were given 0.6% CMC-Na intragastrically ,and other groups were given relevant medicine 10 mL/kg intragastrically ,once a day ,for consecutive 4 weeks. After last medication ,the serum levels of albumin(ALB),total protein (TP),globulin(GLB),ALB/GLB and free fatty acid (FFA)were detected ;TUNEL staining was used to observe the apoptosis of rat hepatocytes. Western blot assay was used to detect the protein expression and phosphorylation level of inflammatory signaling pathway related proteins [Toll-like receptor 4(TLR4),myeloid differentiation factor 88(MyD88), nuclear factor-κB p65(NF-κB p65),NF-κB inhibitor protein(IκBα)] in liver tissue as well as the expression of apoptosis signaling pathway related proteins [B cell lymphoma 2(Bcl-2),Bax,caspase-3]. RESULTS :Compared with model group ,serum levels of TP (except for low-dose group ),GLB and FFA ,the protein expression of TLR 4(except for low-dose group ),MyD88 (except for medium-dose group )and caspase- 3,the phosphorylation levels of NF-κB p65 and IκBα protein were decreased significantly(P<0.05 or P<0.01). The ratio of A LB/GLB in serum and the ratio of Bcl- 2/Bax in liver tissue were significantly increased(P<0.05 or P<0.01),and the phenomenon of hepatocyte apoptosis was improved. CONCLUSIONS :4-hydroxy-2 (3H)-benzoxazolone can ameliorate NAFLD in rats ,and the mechanism may be associated with inhibiting the expression TLR 4/ MyD88/NF-κB signaling pathway-related proteins and apoptosis-related proteins in liver tissues.
