1.Determination of riboflavin in rabbit corneal stroma under three interventions

Backgroud EDTA is an important calcium chelator,commonly used as an ophthalmic additive,its role in promoting corneal penetration of drug may facilitate transepithelial corneal collagen crosslinking.However,there were few reports about the penetration of riboflavin to corneal stroma enhanced by EDTA both home and abroad.Objective To determine the concentrations of riboflavin in corneal intrastromal under different interventions using high-performance liquid chromatography (HPLC).To ensure the efficacy and safety of 0.5% EDTA in riboflavin penetrating to corneal stroma by the transepithelial procedures.Methods Fifteen New Zealand white rabbits were randomized into three groups,corneal epithelium of 5 rabbits were debrided,corneal epithelium of another 5 rabbits were left in situ and 0.5% EDTA-Na2 was topically applied for 1 hour,and the corneal epithelium of last 5 rabbits were just left in situ,after those procedures 0.1% riboflavin + 20% dextran was applied topically to the corneas for 30 minutes(1 drop every 3 minutes).Then,the epithelium in the corneas of EDTA and epi-situ group were carefully removed before all the corneas were punched with an 8.5 mm diameter blade and homogenized subsequently.Ultimately,the riboflavin concentrations were determined by HPLC.Results After treated with riboflavin for 30minutes,the mean concentration of riboflavin in the epi-removed group was (23.54± 1.61)μg/g tissue,the EDTA group was (2.04 ±0.25)μg/g tissue and the epi-situ group was (1.44 ±0.06)μg/g tissue,showing significant difference among them (F =1792.839,P =0.000).There were statistically significant differences between any two groups (t =41.780,7.484,43.408,all at P < 0.05).However,the riboflavin concentration of other two groups were failed to meet the theoretical value 15 μg/g tissue,excepted for epi-removed group.Conclusions The diffusion of riboflavin to the corneal stroma can be fully enhanced by removing the epithelium preoperatively,penetration enhancer EDTA can effectively promote the penetration of riboflavin to the cornea,but it still can not replace the role of removing epithelium on the penetration of riboflavin to the cornea.
3.A two - year's results of iontophoresis -assisted transepithelial corneal cross -linking for progressive keratoconus

AIM: To report a two- year's results of iontophoresis-assisted transepithelial corneal cross-linking (I-CXL) for progressive keratoconus.
METHODS: Thirty - four eyes in 24 patients with progressive keratoconus ( mean age 21. 0 ± 5. 6 years;range: 14-32 years) were treated. After 1g/ L riboflavin-distilled water solution was administered by iontophoresis-assited (current 1mA) transepithelial method for 5min in total, standard surface UVA irradiation ( 370nm, 3mW/cm2 ) was performed at a 1 - cm distance for 30min. The best corrected visual acuity ( BCVA ) measured as LogMAR number, corneal refractive astigmatism, K1, K2, Kmean, Kmax, intraocular pressure, endothelial cell density, the thickness at corneal apex and the thinnest point were measured preoperatively and 2a postoperatively.
RESULTS:At 2a after the procedure, BCVA (LogMAR) improved from 0. 32 ± 0. 25 to 0. 25 ± 0. 19 ( t = 2. 849, P =0. 015). K1 decreased from 47. 12±4. 33 to 46. 06±4. 77 (t =2. 652, P= 0. 015). K2 decreased from 51. 36±5. 59 to 50. 40±6. 16 (t= 2. 121, P= 0. 047). Kmean decreased from 49. 12±4. 76 to 48. 10±5. 25(t = 2. 663, P = 0. 015). Kmax decreased from 57. 57±8. 30 to 55. 91±8. 14 (t = 2. 398, P = 0. 026). The corneal apex thickness decreased from 476. 90±38. 71μ m to 454. 43 ± 40. 86μ m ( t = 2. 853, P = 0. 010 ). The thinnest thickness decreased from 464. 38 ± 39. 92μ m to 433. 86 ±50. 78μ m ( t = 3. 485, P = 0. 002 ). Corneal refractive astigmatism, intraocular pressure and endothelial cell density did not show significant changes.
CONCLUSION: I - CXL for progressive keratoconus is safe and effective which can prevent deterioration of progressive keratoconus within 2a, but further long-term studies are necessary still.
4.Nrf2 as a chemoprevention target in gastrointestinal carcinoma.

Gastrointestinal tract carcinoma is one of the leading causes of cancer-related death in China. Chemoprevention has been considered as a potential approach to control this type of disease. Nuclear factor erythroid 2-related factor 2 (Nrf2) is a redox-sensitive transcription factor that protects cells from oxidative/electrophilic stresses by activating the expression of a battery of cytoprotective genes through the antioxidant response element (ARE). Recently, Nrf2 has emerged as a novel target for chemoprevention. Several natural or synthetic chemicals, which activate Nrf2/ARE signaling pathway, have showed effect in animal models, and promises in many ongoing clinical trials. This review summarizes the recent findings on the regulation of Nrf2/ARE signaling pathway, and the developments in both preclinical and clinical studies.

5.Analysis on the present running status of water-improving project in Anda city,Heilongjiang province in 2008

Objective To understand the running status and distribution of water-improving pmjects in Anda city.Methods The running and management of the water-improving projects were investigated and located the position by Global Positioning Systerm(GPS)in Anda city.Results Among 317 water-improving projects,16.09%were either long-term projects or in poor management or had already stopped usage,77 projeets were broken,accounting for 24.29%;all inadequate supply of equipment and pipeline,83 projects had never been started,accounting for 26.18%;now,106 projects were running,accounting for 33.44%.In only 46 projects,the water fluoride concentration was lower than 1.00 mg/L,accounting for 14.51%,as a result 36 thousand people benefited.Conclusions The running status of water-improving projects was unacceptable,most of them stopped running and endemic fluorosis control was still severe in Anda city.
6.Lacrimal passage probing combining injection of TobraDex eye ointment for treating chronic dacryocystitis

AIM: To evaluate the effect of probing of lacrimal passage combining injecting of US-produced TobraDex eye ointment for the treatment of chronic dacryocystitis.METHODS: For 127 cases (129 eyes) of chronic dacryocystitis, first the pyoid secretion gathered in the lacrimal sac was dashed out, and some TobraDex was injected in the middle of the lacrimal passage once per day, repeated for several times. The lacrimal passage was probed when the secretion of lacrimal sac disappeared essentially. The lacrimal passage and immit TobraDex eye ointment was used once every two days, repeated for 3-4 times.RESULTS: In 126 cases (128 eyes) the lacrimal passage was dredged. Only one case (1 eye) the youthful patient did not recover for the opening ectopia of lacrimonasal duct.During the 3mo-1a random visit the chronic dacryocystitis did not recrudesce in the cases of lacrimal passage dredging.CONCLUSION: It is simple and safe to use the probing of lacrimal passage combining injecting of US-produced TobraDex eye ointment to treat the chronic dacryocystitis. This method has good curative effects and can keep the normal physiological structure of lacrimal passage. It does not need any expensive medical equipment and cost less, therefore is advisable for patients.
7.Evaluation of 99Tcm-DTPA nuclear dynamic inaging in renal occupied disease

Objective To evaluate 99tcm-DTPA nuclear dynamic imaging in distinguishing the renal occupied disease.Methods A total of 164 in-patients with renal occupied disease who underwent surgery were included.According to the pathological diagnosis,119 patients had malignant tumors,and 45 patients had benign diseases.All patients’ imaging was retrospectively analyzed.Application of 99Tcm-DTPA nuclear dynamic imaging in renal occupied disease was compared with ultrasonography (US),computed tomography (CT),magnetic resonance imaging (MRI),intravenous pyelogram (IVP),and positron emission tomography (PET)-CT.Results The accuracy rates of different imaging methods in distinguishing between renal malignant and benign disease were 99Tcm-DTPA (84 %,45 %),US (72 %,64 %),CT ( 91%,92 %),MRI (50 %,67 %),IVP (50 %, 17 %), respectively.The diagnostic accuracy rate of PET-CT for malignant tumors was 67 %.The accuracy rates of 99Tcm-DTPA in distinguishing different phases of renal cell carcinoma were statistically significant (x 2 =83.4, P < 0.01), while the accuracy rates in distinguishing renal cyst from renal angiomyolipoma were not statistically different.With the greater diameter, the diagnostic accordance rate is higher (x 2 =16.05,P < 0.05).Conclusion 99Tcm-DTPA could be used not only to evaluate the renal function quantificationally,but also be helpful to distinguish renal malignant tumor from benign disease.
8.Nrf2 down-regulated cell line H460-N5 with Keap1 over-expression increased sensitivity to anti-cancer drugs.

OBJECTIVETo maked a Nrf2 down-regulated cell line by over-expressing Keap1 in H460 cells to study the role of Nrf2 in drug resistance.
METHODSTransfecting H460 cells with mKeap1-pEGFP and screenig for Keap1 expressing clones by Western blotting with antibodies against Nrf2, HO-1, NQO1 and AKR1C. The cell line with Keap1 over-expression was further confirmed by real-time PCR. The cytotoxicity of H460-N5 to anti-cancer drugs was evaluated by MTS assay.
RESULTMTS assay results showed the enhanced cytotoxicity of anticancer drugs (Oxaliplatin, Doxorubicin and Etopside) to the H460 cell line with keap1 overexpression compared to the control cell line. In H460-N0 cells, the IC(50) values of Oxaliplation and Etopside were 93 micromol/L and 100 micromol/L respectively whereas the IC(50) values of the two drugs were 42 micromol/L and 30 micromol/L correspondingly in H460-N5 cells. A Nrf2 down-regulated cell line H460-N5 and a control cell line with GFP over-expression have been identified.Down-regulation of Nrf2 enhanced the cytotoxicity of Oxaliplatin, Doxorubicin and Etopside. The IC(50) value of Doxorubicin to H460-N0 cell was above 3 mg/L, but that to H460-N5 cell was about 2 mg/L.
CONCLUSIONA Nrf2 down-regulated cell line H460-N5 and a control cell line with GFP over-expression have been identified. Down-regulation of Nrf2 enhanced the cytotoxicity of Oxaliplatin, Doxorubicin and Etopside.

9.Activation of the immunologic function of rat Kupffer cells by the polysaccharides of Angelica sinensis.

AIMTo observe the influence of polysaccharides of Angelica sinensis (ASP) on the immunologic function of rat Kupffer cells.
METHODSNormal rat Kupffer cells were treated with ASP in vitro. Absorbance at 540 nm ( A540) of neutral red absorption and supernatant NO, TNF-alpha in the cells were measured to evaluate the immunologic function of Kupffer cells; LDH leakage was measured to estimate the severity of cellular damage; Rats were given ASP 0.025, 0.1, 0.25 and 1.0 g x kg(-1) ig (qd x 7 d) in vivo. The above indices and ACP of Kupffer cells were measured, sGST and sALT activity were detected as indices of hepatotoxicity.
RESULTSASP markedly enhanced the phagocytic activity, ACP and supernatant NO, TNF-alpha of Kupffer cells both in vitro and in vivo . The increase of sGST was observed after administration of ASP 1.0 g x kg(-1), but the LDH leakage of the hepatocytes was not increased in vitro.
CONCLUSIONASP with suitable dose could activate the function of Kupffer cells. Slight liver injury was caused by ASP 1.0 g x kg(-1) in vivo, which was likely caused by factors, such as NO, TNF-alpha, indirectly.
