1.Application value of FFA and OCT in diagnosis of central retinal artery and vein occlusion

AIM: To investigate the clinical value of fundus fluorescein angiography ( FFA ) and optical coherent tomography ( OCT) in the diagnosis of central retinal vein occlusion.
●METHODS:A total of 47 cases (47 eyes) central retinal vein occlusion were retrospectively analyzed from Jun. 2012 to Dec. 2015 in our hospital ophthalmology center. According to the final diagnosis, the results were divided into 21 cases of central retinal artery occlusion ( group CRAO, 21 eyes) and central retinal vein occlusion ( group CRVO, 26 eyes ) . All patients received FFA and OCT examination within 2wk of onset, and the image data of the two kinds of examination results were analyzed.
● RESULTS: Group of patients with CRAO average macular foveola thickness, angle measuring, filling time determination results were significantly lower than that of the patients with CRVO group average and the differences were significant (P<0. 05).
●CONCLUSION:FFA and OCT images of central retinal artery and vein occlusion have their own characteristics, and the combination of these two images can be used to identify and diagnose the central retinal artery and vein occlusion.
2.Synthesis and biological activities of beta-chain fragments of hemoglobin.

To investigate the angiotensin I-converting enzyme (ACE) inhibitory activity of beta-chain hemoglobin fragments, 17 fragments were synthesized by microwave-assisted solid-phase synthesis method. Wang resin or Trt(2-Cl) resin, Fmoc and HBTU-HOBt were used as solid carrier, N-terminal amino acid protecting groups and coupling reagents, respectively. The ACE inhibitory, alpha-glucosidase inhibitory, antibacterial and antitumor activities of the synthesized fragments were assayed. In vitro, Val-Val-Tyr-Pro-Trp-Thr showed high ACE inhibitory activity (IC50 = 7.42 micromol x L(-1)). The results indicate that there are two active sites in Val-Val-Tyr-Pro-Trp-Thr-Gln-Arg-Phe, one consists of Val-Val-, and the other -Gln-Arg-Phe. Peptides showed high ACE inhibitory activity when the N-terminal was hydrophobic amino acid such as Val and C-terminal tripeptide contained Phe, Trp or Arg. Some of the fragments showed low a-glucosidase inhibitory activity. No antibacterial activity or antitumor activity was detected in vitro. The results indicate that these peptides have a potential antihypertensive effect and possible application in the treatment of hypertension.

3T3-LI preadipocytes were induced to differentiate and 3T3-L1 adipocyte or preadipocytes were incubated with oleate or palmitate overnight.RT-PCR was used to measure adiponectin receptor(AdipoR)1 and AdipoR2 mRNA levels.The results showed that the AdipoRl and AdipoR2 expressions were differentiation- dependent.Oleate only suppressed AdipoR mRNA expression in preadipocyte but not in adipocyte.However,high concentration of palmitate reduced AdipoR mRNA expression in both 3T3-LI preadipocyte and adipocyte.
6.Dynamic analysis of TCM syndrome in 63 patients of SARS.

OBJECTIVETo observe the dynamic characteristics of symptoms and tongue figure in SARS patients, and the relationship between them and laboratory indexes.
METHODSThe dynamic changes of 63 patients, who were hospitalized in authors' hospital from March 11 to April 30, 2003, were observed.
RESULTSThe symptoms mostly often revealed in patients of early stage were fever in 56 patients, cough in 41 and weakness or heaviness of limbs in 40, which in patients of middle or crisis stage were cough in 53 and weakness or heaviness of limbs in 43. The clinical symptoms in patients of recovery stage were atypical, they occasionally manifested as limb weakness or heaviness in 17, chest stuffiness or pain in 13, low fever in 11, cough in 12 and expectoration in 11. In the early stage, patients' tongue was pink or red in tongue tip, with thin-white, white-greasy or yellow-greasy coating; in middle stage, tongue in most patients were red, with white-greasy or yellow-thick-greasy coating; in recovery stage, tongues of patients were mostly pink or red in tongue tip, few were dark purple, with thin-white or white-greasy, occasionally yellow-greasy coating.
CONCLUSIONTCM syndrome of SARS patients has its characteristics and regularity of changes, which was in accordance with the laboratory findings.

7.Clinical importance of preoperative measurement of plasma amino-terminal pro-B-type natriuretic peptide in infants with congenital heart disease.

OBJECTIVETo define cut-off values of plasma amino-terminal pro-B-type natriuretic peptide (NT-ProBNP) for the diagnosis of congenital heart failure (CHF) and evaluate the importance of plasma NT-ProBNP measurement in the assessment of cardiac function prior to heart surgery in infants with congenital heart disease (CHD).
METHODSPlasma levels of NT-proBNP were measured in 120 infants with CHD before heart surgery and in 100 age-matched healthy infants between June 2010 and June 2013. The data were stratified based on the presence or absence of CHF in the whole group of CHD infants and on age (i.e., <1 year and ≥1 year) and time (i.e., before surgery) within the subgroup of CHF infants.
RESULTSOf the 120 infants with CHD, 41 met the criteria for CHF defined in the Ross Classification for Heart Failure in Infants.The cut-off values of plasma NT-ProBNP were ≥498 ng/L for infants of all ages, 557 ng/L for <1 year age group and 452 ng/L for ≥1 year age group, respectively, in the 41 CHF patients. In CHF infants, plasma NT-proBNP was significantly decreased after protecting of cardiac function (P<0.001).
CONCLUSIONSThe cut-off values of plasma NT-ProBNP for CHF differ between infants <1 year and infants ≥1 year. Moreover, plasma NT-ProBNP can be used as an additional parameter in the preoperative assessment of cardiac function in CHD infants.

8.Uptake of 2-NBDG by human breast cancer cells in vitro.

OBJECTIVEThe purpose of this study was to assess the feasibility of fluorescent 2-deoxyglucose analog, 2-[N-(7-nitrobenz-2-oxa-1, 3-diaxol-4-yl)amino]-2-deoxyglucose (2-NBDG), that could be taken up by breast cancer cells highly expressing glucose transporter 1 (GLUT-1). The purpose of this study was to clarify if a fluorescent 2-deoxyglucose analog, 2-[N-(7-nitrobenz-2-oxa-1, 3-diaxol-4-yl)amino]-2-deoxyglucose (2-NBDG), can be taken up by breast cancer cells highly expressing glucose transporter 1 (GLUT-1), and to assess whether it can be used as a targeting imaging agent.
METHODSThe expressions of GLUT-1 mRNA and protein in breast cancer MDA-MB-231 cells were detected by RT-PCR and immunohistochemistry, respectively. The difference of GLUT-1 protein expression between breast cancer MDA-MB-231 cells and MCF-7 cells was compared by Western blot. Secondly, MDA-MB-231 cells which were grown in 6-well plates were incubated with 2-NBDG, and the result of 2-NBDG uptake was analyzed by fluorescence microscopy and flow cytometry. The difference of 2-NBDG absorption in MDA-MB-231 and MCF-7 cells was compared by flow cytometry.
RESULTSThe results of RT-PCR and immunohistochemistry confirmed that MDA-MB-231 cells highly expressed GLUT-1. Furthermore, Western blot revealed that GLUT-1 expression of MDA-MB-231 cells (0.946 ± 0.007) was higher than that in the MCF-7 cells (0.833 ± 0.010). Fluorescence microscopic and flow cytometric analysis showed that 2-NBDG was uptaken rapidly by MDA-MB-231 cells. Addition of 50 mmol/L D-glucose to the media with 2-NBDG reduced its uptake by 46.0%. Moreover, flow cytometry indicated that the fluorescence intensity of MDA-MB-231 cells (25.10 ± 0.57) was higher than that of MCF-7 cells (10.12 ± 0.62) when incubated with 2-NBDG for 20 minutes.
CONCLUSIONThe preliminary data clearly demonstrate that 2-NBDG is taken up and accumulated in breast cancer cells that highly express GLUT-1, and may be used as an optical probe for glucose uptake in hypermetabolic malignant cells.

10.CT fluoroscopic guidance percutaneous acetic acid injection tumor ablation and TACE for the malignant liver tumor

Objective To evaluate the safety and efficacy of CT fluoroscopic guidance percutaneous acetic acid injection(PAI)tumor ablation and TACE for the malignant liver tumor.Methods PAI had been performed after TACE on 78 patients with malignant tumor(hepatocellular carcinoma 70,metastatic adenocarcinoma 8)since May 2005 to June 2007 in Southwest Hospital.All procedures were performed under CT fluoroscopic guidance.According to the different size of lesions,50%-60% acetic acid 2-10ml was injected into the lesions.PAI was repeated once or twice in some lesions based on the imaging.Results The needle could reach targets under CT real time fluoroscopy in all patients.No complication occurred,but with only moderate pain in 42 cases.The size of tumors shrank in 32 cases,remained unchanged 38 cases,and increased in 8 cases.The efficiency was 90%.All 78 patients were followed up for 3 months with all survival and 48 patients for 1 year with 40 survival.Conclusions CT fluoroscopic guidance PAI tumor ablation combined with TACE in the treatment of malignant liver tumor is safe and effective.(J Intervent Radiol,2007, 16:831-833)
