2.Effect of celecoxib on endometrial carcinoma cell proliferation and expression of COX-2 mRNA

Objective To investigate the effect of celecoxib on endometrial carcinoma cell HEC-1-B and expression of the COX-2 mRNA.Methods To investigate the inhibition of different concentration Cele- coxib on the growth of human endometrial carcinoma cell HEC-1-B by the method of cell culture and MTT experiments.Detect the changes of expression of the COX-2 mRNA by RT-PCR.Results Celecoxib could effectively inhibit the growth of the HEC-1-B cell in dose and time dependent manner.Celecoxib could inhib- it the expression of the COX-2 mRNA.The inhibition ratio of expression in COX-2 mRNA of 20?mol/L and 50?mol/L Celecoxib is 25.92 % and 50.81% respectively.Conclusion Celecoxib may inhibit the growth of endometrial carcinoma cell by degrading the expression of the COX-2 mRNA.
3.Effects of estrogen and androgen on expression of metalloproteinase-2 in lachrymal gland in ovariectomized rats

Background The pathogenesis of dry eye is complicated,hormone level is thought to be one of impact factors in the development of dry eye.The regulation of the synthesis process of metalloproteinases(MMPs) in tissue has been reported.However,the effects of hormone on expression of MMP-2 in lachrymal gland is not clear.Objective This study was to investigate the effects of estrogen and androgen on the expression of MMP-2 in lachrymal gland in ovariectomized rats,and explore the role of MMP-2 in dry eye.Methods Sixty-four 3-monthold clean female Wistar rats were randomized into control group(8 rats),sham operation group(8 rats)and experiment group(48 rats).Ovariectomy(OVX) was performed on the rats of experiment group,and only fat tissue of abdominal cavity was cut off in the rats of the sham operation group.After 5 months of OVX,the experimental rats were subdivided into model control group,vehicle group,estrogen and androgen systemic or topical utilization groups and 8 rats for each group.Six weeks after administration of the drugs,the lachrymal gland was obtained.The expression of MMP-2 mRNA in the lachrymal gland was detected by reverse transcription PCR(RT-PCR),β-actin mRNA was used as an internal control,and the expression of MMP-2 protein was detected by Western blot,GAPDH was used as protein loading control.The use and care of the rats complied with the ARVO Statement.Results The expression of MMP-2 mRNA was strongest in the systemic estrogen group and was weakest in the systemic androgen utilization group.A significant difference in the MMP-2 mRNA expression was found among the 8 groups(F=18.60,P<0.01),and the MMP-2 mRNA was significantly higher in the model group than that of the normal control group(0.66±0.10vs.0.47±0.10)(q=3.01,P<0.05).In addition,the MMP-2 mRNA was significantly higher in the systemic estrogen group compared with the model group (0.83 ±0.10 vs.0.66-0.10) (q =2.79,P<0.05) ; while the expression of MMP-2 mRNA was significantly declined in the systemic androgen group in comparison with the model group(0.12±0.04 vs.0.66±0.10)(q=11.41,P<0.01).The MMP-2 protein presented with a strongest expression in the systemic estrogen utilization group and a weakest expression in the systemic androgen utilization groups.The expression level of the MMP-2 protein in the lachrymal gland was significantly different among the 8 groups(F =7.28,P<0.01).The MMP-2 in the model group was significantly higher than that of the normal group(0.55±0.13 vs.0.38±0.08) (q =2.39,P<0.05),and that in the systemic estrogen group was increased in comparison with the model group(0.69±0.12 vs.0.55±0.13) (q =1.85,P<0.05).However,the MMP-2 in the systemic androgen group was significantly lowed in comparison with the model group(0.27±0.07 vs.0.55±0.13) (q =4.32,P<0.01).Conclusions Estrogen may up-regulate the expression of MMP-2 in lachrymal gland,but the effect of androgen is opposite.Hormone level may play an important role in the regulation of the function of lachrymal gland.
6.Infantile Acute Upper Respiratory Tract Infection With Myocardial Injury

Objective To explore the relation of the infantile acute upper respiratory tract infection(AURI) with myocardial injury.Methods Creatine kinase (CK) and its isoenzyme(CK-MB),lactate dehydrogenase(LDH),asparatic acid transminase (AST) were tested with the fully automatic analyser and electrocardiogram(ECG) were examined with AURI in 86 cases as experimental group and 30 cases of health infants as control group.Results The abnormal rates of CK,CK-MB,LDH,AST and ECG in the AURI group were higher than those of the control group.Conclusions There is different degree of myocardial injury in some children with AURI, the clinical doctor should pay more attention in the AURI cases companied with the myocardial injury and to prevent myocardial injury complicaton in these cases . Objective To investigate the value of 99Tcm-methoxyisobutylisonitrile (MIBI) scintigraphy in assessing the preoperative chemotherapy response and multidrug resistance of osteosarcoma.Methods From January 2007 to October 2008, 12 patients (female:4, male:8; mean age:16.3 years,range:8-27 years) underwent early (10min) and delayed (120 min) 99Tcm-MIBI scintigraphy before and after preoperative chemotherapy.Seven cases had osteosarcoma at the distal femurs, 2 at the proximal tibias, 2 at the upper end of humerus and 1 at the fibula.The tumor-to-background ratio (T/B) and washout rate (WR) were calculated.Tumor necrosis was classified according to Huvos criterion after limb salvage surgery.Immunohistochemical staining for P-glycoprotein(gp) was examined.Spearman correlation analysis and t-test were performed.Results According to Huvos criterion, 7 patients were classified as good responders with more than 90% of tumor cell necrosis and 5 as poor responders with less than 90% of tumor cell necrosis.R value (ratio of early phase T/B after and before chemotherapy) was significantly lower in good responders than that in poor responders (0.473 ± 0.21 vs 0.998 ± 0.06, t= 5.342, P= 0.000 ).R value was significantly correlated with the degree of tumor cell necrosis ( rs=- 0.87, P= 0.000 ).WR was significantly higher in patients with positive P-gp expression than that in patients with negative P-gp expression ((38.36 ±18.64)% vs (6.40±5.87)%, t= -3.278, P=0.008).There was significant correlation between the WR and P-gp expression (rs = 0.91, P= 0.001 ).Conclusion 99Tcm-MIBI scintigraphy is a feasible non-invasive technique to assess the chemotherapy response and to detect P-gp expression of osteosarcoma.
9.Brain mechanisms of hypoxic preconditioning.

A Review: A concept of tissue adaptation to hypoxia (i. e. hypoxic preconditioning) was developed and its corresponding animal models were reproduced in 1966s. The methods of model reproduction in rat, rabbit, and mouse in particular and the main results are briefly introduced in this review. The tolerance to hypoxia of preconditioned animals is significantly increased. Regular changes in animals' behavior, neurophysiology, respiratory and circulatory physiology, neuron morphology in vivo and function of brain and spinal cord in vitro are briefly demonstrated. The protective effects in vivo and in vitro of homogenate extract taken from the brain of preconditioned animals, neurochemicals and molecular neurobiological alterations are briefly presented. The essence and significance of tissue adaptation to hypoxia/hypoxic preconditioning are discussed in the review in terms of evolution and practical implication.
