Primary immunodeficiency diseases (PIDs) represent a typical group of rare diseases with high mortality and disability with regard to long-term outcome.Saving life and improving quality of life of the patients with PID remain to be the responsibility of the stakeholders from all aspects of the society.As the central player in the long run of PID campaign,pediatric immunologists should play a critical role in providing key epidemiological data in favor of political decision-making and social resource distribution.
2.Calcium mechanism of endoplasmic reticulum stress and the change of ultrastructures induced by rotenone in PC12 cells

Objective To investigate the calcium mechanism of endoplasmic reticulum stress and the changes of ultrastructure induced by rotenone. Methods Rotenone induced reactive oxygen species(ROS) production in PC12 cells were measured by using flow cytometry(FCM). Relative changes of intracellular calcium concentration were monitored by use of laser scanning confocal microscope. The ultrastructures were observed under transmitting electronic microscope. Results Treatment with different concentrations of rotenone ( 0.1 ?mol/L,1.0 ?mol/L,2.0 ?mol/L and 3.0 ?mol/L )increased the ROS generation in PC12 cells, and fluorescence intensity (FI) value was shown as 1.55?0.17, 2.16?0.10, 1.77?0.20 and 1.41?0.12, respectively. The same concentration of rotenone resulted in the elevation of intracellular calcium,the relative changes of fluorescence intensity about 0.6029?0.0685,1.0902?0.1127,0.7479?0.0820 and 0.5614?0.0870. 3.Mechanism of mitochondrial dysfunction and ultrastructural changes induced by rotenone in PC12 cells

Objective To observe the effects of different doses of rotenone on reactive oxygen species, mitochondrial transemembrane potential as well as mitochondrial ultrastructure in order to investigate the possible mechanism of mitochondrial dysfunction in cultured pheochromaffinoma cells. 4.Treatment of central retinal artery occlusion by thrombolysis via super-selective ophthalmic artery catheterization

Objective To investigate the therapeutic method and effect of thrombolysis via super-selective ophthalmic artery catheterization treating central retinal artery occlusion (CRAO). Methods 9 patients with CRAO were treated by urokinase infusion via super-selective ophthalmic artery catheterization with Seldinger technique. Results In the 9 patients, the visual acuity was improved to different extent in 8, and remained unchanged in 1. No complications was found during the treatment in any patients. Conclusions Thrombolysis via super-selective ophthalmic artery catheterization for CRAO can improve the visual acuity of most of the patients in different degrees. No positive relation exists in clinical therapeutic effect, time of onset, quantity of urokinase and the visual acuity before the treatment. The method of thrombolysis via super-selective ophthalmic artery catheterization for CRVO is safe and reliable.
5.Effect of temozolomide on the glioma invasiveness and its possible mechanism

Purpose To explore the role of tumor necrosis factor-α( TNF-α) in the process of temozolomide ( TMZ) reduce glioma in-vasiveness and its possible mechanism. Methods C6 glioma cells of logarithmic phase were randomly divided into TMZ treatment (10, 30, 60, 120, 180, 240 min group) (n=15), dynamic monitoring content of TNF-αin the culture medium was measured by ra-dioimmunoassay, expression of p53 protein in C6 cells was detected with Western blotting method, and cell apoptosis was used with AnnexinV-FITC. A glioma invasiveness model was established in vitro and glioma invasiveness was determined by crystal violet stai-ning. Results For C6 cells, contents of TNF-αin the nutrient fluid and expressions of p53 protein in C6 cells obviously increased af-ter TMZ treatment and they achieved the peak at 120 min (P<0. 01), followed by decrease gradually. Glioma invasiveness was re-duced after TMZ acted on glioma in vitro. Conclusion TMZ can reduce glioma invasiveness by TNF-α, which this role may be is TMZ promote C6 cells release of TNF-α and increased TNF-α due to glioma cells apoptosis.
6.Preparation and Quality Control of Thermo-sensitive Paclitaxel Liposomes

Objective To prepare long-circulating temperature-sensitive liposomes with paclitaxel( LTSLP ),develop methods for determination of paclitaxel,related substances and monostearoyl phosphatidylcholine( MSPC ) in LTSLP,and the haemolysis of LTSLP in vitro. Methods HPLC-UV methods for paclitaxel content and related substances and HPLC-CAD method for MSPC in LTSLP were established and validated. Spectrophotometric method was used to determine hemolysis in vitro. Results There was a good linear relationship between peak area and concentration within the range of 60. 39-181. 17μg·mL-1 . Recovery and precision of the method for determination of paclitaxel content met the requirements. Specificity,sensitivity,and system suitability for related substances were consistent with requirements. There was a good linear relationship between peak area and concentration within the range of 1. 5-50. 0μg·mL-1 for the determination of MSPC with good specificity,sensitivity and recovery. Paclitaxel contents in three batches of self-prepared LTSLP were between 90. 0% and 110. 0%,single related substances were below 0. 5% and total impurities were below 2. 0%. There was almost no hemolysis in vitro. Conclusion The methods for determining paclitaxel content,related substances and haemolysis can be used to assess the quality of LTSLP. Self-produced LTSLP consistently meet the quality standards.
7.Ultrastructural changes of the wall of femoral vein after simulated weightlessness in rabbits

BACKGROUND: At present, most researches on simulated weightlessness are confined to its effect on the artery. Therefore, the effect on the vein needs to be further studied.OBJECTIVE: To probe into the effect of simulated weightlessness on the ultrastructure of rabbit femoral vein and the remodeling of femoral vein.DESIGN: A completely randomized and controlled trial based on experimental animals.SETTING: Department of Aerospace and Aviation Medical Science, Fourth Military Medical University of Chinese PLA.MATERIALS: The experiment was completed in the Department of Aerospace and Aviation Medical Science, Fourth Military Medical University of Chinese PLA from December 2000 to December 2001. Altogether 24healthy male New Zealand rabbits, weighing 2.0-2.5 kg were obtained.INTERVENTIONS: The model of Head-Down Tilt( -20°) (HDT) simulated weightlessness was established in rabbits. A total of 24 healthy male New Zealand rabbits were randomly divided into control group, 10-day simulated weightlessness group and 21-day simulated weightlessness group, with 8rabbits in each group. The ultrastructure of the rabbit femoral vein was observed under transmission electron microscope.MAIN OUTCOME MEASURES: Changes and degeneration of endothelial cell, mitochondrion, internal elastic membrane, and smooth muscle.RESULTS: Cell organelles in endothelial cell of the femoral vein of HDT rabbits decreased, mitochondrion dissolved and disappeared, vacuolar degeneration in endothelial cell increased and phagolysosome in endothelial cell could be found. Internal elastic membrane became thinner and was broken. The smooth muscle layer became thinner and some smooth muscle cells became degenerated. Cell interstitial substance increased. There was significant difference in changes between 21-day and 10-day simulated weightlessness groups.CONCLUSION: The vascular remodeling of femoral vein occurs with atrophic changes during simulated weightlessness. The longer duration of simulated weightlessness is, the more obviously the structure of femoral vein changes.

Objective To investigate the effects of Epimedium ,Astragalus ,Radix Puerariae on DMT1 expression in the cere‐bral cortex of APPswe/PS1ΔE9 double transgenic mice model of AD .Methods A total of 60 specific‐pathogen‐free male APPswe/PS1ΔE9 double transgenic mice aged 6 months were equally and randomly assigned to model ,Epimedium ,Astragalus ,Radix puerari‐ae ,compound and DFO groups .An additional 10 6‐month‐old C57BL/6J mice served as negative control group .Using immunohisto‐chemistry and molecular biology methods to investigate the effects of a compound combining the effective components of Epimedi‐um ,Astragalus ,Radix puerariae on DMT1 expression in the cerebral cortex of APPswe/PS1ΔE9 double transgenic mice model of AD . Results Immunohistochemical staining results revealed that DM T 1 positive cell did not show in negative control group .DM T1 ex‐pression was higher in model group compared with the negative control group .DMT1 expression was lower in the compound and deferoxamine groups than in the model group .No significant difference was detected in DM T 1 expression between deferoxamine and compound groups .RT‐PCR ,Western blot and immunohistochemical staining results showed no significant difference .Conclusion These compounds can downregulate DMT1 expression and inhibit iron overload in the cerebral cortex of mice with Alzheimer′s dis‐ease ,reduce iron overload induced impairment of the central nervous system .

Objective:To do factor analysis related to post-stro ke anxiety.Method:206 inpatients(male 126,female 80,mean age 63?16 )with stroke were tested with SAS,DNF,MMSE,LES and ADL.Results:T h e rate of anxiety after 1 month of stroke in our sample was 18.4%(38 cases).Mu lt i-factorial analysis showed,many factors including female,younger at onset,sever ity of stroke,poor general physical condition,less compliance with treatment,and other psycho-social factors(heavier family burden,less income,more life events, less social support)were all associated with anxiety after stroke.Conchus ion:Anxiety is a common complication after stroke,its occurrence were asso ciated with a variety of factors including severity of illness and psychosocial factors.
