A long course of systemic treatment with CsA in therapeutic dose to prolong skin allografts may produce severe toxic effects,therefore it is not acceptable for severe burn patients.Prior studies by one of us have reported that topical treatment with CsA will prolong the life of allografts.During the period of treatment,however,blood CsA levels were detected in high concentrations,similar to systemic use.The question occurs whether the immunosuppressive effect of topically applied CsA was due to local or systemic action.Two experiments were carried out.First,two skin grafts of ACI (RT1) ratswere transplanted on both sides of the back of same LEW (RT1) rat (n=56).The bandage was kept on for the first 5 postoperative days.One graft was treated topically daily with CsA 7.5mg/day in olive oil until rejection.At the same time,the other side was treated with olive oil alone as control.,The mean survival time (MST) was 12.38?0.70 and 9.14?0.33 d.,respectively,p

Objective To explore the application value of multi-ISO center planning method in intensity-modulated radiotherapy for carniospinal irradiation.Methods Ten patients treated with craniospinal irradiation with helical tomotherapy were selected.For these patients, new plans were designed with multi-ISO centers planning method on the treatment planning system (TPS) named Eclipse 11.0.Dose distribution to the tumor, OARs and normal tissue, the treatment time and the monitor units (MUs) of the two plans were compared.Results The plans designed in two groups satisfied all clinical requirements.For the tumor target (PTV) , the difference of D95％ between two groups was not statistical significant, while D9s ％ , D2 ％ and HI in HT group were superior to M-ISO group (t =2.822,2.333,4.743, P ＜0.05) , D50％ , D and CI in M-ISO group were superior to HT group (t =5.259,3.685,8.835 ,P ＜ 0.05).The dose of OARs such as cochlea, parotid, submandibular gland, thyroid gland and kidney in HT group was lower than M-ISO group (t =4.365,5.416,2.674,3.077,2.782,2.607,4.659,P ＜0.05) , and the dose of pancreas and small bowel was higher than M-ISO group (t =5.265,5.935, P ＜ 0.05).Differences were not significant for V5 of normal tissue between two groups;while V10, V20 and V36 of normal tissue in M-ISO group were lower than HT group (t =3.57,3.701,2.602, P ＜ 0.05).M-ISO group reduced 41.0％ of the treatment time by average and reduced 94.1％ MUs by average.Conclusions Intensity-modulated radiotherapy for carniospinal irradiation with multi-ISO centers planning method not only met the requirements of clinical dosimetry, but also shorten the treatment time, reduced the damage to the machine.Multi-ISO centers planning method might be promoted as a new design scheme.

Objective To evaluate the role of equivalent uniform dose (EUD) in planning optimization of intensity-modulated radiotherapy (IMRT) for prostate cancer.Methods Ten patients with prostate cancer were randomly selected who treated with IMRT.For these patients,the treatment plans were designed with dose-volume objectives.Based on these plans,new plans were designed through replacing the dose-volume objectives with maximum EUD for rectum,bladder and small bowel,while keeping the dosevolume objectives unchanged.Comparison was made between the new plans and the former cones by paired t-test.Results The conformity index of planning target volume was better with EUD optimization compared to dose-volume (1.00 ± 0.04 ∶ 0.94 ± 0.04,t =3.80,P =0.04).The D53,D30 and Dicm3 for rectum was better with EUD optimization compared to dose-volume (24.4 ± 2.7 ∶ 25.5 ± 2.6,t =-3.82,P =0.004,34.1 ±4.3∶39.1±2.1,t=-3.80,P=0.004 and51.4±1.0∶51.8±0.9,t=-2.42,P=0.039),with V10,V20 for bladder and V10,V20,V30,V40 for small bowel also better with EUD optimization (92.2 ±6.2∶99.4±1.1,t=-4.28,P=0.002;70.7±5.7∶78.7±6.3,t=-3.10,P=0.013 and 62.2±30.2∶74.7 ±30.0,t =-4.18,P =0.002;34.3 ±26.3∶46.5 ±30.9,t =-5.46,P =0.000;17.1 ±17.0∶25.1 ±22.6,t=-3.52,P=0.007;10.6± 11.5∶ 15.6± 16.1,t=-2.64,P=0.030).Conclusions The conformity index of planning target volume is better with EUD optimization compared to dose-volume.And the dose to rectum,bladder and small bowel can be reduced through optimization with EUD optimization compared to dose-volume.

Objective The extract of spinal cord tissue in morphine spared root rat was isolated and purified to look for some neurotrophic active substances. Methods Neurotrophic active substances were isolated and analysed by Sephacryl S 200 HR gel chromatography、high performance liquid chromatography(HPLC)and tissue culture,ect. Results The extract of spinal cord tissue of spared root rat could enhance the neurite growth of chick embryonal dorsal root ganglion(DRG) in vitro.There was also same effect in the extract of spinal cord tissue of morphine\|treated rat.But there was no significant difference in the extract of spinal cord tissue promoting the neurite growth between spared root rat and morphine treated rat.The extract of spinal cord tissue of morphine spared root rat had evident neurotrophic active role.The peak Ⅱ eluate and peak Ⅳ eluate obtained from the spinal cord tissue extract of morphine spared root rat through Sephacryl S 200 HR gel chromatography could promote the neurite growth of DRG.According to the analysis of SDS PAGE,the peak Ⅱ eluate showed one main protein zone with a molecular weight of 65kD and the protein composition of peak Ⅳ eluate was more complicated.The peak Ⅳ eluate of gel chromatography was then furhter isolated by HPLC.It was observed.That the peak A eluate of HPLC could promote the neurite growth of DRG. It was showed by SDS PAGE that the peak A eluate presented two main protein zones with molecular weight of 30kD and 18 kD.Conclusion\ The molecular weight of neurotrophic active substances,which were isolated from the extract of spinal cord tissue of morphine spared root rat,might be 65kD,30kD and 18kD proteins.\;[

Objective To study the relationships among the expression of inhibitors of DNA binding 1 (ID-1) , Ki-67 and Bcl-2 in esophageal squamous cell carcinoma (ESCC) ,and to investigate the potential role of ID-1 in the carcinogenesis of ESCC. Methods One hundred and eighteen cases of surgical resected ESCC specimens and 20 cases of normal tissues ( sampled far from the tumors, as control) were involved. Immunohistochemical technique was applied to detect the expression of ID-1, Ki-67 and Bcl-2. Results The positivity and staining intensity of ID-1 , Ki-67 and Bcl-2 in ESCC were higher than those in normal tissues. Positive immunological reactions of ID-1, Ki-67 and Bcl-2 were found in 86.44% (102/118) , 81.36% (96/118) and 59. 32% (70/118) cases of examined tumor samples, respectively. The expression of ID-1 and Bcl-2 were positively correlated with the histological grades, while the Ki-67 expression showed negative correlation with differentiation degree. No relationship was found among age, sex, lymph node metastasis and the expression of ID-1, Ki-67 and Bcl-2 in ESCC tissues. Conclusion ID-1 expression may be participated in the regulation of apoptosis in ESCC cells, but may not be considered as a biomarker for evaluation of ESCC metastasis.

Objective To observe the impact of transplant nephrectomy on panel reactive antibodies and a secondary renal transplantation.Methods Panel reactive antibodies in 15 patients with a failed renal transplant admitted in our hospital between 2004 to 2007 were measured before transplantation,before and 1 month,6 months,12 months after transplant nephrectomy,and the pathological changes were observed after transplant nephrectomy.Results Panel reactive antibodies was increasing after renal transplantation,and reached the highest level one month after transplant nephrectomy,then grandually got down.New HLA allosensitization sites were discovered after transplant nephrectomy.Large amount of C4d was stained in failed transplant.Conclusion Serum PRA increased after transplant nephrectomy.New HLA allosensitization sites were found,which may be useful in HLA matching.

Objective To compare the efficacy,toxicity and prognosis on treating advanced non-small-cell lung cancer (NSCLC) between using weekly docetaxel (DOC) and using DOC associated with nedaplatin(NDP).Methods 56 cases of NSCLC patients were retrospectively analyzed.Group A included 34 cases who were treated with weekly DOC (35 mg/m2,dl,8).Group B included 22 cases who were treated with DOC associated with NDP (DOC 35 mg/m2,dl,8,NDP 80 mg/m2,d2).One treatment cycle was 21 days.Efficacy,toxicity and prognosis were evaluated after 2 treatment cycles.Results RR of group A was 8.8 ％ (3/34).RR of group B was 27.3 ％ (6/22).DCR of group A was 50.0 ％ (17/34).DCR of group B was 63.6 ％ (14/22).PFS of group A was 2.3 months.PFS of group B was 5.1 months.OS of group A was 8.7 months.OS of group B was 10.5 months.1-year survival rate of group A was 26.5 ％.1-year survival rate of group B was 31.8 ％.The above comparisons were not statistically different (all P ＞ 0.05).In adverse reactions,hematologic toxicity in group B was greater than that in group A (x2 =4.877,P =0.027).Other adverse reactions such as gastrointestinal reactions,fatigue and so on were grade Ⅰ-Ⅱ.Conclusions The treatment of weekly DOC used in advanced NSCLC is safe,effective an low toxicity.There are no significant difference of RR and PFS between single DOC and DOC associated with NDP.

ObjectiveTo explore the clinical and pathological characteristics in diagnosing the renal diseases with diffuse glomerular nodular changes. MethodsThe clinical and pathological data of 127 patients whose renal pathological feature was mainly diffuse glomerular nodular changes were retrospectively analyzed. ResultsThe most common diagnosis was diabetic nephropathy and renal amyloidosis, and sometimes light chain deposit diseases and Mix cryoglobulinemia. ConclusionDiffuse glomerular nodular changes occurred in many diseases. We should make differential diagnosis based on the clinical symptoms, the results of laboratory examinations and the renal pathological characteristics.

Objective To explore the therapeutic mechanisms of nitric oxide donor (SIN 1) in ischemic acute renal failure (IARF). Methods The effects of SIN 1 on intercellular adhesion molecule 1 (ICAM 1) expression and inflammatory cell infiltration in rat kidney after ischemia/reperfusion (I/R) injury were examined using ICAM 1, ?2 integrin polyclonal antibody, and immunohistochemistry. The renal functions were measured simultaneously. Results Progressive increased expression of ICAM 1 was found in kidneys, especially in vasa recta, after IR injury. ?2 integrin positive cells increased simultaneously in the outer medulla. The two indexes reached the peak values at 24 h after I/R injury. SIN 1 infusion at the beginning of reperfusion could remarkably inhibit the enhanced ICAM 1 expression, reduce the local infiltration of inflammatory cells, and ameliorate the renal functions. Conclusion NO donor can inhibit the renal expression of ICAM 1 and the infiltration of leukocytes after ischemia reperfusion injury, which may play an important role in the treatment of ischemic acute renal failure.

Objective To compare dosimetric parameters between automatic three-dimensional conformal radiotherapy (a3DCRT),inverse three-dimensional conformal radiotherapy (i3DRT),inverse intensity-modulated radiotherapy (iIMRT).Methods Ten lung cancer patients with a single target and 10 intracranial tumor patients also with a single target,who were treated in our radiotherapy center from 2014 to 2015,were included in the study.Their image data were transferred to RayStation 4.5 via network,and then the treatment plans for a3DCRT,i3DCRT,and iIMRT were designed for the 20 patients.The dosimetric parameters for planning target volume (PTV) and organs at risk (OAR) were compared between the three plans using multiple dependent variables and two dependent variables.Results For lung cancer patients,iIMRT achieved better results in D98％,D50％,D2％,conformity index (CI),and homogeneity index (HI) than i3DCRT and a3DCRT (P=O.007,0.001,0.002,0.000,and 0.000),and the CI of a3DCRT was superior to that of i3DCRT (P=O.000);there were no significant differences in heart D33,spinal cord Dmax and D1 cm3,and both lungs between the three plans (P=O.702,0.237,0.163,0.739,0.908,0.832,0.886,0.722,0.429,0.840,and 0.702).For intracranial tumor patients,there were no significant differences in dosimetric parameters between the three plans,except that the CI of iIMRT and a3DCRT was superior to that of i3DCRT (P=O.648,0.783,0.256,0.931,0.002,and 0.034);there were no significant differences in whole brain irradiation dose between the three plans (P=0.446,0.755,0.772,0.0266,0.440,0.290,and 0.939).Conclusions For the single target in patients with lung cancer and intracranial tumors,a3DCRT can improve the CI of PTV compared with i3DCRT,and shows no significant dosimetric disadvantage for OARs compared with iIMRT.Considering the simplicity and low cost of 3DCRT,a3DCRT holds promise as a novel radiotherapy technique.
