1.Determination of the Contents of Neferine and Liensinine in Plumula Nelumbini by TLC Scanning

OBJECTIVE:To establish the method for determining the contents of neferine and liensinine in plumula nelumbini.METHODS:Neferine and liensinine in plumula nelumbini were extracted by ultrasound method and determined by TLCs.RESULTS:The average recovery and relative standard deviation were(97.91?2.49)% and (99.28?3.22)%,respectively.The correlation coefficient were 0.996 and 0.999,respectively.CONCLUSION:The method,operated simply and attained the reliable results with good reproducibility,can be used to determine the contents of bioactive alkaloid in plumula nelumbini or other compound preparations.

OBJECTIVE:To study the effect of HP-?-CD on the stability of potassium dehydroandrographolide succina_ te solution.METHODS:The stability of the testing group(potassium dehydroandrographolide succinate solution+HP-?-CD)was determined by a thermostatic accelerated test with the content variation of potassium dehydroandrographolide succi?nate solution as an index and the potassium dehydroandrographolide succinate solution that was free of HP-?-CD as the control.RESULTS:The content variation of the testing group was less as against the control group,the terms of validity for the2groups were0.657y and3.40y respectively.CONCLUSION:The stability of potassium dehydroandrographolide succi?nate solution can be increased by adding HP-?-CD to which.
3.An experimental study on early active mobilization to promote regeneration of flexor tendon sheath

Objective To study the different effects of early active mobilization on flexor ten don sheath regeneration when the sheath has bee n partially removed and when the inju red sheath has been sutured.Methods In 192Leghorns involved in this study,the zone II flexor digital profoun d tendons were partially ruptured at their bilateral 3rd digits.Their te ndon sheathes were partially removed or sutured.All the Leghorns were di vided into four groups randomly:active mo bilization-sheath removed(A 1 ),active mobilization-sheath sutur ed(A 2 ),immobilization-sheath removed(C 1 ),and immobilization-sheath suture d(C 2 ).Their toes were harvested in the1st,2nd,3rd,4th,8th and 12thweeks postoperatively.Then morphologic al observations and biomechanical tests were performed.All data were s tatistically analyzed.Results In the 1st week postoperatively,the tendon sheath structure disappeared in the injured areas in both groups of A 2 and C 2 ,while in groups of A 1 and A 2 ,neogenetic tendon sheath formed in t he operation areas.In groups of C 1 and C 2 dense scar tissues developed around the tendon without neogenesis of tendon sheath or sheath-like str ucture.There was no significant dif ference either morphologically or biomechanically between A 1 and A 2 as well as C 1 and C 2 while significant differences were found between A 1 and C 1 as well as A 2 and C 2 .Conclusions The pathologic transformation afte r sheath repair during active mobilization c an be divided into three stages:The s heath decomposed and degenerated in the1st week postoperatively.Early neogenetic sheath formed in 2weeks.The neogenetic sheath matured in 4to 8weeks.Active mobilization to promo te neogenesis of sheath is a key facto r to prevent adhesion.Only tendon sheath repair cannot prevent adhesion effectively.
4.Effect of modified rabbit defensin 1 on peripheral nerve regeneration

BACKGROUND:Peripheral nerve injury is a common disease in clinic, which severely affects the patients' quality of life. How to promote peripheral nerve regeneration is an issue of concern.OBJECTIVE:To study the effect of modified rabbit defensin 1 on peripheral nerve regeneration and functional recovery. METHODS: Eighteen Sprague-Dawley rats were randomly divided into three groups. The rat sciatic nerves were transected and bridged by biodegenerated chitin conduits, followed by the injection of neurotrophic factor (group 1), modified rabbit defensin 1 (group 2) and normal saline (control group) into the gluteus, respectively, for consecutive 7 days. RESULTS AND CONCLUSION: The sciatic nerve function index in the groups 1 and 2 was higher than that in the control group at 4 weeks postoperatively. The order of motor nerve conductive velocity was as follows: group 1 > group 2 > control group. The diameter regenerated fibers and axons, and the myelin thickness in the group 2 were less than those in the group 1, but were more than those in the control group. These results indicate that the modified rabbit defensin 1 can promote peripheral nerve regeneration, which may be related with the clearance of residual myelin by macrophages and the improvement in nerve regeneration environment.
5.Determination of Carbazochrome Sodium Sulfonate in Carbazochrome Sodium Sulfonate for Injection by HPLC

OBJECTIVE:To establish a method for determination of carbazochrome sodium sulfonate for injection.METH_ ODS:C 18 was used as chromatographic column;the mobile phase consisted of0.12%ammonium dihydrogen phosphate buffer solution-absolute alcohol(925∶75)with detection wavelength at363nm and flow rate at0.8ml/min.The sample size was10?l and the column temperature was25℃.RESULTS:Linear relation was achieved when the concentration of carbzochrome sodium sulfonate was within the range of0.08865mg/ml~0.4432mg/ml(r=0.9999,n=5).The mean recovery rate was99.89%(RSD=1.53%,n=9).CONCLUSION:This method is simple,rapid,accurate,precise,and which can be used as the quality control of carbzochrome sodium sulfonate for injection.
6.Treatment of osteosarcoma by targeting transportation of pHSP70-shPLK1/DOX compounds with bacterial magnetosomes

BACKGROUND:In an additional changing magnetic field, magnetosomes can invade the tumor cells and inhibit their proliferation.OBJECTIVE: To study the effect of targeted delivery of pHSP70-shPLK1/DOX compounds with magnetosome of magnetotactic bacteria on the treatment of osteosarcoma.METHODS: Human osteosarcoma cell U2OS was divided into four groups: group A: magnetosomes of magnetotactic bacteria+pHSP70-shPLK1/DOX complex; group B: magnetosomes of magnetotactic bacteria; group C: magnetosomes of magnetotactic bacteria+pHSP70-shPLK1/DOX complex, combined with magnetic intervention; group D: magnetosomes of magnetotactic bacteria, combined with magnetic intervention. After 12, 24, 48 and 72 hours of culture, the uptake rate of osteosarcoma cells was observed; cell cycle was evaluated by flow cytometry; PLK1 mRNA and protein expression levels were detected by RT-PCR and western blot, respectively; cell proliferation was explored by MTT assay; cell adhesion and invasion were checked by adhesion assay and Transwel chamber assay, respectively; and the apoptosis rate was measured by apoptosis detection Kit.RESULTS AND CONCLUSION: (1) After culture of 12 and 24 hours, the uptake rate was the highest in group D; meanwhile, the uptake rate was the highest in group B after 48 and 72 hours of culture. (2) The ratio of G2/M phase decreased and the ratio of G0/G1 increased gradualy in group A, group C and group D; the G2/M phase ratio of C group was the lowest at each time point, followed by group D, and that of group A was the highest; and the G2/M phase ratio increased gradualy in group B. (3) Folowing 12 and 24 hours of culture, the expression of PLK1 in group B and group D was higher than that in group A and group C (P < 0.05); in addition, the expression of PLK1 in group B was higher than that in the other three groups after 48 and 72 hours of culture (P < 0.05), and the PLK1 level in group D was higher than that in group A and group C (P < 0.05). (4) The proliferation, adhesion and invasion ability in group B were the highest at different time points, but the apoptosis rate was the lowest. Furthermore, the proliferation, adhesion and invasion abilities in group C were the lowest at different time points, and the apoptosis rate was the highest. To conclude, our experimental results show that the targeted delivery of pHSP70-shPLK1/DOX complex by magnetosomes of magnetotactic bacteria can enhance the apoptosis of osteosarcoma cells and inhibit their proliferation and invasion.
7.Clinical study of laparoscopic pelvic lymphadenectomy assisted radical vaginal hysterectomy (LARVH) in the treatment of cervical cancer

Objective To discuss clinical effect and feasibility of laparoscopic pelvic lymphadendenectomy assisted radical vaginal hysterectomy (LARVH) in the treatment of cervical cancer. Methods Laparoscopic pelvic lymphadendenectomy assisted radical vaginal hysteretomy was performed in 38 patients with cervical cancer. The operative quality and postoperative recovery effects were analyzed. Results All 38 patients were successfully performed with LARVH and no complications. The radical vaginal hysteretomy were performed in all of patients, 21.4±2.2 pelvic lymph nodes were removed. The average duration of surgery was (240±25.6) min and the average hemorrhage was (340±40.2) ml. There were 7 patients who needed blood transfusion during operation. After operation, the mean time for passage of gas by anus was (2±0.26) days, and the mean recovery time of bladder function was (10.5±1.3) days. Patients were discharged after (9.2±1.1) days on average. 38 patients had no complication. Conclusions For the treatment of uterine neoplasm, LARVH decreased operative damage with more short recovery time, same operative extent as laparotomy. It is an ideal way to treat uterine neoplasm currently.

AIM: To investigate the effects of lidocain and gentamycin on inflammation reaction and wound infection of explosive wound in limb arthrosis after seawater immersion. METHODS: The explosive wound were made in 48 New Zealand white rabbits. All rabbits were randomly divided into 4 groups: lidocain, gentamycin and glutin sponge bound up and immersed in seawater 1 h group (LGGS group, n=12); bactroban and glutin sponge bound up and immersed in seawater 1 h group (BGS group, n=12); 12 layers common gauze bound up and immersed in seawater 1 h group (GS group, n=12) and 12 layers common gauze bound up 1 h group (G group, n=12). The changes of blood WBC, CRP, IL-1, IL-6, TNF-?, MDA and SOD in each group were tested. The bacterial culture was performed in each group after explosion 1 h, 3 d, 7 d and 1 month. Objective To investigate the design and clinical result of the modified medial fasciocutaneous flap of lower leg in the different zonation. Methods The length of lower leg was divided into three zonations equally.According to the different zonation of the fasciocutaneous flap,the 45 patients were divided into three groups from Jan.2005 to Feb.2008.The defects of the patients were repaired with the flaps.The sizes of the flaps ranged from 5 cm×3 cm to 25 cm×10 cm.Results The flaps survived completely in 43 cases.The distal sides of flaps necrosed partially in 2 cases in the upper third of the lower leg.The necrosed part of the flap was repaired after the change dressings.The colour and texture of flaps were excellent,the appearance and function were satisfactory after a follow up of 4-20 months.Conclusion The modified medial fasciocutaneous flap of the lower leg is easy to design and dissect,blood supply is reliable without sacrifice of the major arteries.The flap can be used according to the different location of the fascia pedicel.It is the idea flap to repair the soft tissue defects of the front of knee joint,the leg,the ankle and the foot.
10.Detection of TORCH infection in pregnant women by using reverse phase protein array

Objective To evaluate whether the reverse phase protein array (RPPA) method can be used for detecting TORCH infection in pregnant women .Methods The RPPA method was established for detecting TORCH infection .The positive coinci‐dence rates of TORCH infection detected by the RPPA method and ELISA method in 2000 fresh serum samples from pregnant women were compared for evaluating the feasibility of RPPA in TORCH detection .Results The positive coincidence rates of estab‐lished RPPA and ELISA for detecting TORCH infection was 100 .0% ,91 .1% ,97 .2% ,91 .3% and 93 .0% respectively ,indicating that the detection results of various indexes by RPPA and ELISA had better consistency (P>0 .05) ,but the positive detection rates of RPPA for Rubellavirus ,CMV and HSV‐1 ,2 were higher than those of correspondent ELISA method .Conclusion RPPA method for detecting TORCH infection has the advantages of simpleness ,rapidness ,high sensitivity and strong specificity ,is an effective method of auxiliary diagnosis for bearing and rearing better children in clinical ,and is worthy of being promoted and used in the fu‐ture .
