1.Relationship between vascular endothelial growth factor and microvessel density in placenta and placenta increta

Objective To investigate the relationship between the expression level of vascular endothelial growth factor (VEGF) and microvessel density (MVD) in placenta tissue and placenta increta.Methods Thirty singleton pregnant women who received antenatal care and underwent cesarean section in Fujian Maternity and Child Health Hospital between November 2013 and August 2014,were enrolled in this study.They were divided into placenta previa group,placenta increta group and control group,with ten patients in each group.Placenta tissue was collected from each patient.Expressions of VEGF and its mRNA were detected by immunohistochemistry,Western blot,and real-time polymerase chain reaction.MVD in placenta tissue was measured by immunohistochemistry.Rank sum test,t test,Kruskal Wallis test,one-way ANOVA and Spearman correlation test were used for statistical analysis.Results (1) Gestational age at admission and delivery in placenta previa and placenta increta groups was lower than in control group (all P<0.05).Compared with control group,the placenta previa and placenta increta groups had more blood loss,and longer operating duration and hospital stay (all P<0.05).(2) The expression levels of VEGF and its mRNA in placenta increta and placenta previa groups were higher than in control group (VEGF:0.691±0.032,0.695 ± 0.027 and 0.518±0.025,respectively,F=373.401;VEGF mRNA:1.667±0.661,1.832±0.678 and 0.767±0.269,respectively,F=27.399;both P<0.05).But there was no significant difference between placenta previa and increta groups.(3) There was no significant difference of VEGF expression in increta location,border site and normal site in placenta increta group,but its mRNA was decreasing (2.519± 0.116,1.482 ± 0.232 and 1.000± 0.000,respectively,F=240.827,P<0.05).(4) Expression level of VEGF at the attachment of umbilical cord,upper and lower margin of placenta in placenta previa group was higher than in control group (0.702 ± 0.026 vs 0.528± 0.020,t=12.302;0.698 ± 0.026 vs 0.519±0.035,t=12.715;and 0.685±0.029 vs 0.509±0.010,respectively,t=17.891;all P<0.05).Expression of VEGF mRNA in placenta previa group was higher than in control group (2.080± 0.539 vs 1.024±0.272,t=8.093;1.587±0.757 vs 0.546±0.083,t=2.401;1.828±0.704 vs 0.731 ±0.157,t=4.259;all P<0.05).(5) MVD in placenta increta group and placenta previa group was higher than in control group (171.2± 14.7,155.7± 14.6 vs 147.8±12.3,respectively,F=7.277,P<0.05).(6) Expression level of VEGF in placenta increta group and control group was positively associated with MVD (r=0.825,P<0.05).Conclusions There may be some common mechanisms in the occurrence of placenta previa and placenta increta.Overexpression of VEGF in placenta and abnormal formation of villous vessels may be important factors in the pathogenesis of placenta increta.
2.Effects of HDAC5 on the proliferation and apoptosis of the gastric cancer cell line SGC-7901

Objective: To investigate HDAC5 expression in gastric cancer cell lines and its effects on the proliferation and apoptosis of the gastric cancer line SGC-7901. Methods: The expression patterns of HDAC5 and Twist1 in gastric cancer cell lines and normal gastric mucosal cells were detected by Western blot. The effects of HDAC5 and Twist1 on the proliferation and apoptosis of SGC-7901 cells were analyzed by MTT and flow cytometry, respectively. Results: The expression of HDAC5 and Twist1 in gastric cancer cell lines were significantly higher than that in normal gastric mucosal cells (P<0.05). HDAC5 knockdown significantly down-regulated Twist1 expression,inhibited cell proliferation, and induced apoptosis in SGC-7901 cells, whereas HDAC5 overexpression exhibited an opposite effect (P<0.05). Moreover, Twist1 knockdown significantly inhibited cell proliferation and induced apoptosis in SGC-7901 cells (P<0.05). Conclusion:HDAC5 may promote cell proliferation and inhibit apoptosis in gastric cancer cells by upregulating Twist1 expression, thus promoting the initiation and development of gastric cancer.
3.The value of differential diagnosis of the configuration of QRS complex in lead aVR in patients with inferior wall myocardial infarction

Objective To investigate the value of differential diagnosis of the configuration of QRS complex in lead aVR in patients with inferior wall myocardial infarction. Methods The configuration of QRS in 52 patients with pathological Q-wave both in lead Ⅲ and aVF were analyzed and the result of selective coronary arteriography was compared. Results 13 patients with the configuration of QRS in lead aVR appeared rS ( s), while 10 patients appeared QS(qs) and 29 Q(q)r,correlated with 12,4 and 0 patients with coronary arteriography showed stenosis or occlusion lesion in fight coronary artery or left circumflex artery (χ2 = 35.56, P = 0.000). Conclusions The con-figuration of QRS in lead aVR is helpful to differential diagnosis of the patients with pathological Q-wave both in lead Ⅲ and aVF. Patients with the configuration of QRS in lead aVR appear rS(s) could be diagnosed as old myocardial infarction,but excluded from old myocardial infarction while appearing Q(q)r.
4.Characteristics in the secretion of liver TNF-? induced by infusion of LPS into veins in goats

AIM: To study the role of liver in immune regulation in experimental endotoxemia. METHODS: 17 castrated male goats were subjected to simultaneously installing catheters in jugular, hepatic and portal veins by surgery. Four days later, lipopolysaccharide (LPS) was infused in term of three groups as followings: In group ①, LPS of 20 EU (endotoxin unit, EU)?kg -1 was infused into portal vein; In group ②, LPS of 20 EU?kg -1 was infused into jugular vein and LPS of 1 500 EU?kg -1 infused into jugular vein in group ③. Before and after infusion, blood samples were collected from the three veins through the catheters for 8 h. The plasma levels of TNF-? were measured by RIA. RESULTS: In group ①, the plasma TNF-? levels of hepatic and portal vein rose to peak value at 5 h, but that of the jugular vein did not changed. In group ②, the plasma TNF-? levels in hepatic vein rose to peak value at 3 h. The TNF-? levels of jugular vein rose to peak value at 1 h and the one in portal vein enhanced continuously between 0-8 h. In group ③, the plasma TNF-? levels in jugular, hepatic and portal vein rose to significant peaks at 1 h simultaneously. CONCLUSION: During experimental endotoxemia,liver showed different dynamic characteristics in TNF-? secretion according to the pathway and doses of LPS delivery. [
5.Study on the Correlation Between theⅣPlase of Diabetic Nephropathy and Bailing Capsules Combied with Salvianolate

Objective:To explore the effects of Bailing capsules combined with salvianolate on inflammatory cytokines, renal vas-cular endothelia and renal function in diabetic nephropathy inⅣphase. Methods:Totally 60 patients with diabetic nephropathy in IV phase were randomly divided into the control group (30 cases) and the treatment group (30 cases). The control group was given the conventional and salvianolate therapy, and the treatment group was treated with Bailing capsules additionally. The treatment course was 3 months. The changes of serum creatine(SCr), urean itrogen (BUN), creatinine clearance rate (Ccr), ethelin (ET-1), interleukin-6 (IL-6), tumor necrosis factor (TNF-a) and C-reactive protein (CRP) were detected before and after the treatment. Results:After the 3-month treatment, SCr, BUN, IL-6, TNF-a, CRP and ET-1 were significantly decreased in the two groups with statistical signifi-cance, and Ccr was significantly increased in the treatment group (P<0. 05), and the changes in the treatment group were significant-ly better than those in the control group (P<0. 05). After the treatment, the blood glucose, blood lipids, glycosylated hemoglobin and the other indicators had no significant changes in the two groups (P>0. 05). There was no significant difference in the incidence of adverse drug reactions between the two groups (P>0. 05). Conclusion:The combination of Bailing capsules and salvianolate can re-duce the inflammatory reaction and improve renal vascular endothelial function and renal function in the patients with diabetic nephrop-athy in Ⅳ phase with promising safety and few adverse reactions.
6.Multiple factor analysis of lumbar spine bone mass and densities in adolescents with early ankylosing spondylitis

Aim To investigate the possible roles of age, bodyheight, mass and body mass index(BMI) in Lumbar1-4 bone mineralcontent (L1-4BMC), L1-4 areal bone mineral density(L1-44BMD) and L1-4volumetric bone mineral apparent density(L1-4BMAD) of Chinese adoles-cents with early ankylosing spondylitis (AS). Methods Thirty-one maleChinese adolescent outpatients with early AS were included. Age (y),total body mass (kg), height(cn) and body mass index (BMI, kg/m2)of subjects were obtained. L1-44BMC(g) and L1-4 BMD(g/cm2) were e-valuated by using DEXA, and L1-4 BMAD(L1-44BMC/Area3/2, g/cm3)was subsequently calculated. Correlation and multiple regression analyseswere performed. Results Multiple regression revealed that height ( P =0.000) and BMI(P=0.009) were significantly positively related toL1-44BMC (R=0.759, Radj2=0.545, P=0.000 <0.01), and heightplayed the pivotal roles in significant correlation with L144BMC(R =0. 676 Radj2 = 0. 439, P = 0. 000) . Body mass significantly positivelycorrelated with both L-44BMD ( R = 0. 657, Radj2 = 0. 412, P = 0. 000)and L1-4BMAD (R=0.551, Radj2=0.280, P=0.001) .Therefore,height as well as BMI significantly positively correlated with L144BMC andmass was definitely associated with both L1-4BMD and L1-4BMAD in ado-lescents with early AS in this study. Conclusion Height and mass couldhave significantly positive effects on axial bone mass and densities ofadolescents with early AS.
7.Study on subtype of human papillomavirus infection among aged 25 - 54 reproductive women in Beijing from 2006 to 2008

Objective To investigate subtype of HPV infection among women at age of 25 to 54 years in Beijing. Methods From Sept. 2006 to Dec. 2008, 5552 reproductive women at age of 25 to 54 years classified into each 5-year as group were screened. Each participant completed a questionnaire and a sample of exfoliated cervical cells for liquid-based cytology and HPV subtype gene testing was performed.Results The overall prevalence of HPV was 6.68% (371/5552). High-risk HPV and Low-risk HPV prevalence were 5.76% (320/5552) and 2.00% (111/5552), respectively. The most common HPV types were HPV16 (2.61%, 145/5552), HPV58 (0. 97%, 54/5552), HPV33 (0.85%, 47/5552), HPV43 (0.74% ,41/5552) and HPV56(0.70% ,39/5552). There were two peaks of HPV prevalence: groups of 30 -34 years and 40 -44 years. Conclusion The most common HPV subtypes in Beijing were HPV 16,58, 33 and 43 and HPV prevalence showed a bi-medal age-specific curve.
8.Changes of lung, spleen and kidney aquaporin-1 in rats with Kidney Yang Deficiency: the "water metabolism theory" in traditional Chinese medicine.

OBJECTIVETo explore the experimental basis of the "water metabolism theory" in traditional Chinese medicine by observing the changes of aquaporin-1 in the lung, spleen and kidney.
METHODSRat models of Kidney Yang Deficiency induced by gavage with 2% adenine suspension for 4 weeks were treated with cistanches decoction for 6 weeks. Urinary 17-hydroxy cortisol, urine creatinine, urine osmolality value content, and aquaporin-1 mRNA and protein expressions in the lung, spleen and kidney tissues were detected after the treatment.
RESULTSTreatment with adenine induced Kidney Yang Deficiency in rats by causing a reduction in urinary 17-hydroxy cortisol, urine creatinine and urine osmolality. Aquaporin-1 mRNA expression in the spleen and kidney were down-regulated after adenine treatment. Compared with the rat models, intervention with cistanche significantly increased aquaporin-1 mRNA expression in the lung and kidney tissues. Adenine resulted in increased aquaporin-1 protein expression in the lung, spleen and kidney of the rats, while cistanche intervention lowered its expression in lung and kidney tissues.
CONCLUSIONThe lung, spleen, kidneys are involved in water metabolism, and aquaporin-1 is one of its molecular basis. Cistanche can increase aquaporin-1 expressions, which is also regulated by other factors.

9.Construction and application of medical morphology online picture examination system

At present, the experimental examination of medical morphology courses in most medical colleges and universities still adopt traditional specimen examination or PPT examination, and there are many disadvantages. This paper introduces how to build a system with B/S architecture based on the local area network, and how to set up an examination system to realize the functions of user management, online examination database updating, free examination paper formation, examination running and automatic marking. Through its application in our school in recent years, the medical morphology online picture exam system has effectively promoted the reform of the examination methods, the style of study and the effect of experimental teaching. However, there are still some problems, such as high investment cost and limited examination room.
10.Surface modification of biodegradable polymer/TCP scaffolds and related research.

Under laboratory condition, the compound materials of Poly (DL-lactic-co-glycolic acid)/Tricalcium phosphate [PLGA/TCP(L), with component ratio of 7:3] were fabricated by combining the thermally induced phase separation (TIPS) with solvent-casting particulate-leaching (SCPL) approach. On the other hand, rapid prototyping (RP) technique manufactured PLGA/TCP scaffolds [PLGA/TCP(RP)] were obtained. These two kinds of carriers were coated with collagen type I (Col I). The extracted bovine bone morphogenetic protein (bBMP) was loaded into carriers to establish biomimetic synthetic bones. PLGA/TCP(L) scaffolds, demineralized bone matrices (DBM) of bovine cancellous bone, PLGA/TCP(L) scaffolds, biomimetic synthetic bones and OsteoSet bone graft substitutes were investigated. Scanning electron microscopy revealed that the microarchitecture of PLGA/TCP(RP) scaffolds was much better than that of PLGA/TCP(L) scaffolds. The diameter of macropore of PLGA/TCP(RP) scaffold was 350 microm. The porosities of PLGA/ TCP(L) scaffolds, DBM, PLGA/TCP(RP) scaffolds and OsteoSet bone graft substitutes were 21.5%, 70.4%, 58.6% and 0%, respectively (P<0.01). Modification of PLGA/TCP scaffolds with collagen type I [PLGA/TCP(L)-Col I and PLGA/TCP(RP)-Col I] essentially increased the affinity of the carriers to bBMP. Among these synthetic materials, PLGA/TCP(RP)-Col I-bBMP composite is promising as a novel bone graft substitute due to its advanced fabrication technique, good tri-dimensional microarchitecture and ideal components.
