3.Study of loss of imprinting of insulin-like growth factor Ⅱ and H19 genes in cervical carcinoma

Objective To study the loss of imprinting(LOI) of insulin-like growth factor Ⅱ(IGF2) and H19 genes in cervical carcinomas.Methods Polymerase chain reaction(PCR) method was used on DNA samples extracted from 40 cases of cervical carcinoma and 20 cases of normal control tissues to detect the heterozygosity of IGF2 and H19 genes.The LOI of IGF2 and H19 genes was detected by RT-PCR.Results The heterozygous frequency of IGF2 was 52% in cervical cancer samples.Comparatively,in 20 matched normal samples,13 cases showed the heterozygosity(65%).The biallelic expression of IGF2 was detected in 10 cases among the 21 informative cervical cancer samples(47.6%),however,1 case in 13 heterozygous samples of control group(7.7%)((P0.05).Conclusion LOI and LOH of IGF-2 and H19 genes are involved in cervical carcinomas.LOI of IGF2 and H19 may be involved in the initiation stage of carcinogenesis of cervical carcinoma.

MicroRNA (miRNA) is a kind of naturally occurring short non-coding small molecular RNA,which regulates gene expression at the posttranscriptional level through its sequence complementation with target mRNAs.Researches have shown that different malignant tumors,including diffuse large B cell lymphoma,have specific miRNA expression profiles.Therefore,miRNAs expression level of specimens of body fluids and tissues is expected to be the new indicator for assessing diffuse large B cell lymphoma.
5.Effects of remifentanil on NF-Bκ activity in a rabbit model of myocardial ischemia/reperfusion injury

Objective To investigate the effects of remifentanil on NF-κB activity in a rabbit model of myocardial ischemia/reperfusion (I/R) injury. Methods Fifty New Zealand white rabbits were randomly divided into 5 groups (n = 10 each): sham operation group (group S); group I/R; low dose of remifentanil group (group L); median dose of remifentanil group (group M); high dose of remifentanil group (group H). In group I/R, L,M and H, myocardial I/R was produced by occlusion of left anterior descending artery for 30 min followed by 120 min reperfusion.In group S and I/R,10 min before ischemia,normal saline was infused at 5 ml·l·h-1 via the external jugular vein until the end of 120 min reperfusion. In group L, M and H, remifentanil was infused at 1, 5 and 10ug·kg·min-1 respectively 10 min before ischemia until the end of 120 min reperfusion,and the other procedures were the same as those in group I/R. The myocardial tissues were taken at the end of 120 min reperfusion for determination of NF-κB expression which was used to reflect the activity of NF-κB and microscopic examination. Results The activity of NF-κB was significantly higher in group I/R, L, M and H than in group S. The activity of NF-κB was gradually decreased with the increase in the dose of remifentanil in group L, M and H compared with group I/R. The microscopic examination showed that remifentanil significantly attenuated I/R-induced injury in a dose-dependent manner. Conclusion Infusion of remifentanil reduces myocardial I/R injury through decreasing the activity of NF-κB in a doee-dependent manner.

8.Analysis of clinicopathologic characteristics of 520 patients with lymphoma in Shandong

Objective:To analyze the pathologic types and clinical characteristics of lymphoma in Shandong, China. Methods:The clinical data of lymphoma patients admitted from September 2011 to September 2014 in Shandong Cancer Hospital and the Affiliat-ed Hospital of Shandong Academy of Medical Sciences were retrospectively analyzed. Data included age, sex, pathologic classification, and onset locus of lymphoma. Results:The clinical data of 520 lymphoma cases were collected. The ratio of male to female was 1.2:1, and the median age was 51 years old. Among these patients, 67 cases (12.9%) were Hodgkin's lymphoma (HL), whereas 453 cases (87.1%) were non-Hodgkin's lymphoma (NHL). Among NHL cases, 75.9% were B-cell NHL and 24.1% were T/ natural killer-cell NHL. Nodular sclerosis classical Hodgkin's lymphoma was the most common pathologic type of HL, comprising 52.2%. The age of HL onset did not present twin peaks. In NHL, 245 cases (54.1%of NHL) were diffuse large B-cell lymphoma (DLBCL). The constituent ra-tio of DLBCL was higher than in other countries and regions (18%to 42.7%), in which the ratio of male to female was 0.9:1, and the age of onset was slightly earlier (median age was 54 years old). Lymph nodes in the cervical region were the most commonly involved in the disease, and NHL extranodal onset was frequently observed. Conclusion:Among the 520 cases of lymphoma, DLBCL is the most common subtype. Most subtypes show male predominance, but female predominance was observed for DLBCL. The commonly involved sites of extranodal lymphoma are the nasal cavity and gastrointestinal tract.
9.Effects of Solanine on the human colon cancer in chicken embryo transplantation model angiogenesis

Objective To establish chicken embryo transplantation model of human colon cancer and to research the effect of so‐lanine on angiogenesis .Methods Cases with chicken embryos were divided into the low‐,mid‐and high dose solanine group and con‐trol group ,with 10 cases in each groups ,and then the cultured human colon cancer cell line HT‐29 cell lines were inoculated to the chicken embryo villus allantois membrane (CAM ) .We observed the characteristics of the transplanted tumor in CAM angiogenesis by the stereo microscope .Image analysis software of Image‐pro plus 6 .0 and immunohistochemical method were used to observe the effect of different dose of solanine on angiogenesis .Results HT‐29 cell lines were inoculated to CAM 3-5 days ,a large number of blood vessels concentrated in tumors ,growing into or acrossing the surface of tumors .While tumors also rapidly growed .We took photo on the 5th day after receiving medicine and did imaging analysis .Then we calculated the area of angiogenesis in experimental group ,which was significantly lower than that of the control group ,quantitatively in a dose‐dependent manner .There were signifi‐cant differences among the groups(P<0 .01) .Microvascular density of 3 different dose of solanine was significantly lower than that of the control group by immunohistochemical method ;the expression of Ki‐67 antigen index decreased gradually ,which was highest in the control group ,and there were significant differences among the groups (P<0 .01) .Conclusion Solanine could inhibit angio‐genesis induced by human colon cancer HT‐29 cell lines obviously ,thus inhibiting the growth of tumor and providing an important basis for the treatment of anti‐tumor angiogenesis .
10.Simultaneous Determination of Four Constituents in Roots and Knotty Rhizome of Glycyrrhiza uralensis by HPLC

This study was aimed to establish an HPLC method for the determination of liquiritin, isoliquiritin, liquiritigenin and glycyrrhizic acid in roots and knotty rhizome of Glycyrrhiza uralensis. The analysis was performed on a Diamonsil C18 column (250 mm í 4.6 mm, 5 μm) by using a gradient elution with mobile phase of water, phosphoric acid, acetonitrile at the flow rate of 1.0 mL·min-1. The detection wavelength was 276 nm (0~18 min), 360 nm (18~24 min), 276 nm(24~30 min), and 250 nm (30~65 min). The column temperature was set at 30℃. The results showed that the linear range of iquiritin, isoliquiritin, liquiritigenin, glycyrrhizic acid was 0 . 108 5~1 . 085、0 . 016 8~0 . 168、0 . 0049 4~0 . 049 4、0 . 407~4 . 07μg , respectively . The average recoveries of four constituents were 96.61%~100.89%, with RSD ≤ 0.81%. The contents of four constituents in roots of five batches were 0.513%, 0.072 9%, 0.048 4%, and 1.945%, respectively. Contents of four constituents in knotty rhizome from two batches were 0.456%, 0.063 6%, 0.036 2%, and 1.630%, respectively. It was concluded that there was good linear relationship between the response and concentration. Contents of four constituents in knotty rhizome were basically the same as those in the roots. The knotty rhizome can be used as raw material for the extraction of active components.
