Acute pancreatitis is one of the common clinical diseases in emergency.The severe acute pancreatitis-associated ascetic fluid is always found in clinic.Its roles related multiple organ dysfunction(MOD)in acute pancreatitis is being understood.In this article,the possible compositions of the ascites,the mechanism of its formation,and its effect on body are recited,in order to better understand the mechanism,the prevention and the treatment of the acute pancreatitis and its compications related MOD.
2.Study of loss of imprinting of insulin-like growth factor Ⅱ and H19 genes in cervical carcinoma

Objective To study the loss of imprinting(LOI) of insulin-like growth factor Ⅱ(IGF2) and H19 genes in cervical carcinomas.Methods Polymerase chain reaction(PCR) method was used on DNA samples extracted from 40 cases of cervical carcinoma and 20 cases of normal control tissues to detect the heterozygosity of IGF2 and H19 genes.The LOI of IGF2 and H19 genes was detected by RT-PCR.Results The heterozygous frequency of IGF2 was 52% in cervical cancer samples.Comparatively,in 20 matched normal samples,13 cases showed the heterozygosity(65%).The biallelic expression of IGF2 was detected in 10 cases among the 21 informative cervical cancer samples(47.6%),however,1 case in 13 heterozygous samples of control group(7.7%)((P0.05).Conclusion LOI and LOH of IGF-2 and H19 genes are involved in cervical carcinomas.LOI of IGF2 and H19 may be involved in the initiation stage of carcinogenesis of cervical carcinoma.
3.Regulatory effect of protein tyrosine phosphatase non-receptor type on the cardiac Kv11.1 (HERG) potas-sium channel

Objective Cardiac HERG potassium channel currents can cause long QT syndrome .The function of the cardiac HERG potassium channel is regulated by tyrosine kinase and phosphatase , and protein tyrosine phosphatase non-receptor type 11 ( PTPN11) negatively regulates the HERG potassium channel .This study aimed to investigate the effect of PTPN 11 on the electrophysio-logical characteristics of the HERG channel . Methods The plasmids of pcDNA3.1-PTPN11-EGFP were constructed by PCR technique and transfected or cotransfected with the pcDNA 3.1-PTPN11-EGFP plasmid into HEK293 cells using Lipofectamine 2000.The patch clamp technique was employed to record the HERG channel currents in the control group ( HEK293 cells transfected with pcDNA3.0-HERG-EGFP), PTPN11 overexpression group (pcDNA3.0-HERG and pcDNA3.1-PTPN6-EGFP cotransfected HEK293 cells), and PTPN11 overexpression with PAO group . Results The pcDNA3.1-PTPN11-EGFP plasmid was successfully constructed .Green fluorescence was observed in the HEK293 cells transfected with pcDNA3.0-HERG-EG-FP or cotransfected with pcDNA3.0-HERG and pcDNA3.1-PTPN11.The maximum densities of pulse and tail currents were significantly decreased in the PTPN11 overexpression group as compared with the control ([31.85 ±5.54] vs [45.92 ±3.18] pApF, P<0.05;[73.82 ±11.31] vs [108.43 ±7.98] pApF, P<0.05) but markedly in-creased in the PTPN11 overexpression with PAO group ([48.08 ±4.32] pApF;[120.06 ±8.02] pApF) (P<0.05).The time con-stant of deactivation was significantly higher in the PTPN 11 overexpression group than in the control ([622.16 ±46.49] vs [440.70 ± 49.49] ms, P<0.05). Conclusion The overexpression of PTPN11 decreases HERG potassium channel currents , which can be re-versed by PAO.This finding provides a theoretical basis for the application of certain regulatory enzymes in the HERG channel as a treat -ment of long QT syndrome .
4. Preparation of adeno-associated virus serotype 8 using baculovirus system

Objective: To set up a baculovirus system for preparing recombinant adeno-associated virus serotype 8 (rAAV8) and to preliminarily examine the infection viability of the prepared virus in vitro. Methods: The rAAV8-EGFP was prepared using baculovirus system and was subsequently extracted and purified by high performance liquid chromatography. The viral titer was determined by real-time quantitative PCR and its infection viability for HEK-293 cells was examined by observing the EGFP reporter. Results: Real-time quantitative PCR showed that high titer of rAAV8-EGFP was prepared (1.5×10 13vg/bottle[100 ml medium]) and strong expression of EGFP was observed in HEK-293 cells infected with rAAV8-EGFP. Conclusion: The baculovirus system is capable of producing rAAV8 with high titer and infection viability, paving a way for future research.
5. Construction of adenovirus vector harboring RANTES regulated by oxygen-dependent degradation domain and its chemoattractant activity in vitro

Objective: To construct an adenovirus vector harboring the human RANTES gene regulated by oxygen-dependent degradation domain (ODD) and to observe its chemoattractant activity in vitro. Methods: The human RANTES gene was fused with ODD by PCR and the recombinant adenovirus was used to construct SG511-CCL5-ODD with the Gateway System. Viral replication experiments were performed to evaluate the selective replication ability of SG511-CCL5-ODD. The expression of RANTES protein was determined by ELISA under normal and hypoxia condition. Chemotactic test was used to analyze the chemoattractant ability of the expressed RANTES in liver cancer cells. Results: A recombinant adenovirus SG511-CCL5-ODD was constructed successfully. Cells infected with the recombinant virus expressed RANTES selectively. The expression of RANTES protein in the transfected liver cancer cells was higher under hypoxia condition than under normal condition (P<0.05), indicating ODD can effectively regulate RANTES protein expression. Chemotactic test showed that liver cancer cell infected with SG511-CCL5-ODD had the ability to recruit NK92 cells. Conclusion: Recombinant vector SG511-CCL5-ODD can effectively infect liver cancer cell line HepG2 and Hep3B, and can express RANTES protein under the regulation of ODD, demonstrating a chemoattractant activity in vitro.

AIM:To investigate the effect of fluvastatin on the expression of serum and glucocorticoid inducible kinase 1 ( SGK1) and connective tissue growth factor ( CTGF) induced by aldosterone ( Ald) in rat mesangial cells (GMCs). METHODS:GMCs were divided into (1) control group; (2) aldosterone group with different concentrations and times; (3) Ald (10 -7 mol/L) + spironolactone (10 -9 mol/L) group; (4) Ald (10 -7 mol/L) + LY294002 (20 ?mol/L) group; (5) Ald (10-7mol/L) +SB203580 (20 mmol/L) group; (6) the group of Ald (10-7mol/L) + fluvas-tatin at different concentrations (10-7,10-6,10-5 mol/L); (7) Ald (10 -7mol/L) + fluvastatin (10 -5mol/L) + mevalonate (10 -4 mol/L) group; (8) Ald (10 -7 mol/L) + fluvastatin (10 -5 mol/L) + FPP (farnesyl pyrophosphate,10-4 mol/L) group; (9) Ald (10 -7mol/L) + fluvastatin (10 -5 mol/L) + GGPP (geranylgerany pyrophosphate,10 -4 mol/L) group. The protein levels of SGK1 and CTGF were determined by Western blotting. The levels of fibronection (FN),monocyte chemoattractant protein-1 (MCP-1) and intercellular adhesion molecule-1 (ICAM-1) in the supernatants were determined by enzymelinked immunosorbant assay (ELISA). 7.Clinical research of chronic pelvic cavity pain syndrome treated with acupoint catgut embedding therapy.

OBJECTIVETo explore the clinical efficacy of acupoint catgut embedding therapy on chronic pelvic cavity pain syndrome differentiated as kidney deficiency and stagnation of damp heat, and explore the impacts on plasma P substance (SP), plasma beta-endorphin (β-EP).
METHODSOne hundred and eighty cases were randomly divided into a catgut embedding group (90 cases) and a western medication group (90 cases). In the western medication group, tamsulosin capsules 0. 2 mg were prescribed for oral administration, once a day; indometacin sustained release tablets, 25 mg, three times a day. Totally, the oral administration for 8 weeks was required. In the catgut embedding group, the acupoint catgut embedding therapy was applied to Qugu (CV 2), Shenshu (BL 23), Zhibian (BL 54), Huiyin (CV 1) and Sanyinjiao (SP 6), once every two weeks; the treatment of 4 weeks made one session, and two sessions were required. Before and after treatment, TCM symptom score, NIH-CPSI (the National Institute of Health Chronic Prostatitis Symptom Index) score, lecithin body numbers in prostatic fluid, score in SAS (self-rating anxiety scale), score in SDS (self-rating depression scale), the levels of SP and β-EP, etc. were observed in the two groups, and the clinical efficacy was assessed in the two groups.
RESULTS(1) Ten cases were dropped in either group. The total effective rate was 91. 25% (73/80) in the catgut embedding group, higher than 78. 75% (63/80) in the western medication group (P<0. 05). (2) After treatment, TCM symptom score, total score in NIH-CPSI, pain score and the scores in SAS and SDS were all reduced as compared with those before treatment in the two groups (all P<0. 05). After treatment, TCM symptom score, total score and pain score in NIH-CPSI, and the scores in SAS and SDS in the catgut embedding group were both lower than those in the western medication group (all P<0. 05). (3) After treatment, the lecithin body numbers were both increased as compared with those before treatment in the two groups (both P<0. 05), and the result in the catgut embedding group was higher than that in the western medication group (P<0. 05). (4)After treatment, the SP level was lower than that before treatment in the two groups (both P<0. 05); the level of p-EP was increased as compared with that before treatment (both P<0. 05). The SP level in the catgut embedding group was lower than that in the western medication group (P<0. 05); the level of β-EP was higher than that in the western medication group (P<0. 05).
CONCLUSIONThe acupoint catgut embedding therapy apparently relieves the clinical symptoms of chronic pelvic cavity pain syndrome differentiated as kidney deficiency and stagnation of damp heat as well as the condition of anxiety and depression, increases lecithin body numbers in prostatic fluid and β-EP level and reduces SP level.

8.Quality Control of Shuangshengen Capsules

Objective:To establish a quality control method for Shuangshengen capsules ( Salvia miltiorrhiza, Panax ginseng and Radix pueraria) . Methods:TLC was performed to identify Salvia miltiorrhiza, Panax ginseng and Radix pueraria. HPLC was used to determine the contents of puerarin and ginsenoside Rg1 . Results:The spots obtained from the test solutions had the same color as the reference solution at the same position. The spots on the TLC plates were clear without any interference. The calibration curve was line-ar within the range of 14. 42-115. 36 ng for puerarin, and the average recovery was 99. 94%(RSD=0. 13%, n=5). The calibration curve was linear within the range of 0. 1-0. 8 μg for ginsenoside Rg1, and the average recovery was 99. 85%(RSD=0. 42%,n=5). Conclusion:The method is reliable, simple and reproducible, and suitable for the quality control of Shuangshengen capsules.
10.Study on Bioflocculant Fermentation in Waste Residue of Chinese Medicine

Normal bacteria purification method was used in this study. Pseudomonas trivialis and Neurospora te-trasperma were screened from residues of traditional Chinese medicine (TCM). The fermentation conditions and flocculating function were also studied. The results showed that the selected strains grow well in the residues of TCM and their products can flocculate kaolin suspension solution. Based on these, we explored effects of various factors on the flocculated results in order to reveal the best fermentation condition among different combinations.
