1.Effect of water extracts of Alpinia oxyphylla Miq. fruit on memory impairment in cerebral ischemia rats

Objective This report investigate the intervention effect of the water extract of Alpinia oxyphylla Miq. fruit (AOF)on memory impairment and the mechanism in cerebral ischemia rats. Methods 48 Rats were randomly divided into sham-operated group(n=12), ischemia group(n=12), AOF group Ⅰ( n= 12)and AOF group Ⅱ (n= 12). The model of transient cerebral ischemic/reperfusion was made by bilateral occlusion of common carotid arteries in rats and combination with reducing blood pressure with an abdominal injection of so-dium nitroprusside. Learning-memory ability was observed by step through test. The content of NO and the activi-ties of NOS were measured in hippocampus. Results In step through test,the latency in ischemia group[(143.8±65.2)s]significantly decreased, the number of errors (8.9±4.2 ) significantly increased compared with sham-operated group [latency: (257.2±67.1 ) s; number of errors: (1.7±1.1 ), P<0.01 ]. The latency in AOF group Ⅰ and AOF group Ⅱ[(186.5±46.2) s, (193.4±43.7 ) s ] significantly increased, the number of errors (6.1±2.9,5.2±2.1 ) significantly decreased compared with ischemia group(P<0.05, P<0.01). In hippocampus, the content of NO and the activities of NOS in ischemia group [(56.53±27.42) nmol/mg prot, (17.23±5.64) nmol/mg prot] significantly increased compared with sham-operated group[ (40.02±17.9 ) nmol/mg prot, ( 10.46±6.15)nmol/mg prot], and in AOF group Ⅰ and AOF group Ⅱ [content of NO:group Ⅰ (46.60 ±20. 26)nmol/mg prot,group Ⅱ (42.38±21.23) nmol/mg prot ;activities of NOS:group Ⅰ (13.98±5.13 ) nmol/mg pint,group Ⅱ(13.61±5.27) nmol/mg prot] significantly decreased compared with ischemia group(P<0.05). Conclu-sion AOF could significantly ameliorate the memory impairment in cerebral ischemic rats. Its effects may be in-volved in the decrease of content of NO and activities of NOS in hippocampus.
2.Pharmaceutical Care in the Management of Nausea and Vomiting Induced by Adjuvant Chemotherapy for a Postoperative Patient with Gastric Cancer

Objective To provide reference for clinical pharmacist participating in management of nausea and vomiting induced by tumor chemotherapy. Methods The process of pharmaceutical care for a patient with severe vomiting caused by adjuvant chemotherapy after gastric cancer operation was described. Antiemetic application and drug adverse reactions were analyzed. A new treatment plan was given by clinical pharmacist. Results The suggestions were adopted by clinician. The vomiting was controlled and drug adverse reactions were dealt with. Conclusion To reduce the risk and improve the income of antiemetic,clinical pharmacists should pay more attention to clinical practice guideline,drug interaction and adverse reactions, provide the most suitable suggestions for clinicians according to pharmacology and evidence-based medicine.
3.Advances in Study on Relationship Between LncRNA and Inflammatory Diseases

Long non-coding RNA (lncRNA)is a non-coding RNA which lacks a complete open reading frame (ORF)and plays an important role in biological processes such as cell proliferation,differentiation and apoptosis. At present,expression of lncRNA has been found to be abnormal in inflammatory reaction and inflammatory diseases. It might be involved in the development and progression of inflammatory diseases via regulating the expression of multiple genes and activation of signaling pathways. This article reviewed the progress in study on relationship between lncRNA and inflammatory diseases.
4.Expression and purification of the soluble envelope protein of Japanese encephalitis virus

Objective To express and purify the envelope ( E) protein of Japanese encephalitis virus (JEV) in soluble form.Methods Various prokaryotic expression vectors , host strains and induction conditions including time and temperatures were screened to obtain an optimum prokaryotic expression system for JEV E protein in soluble form .The expressed protein was purified by using nickel column chromatography and gel filtration chromatography .Results The soluble JEV E protein , accounting for 23% of the totally bacteria soluble protein was effectively expressed by using the recombinant plasmid PBCX -E406 at low tem-perature.The purity of the expressed protein reached up to 85%after the purification by using nickel column and gel filtration chromatography .Conclusion Soluble JEV E protein was successfully expressed and puri-fied in a simple and efficient way .It would provide a useful tool for further investigation on JEV infection , attenuation mechanism of JE live vaccine strain SA 14-14-2 and the quality control of JE vaccine .It can also be used for the development of diagnosis assay for JEV .
5.DNA vaccine construction and expression of encoding gene of 18 000 outer membrane protein of human Helicobacter pylori

Objective:To construct a recombinant eukaryotic expression vector containing gene encoding 18 000 outer membrane protein from human Helicobacter pylori(H.pylori) and be expressed in COS-7 cell,and lay the foundation for the exploiting DNA vaccine of Hp.Methods:The target genes encoding 18 000 outer membrane protein were acquired from the prokaryotic expression vector pET32a(+)/528 by XholⅠ,KpnⅠ digestion simultaneously,in the same way,the pcDNA3.1 was digested by XhoⅠ,KpnⅠ digestion simultaneously,and the objective genes and pcDNA3.1 were extracted out of agarose electrophoresis with gel kit,and connected by T4 ligase.The recombinant vector pcDNA3.1/528 was used to select and transform,meanwhile express in COS-7 cell.The expressions of recombinant eukaryotic vector in COS-7 cell were investigated by reverse transcriptive-polymerase chain reaction and Western.Results:The gene had inserted into eukaryotic vector was the target gene encoding 18 000 outer membrane protein by enzyme digestion analysis,and objective gene was amplified from COS-7 cell transfected with pcDNA3.1/528 by RT-PCR,the Western showed that recombinant eukaryotic vector could be expressed in COS-7 cell.Conclusion:The recombinant eukaryotic vector pcDNA3.1/528 was constructed and expressed in COS-7 cell successfully.The results obtained lay the foundation for research on development of H.pylori DNA vaccine.

The paper proposed a new method to design and fabricate a flexible neural microelectrode arrays (MEA) for retinal prosthesis, the ion-beam technology was introduced to decrease the width of conductive wires and the distances between wires, a high density MEA (120 microelectrodes with a matrix of 10 x 12) was fabricated on a single layer of polymer from this. The MEA was proved to possess a fine electrochemical property. In vitro test, the average impedance of MEA on 1 kHz was 16 k omega +/- 2 k omega and the average phase difference was -85 degrees +/-30 degrees.

7.Effect of yanggan qinggan pinggan technique on the changes of blood pressure and angiotensin Ⅱ in patients with primary hypertension

BACKGROUND: Early clinical researches suggest that mediodespidine capsule can decrease blood pressure of patients with primary hypertension on the basis of yanggan qinggan pinggan technique.OBJECTIVE: To observe the effect of mediodespidine capsule on primary hypertension and angiotensin Ⅱ (Ang- Ⅱ ) of patients in the types of yin deficiency, yang excess and liver fire.DESIGN: Randomized controlled case observation.SETTING: Department of Cardiology, First Affiliated Hospital, Hunan University of Traditional Chinese Medicine.PARTICIPANTS: Patients were selected from the Inpatient Department and Out-patients Clinic of the First Affiliated Hospital of Hunan University of Traditional Chinese Medicine between April 1999 and April 2000. Inclusion criteria: Patients, who were diagnosed with the criteria of primary hypertension, who belonged to the types of yin deficiency, yang excess and liver fire, who could endure this test, who were coincidence with the criteria of mental-stress test, were all consent. Exclusion criteria: Patients who had severe congestive heart failure, atrophic arthritis and unable tolerance were excluded. There were 60 cases in total, including 29 males and 31females. All patients were divided into mediodespidine group and verapamil group with 30 in each group on the basis of randomly digital table.METHODS: Patients who had positive results of awaken mental-stress test were involved in this study. Cases in mediodespidine group were treated with mediodespidine capsule, which was made of tall gastrodia tuber, spine date seed and fourstamen stephania root at the dosage of 0.43 g/pill and were provided by the First Affiliated Hospital of Hunan University of Traditional Chinese Medicine (wei yao ji zi: 05-047, batch number: 990503), five pills each for 3 times a day. Cases in verapamil group were treatedwith verapamil (40 mg/pill, Shanghai Huanghe Liya PharmaceuticalCompany Limited,batch number: 980301B) 80 mg each for 3 times a day. The course in both groups was 4 weeks. ① Effect of blood pressure: Average value of blood pressure tested for 3 successive days were regarded as the blood pressure before treatment, and average value of blood pressure tested for 3 times after treatment was regarded as the blood pressure after treatment. ② Effect of symptom:Scores were recorded as four levels according to main symptoms, such as vertigo, headache, pain and weakness of waist and knees, before and after treatment. ③ Values of Ang- Ⅱ were measured with radioimmunoassay before and after cold pressor test. Effect on Ang- Ⅱ was evaluated with effect of decompression, effect of clinical symptom and criteria of clinical effect.MAIN OUTCOME MEARUSES: Clinical symptoms, values of blood pressure, heart rate and Ang- Ⅱ before and after cold pressor test.RESULTS: A total of 60 patients with primary hypertension were involved into the final analysis.① Totally effective rate of clinical symptoms of patients in mediodespidine group was higher than that of patients in verapamil group, but the effect of decompression was not significant between the two groups (P > 0.05). ② There was remarkably significant difference of heart rate in both groups before and after treatment (P < 0.01), especially the decrease in verapamil group was great, and there was significant difference (P < 0.05). ③ Values of systolic pressure and diastolic pressure were decreased in both groups after cold pressor test (P < 0.01), but there was no significant difference between the two groups (P > 0.05). ④ There was no significant difference in clinical effect between the twogroups (P > 0.05).⑤ After treatment, concentration of Ang- Ⅱ at basic level in both groups was decreased, and there was significant difference between them (P < 0.01);concentration of Aug-Ⅱ was decreased in both groups after cold pressor test as compared with that before treatment, but there was no significant difference between them (P > 0.05).CONCLUSION: Mediodespidine capsule can decrease blood pressure stably, and the effect is stronger than that of verapamil at the aspect of decompression and improving symptoms. Blood pressure of patients with primary hypertension in the types of yin deficiency and yang excess or liver fire can be decreased by decreasing the level of plasma Ang-Ⅱ to a certain degree.
8.Effects of hydrogen sulphide on the proliferation and apoptosis of human fetal lung fibroblasts during hypoxia

Aim To investigate the effects of endogenous and exogenous hydrogen sulfide(H_2S)on the proliferation and apoptosis of human fetal lung fibroblasts.Methods Human fetal lung fibroblasts were cultured under 2% O2~93% N_2~5% CO_2 for 24 h to produce hypoxia.Cells were divided into 6 groups:(1)Hypoxia group(N_2);(2)N_2+600 μmol·L~(-1) NaHS group;(3)N_2+1 200 μmol·L~(-1) NaHS group;(4)N_2+6 400 μmol·L~(-1) NaHS group;(5)N_2+400 μmol·L~(-1) cysteine(Cys)group;(6)N_2+200 μmol·L~(-1) S-adenosyl-L-methionine(SAM)group.After they were cultured for 24 h, MTT assay was used to evaluate the cell proliferation, and flow cytometry was used to detect cell apoptosis.Results Compared with N_2 group, 600 and 1 200 μmol·L ~(-1) NaHS(H_2S donor)significantly reduced proliferation induced by hypoxia of human fetal lung fibroblasts(P <0.01)without effects on apoptotic rates of cells(P >0.05)and 6 400 μmol·L~(-1) NaHS increased apoptosis of human fetal lung fibroblasts during hypoxia significantly(P <0.05), although no effects were found on proliferation of cells(P >0.05).In addition, Cys, substrate of cystathionine β-synthetase(H2S synthase, CBS) or SAM(activator of CBS)did not affect proliferation of human fetal lung fibroblasts induced by hypoxia(P >0.05), whereas apoptotic rates were increased significantly compared with that of N_2 group(P <0.05).Conclusions Endogenous and exogenous hydrogen sulfide can inhibit proliferation induced by hypoxia and promote apoptosis of human fetal lung fibroblasts, suggesting endogenous hydrogen sulfide may play a protective role through lung fibroblasts by inhibiting the pulmonary vascular structural remodeling caused by hypoxia.
9.Preliminary study of 3D arterial spin labeling in diagnosis of ischemic cerebrovascular disease

Objective To investigate the feasibility and clinical application value of 3D arterial spin labeling (3DASL) technique in diagnosis of ischemic cerebrovascular disease.Methods Experiment 1:Controlgroup (n=30) and brain infarction group (n=20) were established.All patients were routinely examined including 3DASL and dynamic susceptibility contrast (DSC) enhanced series.The analysis of the control group was performed using signal intensity measurement in three paired regions of interest (ROI) which were selected in left hemisphere centrum semiovale and the opposite hemisphere mirror regions respectively.The ROIs of brain infarction group were selected in the lesion areas and their mirror region.The cerebral blood flow (CBF) of each ROI was measured with 3DASL and DSC respectively.The CBF ratio to each ROI and its mirror ROI were calculated.Data were compared using paired-t test.Experiment 2:Acute cerebral infarction (n=30) and transient ischemic attack (TIA) group (n=30) were established.All patients were routinely examined including 3DASL and diffusion weighted imaging (DWI).The positive rate and the area of the lesions were calculated respectively with 3DASL and DWI and compared.Results Experiment 1:Both the control and brain infarction group demonstrated agreement between 3DASL and DSC.The CBF ratios of the control group were 1.02±0.18 and 1.06±0.24 respectively and revealed no statistically significant between that of 3DASL and DSC (P>0.05).The CBF ratios of brain infarction group were 0.50±0.12 and 0.48±0.18 respectively,and also revealed no statistically significant between two methods (P>0.05).Experiment 2:The positive rate of acute cerebral infarction were 100% with both DWI and 3DASL.The areas of the lesion detected by both methods were:SDwI<S3DASL,n=22; SDWI ≈-S3DASL,n=8; SDWI>S3DASL,n=0.The positive rate of TIA by DWI was 0%,and 70% by 3DASL.Conclusion The noninvasive,safe and repeatable 3DASL technique is almost as sensitive as DSC in the measurement of CBF.However,and it is more sensitive than DWI in diagnosis of TIA.Combining 3DASL and DWI will be helpful in early diagnosis of TIA,finding ischemic penumbra and estimating the blood perfusion of ischemic cerebrovascular disease.
10.Application of right heart contrast echocardiography in patients with transient ischemic attack of unknown cause

Objective To explore the application value of right heart contrast echocardiography in patients with transient ischemic stroke (TIA) of unknow cause.Methods Totally 120 patients with TIA (TIA group) and 60 cases of normal healthy volunteers (normal group) were enrolled.Two groups underwent right heart contrast echocardiography to observe whether there was a foramen ovale arteriosus (PFO) or not.Diagnostic criteria were left ventricular internal appearing micro bubble≥5 that came from the right atrium to left atrium in three cardiac cycles.Results Small and medium amount of right to left shunt had no significant difference between TIA group and normal group (both P>0.05),and large amount of right to left shunt had significant difference between the two groups (P<0.01).Conclusion Contrast echocardiography can effectively diagnose of PFO,and amount of right to left shunt of PFO has relationship with the onset of TIA.
