1.Clearance effect of regulation of serine/threonine protein kinase-1 signaling pathway by insulin on pulmonary edema in mice with acute lung injury

Objective To investigate the mechanism of insulin alleviating pulmonary edema in mice with acute lung injury(ALI) by serine/threonine protein kinase-1(SGK1).Methods 32 male adult C3H/HeN mice were randomly divided into control group(only pumped with the same amount of normal saline as the treatment group),ALI group(continuously pumped with the same amount of normal saline as the treatment group after modeling),treatment group [continuously pumped with 0.1 U/(kg·h) of insulin through jugular vein after establishing ALI model] and SGK1 siRNA group[continuously pumped with 0.1 U/(kg·h) of insulin and given SGK1 siRNA(75 μg SGK1 siRNA diluted in 100 μL saline) simultaneously after establishing ALI model] with 8 mice in each group.After 8 h,the mice were killed for arterial blood gas analysis(1 h after establishment of the model) and the changes of plasma glucose levels were detected(0,1,4and 8 h);The bronchoalveolar lavage fluid(BALF) was collected to detect the content of total protein,and the alveolar epithelial permeability and lung water content were measured;The pathological changes of lung tissue and apoptosis of lung epithelial cells were observed;The protein expressions of alveolar epithelial sodium channel(ENaC) and α_1-Na~+,K~+-ATPase and the phosphorylation level of SGK1 were determined by Western blot.Results There was no significant difference in plasma glucose level of ALI and treatment group at 0,1,4 and 8 h after insulin infusion(t=1.330 0,0.986 0,0.565 7 and 0.724 3,P=0.204 7,0.340 7,0.580 6 and 0.480 8,respectively).Compared with ALI group,the partial pressure of oxygen in arterial blood in treatment group increased significantly(t=6.026,P <0.000 1),while the BALF protein content,alveolar epithelial permeability,lung water content and lung epithelial cells apoptosis decreased significantly(t=7.39,5.286,5.651 and 3.312,P <0.000 1,=0.000 4,=0.000 2 and=0.007 8,respectively),and the expression of α-ENaC and α_1-Na~+,K~+-ATPase and the phosphorylation level of SGK1 in lung tissue significantly increased(t=26,18.67 and 8.547,P <0.000 1,<0.000 1 and=0.000 1,respectively);Compared with the treatment group,the BALF protein content,alveolar epithelial permeability,lung water content and lung epithelial cells apoptosis increased significantly in SGK1 siRNA group(t=5.964,3.449,3.148 and 3.520,P=0.000 2,0.006 2,0.010 4 and0.016 9,respectively),while α-ENaC and α_1-Na~+,K~+-ATPase protein expression and SGK1 phosphorylation level decreased significantly(t=13,9.874 and 7.741,P <0.000 1,<0.000 1 and=0.001 5,respectively).Conclusion Exogenous insulin can alleviate the pulmonary edema in ALI mice,which might be mediated via up-regulation of the expressions of α-ENaC and α_1-Na~+,K~+-ATPase by SGK1.
2.Discussion on how to promote the work about cadaver donation

Cadaver donation has far-reaching social significance, but currently it cannot satisfy the practical need. Through making stochastic visits to part of dwellers in Chongqing urban districts, to find out the main obstacles to cadaver donation. Then compared to the similar activities like blood donation, we learn about the experiences of such public service activities to get further improvement of cadaver donation.
3.Effect of yiqixue recipe on expressions of Bcl-2,Bax and Caspase3 in ovarian granulosa cells of controlled ovarian hyperstimulation mice

Objective To assess the effects of Jinghou Zengzhi Recipe ,a Chinese prescription with the action of tonifying Qi and blood,on the expressions of Bcl-2,Bax and Caspase3 in ovarian granulosa cells of con-trolled ovarian hyperstimulation(COH)mice. Methods A model of COH mice was prepared and 30 female KM mice were divided into blank group,model group and treatment group. The expressions of Bcl-2,Bax and Cas-pase3 were detected by Real-time Quantitative PCR and Western Blot. Results The mRNA and protein levels of Bax and Caspase3 were lower(P<0.05),while the protein levels of Bcl-2 were significantly higher(P<0.05)in the treatment group than those in the model group. There was no significant differences between the treatment group and blank group in the mRNA and proteins. Conclusion Jinghou Zengzhi Recipe can inhibit the ovarian granulo-sa cellular apoptosis of COH mice through increasing the expression of Bcl-2 protein as well as decreasing the ex-pressions of Bax and Caspase3 mRNA and proteins to nearly the natural level,which improves the quality of ovari-an follicle.
4.Effect of Yiqixue Buganshen Recipe on the level of serum VEGF in high-risk patients with ovarian hyper- stimulation syndrome

Objective To investigate the clinical effect of the Yiqixue Buganshen Recipe(YBR)on the level of serum VEGF in high-risk patients with OHSS. Method 298 infertile women with high-risk OHSS in the reproductive centre of our hospital from June to November ,2016 were enrolled into the traditional Chinese medicine (TCM) group (n = 148) and the control group(n = 150). 145 infertile women without high-risk OHSS were enrolled into the normal group. All the patients received the long protocol treatment ,and patients in the TCM group received the YBR treatment. Results Patients have higher quality embryo rate ,lower moderate and more severe OHSS rate in the TCM group compared with the control group(P<0.05). Patients in the normal group have lower serum VEGF compared with patients in the other 2 groups on the all three days. Patients in the TCM group has lower serum VEGF on the day of embryo transfer compared with patients in the control group(P<0.05). Conclusions The increment of the serum VEGF level is closely related to OHSS. The YBR may reduce the rate of moderate and severe OHSS by reducing the serum VEGF level on the day of embryo transfer ,which may prevent the moderate and severe OHSS of the high-risk patients.
5.Epidemiological analysis of elderly cases with the 2009 influenza A (H1N1) in Beijing

Objective To analyze epidemiological characteristics of elderly cases with influenza A (H1N1) in Beijing. Methods Descriptive epidemiological methods were used to describe epidemiological characteristics of elderly cases with 2009 influenza A (H1N1) in Beijing. Results The 321 laboratory-confirmed elderly cases with influenza A (H1N1) were reported in Beijing, and the morbidity was 13.2/100 000. The peak of infection occurred during November and December, the cases in this period accounted for 84.7% of the whole year, and 53.0% of them were reported in suburb areas, with the highest morbidity (19.2/100 000) in people beyond 85 years, and the morbidity increased with age (x2 = 7.24, P<0.01). The mild cases accounted for 63.6 %, severe and critical cases accounted for 36.4%. No significant difference was found between severity and BMI (x2=8.14, P=0.52). Severity was associated with number of chronic diseases (x2= 123.0, P<0. 01). Conclusions The H1N1 morbidity and proportion of severe cases are high among the elderly in Beijing, more attention should be paid to this population for influenza A (H1N1) prevention and control.
6.Effect of Yiqixue Buganshen Recipe on Endometrial Integrinβ3 Expression and Pinopode Formation in Mice with Embryo Implantation Dysfunction

Objective To observe the effect of Yiqixue Buganshen Recipes(YBR), the compound recipes of Chinese herbal medicine with the actions of replenishing Qi and blood and tonifying liver and kidney, on the endometrial integrin β3 of the mice with embryo implantation dysfunction, and to explore the molecular mechanism of YBR in improving embryo implantation. Methods Ninety mice were divided normal group, model group, and YBR group, 30 mice in each group. YBR group was given Jinghou Zengzhi granules 8mg/kg for promoting post-menstruation proliferation and Cuhuangti granules 8mg/kg for promoting the formation of corpora luteum. At 8∶00 am of pregnant day 4 (Pd4), subcutaneous injection of mifepristone was used for inducing embryo implantation dysfunction in the model group and YBR group. Twenty mice from each group were executed 12 hours after subcutaneous injection of mifepristone, and the uterus was isolated for the detection of mice endometrial integrin β3 mRNA and protein expression with real-time RT-PCR and Western blotting method respectively. Ten mice from each group at Pd4 were executed, and the uterus was isolated and then fixed in 2.5% glutaraldehyde solution for the detection of the formation of pinopode in mice endometrium under scanning electron microscope. Results The detection by real-time PCR and Western blotting method revealed that integrinβ3 mRNA and protein expression levels of the model group were significantly lower than those of the normal group (P<0.05), and YBR group had higher integrin β3 mRNA and protein expression levels than the model group(P<0.05). The formation of pinopodes in the model group was less than the normal group and YBR group, but the number of formed pinopodes in YBR group was similar to that in the normal group. Conclusion YBR can up-regulate the expression of integrin β3 in the endometrium of mice with embryo implantation dysfunction, and have positive regulatory effect on the formation of pinopodes on Pd4.
7.Effect of Yiqixue Recipe on Ovary Apoptosis and Expression of Bim in Ovarian Granulosa Cells of Con-trolled Ovarian Hyperstimulation Rats

Objective To assess the effects of Jinghou Zengzhi Recipe(JHZZR),a Chinese prescription with the action of tonifying Qi and blood ,on the ovary apoptosis and expression of Bim in ovarian granulosa cells (GCs) of controlled ovarian hyperstimulation(COH) rats , and analyze the possible therapeutic mechanism. Methods A model of COH rats were prepaered and 30 female SD rats were randomly divided into 5 groups , including control group,positive control group,low,medium and high concentration group in six rats in each group. The apoptosis index(AI)in ovarian GCs were detected by TUNEL ,and the expression of Bim by qPCR. Results The AI of ovarian GCs in high and medium concentration group were obviously lower(P < 0.05)than that in the positive control group,and there is no significant differences(P > 0.05)comparing with control group. The mRNA levels of Bim were lower(P<0.05)in all concentration group than those in positive control group ,but still higher(P<0.05)than that in control group. And there is no significant differences(P>0.05)during the three concentration groups in Bim mRNA. Conclusion JHZZR can inhibit the ovarian GCs apoptosis of COH rats through decreasing the expression of Bim mRNA ,which improve the quality of ovarian follicle.

Objective To Investigation the quality of marrow smear purchased by CDUTCM.Methods The quality and the typ-icality of marrow smears purchased during 2015 -2016 were collectively examined,and then decided whether these smears fit the blood cell morphology experimental teaching requirement.Results Of all the 960 marrow smears purchased these two years, 49.7% failed in smear made or stained,and 16.0% failed to meet the teaching requirements in the typicality of marrow cells.Con-clusion Teaching marrow smears,being different from clinic ones in their preparation and morphological diagnosis,must be of great quality in sustaining and of better typicality in their cell features.

Objective To determine the effects of Tempol, one of the nitroxides, in the presence of ultraviolet-AI (UVA1, 340 nm -400 nm) on superoxide enzyme (SOD) activity, lipid peroxidation, and expression of collagen Ⅰ, collagen Ⅲ and matrix metalloproteinases (MMP)-1, MMP-3 in human dermal fi broblasts in vitro. Methods Fibroblasts were irradiated by a single exposure to UVA1 and at the same time incubated with or without Tempol, and detected 24 h later. SOD activity and lipid peroxidation, as shown by accumulation of malondialdehyde (MDA), were detected by biochemical assay. 10.Analysis of clinical features in patients with influenza A/H1N1

A total of 3499 cases of influenza A (H1N1) were included in this study for analysis.Epidemiological and clinical data of these cases were input into EpiData software and analyzed by SPSS software. Throat swabs were collected from the cases and detected for nucleic acid of influenza A ( H1N1 )virus using real-time polymerase chain reaction (RT-PCR) with fluorescence quantitative method, and time of viral excretion and clinical features of the cases were analyzed. Results showed that 0. 37% of the cases were in-apparent and asymptomatic and the most common symptom of the cases was fever (86. 77% ).Throat swabs converted to negative on the sixth day of onset in average, and no factors related to the time of conversion was found with logistic regression analysis.
