1.The Effect of Methylprednisolone on the Quality of Life in Advanced Lung Cancer Patients with Dyspnea

Objective To analyze the changes of the quality of life of patients with advanced lung cancer accompanied with dyspnea before and af?ter methylprednisolone therapy,so as to investigate the effects of methylprednisolone on the quality of life of the patients. Methods Totally 134 ad?vanced lung cancer patients with dyspnea were divided into two groups,the methylprednisolone group and the control group,according to whether they consented to the use of hormone therapy. All of them were followed with visual analog scale(VAS),the quality of life questionnaire?core 30 (EORTC QLQ?C30)and the quality of life questionnaire?lung cancer module 13(QLQ?LC13)questionnaire. Results At one hour after treatment compared to before treatment,the VAS scores for both groups were significantly decreased(P<0.01),and the score for the methylprednisolone group were lower than the control group(P<0.05). At two weeks after treatment compared to before treatment,social function,emotional function, and overall quality of life scores for both groups were decreased(P<0.05). But the scores of pain,dyspnea,insomnia,loss of appetite,constipation, dysphagia were increased(P<0.05). Compared to the methylprednisolone group,the dyspnea and pain score for the control group was higher(P<0.05). Conclusion Methylprednisolone may improve the quality of life of advanced lung cancer patients with dyspnea in short term and slow down the deteriorating process of their quality of life.
2.Effect of mycophenolate mofetil on the immune function in patients with systemic lupus erythematosus

Objective To observe the effect of mycophenolate mofetil (MMF),a new type of immunosuppressant,on the immune function in patiens with systemic lupus erythematosus (SLE).Methods The changes of serum lL-6,IL-8,TNF-α,sIL-2,ANA,A-dsDNA and subsets of lymphocytes were observed in patients with SLE before and after treatment with MMF and CTX by ELISA,indirect immunofluorescent assay and flow cytometry,and the effect of MMF and CTX on the functions of hematopoiesis,liver and kidney were also observed.Results Three months after treatment with MMF,the serum levels of IL-6,IL-8,TNF-α,sIL-2,ANA,and A-dsDNA in patients with SLE were significantly decreased,CD3+,CD4+ and CD4+CD45RA+ cells were significantly increased,and CD8+,CD4+CD45RO+,CD8+CD45RA+,CD8+CD45RO+ cells were significantly decreased.No remarkable injury was observed on the functions of hematopoiesis, liver and kidney after treatment with MMF.Conclusion MMF can inhibit T and B lymphocytes selectvely and is a new type of immunosuppresant to treat SLE with less toxicity and side-effect.

miR-205 is differentially expressed in tumor tissues and is closely related to tumorigenesis and tumor metastasis.miR-205 can regulate the biological behaviors of tumor cells such as cell proliferation,differentiation,in situ invasion and distant metastasis by post-transcriptional regulation through binding to targeted genes.Additionally,further research of miR-205 may be helpful for tumor diagnosis,targeted therapy and prognosis.

5.Effect of Endotoxin Tolerance on Cytokine

Objective To further determine the inflammation?associated cytokine changes during endotoxin(LPS)tolerance. Methods Kun?ming mice were divided into groups. The endotoxin tolerance mouse model was established with repeated injection of LPS. Serum levels of tumor ne?crosis factor?α(TNF?α),interleukin?10(IL?10)and IL?17 were determined by ELISA assay. Results The serum concentrations of TNF?α,IL?10 and IL?17 were 2 261.49±1 090.28 pg/mL,7 780.42±2 320.04 pg/mL,and 27.28±13.76 pg/mL in the LPS control group,which were increased significantly compared to the normal healthy control group. The serum concentrations of TNF?α,IL?10 and IL?17 were 1 174.19±450.91 pg/mL, 1 293.26±520.85 pg/mL,and 22.26±11.44 pg/mL in the endotoxin tolerance group. Levels of TNF?αand IL?17 were further decreased by dexameth?asone treatment during LPS tolerance inducing,while the IL?10 level did not change significantly. Conclusion Compared to the status of endotoxin inflammation,levels of TNF?αand IL?10 decreased significantly,while levels of IL?17 were not significantly changed in the status of endotoxin toler?ance. Dexamethasone could further suppress TNF?αand IL?17 levels during endotoxin tolerance inducing.
6.Effects of PPARγ gene expression on cell migration, invasion, and proliferation in endometrial cancer cells

Objective To observe the effects of differentially expressed peroxisome proliferatoractivated receptor γ (PPAR γ) on cell migration,invasion and proliferation of endometrial cancer cells.Methods Two endometrial cancer cell lines ECC-1 (ER positive) and KLE (ER negative) cells were used in this study.To up or down regulate PPARγ expression,the transient transfection by using PPARγ expression vector (PPARγ expression vector group) and PPARγ small interference RNA (PPARγ siRNA group) were done.The negative control groups were cells transfected by nonsence sequence siRNA (siRNA non sence sequence group) or empty vector (empty vector group).At the same time,cells only added with liposome were used as blank control group.Then,quantitative real time (RT)-PCR and western blot were used to detect PPARγexpression both in mRNA and protein levels.To assess the expression levels of Wnt signaling pathway,western blot was performed to analysis protein levels of β-catenin and C-myc.The effects on cell migration,invasion and proliferation using in vitro transwell migration,invasion assays and cell counting kit-8 (CCK-8) assay were further be examined.Results After transfection for 48 hours,quantitative RT-PCR and western blot showed that PPARγmRNA (5.18 ± 0.99,4.54 ± 0.89) and protein (1.45 ± 0.12,1.30 ± 0.13) expression levels significantly increased and the protein levels of β-catenin (0.44 ± 0.06,0.46 ± 0.04) and C-myc (0.42 ± 0.08,0.30 ± 0.11) decreased in PPAR γ expression vector group,while in PPARγ siRNA group,PPARγ mRNA (0.48 ± 0.08,0.53 ± 0.11) and protein (0.41 ±0.04,0.49 ±0.05) expression levels decreased and the protein levels of 3-catenin (1.18 ±0.12,0.89 ±0.07) and C-myc(0.91 ±0.08,0.77 ±0.12) increased significantly compared with control groups (all P < 0.05).In vitro migration and invasion assay indicated that the migratory and invasive cell numbers of PPARγ expression vector group (ECC-1:129 ± 9,63 ± 12 ; KLE:119 ± 9,68 ± 16) were significantly decreased,while the migratory and invasive cell numbers of were PPARγ siRNA group (ECC-1:201 ± 14,142 ±9 ; KLE:170 ± 11,138 ± 7) increased significantly compared with those in control groups(all P < 0.05).CCK-8 assay showed that A values (0.66 ±0.14,0.78 ±0.06) in PPARγexpression vector group were lower than those in control groups,and in PPARγ siRNA group,A values (1.42 ± 0.16,1.23 ± 0.04) were higher than those in control groups,and there were statistically significant difference among them (all P < 0.05).Conclusion Up-regulated PPARγ gene expression could inhibit endometrial cancer cell migration,invasion and proliferation abilities,and down-regulated PPARγ gene expression could promote endometrial cancer cell migration,invasion and proliferation abilities.
7.Abnormity rates of blood lipids and uric acid and their relationship analysis in health examination population in Yangzhou city

Objective To investigate the abnormity of blood lipids and uric acid ,and to analyze their relationship in health exami-nation population over 18 years in Yangzhou .Methods Totally 28 202 individuals receiving the health examination from October 2011 to August 2012 in the physical examination center of this hospital were enrolled in our study .Total cholesterol(TC) ,triglycer-ide(TG) ,low density lipoprotein cholesterol(LDL-C) ,high density lipoprotein cholesterol(HDL-C) and blood uric acid(UA) were detected ,and the relationship between age and sex with the abnormity of blood lipids and uric acid and the relationship between UA levels and blood lipids were analyzed .Results Among the detection population ,the detection rates of abnormal TG ,abnormal TC , abnormal HDL-C ,abnormal LDL-C and hyperuricemia were 31 .5% ,32 .1% ,15 .9% ,18 .4% and 17 .3% respectively .Compared with females ,males had the higher rates of abnormal blood lipids and UA (P<0 .05) .The rate of abnormal blood lipids was in-creased with age increase before 50 years old ,the > 50 -70 years old group had the highest abnormity rate of blood lipids ,then which was gradually decreased after 70 years old .The morbidity rate of hyperuricemia was increased with age .Compared with nor-mal UA group ,the hyperuricemia group had the higher rate of abnormal blood lipids (P<0 .01) .Conclusion The health examina-tion population over 18 years have the higher abnormity rate of blood lipids and uric acid ,which is significantly correlated with age and sex ;the increase of uric acid has certain correlation with abnormal blood lipids metabolism .Both blood lipids and uric acid are the risk factors of cardiocerebralvascular diseases ,rational dietary structure and periodic physical examination have the important significance to prevent the occurrence of these diseases .
8.Clinical analysis of different intraperitoneal hyperthermic chemotherapy drugs in the treatment of advanced ovarian cancer

Objective To explore short term curative effects and adverse reactions of two different kinds of intraperitoneal hyperthermic chemotherapy (IPHC) drugs in the treatment of advanced ovarian cancer after cytoreductive surgery.Methods 76 patients with advanced ovarian cancer in the Second Affiliated Hospital of Zhengzhou University from July 2013 to November 2015 were divided into two groups:single-drugs group of 36 patients (IPHC with 5-fluorouracil after cytoreductive surgery combined with intravenous chemotherapy),combined treatment group of 40 patients(IPHC with 5-fluorouracil and carboplatin after cytoreductive surgery combined with intravenous chemotherapy).Short term curative effects,postoperative clinical indicators and adverse reactions of chemotherapy in two groups were compared and analyzed.Results The CA125 effective rates in single-drugs and combined treatment group were 86.11% and 95%,and the difference showed statistically significant differences (P > 0.05).The ascites remission rates in single-drugs and combined treatment group were 97.22% and 97.5%,and the difference between two groups showed no statistically significant differences (P > 0.05).Adverse drug reactions showed statistical difference in distribution of the bone marrow,liver damage and gastrointestinal toxicity.No statistical difference were found between the two groups in terms of distribution of renal damage and cardiovascular system damage.Conclusion IPHC after cytoreductive surgery in the treatment of advanced ovarian cancer is an effective means as adjuvant chemotherapy.The short-term curative effect of combined treatment group is obvious and adverse reactions can be tolerated.IPHC can be applied according to the patient's specific clinical situation.
9.Acupuncture method of "Huoxue Sanfeng, Shugan Jianpi" for morning blood pressure in patients with cerebral infraction combined with essential hypertension: a randomized controlled trial.

OBJECTIVETo observe the clinical effects of acupuncture method of "Huoxue Sanfeng, Shugan Jianpi" (activating blood and eliminating wind, soothing liver and strengthening spleen) on morning blood pressure in patients with cerebral infraction combined with essential hypertension.
METHODSSixty-eight patients were randomly divided into an observation group and a control group, 34 cases in each one. The patients in the two groups were treated with acupuncture method of "Xingnao Kaiqiao" (consciousness-restoring resuscitation) and oral administration of nifedipine. In addition, patients in the observation group were treated with acupuncture method of "Huoxue Sanfeng, Shugan Jianpi" that met the criteria of standard manipulation, in which bilateral Renying (ST 9), Quchi (LI 11), Hegu (LI 4), Zusanli (ST 36) and Taichong (LR 3) were selected. The treatment was given once a day, five times a week, for totally six weeks. The improvement and control rate of morning blood pressure in the two groups were observed.
RESULTS(1) After treatment, the morning blood pressures were decreased significantly in the two groups (all P < 0.05); after 15 treatments, the reduction of systolic pressure and diastolic pressure in the observation group was superior to that in the control group, but the difference was not significant (both P > 0.05); after 30 treatments, the reduction of systolic pressure and diastolic pressure in the observation group was significantly superior to that in the control group (both P < 0.05). (2) After 30 treatments, the control rate of morning blood pressure in the observation group was significantly higher than that in the control group [82.4% (28/34) vs 58.8% (20/34), P < 0.05].
CONCLUSIONAcupuncture method of "Huoxue Sanfeng, Shugan Jianpi", characterized with standard manipulation criteria, can effectively control morning blood pressure in patients with cerebral infraction combined with essential hypertension.

10.Effect of oral administration of type Ⅱ collagen peptide-cholera toxin B subunit complex on collageninduced arthritis

ObjectiveThe aim of this study is to investigate whether oral administration of collagen Ⅱ peptide (250-270)[C Ⅱ (250-270)]-cholera toxin B subunit (CTB)complex could effectively set up oral immune tolerance to collagen-induced arthritis (CIA) in mice. MethodsDBA/1 mice were divided into Ⅰ, Ⅱ, Ⅲ and Ⅳgroups. Group Ⅰ was normal control group. Collagen type Ⅱ emulsified in Freund's complete adjuvant were injected to mice of groups Ⅱ , Ⅲ and Ⅳ twice from the base of the tail. Mice of group Ⅲ were fed with C Ⅱ (250-270)-CTB covalent complex twice after the arthritis was developed. Mice of group Ⅳ were fed with C Ⅱ(250-270) and CTB mix at the 14th day after primary immunization. Visual scores and histopathologic scores of arthritis were recorded. The frequencies of arthritis between the groups were compared usingFisher's exact test. The clinical and histological severity of arthritis were analyzed by ANOVA.Results The frequencies of arthritis in groups Ⅰ , Ⅱ , Ⅲ and Ⅳ were 0, 100%, 100% and 25% respectively. Average accumulative scores of arthritis were 0, 5.0±1.7, 10.8±2.8 and 1.0±2.0 respectively. Average accum-ulative histopathological scores of arthritis were 0, 16±8, 32±13 and 7±6 respectively. Conclusion Oral administration of C Ⅱ (250-270) and CTB mix in arthritis mice after C Ⅱ immunization can suppress the onset and severity of arthritis. Oral administration of C Ⅱ (250-270)-CTB covalent complex in the acute stage of arthritis can accelerate arthritis.
