1.Clinical characteristics of hereditary spastic paraplegia with thin corpus callosum

Objective To explore the clinical characteristics of hereditary spastic paraplegia with thin corpus callosum(HSP TCC).Methods Clinical data of 4 patients with HSP TCC were analysed retrospectively.Results 4 patients were at the onset during youngsters,they revealed mental impairment,walk of spasticity,spasticity of the lower extremities,slowly progressive weakness and hyperreflexia, extensor plantar responses and morbid indication for positive. Sensory impairment was not observed. 2 cases showed ataxia and sphincter disturbance;1 case showed upper limb spasticity and muscular atrophy. Cranial MRI revealed an extremely thin corpus callosum on sagittal image.Conclusion Main clinical characterizations of HSP TCC were slowly progressive spastic paraparesis, mental impairment during youngsters, cranial MRI showed extremely thin corpus callosum.
2. Expression of GTF2IP23 in breast cancer and it mediated regulation of GTF2I

Objective:
To investigate the expression level of antisense transcript of pseudogene, general transcription factor Ⅱi psedugen23 (GTF2IP23), in breast cancer and its effect on the host gene general transcription factor Ⅱi (GTF2I).
Methods:
The expressions of GTF2IP23 and GTF2I were detected in 40 cases of invasive breast cancer tumors and their counterparts by using quantitative real-time polymerase chain reaction (qRT-PCR). The effects of GTF2IP23 on the expression of GTF2I gene and cell proliferation and migration were analyzed by overexpression of GTF2IP23 in breast cancer cells.

【Objective】 To investigate the effect of HLA-G expressed in platelets on Tax protein of human T cell leukemia type 1 virus (HTLV-1). 【Methods】 Platelets were isolated from anticoagulant whole blood, and HLA-G molecule on platelet membrane was detected by flow cytometry. The content of secretory HLA-G before and after platelet lysis was detected by ELISA, HTLV-1 human lymphoma cells MT2 were cultured with platelet lysate (PL). The effect of HLA-G in platelets on the expression of HTLV-1 protein Tax was evaluated by Western blot (WB). 【Results】 Membrane type mHLA-G was highly expressed on the surface of platelet membrane. The expression of secretory sHLA-G (ng/mL) increased after platelet lysis (15.73±1.01) vs (6.65±0.47), the expression of sHLA-G increased with the increase of platelet concentration in a dose-dependent manner. Compared with fetal bovine serum, PL significantly promoted the high expression of HLA-G protein and HTLV-1 virus tax protein in MT2 cells, and the addition of anti-HLA-G antibody to PL could effectively inhibit the expression of Tax and HLA-G protein. 【Conclusion】 High expression of immune tolerance molecule HLA-G on platelets can induce high expression of HTLV-1 protein Tax in human lymphoma cell MT2, which contributes to viral infection.
4.Antimetastatic effects of cordycepin and its molecular mechanism in human hepatoma MHCC97H cells in vitro and in vivo

The objective of this study was to examine the antimetastatic effects of cordycepin and elucidate its molecular mechanism using MHCC97H cells in vitro and in vivo. Cellular proliferation was detected with MTT assay. The migration and metastatic potential were measured with scratch wound healing as well as transwell migration assays in vitro. Protein expression was detected by Western blotting. Antitumor and antimetastatic effects of cordycepin were evaluated by subcutaneous xenograft and lung metastatic model in vivo. The results demonstrated that cordycepin significantly inhibited MHCC97H cells proliferation and metastasis which was due to the down-regulation of AKT, p-AKT, p-GSK-3β, β-catenin, N-cadherin, MMP-7, MMP-9 and up-regulation the expression of E-cadherin. Furthermore, cordycepin inhibited tumor growth and metastasis in a dose-dependent manner in vivo. Cordycepin(40 and 20 mg·kg-1)and 5-fluorouracil group significantly inhibited the tumor weights to 0.38 ± 0.04, 0.61 ± 0.08 and 0.65 ± 0.07 g, respectively, comparing with the control group(1.52 ± 0.46 g)(P < 0.01), but not 10 mg·kg-1 cordycepin group(1.13 ± 0.36 g)(P > 0.05); the lung metastasis nodus numbers showed the same results, which in all group above(48.9 ± 7.2, 67.2 ± 9.4, 73.6 ± 8.6, respectively)were fewer than the control group(123.5 ± 14.5)(P < 0.01), except 10 mg·kg-1 cordycepin(106.4 ± 11.3)(P > 0.05). Collectively, cordycepin inhibited MHCC97H cell proliferation and metastasis in vivo and in vitro.
5.Clinical characteristics and risk factors in elderly patients with post-stroke seizures

Objective To investigate the risk factors and seizure types of secondary seizures in elderly patients with acute ischemic stroke(AIS),and to screen the effective indicators for predicting Post-stroke seizures(PSS). Methods Elderly patients who were hospitalized in Henan Provincial People's Hospital from January 2019 to June 2020 and diagnosed with AIS for the first time were consecutively collected. The patients were followed up for 1 year and divided into the observation group and the control group according to whether they developed secondary PSS,and the patients in the observation group were divided into subgroups according to the time of secondary PSS. The demographic and clinical characteristics of the patients were recorded,and the National Institutes of Health Stroke Scale(NIHSS) score and Modified Rankin Scale(mRS) score were used to assess stroke severity and outcome at discharge,respectively. Univariate and multivariate Logistic regression analyses were used to identify the independent risk factors for PSS. Results (1)Comparison of general information:There were statistically significant differences in gender,lesion location,hemorrhagic transformation(HT),NIHSS score ≥11,hypertension,cardiac cerebral embolism(CE) and baseline NIHSS score between the observation group and the control group(P<0.05). (2)Results of Multivariate Logistic regression analysis:Cortical infarction(OR=1.702,95%CI 1.172-2.471,P=0.005),HT(OR=2.384,95%CI 1.605-3.434,P<0.001) and NIHSS scores ≥11(OR=1.723,95%CI 1.188-2.500,P=0.004) were independent risk factors for PSS,and the differences were statistically significant. (3)Comparison of early-onset seizures(ES) and late-onset seizures(LS):Focal progression to bilateral tonic-clonic seizures(46.8%) was the most common seizure type in this group and the most common seizure type in the LS group(50.4%). The most common seizure type in ES group was focal seizure(47.8%). In addition,the proportion of negative emotions(65.2%) and mRS Score[2(3,4)]in ES group were higher than those in LS group[37.4%,2(2,3)],and the differences were statistically significant. Conclusion Cortical infarction,HT and NIHSS score ≥11 were independent risk factors for PSS. Focal progression to bilateral tonic-clonic seizures is the most common seizure type and is a common seizure type of LS,The common seizure type of early seizures is focal seizures,which are associated with negative mood and a worse prognosis.
6.Inhibitory effect of galangin on DNA topoisomerases in lung cancer cells.

OBJECTIVE:
To explore the eff ect of galangin on DNA topoisomerases in lung cancer cells A549 and H46 as well on cell growth.
METHODS:
The inhibitory effect of galangin on the growth of A549 and H46 cells was analyzed by MTT method. The effect of galangin on Topo I activity was detected by the agarose gel electrophoresis method. Furthermore, the interaction between galangin and Topo I was evaluated by fluorescence spectroscopy. Finally, the eff ect of galangin on the Topo I structure was discussed.
RESULTS:
Galangin could induce the apoptosis of A549 and H46 cells (IC50 was 0.221 mmol/L and 0.173 mmol/L, respectively). Agarose gel electrophoresis showed that galangin exerted significant inhibitory effect on Topo I activity. Fluorescence spectrum analysis showed that galangin was able to quench Topo I fluorescence, and hydrophobic interaction was the main driving force. Circular dichroism analysis showed that galangin induced Topo I conformation change and increased the content of α-helix, which prevented the formation of active center and in turn led to the decrease in Topo I activity. Molecular simulation results showed that galangin could bind to the active center of Topo I to form hydrogen bonds with the catalytic site at Arg364 and Asn352.
CONCLUSION
Galangin is able to inhibit Topo I activity and to reduce the unwinding rate of single stranded DNNA in tumor cells, which plays an important role in induction of A549 and H46 cell apoptosis.

7.Effect of fluoxetine combined with Shugan Jieyu capsule in the treatment of patients with depression and its impact on heart rate variability

BackgroundDepression is a kind of disease with relatively high suicide risk, which seriously affects the quality of life of patients and their families, and brings a great burden to society. Antidepressants in western medicine are effective, but the improvement of depressive symptoms is relatively limited by single use, and the combination of two antidepressants may increase the risk of adverse reactions in patients. The rational use of Chinese patent medicine and western medicine may play a complementary role, and the safety of Chinese patent medicine is high. ObjectiveTo explore the early clinical efficacy of fluoxetine combined with Shugan Jieyu capsule in the treatment of depression, and to compare the differences in efficacy, safety and influence on heart rate variability between fluoxetine combined with Shugan Jieyu capsule and fluoxetine alone, so as to provide references for clinical medication of depression patients. MethodsFrom December 2015 to June 2016, 64 patients who met the diagnostic criteria of depression in the Diagnostic and Statistical Manual of Mental Disorders, fifth edition (DSM-5) who were hospitalized in the Second Affiliated Hospital of Xinxiang Medical University were selected as the research objects, and were randomly divided into the combined medication group and the fluoxetine group with 32 patients in each group. Patients in both groups were treated with fluoxetine, while patients in the combined medication group were treated with Shugan Jieyu capsule on this basis. Patients in both groups were assessed with Hamilton Depression Scale-24 item (HAMD-24), Hamilton Anxiety Scale (HAMA) and Heart Rate Variability (HRV) before treatment, and were assessed with HAMD-24 and Treatment Emergent Symptom Scale (TESS) at the end of the 2nd, 4th and 6th week of treatment, and HRV was analyzed again at the end of the 6th week of treatment. ResultsThe study ultimately included 60 patients with depression, with 30 cases in the combination therapy group and 30 cases in the fluoxetine group. At the end of the 2nd, 4th and 6th week of treatment, the HAMD-24 score of the combined drug group was lower than that of the fluoxetine group (t=-2.677, -3.960, -4.432, P<0.05 or 0.01). Compared with before treatment, the 24-hour mean standard deviation of normal RR intervals (SDNN), normal low frequency (nLF) and normal high frequency (nHF) in the combined treatment group were higher at the end of the 6th week (t=-73.970, -31.878, -38.721, P<0.01), but significant lower in LF/HF (t=3.525, P<0.01). At the end of the 6th week of treatment, the total effective rate of the combined treatment group was higher than that of fluoxetine group, and the difference was statistically significant (86.67% vs. 70.00%, χ2=18.764, P<0.01). At the 2nd, 4th and 6th week of treatment, there was no significant difference in the number of adverse reactions between the two groups (P>0.05). ConclusionCompared with fluoxetine alone, Shugan Jieyu capsule combined with fluoxetine may be better in clinical efficacy and improvement of heart rate variability in patients with depression, without increasing adverse reactions.
8.LncRNA H19 and Target Gene-mediated Cleft Palate Induced by TCDD.

This study investigated the role of long non-coding RNAs (lncRNAs) in the development of the palatal tissues. Cleft palates in mice were induced by 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD). Expression levels of long non-coding RNA H19 (lncRNA H19) and insulin-like growth factor 2 (IGF2) gene were measured by quantitative real-time polymerase chain reaction (qRT-PCR). The rate of occurrence of cleft palate was found to be 100% by TCDD exposure, and TCDD could cause short upper limb, cerebral fissure, webbed neck, and short neck. The expression levels of lncRNA H19 and IGF2 gene specifically showed embryo age-related differences on E13, E14, and E15 in the palatal tissues. The expression levels of lncRNA H19 and IGF2 gene showed an inverse relationship on E13, E14, and E15. These findings demonstrated that lncRNA H19 and IGF2 can mediate the development of mouse cleft palate.

9.Mannan-Binding Lectin Inhibits Candida Albicans-Induced DC Maturation and Cytokine Secretion.

OBJECTIVETo investigate the effects of mannan-binding lectin (MBL) on the maturation and cytokine secretion of human dendritic cells (DC) induced by Candida albicans (C. albicans).
METHODSThe plastic-adherent mononuclear cells were prepared from the blood of healthy adult volunteers. The human peripheral blood mononuclear cells-derived dendritic cells (MNC-DC) were induced by 5-day-culture in medium supplemented with rhGM-CSF and rhIL-4, and then cultured for 2 days in presence or absence of C. albicans at varying concentration of human MBL ranging from 1 to 20 mg/L. DC's shape and characters were observed under inverted microscopy, the expression of CD83 and CD86 on DC was analyzed by FACS. The levels of TNF-α and IL-6 were detected by ELISA. FACS also was used to investigate the interaction of MBL with immature DC(imDC) and C. albicans. Western blot was used to detect C. albicans-induced IκBα phosphorylation and p65/NF-κB translocation in DC.
RESULTSMBL at higher concentrations (10-20 mg/L) down-regulated the expression of CD83 and CD86 on the monocyte-derived dentritic cells(MoDC) induced by C. albicans, and inhibited the production of TNF-α and IL-6 induced by C. albicans. FACS showed that MBL could not only bind to C. albicans but also bind to imDCs in a Ca2+-dependent manner. Western blot showed that MBL could decrease the phosphorylation of IκBα and the nuclear translocation of p65/ NF-κB.
CONCLUSIONMBL may inhibit TNF-α and IL-6 production induced by C. albicans in DC through NF-κB signaling pathways, suggesting that MBL can play some roles in the regulation of C. albicans-induced immune response.

10.Relationship between parenting rearing behavior and mobile phone dependence among college students: the mediating role of emotional intelligence

ObjectiveTo investigate the prevalence rate of mobile phone dependence, and to analyze the mediating role of emotional intelligence between parenting rearing behavior and mobile phone dependence among college students, so as to provide references for relieving the mobile phone dependence among college students. MethodsStratified cluster sampling method was applied to enroll 1 200 students from three colleges in He'nan province, and the selected individuals were assessed using Parental Bonding Instrument (PBI), Mobile Phone Addiction Index (MPAI) and Emotional Intelligence Scale (EIS). Then the relationship between parenting rearing behavior and mobile phone dependence was discussed, and the mediating effect of emotional intelligence was explored using AMOS path analysis. ResultsOf the students who completed the survey, 597 out of 1 090 (54.77%) suffered from mobile phone dependence. The scores of care and encouragement of behavioral freedom in PBI were negatively correlated with the total score and each dimension score of MPAI (r=-0.316~-0.197, P<0.01), the denial of psychological autonomy score in PBI was positively correlated with the total score and each dimension score of MPAI (r=0.206~0.258, P<0.05 or 0.01). EIS score was negatively correlated with total score and each dimension score of MPAI (r=-0.317~-0.219, P<0.01). The indirect effect sizes of maternal care, encouragement of behavioral freedom and denial of psychological autonomy on mobile phone dependence through emotional intelligence were 47.98%, 47.00% and 42.93%, respectively. The indirect effect sizes of paternal care, encouragement of behavioral freedom and denial of psychological autonomy on mobile phone dependence through emotional intelligence were 47.99%, 48.71% and 44.70%, respectively. ConclusionEmotional intelligence partially mediates the relationship between parental rearing behavior and mobile phone dependence.
