1.THE CENTRAL CONNECTIONS OF THE HYPERSTRIATUM VENTRALE PARS CAUDALE NUCLEUS IN CARDUELIS SPINUS——A HRP METHOD STUDY

The efferent and afferent projections of the hyperstriatum ventrale, pars caudal nucleus (HVc) in carduelis spinus were investigated using anterograde and retrograde HRP tracing techniques. After injecting HRP into the HVc, anterogradely labelled fibers and terminals were observed in the robust nucleus of the archistriatum (RA), tractus archistriatum dorsalis (DA) and the area X of lobus parolfactorius. The dense retrogradely labelled cell bodies were found in medial magnocellular nucleus of the anterior neostriatum (mMAN), nucleus interfacialis (NIf) of midneostriatum, telencephalic auditory area--field L of the neostriatum, and locus ceruleus (LoC) of pons. All these projections were ipsilateral. Based on the present and previous studies, the authors got the following understanding: 1. The HVc projects to the high vocal control centre--RA. This proved that the HVc is also a high vocal control nucleus. 2. The HVc receives afferent projections from the telencephalic auditory area--field L. It indicates that there are direct connection between the two high centres of audition and vocal control nuclei in telencephalon. 3. This study demonstrated that the HVc receives afferent projection from the LoC for the first time. According to the fact, we presume that HVc may be involved in the neural regulations of vegetative function and emotional reactions. 4. The HVc also receives afferent projections from the mMAN and NIf of neostriatum, and from the Uva of the thalamus. On the other hand the HVc projects to area X. Since the mMAN, NIf, Uva and area X are vocal learning and memory centres, therefore the HVc may be also involve in vocal learning and memory functions.

Objective To investigate the effects of Atorvastatin calcium on the incidence of macular edema after phacoemulsification in diabetic patients.Methods Forty two eyes of 42 cataract patients with diabetes and hypercholesterolemia who underwent phacoemulsification surgery were divided into interventional group (23 patients) and non-interventional group (19 patients) by random number table methods.The blood glucose and pressure of patients in two groups was controlled strictly before and after surgery.10 mg Atorvastatin calcium per day was delivered one day after cataract surgery for the patients of interventional group and used for 24 weeks.No lipid-lowing agent was provided to the patients of non-interventional group.The main outcome measures included the best corrected visual acuity (BCVA),central retina thickness (CRT),total cholesterol (TC),low density lipoprotein cholesterol (LDL-C) and high density lipoprotein cholesterol (HDL-C).No significant difference was shown in the BCVA,CRT,TC,LDL-C and HDL-C in two groups before phacoemulsification surgery (t=1.251,1.257,1.031,1.042,1.461;P>0.05).At the end of the 24 weeks after surgery,the efficacy evaluation and comparative analysis were performed.The analysis included the BCVA,the incidence of macular edema,CRT,TC,LDL-C and HDL-C.Results The BCVA was no significantly different between two groups one day after surgery (t=1.523,P>0.05).But 4,12,24 weeks after phacoemulsification surgery,the BCVA in interventional group was better than that in non-interventional group(t=3.920,3.012,7.025;P<0.05).24 weeks after the operation,macular edema was occurred in 2 eyes (8.69%) in interventional group and 4 eyes (21.05 %) in non-interventional group.Significance difference was found between two groups (x2 =4.896,P<0.05).There was no significance different of the CRT between two groups one day after operation (t=1.501,P>0.05).Whereas,the significance difference of the CRT was occurred in two groups 4,12,24 weeks after surgery(t=4.673,7.583,9.035;P<0.05).Comparing with that in non-interventional group,the level of TC (t =7.043,7.930,8.611) and LDL-C (t =9.374,9.554,10.856) in interventional group was significantly decreased 4 to 24 weeks after operation (P<0.05).But no significance different of HDL-C was shown in two groups 4,12 and 24 weeks after surgery (t=1.057,1.127,1.295;P> 0.05).Conclusion The treatment of Atorvastatin calcium effectively reduced the incidence rate of macular edema in hypercholesterolemia patients with good glycemic and hypertension control after phacoemulsification surgery.
4.Research on innovative tutor system with discipline crossing to train young teachers

To explore innovative training model for young teachers in university,literature and practical data in university were analyzed.A new concept of research tutor system was put forward in which young teachers and tutors were linked with scientific research project and common interests.Tutors in one subject trained the young teachers from another subject.In this model,a new relationship with equality and free-talk between tutors and students was established and the advantage of talent group effect was ascended.Discipline crossing,multiple communication and interactive learning may be promoted and the number of innovative young teachers and scientific research achievements may increase.
5.Determination of Vancomycin in Human Serum by HPLC

OBJECTIVE: To establish an HPLC method for the determination of the concentration of vancomycin in human serum. METHODS: Serum samples were centrifuged after serum protein was precipitated by 20% metaphosphoric acid. The separation was performed on Hypersil C18 column with mobile phase consisted of acetonitrile-0.012 5 mol?L-1 potassium dihydrogen phosphate buffer (10 ∶ 90) at flow rate of 0.8 mL?min-1. The detection wavelength was set at 236 nm and column temperature was set at 25 ℃. RESULTS: The average relative recovery rate of vancomycin in low, medium and high concentrations (2.5,30.8,150.4 mg?L-1) were 99.8%, 101.1% and 98.9% respectively. The inter-day and intra-day RSD were less than 2.5%(n=5). The limit detection of vancomycin in serum was 2.0 mg?L-1. The linear range was within 2.0~170.0 mg?L-1(r=0.999 8). CONCLUSION: The method is simple, accurate, and rapid for the monitoring and phamacokinetic studies of vancomycin in human serum.
6.Effects of three kinds of calcium activator on the proliferation of cultured vascular smooth muscle cells in rats

AIM: To investigate the effects of intracellular free calcium ([Ca 2+ ]i) from different resources on the proliferation mediated by mitogen activated protein kinase (MAPK) in vascular smooth muscle cells (VSMCs). METHODS: Cultured VSMCs were used in all experiments. Calcium influx was stimulated by angiotension Ⅱ(AngⅡ). The release of intracellular calcium stores was induced by inositol trisphosphate(IP 3) and ryanodine(RY). MAPK activity was measured by [?- 32 P]-ATP incorporation , MAPK protein expression by western blot, VSMCs proliferation by -Leucine(-Leu) and -Thymidine(-TdR) incorporation. RESULTS: Compared to the control VSMCs, AngⅡ, IP 3 and RY significantly increased [Ca 2+ ]i concentration , activity of MAPK and its protein content in VSMCs. Objective To explore the role and correlation of peroxisome proliferator-activated receptor γ (PPAR-γ) and Toll-like receptor 4 (TLR4) on pulmonary vascular remodeling of chronic obstructive pulmonary disease(COPD).Methods Ninety male patients who underwent surgery for squamous cell carcinoma were enrolled as our subjects.All patients were divided into COPD group and uon-COPD group based on lung function,and 45 cases in each group.Peripheral lung tissues without tumor infiltrated after lobectomy were taken to assess the degree of arterial inflammation,percentage of wall thickness to vessel diamater (WT%) and percentage of wall area to total vascular area(WA%) were measured through Hematoxylin-Eosin(HE) staining under light microscope.The expression of PPARγand TLR4 were determined by immunohistochemistry.Results The distribution of WA% and WT% in COPD group were (43.98 ±6.43)% and (27.37 ±3.34)%,higher than that of non-COPD group ((26.09 ± 2.82) %,t =-13.949,P =0.000 ; (15.57 ± 1.75) %,t =-7.140,P =0.000).The expression of PPAR-γand TLR4 in COPD group were (10.74 ± 8.81) %,(3 1.41 ± 14.67) % respectively,and (28.22 ± 15.08)%,(4.67 ± 4.47)% in non-COPD group.The differences were statistic significantly(t =5.483,P =0.000; t =-9.555,P =0.000).And there was negative correlation between the expression of PPARγand TLR4 (r =-0.404,P < 0.01).Conclusion The pulmonary vascular of COPD patients showed the obviously inflammatory cell infiltration,fibrosis and proliferation,and PPAR-γ and TLR4 participate in the regulation of pulmonary vascular remodeling.
8.Role of microenvironment on in vitro differentiation of mesenchymal stem cells into cardiomyogenic cells

Objective To find the cells in myocardium controlling in cardiomyogenic differentiation of marrow derived-mesenchymal stem cells(MSCs).Methods Rat marrow derived-MSCs,cardiomyocytes(CM)and endothelial cells(EC)were isolated and cultured,then MSCs were incubated with the CM or EC or cultured alone after labeled by 5-Bromo-2-deoxyuridine(BrdU)for 72 hours.The cells were identified by morphology,immunocytochemistry and double immunocytochemistry.Results More than 95% of the cells isolated and cultured belonged to MSCs,CM or EC.Incubated with CM,most MSCs labeled by BrdU showed cardiomyocyte-like morphology.After 4 weeks of cultivation,positive staining of double immunocytochemistry of BrdU and Sarcomeric Actin or Connexin-43(CX-43)were observed in 44% cells.However,there was no obvious change in morphology and no positive staining of double immunocytochemistry was recorded in MSCs incubated with EC or cultured alone after 1、2、3 and 4 weeks of cultivation.Conclusion MSCs can differentiate into cardiomyogenic cells in cardiomyocyte microenvironment and cardiomyocytes may play a crucial role in the differentiation.
9.Investigation of Irrational Infusion Prescriptions for Outpatients from 2005 to 2007 in Wuhan Children's Hospital

OBJECTIVE: To investigate the irrational use of the infused drugs in our out-patient clinic.METHODS: In this retrospective study,the infusion prescriptions in our outpatient clinic from 2005 to 2007 were randomly selected for a statistical analysis of the irrational ones based on our knowledge on clinical pharmacology and literature.RESULTS: The problems manifested by the total 81 172 prescriptions were unsuitable solvent,improper dosage regimen,irrational drug combination and repeated drug of medicines etc.CONCLUSION: The use of infused antibiotics in our out-patient clinic were rational on the whole,however,there were some problems which remain to be solved and improved.
10.Clinical features and drug sensitivity of lower respiratory tract infection induced by Streptococcus pneumoniae in children

Objective To explore clinical features and drug sensitivity of Streptococcus pneumoniae (SP) isolated from pediatric patients with lower respiratory tract infection, and to provide evidence for clinical use of antibiotics. Methods A total of 6 358 clinical SP isolates from children with lower respiratory tract infection from January 2008 to December 2012 were col-lected and retrospectively analyzed. The antibiotic sensitivity was done by Kirby-Bauer method and E-test, and all results were in strict accordance with the rules of CLSI. Results The isolated SP strains were mainly from different departments of pediatrics. All clinical cases with SP infection mainly included pneumonia and bronchitis. The resistance rates of 6 358 SP strains to penicil-lin, cefotaxime, erythromycin, clindamycin, cotrimoxazole, tetracycline, chloramphenicol, levolfoxacin, vancomycin were 5.0%, 12.9%, 98.7%, 96.0%, 92.2%, 7.3%, 5.6%, 0.2%and 0.0%respectively, and the resistance rate to penicillin and cefotaxime was signiifcantly different in every years (all P<0.05). The resistance rates of the 318 penicillin-resistant SP strains to the above anti-biotics were 100.0%, 42.6%, 100.0%, 100.0%, 99.2%, 23.6%, 6.8%, 0.6%, 0.0%respectively, and the resistance rate to penicillin and cefotaxime was signiifcantly different (P=0.001). Conclusions The antibiotic resistance rates of SP strains isolated from children with lower respiratory tract infection were higher to erythromycin, clindamycin, cotrimoxazole and tetracycline, and an increasing rate in drug resistance to cefotaxime was observed in recent years. Appropriate antibiotics should be selected for the treatment of infection according to drug sensitivity.
