OBJECTIVE:To study the inhibitory effects of emodin on the proliferation of human hepatocellular carcinoma SMMC7721 cells. METHODS:SMMC7721 cells were treated with 0(negative control),25,37.5,50,62.5,75,87.5,100μmol/L emodin solution and 100 μmol/L 5-FU for 24 h,48 h,72 h. The optical density value of cells was detected,and inhibition rate was calculated. SMMC7721 cells were treated with 0 (negative control),25,50,75 μmol/L emodin solution and 100 μmol/L 5-FU for 48 h,and cell apoptosis rate,cell cycle and the expression of Bax and Bcl-2 gene were detected. RESULTS:Compared with negative control,the rate of cell proliferation inhibition increased after treated with 25,37.5,50,62.5,75,87.5,100 μmol/L emodin and 100 μmol/L 5-FU,which was positively associated with the concentration and duration. Compared with negative con-trol,the rate of cell apoptosis increased after treated with 25,50,75 μmol/L emodin solution and 100 μmol/L 5-FU;the expres-sion of Bax increased and that of Bcl-2 dereased;50,75 μmol/L emodin solution and 100 μmol/L 5-FU could arrested cells at G0/G1 phase(P<0.05 or P<0.01). CONCLUSIONS:Emodin can inhibit the proliferation of SMMC7721,promote cell apoptosis and in-hibit cell growth.

BACKGROUND: In vitro cultured bone marrow mesenchymal stem cells (BMSCs) can differentiate into neural cells. Some inductors are poisonous and cannot be used in human. OBJECTIVE: To induce the differentiation of rat BMSCs into neuron-like cells with astragalus mongholiens. DESIGN, TIME AND SETTING: The randomized control experiment was performed at the Yunnan Key Laboratory of Faculty of Basic Medicine of Dali University and Stem Cell Research Center of College of Preclinical Medicine and Forensic.Medicine of Sichuan University from January 2002 to January 2005. MATERIALS: Healthy SD rats aged 6-weeks old and weighing 120-130 g were used in this study. METHODS: BMSCs were collected from rat bone marrow by density gradient centrifugation, and then cultured. BMSCs were incubated in serum-free L-DMEM medium containing 0.125 volume fraction of astragalus mongholicus to observe morphologic changes in differentiated cells. Expression of the nestin, neuron specific enolase (NSE) and glial fibrillary acidic protein (GFAP) was detected by immunohistochemistry. MAIN OUTCOME MEASURES: Immunohistochemistry of differentiated cells; The expression of Wnt-1 gene and Ngn-1 gene were detected by semi-quantitative reverse transcdption-polymerase chain reaction during cell differentiation. RESULTS: After passage, BMSCs were fibroblast-like, and had high reproductive activity. After cultured with astragalus mongholicus, BMSC morphous changed. Nestin, NSE and GFAP were positive. The expression of Wnt-1 gene and Ngn-1 gene increased after astragalus mongholicus treatment. CONCLUSION: BMSCs can differentiate into neuron-like cells induced by astragalus mongholicus. Wnt-1 gene and Ngn-1 gene play a positive regulatory effect during the differentiation.

OBJECTIVE:To investigate therapy efficacy of large-dose of methylprednisolone plus therapy for acute myelitis, and it effects on nerve function recovery,pyramidal tract conduction function and sensory disturbance. METHODS:A total of 116 patients with acute myelitis were randomly divided into observation group and control group,with 58 cases in each group. Both group received conventional treatment. Control group was additionally given Dexamethasone sodium phosphate injection 0.4 mg/(kg·d)intravenously,once a day;7 d later,they were given Methylprednisolone tablet 60 mg,orally,qd,gradually decreasing to 5 mg/d. Observation group was given Methylprednisolone sodium succinate injection 1000 mg intravenously,once a day;7 d lat-er,they were given Methylprednisolone tablet 60 mg,orally,qd,gradually decreasing to 5 mg/d. The treatment course in eaeh group was 1 month. The nerve function recovery time,pyramidal tract conduction function,sensory disturbance scores,clinical ef-ficacy and ADR were compared between 2 groups. RESULTS:After treatment,the response rate of observation group was signifi-cantly higher than that of control group(93.10% vs. 79.31%);the time of muscle strength improvement≥grade 2,walking down the ground,urination and defecation function recovery in observation group were all significantly shorter than control group,with statistical significance(P<0.05). Before treatment,there was no statistical significance in lower limb cone conduction latency,am-plitude of pyramidal tract conduction,scores of sensation,sense of pain and touch between 2 groups. After treatment,lower limb cone conduction latency of 2 groups were significantly lower than before,and the observation group was significantly lower than the control group,the amplitude of pyramidal tract conduction,scores of sensation,sense of pain and touch in 2 groups were sig-nificantly higher than before,and the observation group was significantly higher than the control group,with statistical significance (P<0.05). There was no significant difference in the incidence of ADR between 2 groups(P>0.05). CONCLUSIONS:Based on conventional treatment,large-dose of methylprednisolone plus therapy shows good efficacy in the treatment of acute myelitis,which can helps to improve pyramidal tract conduction function of patients with acute myelitis,ease sensory disturbance,promote the nerve functional recovery with good safety.

Objective To investigate the clinical significance of peritoneal and retroperitoneal edema in patients with decompensated cirrhosis(DCC). Methods Forty-four patients were identified with DCC on the basis of clinic and laboratory examinations, except the cases with mesenteric,omental and retroperitoneal edema caused by inflammatory and malignant diseases. The diagnosis of edema depended upon hyperdensity in peritoneal and retroperitoneal. The degree of edema was divided into minor, middle, and sever types based on the extent of edema. Ascites, varices, serum albumin(ALB) levels, and hyaluronicacid(HA) levels were also documented. Correlations between the laboratory and CT findings were analyzed. Results The severity of peritoneal edema was correlated with decreasing serum ALB( r s =0.708 8, P 0.05). Conclusion CT findings of the edema in peritoneal and retroperitoneal may indicate the severity of the liver cirrhosis.

The female sheep reproductive tracts were freshly collected from a local meat processing plant and used as experimental materials. Two antibacterial peptides were isolated and characterized from female sheep reproductive tracts by two consecutive chromatographic steps. The peptide isolation procedures included acetic acid extraction, dialyzed, gel filtration chromatography on Sephadex G-50, and reverse phase high-performance liquid chromatography (RP-HPLC). Their molecular mass were 4 820.47 u and 4 012.5 u, respectively, analyzed by MALDI-TOF-MS. The partial N-terminal amino acid sequences of two peptides were determined as AYVLDEPKP and YDSGA, respectively, by Edman degradation. The antimicrobial activity was tested during each purification step by the radial diffusion plate assay and broth microdilution method. These two peptides showed good antimicrobial activities against reference strains of G~+(S. Aureus ATCC2592 and Streptococcu ATCC55121), G~-(E. Coli ATCC25922) and fungi(C. Albicans ATCC2002). The peptides did not show active hemolytic activity against rabbit blood red cells and had no significant effects on human blood coagulation system. The discovery of antibacterial peptides in sheep reproductive system reveals that antibacterial peptides may play a role in innate immunity against microorganisms in a wide range of animal species.

Objective To investigate the the effect of Amiodaroneo on acute myocardial infarction (AMI) and atrial fibrillation (AF) in elderly patients,and provide the basis for the treatment.Methods A total of 108 patients with AMI and AF in our hospital from January 2009 to June 2013 were collected and randomly divided into control and experimental groups,each group of 54 cases.The control group was treated with cedilanid and the experimental group were treated with amiodarone.AF control rate,sinus rhythm maintenance rate and adverse reactions of the two groups were analyzed and compared after treatment.Results The AF control rate of experimental group was significantly higher than that of control group in 6 h,12 h,24 h and 48 h after treatment (x2=5.173,5.066,4.757,6.684,P＜0.05).Sinus rhythm maintenance rate of experimental group was increased compared with control group (47 cases vs.24 cases,x2=21.748,P＜0.01).Adverse reaction rates of the experimental group was lower than the control group (x2 =15.621,P＜0.01).Conclusions The treatment of AMI and AF by amiodarone is betterr and safe than cedilanid alone,so it is worth popularizing and applying on clinic.

OBJECTIVE:To approach the correlations of carbamazepine concentration in serum with clinical therapeutic effect,dosage,sex,age and body weight in epileptic children.METHODS:158 monitoring records for serum concentration of carbamazepine in children with epilepsy in our clinical pharmacy department were consulted retrospectively.Data such as serum concentration,sex and age were recorded into data base and were analyzed using SPSS 13.0 software.RESULTS:No significant correlation was noted between serum concentration of carbamazepine in epileptic children and sex.However,serum concentration of carbamazepine was closely correlated with clinical therapeutic effect,dosage,age and body weight.CONCLUSION:The individual difference in the metabolic processes of carbamazepine was concerned with age and body weight,so it's necessary to make individualized regime according to child's specific condition and monitoring results.

The effects of hypertonic saline solution (HS) on prostacyclin (PGI2) and endothelin (ET) were evaluated.30 patients under epidural anesthesia during elective operations were infused with 7.5% HS (n=20) or 5% dextrose (n=10) 4ml/kg before anesthesia.PGI2, ET and hemodynamic indices were measured before and 10, 60 min after infusion.In HS group, the PGI2 increased about 51.4%, ET decreased 49.6% and the aortic compliance increased 37.5%. However.after dextrose infusion, PGI2 increased about 23.7% at 60 min, ET increased but dispersedly, with increased TPR and decreased CO, SV.Systolic pressure were declined over 20% in four out of ten patients.It demonstrated that HS could stimulate the secretion of PGI2 and depress ET. These hormonal changes revealed an important role in the course of decreasing TPR, with improving hemodynamios.
