1.Clinical observation of the efficacy of No. 1 Baiban granules on vitiligo

Objective To observe the efficacy of No. 1 Baiban granules in the treatment of vitiligo using skin confocal laser scanning microscopy. Methods The study was approved by the hospital ethics committee. Nine-two patients with qi stagnation and blood stasis were collected from Department of Dermatology of the Affiliated Hospital of Tianjin Academy of Traditional Chinese Medicine and were randomly assigned to two groups. The experimental group (n=49) received oral Baiban granuleΙ, 3 g, twice a day, transfer factor capsules 6 mg, 3 times a day, 0.1%mometasone furoate cream (excluding the face) for external use, once a day, or 0.1%tacrolimus ointment (Afacial), twice a day for 3 months. The control group (n=43) only received oral transfer factor capsules 6 mg, 3 times a day, 0.1%mometasone furoate cream (except for the face) for external use, once a day, or 0.1%tacrolimus ointment (face), twice a day, treatment for 3 months. The therapeutic effect was observed in two groups. CLSM was used to monitor the compound color and pigment recovery level, and changes of the number of dendritic cells and inflammatory cell infiltration in skin. Results The compound color was improved significantly after treatment in experimental group and control group. The total effective rate was higher in experimental group than that of control group (81.63% vs. 69.77%,χ2=3.947,P<0.05). Results of CLSM observation showed that the pigment recovery of skin lesion, the proportion of dendritic cells and inflammatory cells were significantly higher in experimental group than those of control group (P < 0.05). Conclusion The treatment effect of No. 1 Baiban granules is more obvious for vitiligo of qi stagnation and blood stasis.
2.Preparation and in Vitro Properties of Paeonol Thermo Sensitive Gel

This study aimed at preparing paeonol thermosensitive gel and preliminary exploring its properties in vitro.Tube inversion method was adopted to investigate the effects of concentrations of poloxamer 407 and poloxamer 188 on gelation temperature.Then,viscosity of the gel was detected by rotary viscometer,and in vitro erosion and drug release characteristics of the gel by no film stripping method.As a result,the gelation temperature of poloxamer 407 decreased with the increase of its concentration,while gelation temperature of poloxamer 407 increased with the accelerating concentration of poloxamer 188.The cumulative drug release of paeonol thermo sensitive gel was up to 70% in 320 rin.Gel dissolution and drug release were simultaneously performed without burst release phenomenon.It was concluded that the preparation process of paeonol thermo sensitive gel was simple and easy to use with the overt effect of sustained-release.
3.Dynamic model of neointima proliferation and remoulding of vascular wall after common carotid artery denudated in rabbits

Objective To study the mechanism of restenosis following percutaneous transluminal coronary angioplasty(PTCA),and to replicate a dynamic model of cell proliferation and remoulding of vascular wall at different time points in rabbits after intimal injury.Methods The model of restenosis in common carotid artery was established by balloon injury in 70 rabbits.The indexes such as lumen area,thickness and area of intima and media,and cross sectional area bounded by the external elastic lamina(EELA) were respectively measured by computer image analysis technology at the 1st,3rd,5th, 7th,14th,28th and 35th day after the injury.Results Endothelial cells were denudated at the 1st day after injury.The proliferation of vascular smooth muscle cell(VSMC) was detected on the surface of lumen at 3 days after injury.At the 7th day after injury,the neointima was formed and continuously thicken.The thickness and area of the neointima as well as extracellular matrix were gradually increased after 14 days,and were maximal after 35 days.The thickness and area of media were also gradually increased during 3~14 days and decreased after 28 days.Compared with non-injured vessel,the medial area was obviously increased at the 14th day.The lumen area was decreased at the 5th~7th day after injury and was obviously less than that of non~injured vessel after 14 days.The EELA was gradually increased at the 1st~7th day after injury,and reached its maximum at the 14th day.The EELA was declined gradually after 28 days.Conclusion The progress of restenosis(RS) can be simulated through the model of restenosis in common carotid artery of rabbit established by balloon injury.The intimal proliferation and vascular remodeling are the leading pathogenesis of restenosis.
4.Rapid detection of chromosomal aneuploidies by multiplex ligation-dependent probe amplification

Objective To test whether multiplex ligation-dependent probe amplification(MLPA)could be used for the prenatal detection of the most common aneuploidies of chromosomes 13,18,21,X,and Y.Methods 34 cases including 22 blood samples(12 with trisomy 21,1 with monosomy X,one male witll extra Y and 8 healthy persons),4 cord blood samples with Down syndrome and 8 amniotic fluid samples ( 1 with trisomy 21 and 7 normal fetuses)were recruited into this study.All samples were confirmed by karvotype analysis. DNA was extracted from blood and amniotic lysate was incubated with proteinase K.MLPA was used to determine the relative copy numbers.Results The resuhs were available within 48 h and were concordant with karyotype analysis in all but one case of amniotic fluid that was suggested to be triploid sample 69,XXY by MLPA or contaminated by maternal blood.This sample actually was found containing a number of red blood cells after centfifugation in test. In total,the concordance rate with clinical characteristics was 97.1%.The Ratio values of 13,18,21,X in normal samples were approaching 1.0 except chromosome Y having slightly higher variation in relative copy number.The difference of ratio means between the normal and trisomy 21 samples was statistically significant by one-way ANOVA(F=298.906.P=0.000).Conclusion Computer assisted MLPA with high sensitivity is a rapid,simple,automatic and reliable method for detection of common chromosomal aneuploidies.

This review systematically introduces the functional connections among cardiovascular centers from spinal cord to cortex, and the mechanisms underlying pressor or depressor response of these cardiovascular centers, including the pathways, transmitters and receptors involved. The pressor or depressor response of these cardiovascular centers is mainly mediated by RVLM-sympathetic vasoconstrictor nerve system.

6.An Epidemiological Survey of Visceral Leishmaniasis in Hamangou Coal Mine Area of Korla City, Xinjiang

Objective To investigate the epidemiological status of visceral leishmaniasis in Hamangou coal mine area of Korla City of Xinjiang Uygur Autonomous Region.Methods Based on a hint of possible existence of patients, a retrospective survey was carried out house by house to find cases with suspected signs/symptoms of the disease.Meanwhile, a survey on current status was conducted, including physical examination(liver and spleen palpation) to those less than 15 years-old, leishmanin skin test and rk39 immunochromatographic strip test for part of the residents.Bone marrow smears were examined for the cases with clinical signs/symptoms and positive rk39 strip test.Sandflies were collected using routine methods in and around the area, identified, and dissected to find infection of promastigotes.Results Leishmanin skin test was performed in 185 people with a positive rate of 21.1%(39/185), 39 out of 140 local residents who have lived there for more than 6 years showed positive(27.9%) , while all residents who have lived less than 6 years and children under 5 years old were negative.Of the 81 children under 15 years old with a negative skin test, one showed positive for rk39 strip test, and leishmania body was found in the bone marrow smear of this case, so confirmed as visceral leishmaniasis.12 sandfies were identified as Phlebotomus alexandri, and natural infection with promastigotes was found in one sandly.Conclusion The investigation confirms that visceral leishmaniasis is endemic in the Hamangou coal mine area.
7.Molecular typing of Brucella abortus strains in Xinjiang

Objective To identify molecular typing of Brucella abortus isolates in Xinjiang,and determine the identification ability of multiple locus variable-number tandem repeat analysis (MLVA).Methods The optimized Brucella AMOS-PCR was used for identification of Brucella (n =7) genus and species in Xinjiang from 2010-2015,and MLVA-16 was used to further identify the isolates.Results were compared with the data of the Brucella standard strain provided by the http://mlva.u-psud.fr database.Cluster analysis was carried out with Bionumerics 6.6.Results The results of AMOS-PCR and MLVA-16 were identical,all were Brucella abortus.Further classification results of the MLVA-16 showed that the strain in Xinjiang was type 3 of Brucella abortus,which was basically the same as that of the domestic Brucella.Conclusions The molecular typing of isolates separated in Xinjiang is type 3 of Brucella abortus.MLVA can identify Brucella at the level of species,and highly sensitive to Brucella biotype and isolates differences,which provides a basis for the traceability and evolution of brucellosis epidemic strains.
8.Correlation between the vulnerability of carotid plaque and ischemic stroke

Objective To analysis the correlation between the vulnerability of carotid plaque and ischemic stroke and to explore the clinical significance of assessing vulnerable plaques accurately.Methods A total of 64 patients,who underwent carotid endarterectomy (CEA) in First Affiliated Hospital of Soochow University from October 2014 to February 2016,were classified into ischemic and non-ischemic stroke group according to whether ischemic stroke symptoms occurred during the last 6 months before surgery.To judge the stable and vulnerable plaques,the patients underwent carotid doppler ultrasonography (CDU) before surgery,and the carotid plaques were evaluated with HE staining after CEA.We also analyzed the risk factors of plaque formation and the relevance between the plaque characteristic and ischemic stroke with chi-square test.Kappa test were used to analyze the consistency of CDU and pathology.Results Of the 44 cases in ischemic stroke group,CDU identified 81.8% (36/44) vulnerable plaques and 18.2% (8/44) stable plaques,while pathology confirmed 86.4% (38/44) vulnerable plaques and 13.6% (6/44) stable plaques.Of the 20 cases in non-ischemic stroke group,CDU identified 35% (7/20) vulnerable plaques and 65% (13/20) stable plaques,while pathology confirmed 40% (8/20) vulnerable plaques and 60% (12/20) stable plaques.In both CDU and pathology,vulnerable plaques in ischemic stroke group were obviously higher than that in non-ischemia group (x2=13.67,P < 0.001;x2=14.62,P < 0.001).The results of CDU agreed well to that of pathology results (Kappa=0.669,P < 0.01).Conclusions The vulnerability of plaques were closely related to the ischemic stroke.CDU is a reliable examination method to identify vulnerable plaques.The accurate assessment of vulnerability of plaques has a great significance in preventing ischemic stroke.
9.Analysis of drug sensitivity test results of Brucella from bovine and sheep in Xinjiang

Objective To explore the drug sensibility of Brucella from bovine and sheep in Xinjiang.Methods Using paper diffusion method,19 drugs of 8 kinds of antibiotics including aminoglycosides,macrolides,sulfonamides,tetracyclines,β-lactams,fluoroquinolones,chloramphenicols and rifamycins,were tested.Drug sensitivity test was conducted on 57 Brucella strains isolated from bovine and sheep in Xinjiang from 2010 to 2016.Results The 57 Brucella strains were highly sensitive to doxycycline,tetracycline,streptomycin,tobramycin,gentamicin,amikacin,amoxicillin,ofloxacin,fleroxacin,ciprofloxacin and chloramphenicol,with the sensitivity rates were all higher than 90%;and they were highly resistance to azithromycin,clarithromycin and bactrim,with the drug resistance rates were all higher than 80%.Conclusion Brucella from bovine and sheep in Xinjiang is sensitive to tetracyclines,aminoglycosides,β-1actams,fluoroquinolones and chloramphenicols.
10.Antiapoptotic Mechanism of Insulin in Reoxygenation-induced Injury in Cultured Cardiomyocytes of Neonatal Rats

To examine the protective effect of insulin on reoxygenation-induced injury and explore the underlying mechanisms, the model of anoxia/reoxygenation (A/R) injury was established by inducing anoxia for 2 h and reoxygenation for 4 h in cultured cardiomyocytes of neonatal rats. The rats were randomized to four groups receiving vehicle, insulin, LY294002, insulin plus LY294002at the onset of reoxygenation after 2 h of anoxia. At the end of reoxygenation of 4 h, activity of lactate dehydrogenase (LDH) and content of malondialdehyde (MDA) were spectrophotometrically determined, apoptosis of cardiomyocytes were detected by using TUNEL and DNA Ladder, and Western blotting was employed to examine the expression of phosphorylated Akt in all groups. Our results showed that compared with vehicle-treated group, activities of LDH, contents of MDA, apoptosis index (AI) were significantly decreased, and expression of phosphorylated Akt was in creased significantly in insulin-treated group. However, changes in LDH, MDA, AI and phospho rylated Akt resulting from insulin were attenuated or abolished by LY294002 (PI3K inhibitor).These data strongly suggest that early administration of insulin at reoxygenation protects cardiomyocytes from reoxygenation-induced apoptosis through PI3K/Akt signaling pathway.
