1.Osteogenetic ability of mouse spermatogonial stem cells cultured in vitro

BACKGROUND:Spermatogonial stem cells are a kind of adult stem cells, which have self-renewal and differentiation potential, and can be differentiated into specific cells in vitro, suggesting that the spermatogonial stem cells may be possibly differentiated into osteoblasts. But the related research has not been reported. OBJECTIVE:To observe the biological characterization and osteogenic process of mouse spermatogonial stem cells cultured in vitro. METHODS:Spermatogonial stem cells were obtained from the testicle of mice aged 15-20 days, and were cultured on the feeder layer from bone marrow stroma cells in vitro. When cultured for 3 days, the cells were cultured in the conditioned medium (experimental group) and basic medium (control group). The cells proliferation capability and osteogenic property were examined by phase-contrast microscope, alkaline phosphatase activity and type I col agen immunofluorescence staining. RESULTS AND CONCLUSION:Spermatogonial stem cells proliferated faster in the experiment group than in the control group. cells grew rapidly in colony-like shape in the conditioned medium at 3-6 days, the three-dimensional feeling enhanced, cellmass and clusters continued to increase in size, the extracellular matrix was increased in number and the cytoplasmic bridge was not obvious. After culture for 15 days, cells in the two groups were positive for alkaline phosphatase staining that the cytoplasmic membrane was dyed black. Under the fluorescent microscope, green fluorescence was visible in the experimental group, suggesting the cells in the experimental group was positive for type I col agen, but negative in the control group, which is similar with the biological characteristics of osteoblasts. These findings indicate that spermatogonial stem cells possess the osteogenic capability under induction conditions, which are expected to provide seed cells for bone tissue engineering.
2.Value of MRI in qualitative and quantitative diagnosis of bone marrow infiltration in lymphoma

Objective To evaluate the value of MRI in qualitative and quantitative diagnosis of bone marrow involvement in patients with lymphoma. Methods 28 diagnosed lymphoma with bone marrow infiltration and 31 healthy spines as controls were included.MRI performance and the signal intensity ratio on T1WI (SIR1) of the spine bone marrow and cerebrospinal fluid were analyzed and statistical analyses were performed.Results Qualitative diagnosis among these patients,was shown than MRI results indicated that 27 cases had abnormal signal. The sensitivity of MRI was 96.4 %. There were four main patterns of marrow infiltration in MRI,containing nodular pattern 21.4 % (6/28),scattered pattern 53.6 % (15/28),mottled pattern 14.3 % (4/28) and uniform pattern 7.1% (2/28) respectively.By quantitative diagnosis the SIR1 of study group (1.251±0.253) was apparently lower than that in the control group (2.625±0.434) with statistical significance (t =15.022,P < 0.001).The results of multiple comparisons showed that the SIR1 of mild degree (1.390±0.172),moderate degree (0.982±0.790) and severe degree (0.908±0.122) patients with lymphoma compared with the normal controls had significant differences (all P =0.000),and difference between mild and moderate degree had statistical significance (LSD,P =0.012),so did the difference between mild and severe degree (LSD,P =0.025).However,no significant difference could be seen between moderate and severe degree (LSD, P =0.757). Positive linear correlation was existed between the persentages of SIR1 and tumor cells in bone marrow (r =-0.765, P < 0.001). Conclusion As a noninvasive and direct-viewing technique, MRI presents a global view of bone marrow with high sensitivity in detecting bone marrow involvement in lymphoma. To a certain extent, it could be possible to estimate the infiltration degree and evaluation of the tumor burden in bone marrow by quantitative measurement of MRI.
3.Elevated radial arterial augmentation index in hypertensive patients with diastolic dysfunction

Objective To investigate the correlation between augmentation index (AI) of the radial artery and diastolic heart function in patients with hypertension.Methods Echocardiographs were obtained for 305 patients with hypertension.AI,pulse wave velocity (PWV) of peripheral arteries and serum pro-brain natriuretic peptide (proBNP) levels were determined.Correlations and receiver operating characteristic (ROC) curves were drawn between AI values and impaired diastolic function.Results AI levels were significantly increased in patients with impaired diastolic function diagnosed by ultrasound.Assessment of diastolic heart function based on proBNP levels revealed that AI and aortic pulse wave velocity were significantly elevated in patients with impaired diastolic function.The operating curve indicated that AI may be a more accurate and efficient index for the evaluation of impaired diastolic function compared to pWV.Correlation analysis also showed that proBNP levels had altered in parallel with changes in AI and PWV.After adjusting for various factors including age,gender,blood pressure and blood lipid,a positive correlation was observed between proBNP and AI with a correlation coefficient of 0.3697 (P =0.003).However,no correlation between proBNP and aortic PWV was seen after adjustment.Conclusions Changes in radial AI levels may reflect parallel changes in diastolic cardiac function in patients with hypertension,suggesting that AI may be utilized as a non-invasive clinical indicator of diastolic heart function.
4.Advanced orbicularis oculi muscle flap for eyelid defect.

OBJECTIVETo investigate the application and therapeutic effect of advanced orbicularis oculi muscle (OOM ) flap for eyelid defect.
METHODSUni-pedicle or bi-pedicle advanced OOM flaps were designed according to the location, depth and size of the eyelid defects. The resulted wounds in the donor sites were closed directly. The flap size ranged from 1.5 cm x 0.5 cm - 6.0 cm x 3.5 cm.
RESULTS120 cases were treated. All the flaps survived except for 3 flaps with epidermis necrosis at the end of flaps, which healed after dressing. The patients were followed up for 3 -36 months with inconspicious scar in donor sites. The flap color, texture had a good match with surrounding skin.
CONCLUSIONSThe OOM flap is ideal for eyelid defect with reliable blood supply, satisfied color and texture. The wound at donor site can be closed directly with less morbidity.

5.Imaging features of primary mediastinal cysts with clinical and histopathologic correlation

Objective To characterize the imaging features of mediastinal cysts and gain a better understanding of atypical manifestations of various mediastinal cysts and improve the level of diagnosis. Methods The CT and/or MR images(CT n=28, MRI n=26, CT and MRI n=10), surgical information, and pathologic material in 44 histopathologically proved cases of mediastinal cyst were retrospectively reviewed. Results The mediastinal cysts were located in the anterior mediastinum(n=13), middle mediastinum(n=18), anterior-middle mediastinum (n=2) and posterior mediastinum(n=11). Bronchogenic cysts and pericardial cysts were atypically located in 7/20(35%)and 5/6 respectively. The density of 42.9% cysts on CT was close to that of water. Each of the cysts had a high signal intensity that was equal to or greater than cerebrospinal fluid on T_2-weighted MR images. One cyst showed marked loss of signal on MR hydrography. The signal intensities of the cysts were variable on T_1-weighted images. The signal intensity of MRI was not homogeneous in 5 and the reasons were different. Fourteen cases were misdiagnosed. Conclusion 1.The cysts with soft-tissue attenuation on CT in the anterior and posterior mediastinum may be easily misdiagnosed as neoplasm. Pericardial cysts located in the paratrachea may be easily misdiagnosed as bronchogenic cysts .2. MRI can be used to diagnose the cysts with high density on CT.3. Heterogeneous signal on MRI and loss of signal on MR hydrography might be the reasons for misdiagnosis.
6.Blood glucose and glycosylated hemoglobin levels of type 2 diabetes at different altitudes and different races in Yunnan Province

Objective To investigate the effects of altitude,race,gender and age on blood glucose and glycosylated hemoglobin(HbA1 c) of type 2 diabetes.Methods A total of 410 patients with type 2 diabetes (T2DM) who resided at different altitudes in Jinghong,Kunming and Lijiang city for over 5 years and received antidiabetic treatment without adjustments for over 3 months were enrolled.To compare blood glucose,hemoglobin (Hb) and HbA1 c levels of T2DM among different altitudes,different races,different genders and different ages,The correlation between races and HbA1c was evaluated with Pearson's correlation analysis.Results No significant differences were observed in fasting blood glucose (FBG) (P =0.49,P =0.1,P =0.09,P =0.3),2 h postprandial blood glucose (2 h PBG) (P =0.33,P =0.4,P =0.7,P =0.1),and HbA1 c (P =0.47,P =0.28,P =0.2,P =0.07) levels between patients at different altitudes,different races,different genders and different ages.However,there were significant differences in Hb (P <0.01).There was no significant correlation between races and HbA1c levels(r =0.003,P =0.6).Conclusions There were effects of altitude,race,gender and age on hemoglobin of type 2 diabetes,but no effects on glycosylated hemoglobin.
7.Antitumor effect of enhancer binding protein C/EBPα on the leukemic BALB/c nude mice

Objective To explore the role of tumor inhibition of enhancer binding protein C/EBPα in the leukemic mice. Methods BALB/c nude mice were randomly divided into three groups. Three kinds of cells including pEGFP-C/EBPα-K562 cells, pEGFP-K562 cells and K562 cells as the control were injected into mice separately through the subcutaneous and tail vein, and subcutaneous tumors and leukemic models were formed. The changes of tumors were observed and the apoptosis of cells was detected by TUNEL; The capacity of proliferation of leukemia cells was observed in the bone marrow and the peripheral blood by Wright-Giemsa staining. The expression of genes of related to proliferation was detected by RT-PCR. Results The quality and the max diameter of tumors in the pEGFP-C/EBPα-K562 group were smaller than that of pEGFP-K562 group and K562 control group [(2.4±0.1) g vs (5.1±0.3) g and (5.7±0.4) g, both P <0.05; (11+2)mm vs (19+3) mm and (23+3) mm, both P <0.05]. More apoptosis cells were found in the pEGFP-C/EBPα-K562 group leukemic cells were found in the peripheral blood of leukemic models, and the proliferation of leukemic cells in the pEGFP-C/EBPo-K562 group were lower than that of other groups, accompany by the conspicuous cell differentiation. p53 was significantly elevated by RT-PCR, while down-regulated of c-myc.Conclusion Enhancer binding protein C/EBPα promote the apoptosis of cells and inhibit the proliferation of leukemia cells in leukemia mice, and further induce the cell differentiation. The inhibition of enhancer binding protein C/EBPα in the leukemia may have effect through the regulation of related genes.
8.Effects of disc repositioning and condyle restoration in the treatment of type Ⅲ traumatogenic temporomandibular joint ankylosis

Objective To investigate the potential role of disc repositioning and condyle restoration in the treatment of type Ⅲ traumatogenic temporomandibular joint (TMJ) ankylosis. Methods Eight patients including four females and four males at age range of 7-22 years (mean 13.6 years) were enrolled in this study. The patients suffered from traumatogenic TMJ ankylosis for 1-12 years. The preoperative interincisal opening distances ranged from 2 mm to 10 mm. During surgery, the traumatogenic callus of the lateral condyle process was removed, the condyle process was formed, and then the dislocated disc was sutured to the articular capsule or soft tissues around. Results All patients were followed up for 6-38 months and the last follow-up examination showed that the average interincisal opening distance was 30 mm. No recurrence or TMJ symptoms were found during the period of follow-up. Conclusions Disc repositioning and condyle restoration has the advantages of simple procedures, minor trauma and little recurrence and proves to be a feasible and effective method for the treatment of type Ⅲ traumatogenic TMJ ankylosis.
9.Construction of Recombinant Pichia pastoris Expressing Human SOD

Objective To optimize the expression of active human Cu,Zn-superoxide dismutase (SOD) in recombinant Pichia pastoris (P.pastoris).Methods A recombinant human SOD expression construct was generated and expressed in P.pastoris.The culture time,concentration of copper ions in the medium,and conditions for methanol induction were optimized,and SOD was preliminarily purified.Results Five P.pastoris recombinants that stably expressed human SOD were obtained.SOD could be purified from the culture supernarants of the recombinant strains by chromatography.The amount of SOD secreted into the supernatant was high with optimal activity,when the incubation time was 48 h,copper ion concentration in the medium was 0.5%,and methanol concentration was 1%.Conclusion An effective method for the expression of soluble human Cu,Zn-SOD in P.pastoris was established,and the enzyme produced using this method exhibited high activity.This method provides a foundation for further research on the production of human SOD by fermentation.
10.Bibliometric analysis of vertigo-related histopathology,vestibular rehabilitation,physical rehabilitation,and Chinese and Western medical treatments

BACKGROUND:Vertigo is closely related to clinical neurological disorders.When neurons are damaged or dead,it may lead to abnormalities in the vestibular system and trigger vertigo symptoms.Therefore,it is necessary to explore and analyze the hotspots related to vertigo that are common in clinical neurology. OBJECTIVE:To analyze the vertigo-related histopathological changes in clinical neurology and the research hotspots worldwide using bibliometric methods. METHODS:The WanFang database and Web of Science core set database were searched by the first author to retrieve the research-related literature published from 2014-2023 on the treatment of common vertigo in clinical neurology.A bibliometric analysis of the number of publications,country/region,institution,keywords,co-cited literature,and highly cited literature was peformed using VOSviewer_1.6.19 software to summarize the research hotspots in this research field. RESULTS AND CONCLUSION:Web of Science core set database had the highest number of 174 publications in this field in 2022,and WanFang database had the highest number of 133 publications in this field in 2020.The top 3 countries with the highest number of publications are the United States,Germany,and China.The University of Munich,Germany is the international institution with the highest number of publications in this field,while Chengdu University of Traditional Chinese Medicine is the Chinese institution with the highest number of publications in this field.The results of keyword analysis showed that the research hotspot diseases in this field in China are mainly Meniere's disease,cervical vertigo,senile vertigo,benign paroxysmal positional vertigo,isolated vertigo,and hypertensive vertigo,and the treatments include acupuncture,rehabilitation,medication(gastrodin,Banxia Baizhu Tianma Tang),and manipulative reduction.International research hotspot diseases in this field mainly include benign paroxysmal positional vertigo,vestibular disorders in new coronavirus cases,Meniere's disease,vestibular schwannoma,acoustic neuromas,and vestibular migraines,etc.,and the hotspot treatments are antivertiginous medications,antidepressant and anxiolytic treatments,and microsurgery.The results of literature co-citation analysis showed that for acute vestibular syndrome with persistent vertigo as the main symptom,three-step bedside ophthalmoscopy(HINTS:Head-Impact-Nystagmus-Strabismus Test)is more sensitive than early MRI in the diagnosis of combined strokes in patients with acute vestibular syndrome,which is the most peer-recognized method of detecting strokes in vestibular syndrome,whereas hormonal therapy is more effective to treat vestibular neuritis patients with paroxysmal vertigo as the main symptom.The results of highly cited literature analysis showed that,in the hot literature included in WanFang database in the past 10 years,acupuncture at Fengchi point and the acupuncture method of inducing resuscitation to improve posterior circulation ischemic vertigo have achieved certain results.The literature published in the past 3 years has indicated that Ginkgo biloba leaf extract+gastrodin,acupuncture+Banxia Baizhu Tang,betahistine+gastrodin,vestibular rehabilitation training+Epley Maneuver,all can improve the vertigo symptoms to different degrees.While there were no featured anti-vertigo drugs indicated in the literature in the Web of Science core set data in the recent 10 years,and most of them are based on traditional anti-vertigo drugs and microsurgery.However,there are a few case reports in the international literature in the last 3 years that found that COVID-19 infection may lead to vestibular neuritis and vertigo symptoms.The onset and progression of vertigo may be closely related to neuronal damage and regeneration.For example,viral infections,inflammatory stimuli,or other pathologic factors may lead to neuronal damage or death,thereby affecting the function of the vestibular system.Vertigo-related diagnosis and treatment standardization guidelines have been published both domestically and internationally.Currently,international guidelines recommend the combination of vestibular rehabilitation and physical rehabilitation for the treatment of vertigo,and Chinese guidelines recommend the combination of Chinese and Western medicine,reduction and acupuncture.However,the level of evidence is not very high,so a large number of large-sample,multicenter randomized controlled trials on anti-vertigo treatment are needed in the future.
