1.Effects of hydroxyethyl starch on leukocyte activation and vascular permeability in endotoxic rats

Objective To investigate the effects of hydroxyethyl starch(HES 130/0.4)on leukocyte activation,mast cell degranulation and vascular permeability in rat mesentery during the early phase of endotoxemia.Methods Thirty-six male Wistar rats weishing 200-250 g were randomly divided into 3 groups(n=12 each):Ⅰ normal control group received only normal saline(NS);Ⅱ LPS group and Ⅲ HES group received LPS 2 mg/kg iv followed by iv HES infusion at 16 ml·kg-1·h-1 for 60 min(HES group)or equal volume of NS(LPS group).The animals were anesthetized with intramuscular 20% urethane 1 ml/100 g.The abdomen was opened and the mesentery of small intestine was pulled out and placed on transparent observation plate at constant temperature and moisture and examined under microscope for microcirculatory changes.Leukocytes rolling along,adhering to and emigrating from mesenteric venules,mast cell degranulation and FTTC-albumin effux from venules were examined before(baseline)and during the 90 min after LPS administration.The expression of CD11b and CD18 on the leukocytes was determined using flow cylometry.Results The number of leukocytes rolling along,adhering to and emigrating from venules and degranulated mast cells were signitlcanfly increased in LPS group as compared with control group.These LPS-induced changes were significantly inhibited in HES group.The albumin effiux Was enhanced and CD11b/CD18 expression upregulated in both LPS and HES groups.Conclusion Hydroxyethyl starch(130/0.4)can ameliorate the increased vascular permeability and microcirculatory disturbance in rat mesentery during the early phase of endotoxemia.
2.HAN Xinmin's Experience in Treating Tic Disorders from View of Wind-phlegm

Objective]The paper summarizes the academic thoughts and clinical experience of Professor HAN Xinmin for treating Tic Disorders from view of wind-phlegm. [Methods] By learning from Professor HAN Xinmin, recording the relevant cases and analyzing typical cases, to sum up the knowledge of etiology and pathogenesis, treatment characteristics, as well as list one proved case. [Results]Professor HAN Xinmin holds that this disease origins from liver, involving lung, spleen and kidney. The key pathogenesis is the internal stir of wind and phlegm. Treatment follows the principles of dispelling pathogenic wind and eliminating phlegm, along with suppressing hyperactivity liver qi, dispersing lung qi, regulating spleen and tonifying kidney. The self-ordained Chinese herbal medicine prescription has achieved a satisfactory clinical curative effect. [Conclusion]Therapies of Professor HAN Xinmin on treating Tic Disorders have significant effect, which have the value of popularization and application.

4.Effects of parenting style and behavior on children with allergic asthma

Objective To explore the effect of parenting style and behavior on children with allergic asthma.Methods From July to December in 2016,a total of 60 children with allergic asthma,who were from Jiangsu Province Hospital of Traditional Chinese Medicine and Jiangsu Research Institute of Traditional Chinese Medicine,were selected as the asthma group.Another 60 normal children from kindergarten and primary school were selected as the healthy group.Achenbach child behavior checklist (CBCL) and parenting style questionnaire were used to evaluate the behavior and parenting style of the two groups.The Fractional exhaled nitric oxide (FeNO) of the two groups were also collected.Results (1) The scores of dotage,autocracy,inconsistence of parenting style in asthma group ((14.95±4.95),(22.23±4.69),(13.43±3.80))were higher than those in healthy group((11.68±3.69),(16.47±4.16),(11.07±3.32))and the differences were significant (all P<0.05).The scores of democracy,indulgence in asthma group ((38.92 ± 5.37),(18.12± 4.54)) were lower than those in healthy group((41.35±3.62),(19.88±3.41)),and the differences were statistically significant(all P<0.05).(2) The scores of attention,socialization,aggression and withdrawal of child behavior in asthma group ((7.35±3.55),(3.85±2.17),(11.57±5.30),(3.40±2.65))were higher than those in healthy group((3.38±2.81),(2.52± 1.52),(6.98±4.77),(2.02± 1.63)),the differences were statistically significant(all P<0.01).There was no significant difference between the two groups in the dimension of discipline(P>0.05).(3) The FeNO index of asthma group((32.27± 14.27) × 10-9 mol/L) was higher than that of healthy group ((11.10± 1.88) × 10-9 mol/L) and the differences were statistically significant (P<0.01).(4)Logistic regression analysis showed that the attention(B=0.38,OR=1.47,95% CI =1.27-1.70,P<0.01),dotage(B=0.19,OR=1.20,95% CI =1.06-1.37,P<0.01) and autocracy(B=0.24,OR=1.27,95% CI =1.14-1.42,P<0.01)had significant effects on allergic asthma.Conclusion Attention,dotage and autocracy are the risk factors to influence the control level of allergic asthma.This should arouse the attention of clinicians and parents.
5.Comparison of the behavior characteristics in SHR, WKY and SD rat models of attention deficit/hyperactivity disorder

Objective To compare the differences in behavior characteristics among SHR, WKY and SD rat models of attention deficit/hyperactivity disorder (ADHD), and explore an ideal control model of SHR rats.Methods Using open field test to analyze the rat movement distance, speed, wearing numbers and the number of grooming to evaluate the spontaneous movement in SHR, WKY and SD rats.Using the Morris water maze to test the learning and memory ability among the three rat groups.Results The result of open field test showed that the SHR rats had significantly increased (P< 0.01) total amount of exercise, average speed and wearing numbers than WKY and SD rats.Compared with the WKY rats, SD rats had a significantly higher movement distance (P< 0.01), slightly higher movement speed and wearing number (P< 0.05).In the Morris water maze hidden platform period test, the SD rats had a significantly longer latency than the SHR rats (P< 0.05).SD rats showed longer latency distance on the first, third and fourth days of training, as compared with the SHR rats (P< 0.05 or P< 0.01).Compared with the WKY group, SD rats showed a shorter latency distance in each training time (P< 0.05 or P< 0.01).In the probe trial period, the SD rats showed shorter time and distance ratio to the target quadrant than SHR rats (P< 0.05), while significantly longer than the WKY rats (P< 0.05 or P< 0.01).Conclusions There are significant behavioral differences between SHR and WKY rats, showing certain disadvantages in comparison of the two types of rats.To add SD rats as a control group for SHR rats can improve the comparability of behavior characteristics of SHR rats, and to get more objective evaluation of the behavior characteristics of SHR rats.
6.Prevention and treatment of severe complications of thoracoscopy

Objective Study the prevention and early diagnosis of severe complications of thoracoscopy. Methods 400 patients,(18~81)years,ASA Ⅰ-Ⅲ,underwent thoracoscoy,including 320 pulmonary bubble resections and 80 thorax,mediastinum or lung tumor biopsy or resections.All patients were anesthetized with one-lung ventilation using a double-lumen tube and monitored continuously in circulatory and respiratory functions with a multi-runctional monitor. Results Severe intraoperative complications occurred in 8 patients,including 5 reexpansion pulmonary edema,1 complete occlusion of double-lumen tube due to massive pleural effusion,1 massive bleeding and 1 cardiac arrest due to bilateral pneumothorax.All complications were successfully treated and the patients were cured and discharged. Conclusions Thoracoscopy must be well prepared.Applying one lung anesthesia with double-lumen tube,careful observation during operation,early recognition and treatment of complications may contribute to assuring the safety of this procedure.
7.Prevention and treatment for complications of cardiac myxoma excision in 215 cases

Objective To summarize the prevention and treatment experience of complications of cardiac myxoma excision.Methods Cardiac myxoma excision were performed in 215 cases under general anesthesia,cardiopulmonary bypass and cardiac arrest condition.Results Two cases died from severe low cardiac output syndrome which eventually caused multiple organ failure.The remaining 213 cases were cured and discharged.Among 213 cases,low cardiac output syndrome occurred in 35 cases,re-open chest for bleeding was performed in 8 cases,30 cases in arrhythmia (including paroxysmal supraventricular tachycardia in 20 cases and ventricular premature in 10 cases),no vital organs embolism occurred.Follow-up was done to 200 patients for 1-8 years with a follow-up rate of 93.9% (200/213).In 200 cases,1 case died from lung cancer,1 case recurred and was cured after re-operation.The remaining patients had no obvious abnormalities.Conclusion As the particularity of surgical treatment of cardiac myxoma,it is essential to take prevention measures to reduce complications of cardiac myxoma excision in perioperative periods.
8.Biliometric Analysis on TCM Therapy for ADHD in Recent Ten Years

Objective To discuss the research status of TCM therapy for ADHD in China in recent ten years; To provide references for clinical workers. Methods Clinical research papers about TCM therapy for ADHD in CNKI, VIP, and CBM from 1st January 2005 to 1st April 2015 were retrieved. Metrology analysis was conducted from the aspects of publication time, diagnostic criteria, contrasted medicine, and types and features of TCM therapy. Results By eliminating duplication literature, 670 papers were screened out. After the screening of exclusion criteria, 148 papers were included. DSM-Ⅳ and CCMD-3 were the most commonly used diagnostic criteria in the 148 papers; internal therapy was the main therapy in TCM therapy; decoction was the main dosage form; liver-kidney yin deficiency syndrome and heart-liver fire syndrome were the main patterns of syndrome; Acori Tatarinowii Rhizoma, Rehmanniae Radix Praeparata and other 19 Chinese medicinal herbs were the main Chinese mateia medica; Xiaoer Zhili Syrup was the most commonly used Chinese patent medicine; acupuncture and moxibustion and auricular point therapy were the main external therapy, and external combined with internal therapy had the best efficacy. Conclusion TCM therapy for ADHD is with high effectiveness. External combined with internal therapy has the best efficacy.
9.An optimized extraction protocol of prefrontal cortical and striatal synaptosomes from SHR rat

Objective To introduce an improved extraction method of prefrontal cortical and striatal synaptosomes from SHR rat. Methods Synaptosomes were prepared from SHR rat brain tissue by Percoll density gradient centrifugation.Transmission electron microscopy was used to assess the morphology and structural integrity of the synaptosomes.Results The obtained synaptosomes showed oval structures surrounded by an intact membrane.Presynaptic components contained one or more mitochondria and a large number of synaptic vesicles.The synaptic clefts were clearly visible, and prominent part of the characteristic compact structure was clear, complete and with higher electron-density. The synaptosome presynaptic membrane, synaptic cleft, and postsynaptic membrane were well preserved, and the synaptosomes were densely distributed, showing typical morphological characteristics of synaptosomes.Conclusions The results of our study improved the traditional preparation method and provide a less time-consuming, highly productive protocol for preparation of structurally typical and intact synaptosomes, suitable for further research on neuroscience and neurological diseases.
10.Glycosynthases: a Novel Efficient Synthetic Tool for Oligosaccharides

Oligosaccharides are one of the essential physiological constituents of glycoproteins and glycolipids on mammalian cell surfaces and microbial metabolites. They have considerable potential as therapeutics but are only now slowly assuming this important role. One of the reasons for their slow development has been the considerable difficulty in synthesizing oligosaccharides on the scale necessary for their clinical evaluation. Classical chemical and enzymatic methods both have limitations in synthesizing large-scale oligosaccharides. In recent years, the rapid progress on molecular biotechnology has promoted the development of retaining glycosidases in oligosaccharides synthesis, which led to the production of a novel class of enzymatic activities termed the glycosynthases. These new enzymes are retaining glycosidase mutants in which the catalytic nucleophile has been converted to a non-nucleophilic residue,synthesizing oligosaccharides in high yields ( the highest yields reach 99%) without any hydrolysis. Furthermore thioglycoligases and thioglycosynthases have been developed subsequently in the past three years. Glycosynthases can be screened in high-throughput assay by the two-plasmid system and the yeast three-hybid system respectively. Their activity can be significantly enhanced by substituting alternative residues for nucleophile, additional random mutations and optimizing reaction conditions. Their regioselectivity can be modified through changes in receptors.
