1.Role of serum leptin and tumor necrosis factor-? in malnutrition of male chronic obstructive pulmonary disease patients

Objective: To explore the function of serum leptin in COPD patients with malnutrition, and to investigate the relationship between leptin and TNF-?. Methods: A total of 81 subjects (47 COPD patients and 34 control subjects) participated in this study. The 47 COPD patients were divided into 2 groups: group COPD Ⅰ (patients without malnutrition during stable disease, n=29), group COPDⅡ(patients with malnutrition during stable disease, n=18).To eliminate the effect of sex differences, all the patients and controls were male. Body mass index (BMI), percent ideal body weight (IBW%), triceps skin-fold thickness (TSF), mid-upper arm circumference (MAC), mid-upper arm muscle circumference(MAMC),serum leptin and tumor necrosis factor-?(TNF-?) levels, serum prealbumin (PA), serum transferrin (TF), serum albumin(Alb),total lymphocytes count (TLC), forced expiratory volume in one second (FEV_1), maximal inspiration pressure(MIP)and maximal expiration pressure(MEP)were measured in all participants. Leptin levels were measured by radioimmunoassay. TNF-? levels were measured by ELISA. The between group difference and correlation of these parameters were analysed. 2.Immunomodulatory Mechanism of Kangxian Yixin Prescriptions on Autoimmune Injury Mice Based on Th17/Treg Cell Balance

ObjectiveTo investigate the immunomodulatory mechanism of Kangxian Yixin prescription (KYP) on autoimmune injury mice and its relationship with the T helper 17 (Th17)/regulatory T cell (Treg) balance. MethodSixty healthy 8-week-old male BALBc mice were randomly divided into a normal group and an experimental group at a ratio of 1∶5. On the 0th, 7th, and 28th days, 0.2 mL of porcine cardiac myosin emulsion (containing 0.1 mg of porcine cardiac myosin) was subcutaneously injected into the groin, armpit, and back of the mice in the experimental group to induce an animal model of myocardial immune injury. Mice with myocardial immune injury were randomly divided into a model group (Model), a KYP group (20.4 g·kg-1·d-1, ig), and a valsartan group (12 mg·kg-1·d-1, ig). Mice in the control group and the model group received the same amount of normal saline by gavage. After four weeks of intervention, the heart tissues were collected. Hematoxylin-eosin (HE) staining and Masson staining were used to detect pathological damage in heart tissues. Enzyme-linked immunosorbent assay (ELISA) was used to detect the expression of type B-type natriuretic peptide (BNP), anti-cardiac antibody, interleukin-17 (IL-17), and interleukin-10 (IL-10) in the serum of mice, and the expression levels of Th17 cells and Tregs in the spleen were detected by flow cytometry. The protein expression of B-cell lymphoma-2 (Bcl-2) and Bcl-2-associated X (Bax) in heart tissues was detected by Western blot, and the mRNA expression of retinoid-related orphan receptor gamma t (RORγt) and forkhead box P3 (FoxP3) in the spleen was detected by quantitative real-time polymerase chain reaction (Real-time PCR). ResultCompared with the control group, the model group showed worsened pathological damage in heart tissues, elevated serum levels of BNP, anti-myocardial antibody, and IL-17, decreased serum expression of IL-10 (P<0.05), increased expression of Th17 cells and reduced expression of Tregs in spleen tissues (P<0.05), increased protein expression of Bax, diminished Bcl-2 protein expression, elevated Bax/Bcl-2 ratio, up-regulated mRNA expression of RORγt, dwindled mRNA expression of FoxP3, and elevated ratio of RORγt/FoxP3 (P<0.05). Compared with the model group, the KYP group and the valsartan group displayed relieved pathological damage in heart tissues, decreased serum expression of BNP, anti-myocardial antibody, and IL-17, increased serum expression of IL-10 (P<0.05), reduced expression of Th17 cells and increased Tregs in spleen tissues (P<0.05), dwindled protein expression of Bax and elevated protein expression of Bcl-2 in heart tissues (P<0.05), diminished Bax/Bcl-2 ratio, reduced mRNA expression of RORγt, up-regulated FoxP3, and down-regulated ratio of RORγt/FoxP3 (P<0.05). ConclusionKYP may improve myocardial immune damage by regulating the Th17/Treg cell balance.
3.Human papillomaviruses infections among village women in Henan province.

OBJECTIVETo investigate the prevalence of Human papillomaviruses (HPVs) among village women in Henan and to determine its relevant risk factors.
METHODSA population based cross-sectional study on cervical cancer was conducted among village women in Xinmi, Henan. Women aged 20 - 54 who had sexual intercourse experiences were enrolled in this study. Self-sampling and direct-sampling were used in collecting women's vaginal discharge. 13 high-risk HPVs were tested with HC2 for all of the specimens. Then women with abnormal results did colposcopy and biopsy. The biopsy results were regarded as the golden standard.
RESULTSThere were 881 women enrolled in this paper and 881 self-sampling and 880 direct-sampling specimens were collected. The HPVs prevalence rates for the self-sampling and direct-sampling were 13.05% and 12.27%, respectively. Age-specific prevalence rates were 10.57% (20-), 9.60% (25-), 12.00% (30-), 9.52% (35-), 17.60% (40-), 13.74% (45-) and 12.80% (50 - 54). HPV prevalence rates were increased with progression of cervical disease (chi(2) = 200.69, P = 0.00). And HPV prevalence rates were higher in women with more advanced education background (chi(2) = 11.05, P = 0.01). HPV infection rate in women whose husbands have more than one sexual partner was 18.02% and whose husbands have only one sexual partner was 10.88% (chi(2) = 6.37, P = 0.01).
CONCLUSIONThe infection rate of high-risk HPVs in this area is high. The relationship of HPV infection with age has not been observed in this study, but the the sexual activity is the major risk factor for cervical cancer.

4.Study on effect of voltage-gated calcium channel protein in meridian tissue cells exciting conduction.

OBJECTIVETo explore the material basis of conduction along meridian.
METHODSSixty SD rats(30 males,30 females) were randomly assigned into a normal group,an acupuncture group,a verapamil blocking group and a 0.9%NaCl blocking group(control group),15 rats in each one. Fluo 3-AM(calcium fluorescence probe) was injected at the observation part in femoral stomach meridian of foot-(meridian part) and the approaching femoral meridian part(non-meridian part) in the normal group and the acupuncture group,and then incubation was applied. In the verapamil blocking group,verapamil was injected at local meridian part and non-meridian part,and in the control group 0.9%NaCl was injected. Then Fluo 3-AM was injected at the meridian part and non-meridian part in the two groups,and incubation was implemented. Caimaging changes in cells were recorded for more than 20 min after injection of every part in each group respectively. After the above operations in the last three groups,acupuncture was used at "Zusanli"(ST 36) immediately,with electroacupuncture for one min,then Caimaging changes in cells at the meridian and non-meridian parts were recorded for more than 20 min.
RESULTSIn the normal group, Cafluorescence intensity at the meridian part was higher than that at the non-meridian part. In the acupuncture group,after acupuncture Cafluorescence intensity at the meridian part was obviously higher than before,but the change before and after acupuncture was not apparent at the non-meridian part. After verapamil blocking local calcium channel and acupuncture,the Cafluorescence of the meridian part did not strengthen,and the change of that before and after acupuncture at the non-meridian part was not obvious. In the control group,after injecting 0.9%NaCl at local part,Cafluorescence intensities of the meridian and non-meridian parts showed no obvious change,so was that before and after acupuncture.
CONCLUSIONSThe voltage-gated calcium channel at the meridian part is highly correlated with its tissue cells exciting conduction.
5.Epidemiological investigation on a visceral leishmaniasis case in Zhengzhou City of Henan Province

Objective To perform an epidemiological investigation on a case with visceral leishmaniasis in Zhengzhou City, Henan Province, and to identify the source of infection, so as to illustrate the transmission chain and assess the risk of local leishmaniasis transmission. Methods The medical data were collected from a case with visceral leishmaniasis in Zhengzhou City, and the patient’s bone marrow smears were detected by microscopy. Serum anti-Leishmania antibody test and PCR assay were performed among high-risk residents and all dogs in the village where the patient lived. Sandflies were captured using light traps and artificial traps, and the captured female Phlebotomus chinensis was subjected to PCR assay. The internal transcribed spacer 1 (ITS1) gene was amplified with a nested PCR assay using the genomic DNA extracted from visceral leishmaniasis patients, positive dogs and sandflies, and the sequences were aligned with those download from NCBI. In addition, a phylogenetic tree was created based on the ITS1 gene. Results The visceral leishmaniasis patient had recurrent irregular fever, reduced complete blood counts, low hemoglobin, and a large number of Leishmania amastigotes in bone marrow smears, and was therefore diagnosed as visceral leishmaniasis. Both rk39 rapid diagnostic test and PCR assay tested negative among 324 residents living neighboring the patient’s residence, while 21.39% (43/201) dogs were positive for rk39 rapid diagnostic test and 13.93% (28/201) positive for PCR assay. There were 17 female Ph. chinensis tested positive for Leishmania (0.82%) by PCR assay, and the ITS gene sequences from visceral leishmaniasis patients, positive dogs and sandflies shared a 100% homology with L. infantum. The Leishmania species was therefore characterized as L. infantum. Conclusions L. infantum infection occurs in visceral leishmaniasis patients, dogs and sandflies in Zhengzhou City, indicating a complete transmission chain and a high transmission risk of visceral leishmaniasis by L. infantum. Intensified control measures are required to prevent local transmission of leishmaniasis in Zhengzhou City.
