2.Change and its significance of blood level of myocardium zymogram in acute organophosphrus poisoning mice

Objective To investigate the change and its significance of blood level of myocardium zymogram in acute organophosphrus poisoning(AOPP) mice.Methods 152 eight-week-old male mice were divided into 3 groups randomly: normal control group(n=8);normal saline control group(n=10);AOPP group(n=135).The AOPP group mice were assigned by 9 time-points(30 min,2 h,4 h,12 h,1 d,2 d,3 d,4 d and 7 d after poisoning).15 mice were in per cage.The mice in AOPP group were introperitoneal injected Omethoate(50 mg/kg) and Atropine(10 mg/kg).Centhifugalizing the blood and the serum were taken,examined cholineesterase(CHE),creatine kinase(CK),aspartate aminotransferase(AST) and lactate dehydrogenase(LDH).The level of myocardium zymogram of myasthenia mice was compared with normal muscle mice in the AOPP group.Results Compared with normal control group and normal saline normal saline control group,CHE activity was persistent inhibition in the AOPP 30 min ~3 d subgroups(allP
3.Vasorelaxant effect and underlying mechanism of EtOAc extract from Chrysanthemum morifolium in rat thoracic aorta

AIM: To investigate the vasorelaxant effect and mechanism of EtOAc extract from Chrysanthemum morifolium Ramat (CME). METHODS: The effects of CME on the contraction of rat thoracic a orta were examined. RESULTS: CME caused concentration-dependent relaxation of aorta rings precontricted with phenylephrine and K+. The effect in endothelium-intac t aorta was more effective than that in endothelium-deduced aorta. NG-nitro-L- arginine methylester, methylene blue and glibenclamide attenuated the effect of C ME significantly. However, indomethacin, propranolol, tetraethylammonium, BaCl 2, 4-aminopyridine and 5-hydroxydecanoate did not affect CME effect. The effect of SKF-525A combined with L-NAME had no obvious difference with that of L-NAME o n CME-induced relaxation. NOS activity in aorta was increased markedly by CME in vitro. CME did not reduced the contraction elicited by PE in Ca 2+-f ree medium, but reduced the contraction induced by PE in K+-free solution or C a 2+ free following input Ca 2+. CONCLUSION: CME induces both endothelium-dependent and independe nt relaxation. NO and cGMP are likely involved in the endothelium-dependent rela xation, inhibition of voltage-dependent or receptor-operate Ca 2+ channel a nd activation of ATP-sensitive K+ channel contribute in part to the endotheliu m-independent relaxation by CME.
4.Genotypes of Merozoite Surface Protein 2 of Plasmodium falciparum in Different Malaria Endemic Areas *

Objective To determine the genotypes and distribution of MSP2 of Plasmodium falciparum isolates in Yunnan and Hainan provinces, China.Methods The central polymorphic region of MSP2 allele was amplified by the nested PCR for genotyping of P.falciparum. Results The higher degree of polymorphism of MSP2 of P.falciparum was observed in Yunnan and Hainan. Distribution and allele frequencies differed in both provinces, indicating considerable geographical heterogeneity of parasite populations. The mixed infection of different allele type and multiplicity of infection was more frequent in Hainan than in Yunnan.Conclusion There were obvious differences in the distribution and frequencies of MSP2 alleles between Yunnan and Hainan endemic areas. MSP2 is suitable to be used as a marker gene for the genotyping of P.falciparum infection.
5.Observation on curative effect of invigorating spleen qi, bushen fillixi combined with oxaliplatin+CF/5-FU on colorectal cancer

Objective To investigate the therapeutic effect of Jianpi Yiqi, Bushen Fenjing combined with oxaliplatin+CF/5-FU on colorectal cancer.MethodsEighty patients with colorectal cancer who were treated in our hospital from January 2014 to December 2015 were randomly divided into observation group(40cases) and control group (40cases).The control group was given oxaliplatin+CF/5-FU treatment, the observation group in the control group based on the application of Spleen Qi, Bushen fill essence treatment, observed before and after treatment of two groups of serum tumor markers, clinical symptoms and living conditions, changes, record adverse reactions.ResultsThe observation effect of 82.50% was significantly higher than that of the control group (62.50%).The QOL score (37.8±5.6) was significantly higher than that of the control group (33.5±3.1) and the adverse reaction rate was 30.00% (P<0.05).The difference between the two groups was significant (P<0.05).ConclusionThe effect of invigorating spleen qi and tonifying siren solution combined with oxaliplatin+CF/5-FU on colorectal cancer can improve the clinical symptoms and improve the quality of life of patients.
6.Distribution of nerve fibers in endometrium and its clinical significance in adenomyosis

Objective To investigate nerve fibers distribution in endometrium of adenomyosis and their relationship with dysmenorrhea. Methods Endometrial tissue was sampled from 74 hysterectomy specimens including 32 cases with adenomyosis and 42 cases with uterine fibroids. Two-step Envision immunohistochemical staining was used to detect distribution of nerve fibers in endometrium. Highly specific polyclonal rabbit anti-protein gene product 9.5 (PGP9.5) and monoclonal mouse anti-neurofilament protein (NF) were used to demonstrate both myelinated and unmyelinated nerve fibers in endometrium in women with adenomyosis and uterine fibroids. Results The positive rate of PGP9.5 immunoreactive nerve fibers in the functional layer of endometrium of pain patients were with 64%(14/22) in adenomyosis and 67% (10/15) in uterine fibroids. And their density were 0.6(0-9.4)/mm2 and 0.6(0-6.0)/mm2 without reaching statistical difference (P> 0.05). No expression of NF could be detected in the functional layer of endometrium of adenomyosis and uterine fibroids. There were no PGP9.5 immunoreactive nerve fibers in the functional layer of endometrium in non-pain women with adenomyosis and uterine fibroids. Moreover, No NF immunoreactive nerve fibers in the functional layer of endometrium were shown in non-pain patients with adenomyosis and uterine fibroids. PGP9.5 immunoreactive nerve fibers and the nerve density in the basal layer of endometrium were 64%(14/22), 1.1(0-12.0)/mm2 in pain adenomyosis and 50%(5/10), 0.6(0-3.0)/mm2 in non-pain adenomyosis. NF immunoreactive nerve fibers and the density in the basal layer of endometrium were 23%(5/22),(0-0.6)/mm2 in pain adenomyosis and 20% (2/10),(0-1.0)/mm2 in non-pain adenomyosis. PGP9.5 immunoreactive nerve non-pain fibroids. NF immunoreactive nerve fibers and the nerve density in the basal layer of endometrium were 40%(6/15),0(0-0.4)/mm2 in pain fibroids and 15%(4/27),0(0-1.0)/mm2 in non-pain fibroids. There was no statistical different PGP9.5 and NF immunoreactive nerve fibers distribution in basal layer of endometrium between pain adenomyosis and pain fibroids or between non-pain adenomyosis and non-pain fibroids (all P>0.05). However, PGP9.5 immunoreactive nerve fibers density in basal layer of endometrium was higher in pain adenomyosis and fibroids when compared with non-pain adenomyosis and fibroids(P<0.05). Conclusions PGP9.5 immunoreactive nerve fibers might confer the occurrence of pelvic pain, however, NF immunoreactive nerve fibers may not involved in the pathogenesis of pain.
7.Diagnosis and prognosis of 280 patients with pancreatic carcinoma

Objective To evaluate the factors influencing early diagnosis and prognosis in patients with pancreatic carcinoma.Methods The clinical data of 280 patients who had complete follow-up data with pancreatic carcinoma treated from January 2002 to January 2007 were reviewed retrospectively.The medical history and follow-up data were collected from all patients.Survival rate was calculated by the life table method and the Kaplan-Meier estimation.Log-rank test was used for univariate prognostic analysis and Cox regression was used for multivariate prognostic analysis.Results 91.8%of the patients were more than 40 years old and the peak age was 50~73 years old;the major presentations were abdominal pain and jaundice.Major imaging tests included B-ultrasound and CT,the sensitivity was 70.6%,95.3%,respectively;89.3%of patients had combined B-ultrasound and CT examination.The sensitivity of CA19-9 was 81.1%.The median survival time was(7.0±0.5)months.Overall survival rates at 1~5 year survival rates were 28%,9%,6%,2%,and 1%.Univariate analysis suggested that age>65 years old,CA19-9>mean value,TNM Ⅲ or Ⅳ stage,lymph nodes invasion,vascular invasion,and metastasis of two or more organs,non-surgical treatment,KPS score<60 points,weight loss≥5 kg were poor prognostic factors;Cox multivariate analysis showed that treatment modalities,age,TNM stage,KPS score and ascites were independent risk factors for dismal prognosis.Conclusions The age,ascites,tumor stage and treatment modalities affected the prognosis of patients with pancreatic cancer.Early diagnosis and treatment was important to improve the survival time of patients with pancreatic cancer.
8.Protein gene product 9.5-immunoactive nerve fibers and its clinical significance in endometriotic peritoneal lesions

Objective To investigate the association between distribution of protein gene product (PGP) 9.5-immunoactive nerve fibers in peritoneal endometriotic lesions and disease-associated pain symptoms.Methods Thirty two peritoneal endometriotic lesions from patients with endometriosis (16 cases with pain and 16 cases without pain) and matched with 20 peritoneal tissues from patients with uterine leiomyoma without endometriosis were stained immunohistochemically for PGPg.5-immunoactive nerve fibers.Results The positive rate and density of PGP9.5-immunoreactive nerve fibers in peritoneal endometriotic leision were 62% (10/16) and (3.8 ± 1.7)/mm~2 in endometriosis patients with pain, which were significantly higher than 19% (3/16) and (1.7 ± 0.5)/mm~2 in endometriosis patients without pain (P <0.05) and 25% (5/20) and (1.3±0.6) /mm~2 in peritoneal tissues in women without endometriosis (P <0.05).However, no differences were found between endometriosis patients without pain and women without endometriosis (P > 0.05) .Moreover, the density of PGP9.5-immunoreactive nerve fibers in peritoneal lesions in endometriosis patients with pain was positively correlated with the severity of pain (r = 0.855, P < 0.05).In addition, the density of PGP9.5-immunoreactive nerve fibers in peritoneal lesions was statistically higher in endometriosis patients with chronic pelvic pain and(or) dysmenorrhea than those in endometriosis patients with other type of pain(P < 0.05), which was not associated with active lesion, site and staging (P > 0.05).Conclusion It suggested that PGP9.5-immunoreactive nerve fibers might confer the mechanism of pelvic pain with endometriosis.
9.Clinical comparative analysis of surgical resection of the pregnancy by hysterotomy and hysterectomy for cesarean scar pregnancy

Objective To assess the indication and safety of surgical resection of the pregnancy by hysterotomy (SRPH) and hysterectomy for cesarean scar pregnancy (CSP). Methods A retrospective study of women with CSP was conducted at the Women′s Hospital, School of Medicine, Zhejiang University, from Jan. 2003 to Mar. 2016. The women underwent SRPH (SRPH group, n=35) and hysterectomy (Hysterectomy group, n=14) were included. The gestational age (GA), size of gestational mass(GM), level of serum β-hCG, previous treatments and clinical outcomes were analyzed. Results The median GA, the mean size of GM, median serum β-hCG level, median amount of blood loss, rate ot blood transfusion, rate of persistent CSP, and rate of motal status in SRPH group versus Hysterectomy group were 66 versus 84 days, (65 ± 22) versus (92±36) mm, 23755 versus 802 U/L, 400 versus 650 ml, 11%(4/35) versus 13/14, 49%(17/35) versus 12/14, 20% (7/35) versus 14/14, respectively (all P<0.05). In SRPH group, median amount of blood loss was 500 ml in patients with GA≥10 weeks versus 300 ml in patients with GA<10 weeks (P<0.05). Serious complication occurred in 7 patients: severe pelvic inflammation in 1 patient and hematomas in the uterine isthmus in 1 patient in SRPH group; severe pelvic inflammation in 2 patients and hemorrhagic shock and DIC in 3 patients in Hysterectomy group. No blaader damage occurred. Conclusions SRPH is effective and safe for patients with CSP with GA of 9-10 weeks, a diameter of 60-90 mm and stable hemodynamics. Hysterectomy is an alternative to SRPH for patiens in motal status with advanced GA more than 12 weeks.
10.Eye acupuncture in treating cognitive dysfunction after stroke

Objective To observe any effect when eye acupuncture is combined with computer-assisted cognition training to ameliorate cognitive dysfunction after stroke.Methods Stroke patients with cognitive impairment were randomly divided into an eye acupuncture group (n=30),a computer group (n=30) and a combination group (n=30).The eye acupuncture group was given eye acupuncture,the computer group was given rehabilitative cognition training with specialized equipment and the combination group was given both eye acupuncture and the computeraided training.The treatment lasted 2 months.The Loewenstein occupational therapy cognitive asessment (LOTCA)and the modified Barthel index were used to evaluate the patients' cognitive function and daily life ability before and after treatment.Results Before training there was no significant difference in average LOTCA and MBI results among the three groups.After 2 months of training,all three groups had significantly higher scores in both evaluations.Moreover,the combination group's average score on organization ability,perceptual ability,thinking operation ability and concentration,and also their average total score and MBI score were significantly better than those of the other two groups.Conclusions Eye acupuncture combines synergistically with cognitive rehabilitation training to ameliorate cognitive dysfunction after stroke.Together they promote cognition,ADL ability and an early return to normal family and social life better than either alone.
