Interventional pulmonary procedures performed by flexible bronchoscopy has been widely used in the adult clinics.However,pediatric interventional pulmonary procedures are different from adult in anesthesia methods,equipment,technology and so on.At present.it is more limited.This review showed the experiences of pediatric bronchoscopy in Taipei Veterans General Hospital of Taiwan,China.

In order to improve the technique for histochemical demonstration of the different lymphocyte patterns by alpha-naphthyl acetate esterase (ANAE) activity from a single microscopic slide, 10 kinds of comparative experiments were performed. The results showed as follows. The peripheral blood mononuclear cells first fixed in 0.4% glutaldehyde and postfixed in 15% formol calcium gave the best effect on preserving the enzyme activity and on connterstaning of the leucocyte nuclei. During formol calcium fixation, the erythrocytes were dissolved from the diluted blood film with PBS more easily than from the blood smear without any dilution. The lymphocytes were distributed more uniformly and densely in the former specimen than in the latter. When incubated with 0.5 mg alpha-naphthyl acetate/ml medium in hexoazotized pararosaniline for 1(1/2)～2 hrs at 37℃, the distinct ANAE patterns of circulating lymphocytes were more readily identified.In 28 healthy adults the differential counts of the lymphocyte ANAE patterns from the diluted blood specimen was similar to those found from the specimen of isolated lymphocytes. The focal pattern was about 56%, the diffuse pattern about 25% and the negative pattern about 19%; while the isolated lymphocyte specimen was compared with the diluted blood film from the same individuals, the focal pattern was decreased insignificantly, the diffuse pattern increased and the negative pattern decreased significantly.

Objective: Combining spatial independent component analysis (sICA) with temporal correlation analysis to investigate the functional connectivity of human brain using resting state fMRI. Methods: First, activated area was localized by performing sICA on the data from block design run, then one of the activated brain areas was chosen as a region of interest (ROI)and low frequency correlations between ROI and other regions were calculated in resting state to detect the functional connectivity networks. To validate the method, neural connectivity to primary motor cortex was assessed using this method during a resting state. Results: Functional connectivity network of motor cortex was detected, including primary motor cortex (M1), supplementary motor area (SMA), primary sensory cortex (S1), dorsal premotor cortex (PMd) and posterior parietal somatosensory association area (PSAAp). The connectivity implied by the resting state correlation was far more similar to the connectivity established by non-imaging methods. Conclusion: Functional connectivity of human motor primary cortex was investigated by combining sICA with temporal correlation using resting fMRI data. It provided a simple and noninvasive method for the research of brain functional connectivity.

Objective To investigate the serum high-mobility group box protein 1(HMGB1)level in sepsis patients with different syndrome patterns,and to explore the correlation of HMGB1 level with defense-Qi-nutrient-blood syndrome differentiation and Acute Physiological and Chronic Health Evaluation Ⅱ(APACHEⅡ)score.Methods Twenty-four sepsis patients were differentiated as Qi system syndrome(QS group,N=11)and nutrient-blood system syndrome(NB group,N=13).Eight healthy volunteers served as the normal control.APACHEⅡ score was evaluated,serum HMGB1 level was detected by Western blot method,and the correlation of HMGB1 level with defense-Qi-nutrient-blood syndrome differentiation and APACHEⅡ score was analyzed.Results APACHEⅡ score was higher in NB group than that in QS group,the difference being significant(P

AIM:To establish a method for detecting three aconitum alkaloids(aconitine,mesaconitine,hy-paconitine) in Zhentong Huoluo Tincture(Radix Aconiti,Radix Aconiti kusnezoffii,Radix et Rhizoma Rhei,Rhizoma Pinelliae,Rhizoma Arisaematis ect.).METHODS:A ZOBAX Extend-C_ 18(250 mm?4.6 mm,5 ?m) column was used to determine the aconitum alkaloids in Zhentong Huoluo Tincture.The mobile was gradient elution,0.1% die-thylamine(adjust pH 9.0 with phosphoric acid) as mobile A;0-20 min A∶B(63∶37),20-45 min A∶B(63∶55∶37→45),at the rate of 1 mL/min.The temperature of column was 30 ℃,and the wavelength was at 232 nm.RESULTS:The linearity of the aconitine in 9.74 ng-974 ng was 0.999 9;that of the mesaconitine in 23.86-2 386 ng was 0.999 9 and that of the hypaconitine in 29.94 ng-2 994 ng was 0.999 9.The RSD of precision for these three were 1.50%、0.68% and 0.49%,respectively.The average recoveries and its RSD were 100.2%(RSD 3.2 %)、102.9%(RSD 2.2%) and 98.2(RSD 2.0%),respectively for aconitin,mesaconitine and hypaconitine.The reproducibilities were all less than 2.0%,and the sample solution was stable in 24 h.CONCLUSION:The method is simple,accurate and has good stability,can be used for the quantity control in the product of Zhentong Huoluo Tincture.

The met-enkephalin (MEK), leu-enkephalin (LEK) and ?-endorphin (?-EP) immunoreactivities were detected in the circulating lymphocytes of 64 guinea pigs by PAP immunohistochemical method under the conditions of electroacupuncture, naloxone or exogenous opioid like peptides (OLP). Subsequently the ?-naphthyl acetate esterase (ANAE)was demonstrated on partial immunostained lymphocyte samples. The results showed that the OLP immunoreactivity with different intensity was found in approximate 96％ of the lymphocytes. There was a positive correlation between the ANAE-focal pattern lymphocytes and MEK-IIRL (intense immunoreactivity lymphocytes)or ?-EP-IIRL; whereas there was no significant correlation between the ANAE-focal pattern lymphocytes and LEK-IIRL. The MEK-IIRL, LEK-IIRL and ?-EP-IIRL were all increased markedly in the guinea pigs which underwent electroacupuncture at "Zusanli" points. There was no significant difference in MEK-IIRL, LEK-IIRL and ?-EP-IIRL, when the acupuncture group, naloxone group and control group were compared with each other. However, the LEK-IIRL, MEK-IIRL and ?-EP-IIRL could be depressed by naloxone, when the lymphocytes were preincubated with 10~(-7)mol/L of MEK, LEK or ?-EP in vitro. Hence, the naloxone group had a significant difference in MEK-IIRL, LEK-IIRL or ?-EP-IIRL when compared with the acupuncture group. A high peak of the lymphocyte MEK-IR among the range of 10~(-10)—10~(-8)mol/L of MEK preincubation in the acupuncture group when compared with the control group. It suggests that electroacupuncture may enhance activity of the unbound MEK receptor.

To screen potential human soluble protein tyrosine phosphatase 1B (PTP1B) inhibitors, a high-throughput screening (HTS) model in 384-well microplate with total volume of 50 microL was established. Recombinant PTP1B was cloned and expressed in E. coli. with its specific substrate 4-nitrophenyl phosphate disodium salt hexahydrate (PNPP). The HTS model was based on enzyme reaction rate with enhanced sensitivity and specificity (Z' = 0.78). A total of 24,240 samples were screened, among them 80 samples with inhibition greater than 70% were selected for further rescreening. Finally, six compounds with high inhibitory activity were identified, whose IC50 values were 21.58, 18.39, 15.37, 11.92, 37.27, and 36.61 microg x mL(-1), separately. The results indicated that the method was stable, sensitive, reproducible and also suitable for high-throughput screening.

As a summary of the research progress of chemical modification of lead compounds by saccharides and their derivatives, this article studies the linkage style of saccharides with lead compounds and bioactivity changes of the modified lead compounds. The result shows that the modified lead compounds can decrease the toxicity and side effect efficiently, and enhance the pharmacal effect. Therefore, it is a promising research project to modify lead compounds by saccharides.
