1.Clinical Observation on Treatment of 32 Cases of Urinary Calculi with Lithagogue Decoction Combined with Western Medicine

Objective To observe clinical effect of treating urinary calculi with Lithagogue decoction combined with western medicine. Methods 32 cases patients with urinary calculi were randomly recruited into a control group and a treatment group. The control group was treated with anisodamine, ivgtt, and progesterone, im, and the treatment group was treated with lithagogue decoction based on the control group. The clinical effect was observed in both groups. Results The clinical effect was 93.8% and 68.7% in the treatment group and the control group respectively, the difference showing statistic significance (χ~2=3.459, P<0.05) .Conclusion The treatment of urinary calculi with Lithagogue decoction combined with western medicine is better than with western medicine only.
2. Preparation of cRGD-modified harmine hydrochloride stealth liposomes

OBJECTIVE: To prepare cRGD-modified harmine hydrochloride long circulating liposomes, investigate its characteristics in vitro, and finally establish its best preparation method. METHODS: Firstly the content determination method was established, and secondly harmine hydrochloride liposomes were prepared through reverse phase evaporation method. Both active and passive drug loading methods were investigated to entrap the drug. Thirdly, cRGD-DSPE-2000 was prepared by conjugation method, and then cRGD modified harmine hydrochloride long circulating liposomes were prepared. Finally the particle sizes, Zeta potential and the entrapment efficiency were determined. The release of the liposomes in vitro was measured. RESULTS: The standard curves all had good linearities except for the one using 100% methanol as solvent. The particle sizes of the liposomes prepared by passive loading method, active loading method, and long circulating cRGD modified liposomes were 227.2, 246.3 and 241.9 nm, respectively, and the Zeta potentials were about 20-30 mV. The entrapment efficiency were (36.78 ± 6.82)%, (81.77 ± 7.61)%, and (80.02 ± 1.27)%, respectively. The release of cRGD liposomes was slower than the liposomes without cRGD and HM solution. CONCLUSION: cRGD-modified long circulating harmine hydrochloride liposomes can be made through reverse phase evaporation method and active loading method. KEY

4.Rotating hinge prosthesis knee replacement for the treatment of malignant bone tumors around the knee

[Objective]To study the operative methods and effects of limb salvage with rotating hinge prosthesis replacement for malignant tumors around the knee.[Method]Limb salvage surgery with rotating hinge prosthesis replacement were performed in 14 patients with malignant bone tumors around the knee.There were 9 cases in distal femoral,5 cases in proximal tibia.[Result]Forteen patients were followed up for 17~65 months(averaged,4 months).The satisfactory rates were 85.7%,the extension and flexion spheres of knee activity were respectively 0? and 90?~130?(averaged,110?).There was recurrence in one patient after operation,infection one;prosthetic loosening one.[Conclusion]Application of rotating hinge prosthesis knee replacement in limb salvage treatment of malignant tumor around the knee is an effect method.It is an effect method for the treatment of malignant tumor of the knee by using rotating hinge prosthesis.It can decrease the strength of the bone-prosthetic and the incidence of the occurrence of fatigue fracture and the loosening of prosthetic.
5. Transport of progesterone and its proliposome in Caco-2 cells monolayer

OBJECTIVE: To study the absorption mechanism of progesterone and its proliposome in gastrointestinal (GI) tract. METHODS: Caco-2 cells monolayer was employed to investigate the transport of progesterone and its proliposome, and the effect of transport time, the drug concentration and inhibitors (cyclosporine and verapamil) on progesterone transport was further conducted. RESULTS: The apparent permeability coefficient of progesterone changed with the variety of the drug concentration, and inhibitors could improve the accumulative transport flux and the apparent permeability coefficient of progesterone. Liposome could also increase the transport of progesterone. But the transport of progesterone liposome was complied with a passive diffusion. CONCLUSION: The absorption process of progesterone is affected by P-gp, which may lead to its low bioavailability. As progesterone belongs to the second kind drugs in biopharmaceutical classification system, its bioavailability can be improved as the solubility is increased. Liposome can also improve its absorption and bioavailability through changing its absorption mechanism, which means the transport could be a passive diffusion. 6. Analysis of screening methods for DNA mismatch repair gene deletion in colorectal cancer

Objective: Here, we aimed to analyze the two most commonly used methods for screening DNA mismatch repair (MMR) gene deletions in colorectal cancer to establish a more cost-effective strategy. Methods: A total of 223 patients with colorectal cancer were recruited from the First Affiliated Hospital of Xinjiang Medical University for this study from September 2018 to September 2019. Using the Ventana BenchMark ULTRA automatic immunohistochemistry platform, the expression levels of MLH1, MSH2, PMS2, and MSH6 proteins were assessed, and the microsatellite instability (MSI) status of the tumors was then determined through PCR capillary electrophoresis. Results: Among the 223 patients with colorectal cancer, 27 (12.1%) had MMR deficiency (dMMR) and 196 (87.9%) had MMR proficiency (pMMR). The missing rates of MLH1, MSH2, MSH6, and PMS2 were estimated as 9.0% (20/223), 1.8% (4/223), 2.7% (6/223), and 9.4% (21/223), respectively. The sensitivity and specificity of the two-antibody test employing antibodies against only PMS2 and MSH6 for screening dMMR colorectal cancer were the same as those of the four-antibody test. Twenty-seven cases exhibited high microsatellite instability (MSI-H) (12.1%) and 196 cases exhibited microsatellite stability (MSS) (87.9%). However, no case exhibited low microsatellite stability (MSI-L). The sensitivities of BAT-25, BAT-26, NR-21, NR-24, NR-27, and MONO-27 were 88.9%, 92.6%, 96.3%, 70.4%, 92.6%, and 77.8%, respectively. When MSI was defined using three markers, NR-21, NR-27, and BAT-26, with at least one of the three exhibiting instability, the results were the same as those obtained using the six-marker group. Conclusions: The proposed two-marker detection strategy provides a simple, reliable, and low-cost method for the identification of dMMR/MSI in colorectal cancer.
7. Design and synthesis of 2-aryl-4-(trans-4-hydroxycyclohexanylamino)-5-ethyl- 7H-pyrrolo2, 3-dpyrimidines and Mer tyrosine kinase inhibitory activity evaluation

Objective: To design and synthesize a series of 2-aryl-4-(trans-4-hydroxycyclohexanolamine)-7H-pyrrolo[2, 3-d] pyrimidine derivatives and test their inhibitory activity against Mer tyrosine kinase(MerTK)and tumor cell proliferation. Methods: 2, 4-Dichloro-7H-pyrrolo[2, 3-d]pyrimidine(1)was used as starting material to synthesize the target compounds via iodination, protection with p- Tosyl(Ts), nucleophilic substitution, Stille coupling, vinyl reduction, Suzuki coupling and deprotection of Ts. The MerTK inhibitory activity was tested by the Kinase-Glo Plus luminescence method. The antiproliferation activity was assayed using MV- 4-11, A549, MDA-MB-231, KB, and KB-vin tumor cell lines by the CCK8 and SRB methods. Molecular docking of MerTK and 6h was conducted using the DS3.0 software. Results: Nine compounds were synthesized, and their structures were confirmed by 1H NMR and MS. Compound 6h exhibited certain inhibitory effect on MerTK, with the(6.7±0.3)μmol/L of IC50 value, and could selectively inhibit the growth of MV-4-11 tumor cells, with the(6.6±1.1)μmol/L of GI50 value that was approximately 3-fold to 6-fold more potent than the GI50 value of 6h on the other tested tumor cells. Molecular docking showed that 6h could interact with MerTK on the binding site of UNC569 and overlapped well with the original ligand UNC569, but its binding energy was higher than the binding energy of UNC569. Conclusion: Compound 6h showed a selective inhibitory effect on the MV-4-11 cell growth, which might be further investigated via more biological experiments to explore whether the inhibitory effect is related to the inhibition of MerTK by 6h. The molecular docking results in the present study have suggested that further structural modification on the 2 and 5 position of 7H-pyrrolo[2, 3-d] pyrimidine skeleton could likely improve the MerTK inhibitory activity.
8.Comparison of immunogenicity between recombinant flagellins C and B of Salmonella abortus equi.

To evaluate and compare of the immunogenicity differences of flagellins FliC and FljB of Salmonella abortus equi, and lay the experimental foundation for the further utilization of the two recombinant proteins, FliC and FljB recombinant proteins were induced, expressed and purified. The purified FliC and FljB were used to immunize mice separately. The antibody level, titer and subtype of mice serum were detected after immunization. Immune-related receptors and histopathological changes were observed in immunized mice after challenged. The recombinant proteins FliC and FljB were successfully induced and expressed. Proteins of about 52 kDa and 42 kDa were purified. High levels of specific IgG antibodies were induced in mice immunized with these two proteins, the antibody level of FljB-immunized group was higher than that of FliC-immunized group, and IgG1 was the dominant subtype of antibody. The challenge protection rate of the FljB-immunized group was 87.5%, higher than that of FliC immunized group. Bacterial loads and observation pathological of FljB-immunized group were better than that of FliC immunized group, the levels of TCR2, TCR4, MHC-I and TCR induced by FljB-immunized group were higher than those of FliC-immunized group. The of immune response induced by FljB group was better than that of FliC group.
9. The anti-inflammation effects of Aesculetin from Viola tianshanica Maxim via NF-kB and MAPK signaling pathways in RAW 264.7 ceils stimulated by LPS and its mechanism

Aim To investigate the anti-inflammatory effects of Aesculetin from Viola tianshanica Maxim in LPS-stimulated RAW 264.7 cells and the underlying mechanism.Methods RAW 264.7 cells were divided into control group, model group( LPS), 0.16, 0.8, 4, 20 μmol·L-1 AESN groups( different concentrations of AESN + LPS)and positive control group(10 μmol·L-1 Indomethacin+LPS).LPS(1 mg·L-1)was used to stimulate RAW 264.7 cells for 24 h to establish inflammatory model.MTS assay was used to detemine cytotoxicity of Aesculetin in RAW 264.7 cells.Griess method was used to detect NO secretion in LPS-stimulated RAW 264.7 cells.ELISA was applied to determine the contents of TNF-α, IL-6 and IL-1β in cell culture supernatant.qRT-PCR was employed to detect the mRNA expression levels of TNF-α, IL-6, IL-1β and iNOS.Immunofluorescence assay was used to evaluated the protein expressions of iNOS, p-NF-κB p65, IκBα, p-p38 and p-ERK1/2.Enzyme assay was used to detect the inhibition activity of Aesculetin on cyclooxygenase 1/2(COX 1/2).Results Aesculetin significantly inhibited the secretion of inflammatory mediator NO, mRNA and protein expression of iNOS in LPS-induced RAW 264.7 at 0.16, 0.8, 4 and 20 μmol·L-1.The contents of TNF-α, IL-6 and IL-1β in supernatant significantly decreased, and the mRNA expression levels of TNF-α, IL-6 and IL-1β were also reduced by Aesculetin.Aesculetin also obviously inhibited the protein degradation of IκBα and inhibited the nuclear translocation of p-NF-κB p65, p-p38, p-ERK1/2.In addition, Aesculetin had significant inhibitory activities on COX-1 and COX-2, and the IC50 was 28.1 μmol·L-1, 2.3 μmol·L-1, respectively.Conclusions AESN has good anti-inflammatory effect, and its mechanism is closely related to the inhibition of NF-κB and MAPK signaling pathways
10.Analysis on monitoring results of heavy metals in rural soils in Bole of Xinjiang, 2017-2019

Objective To monitor heavy metals Pb, Cd, Cr, As, and Hg in agricultural land in Bole of Xinjiang, in order to understand the overall pollution status of rural soil environment and determine the environmental health hazards in rural areas of Bozhou, Xinjiang. Methods From 2017 to 2019, 4 administrative villages in 5 townships in Bole City in Bozhou Region were selected as monitoring points. The 5 to 20 cm deep topsoil of monitoring points was collected for a total of 60 samples. According to "Environmental Quality Standard for Soils" (GB 15618-2018), the pollution levels of Pb, Cd, Cr, As and Hg were evaluated by single index method and Nemerow index method. Results The content of Pb, Cr, As, and Hg in the heavy metals in the soil of Bozhou region from 2017 to 2019 were all lower than the screening value and regulatory value of soil pollution risk for agricultural land, while the content of heavy metal Cd in 13 soil samples (21.67%) exceeded the standard. The content difference in Pb, Cd, Cr, and As was statistically significant (P<0.05), while the difference in Hg was not statistically significant (P>0.05). The single pollution index Pi of Pb, Cr, As and Hg was less than 1, indicating a non-pollution state. However, the single pollution index Pi of Cd from 2017 to 2018 was higher than 1, with the highest value reaching 2.33. In 2017 and 2018, the comprehensive pollution index of rural soil samples was 1.67 and 1.06, and the evaluation result was light pollution, while in 2019, the comprehensive pollution index was 0.72, and the evaluation result was still clean. Conclusion The overall quality of rural environmental soil in some rural areas in Bozhou, Xinjiang was generally acceptable, but the heavy metal Cd pollution in some aeras deserves attention.
