Objective:After construction and identification of HBsAg gene recombinant backbone adenoviral vector,it is to prouduct HBsAg gene recombinant adenoviruses by packing PAd-Easy-1-HBs in 293 cells. Methods:The gene of interest was amplified from plasmid pEcob-6 PCR, the gene of interest which contained HBsAg gene was cloned into on adenoviral shuttle vector pAd-track-cmv. The pAd-track-cmv-HBs was linearized by digesting with restriction endonuclease Pme-1, and subsequently cotransformed into E.coli BJ-5183 cells with an adenoviral backbone vector pAd-Easy-1, Homologous recombinants were performed in bacterial cells. Finally, the linearized backbone adenoviral vector was transfected into adenoviruses packing cells lines,e.g. 293 cell by lipofectamine transfection. Transfections and viral productions can be minotored by green fluorescent protein(GFP). The expression of HBsAg in supermatant was investigated by ELISA. It was a certain HBsAg vaccine to amplify recombinant adenoviruses by repeating the infevtion cell to collect the viral supermatant. Results:GFP expression was visible by fluorescence microscopy after transfection. Adenoviral titer was monitored by GFP expression. GFP expression was visible after repeating the infection cell using the viral supernatant in more than 90 percent of the cells. The HBsAg also expressed in supermatant. Conclusion:HBsAg gene recombinant adenoviral backbone vector has been constructed successfully. HBsAg gene recombinant adenoviruses have been producted by packing in 293 cells. The study provides the possibility of further researches on the development of new anti-HBV vaccines.

Medical Journal of Chinese People's Liberation Army 1982;0(03):-
Objective To investigate the effects of different astragalosides(AST) and hydro-soluble astragali polysaccharide(APS) of Astragalus membranaceus on inducing erythroid differentiation of human leukaemic K562 cells. Methods APS and AST were extracted by alcohol or water from Astragalus membranaceus.K562 cells were treated with APS,AST and sodium butyrate(BA) respectively. The proportion of benzidine-positive cells was examined after 1-4 days culture.MTT assay was performed for evaluating the proliferation effects of APS,AST and BA on K562 cells. Results The percentages of benzidine-positive cells induced by APS,AST and BA were 13.2%,2.9% and 17.5%,respectively.The kinetic characteristics of K562 cells induced by different levels of APS(1,2,4,8mg/ml) indicated that 4 mg/ml APS was sufficient to induce K562 cells to turn to be benzidine-positive.The results suggested that APS,other than AST,could induce the K562 cells towards erythroid differentiation.Compared with BA,the percentage of benzidine-positive cells induced by APS was lower at 24h,48h and 72h(F=237.44,P=0.00),while the total number of benzidine-positive cells was higher at 96h(F=322.25,P=0.00).The results of MTT assay for Absorbance(A) showed that APS and AST had no inhibitory effects on growth of K562 cells(P=0.28,P=0.11),while BA showed an obvious inhibitory effect on K562 cells(P=0.00).Conclusion Astragalus membranaceus has pharmacological effects to induce the K562 cells towards erythroid differentiation, and its valuable constituents are contained in hydro-soluble astragali polysaccharide(APS).The induction of differentiation is most evident with a dosage of 4 mg/ml APS.The cell growth curve reveals that APS has no inhibitory effect on K562 cells.

SUMMARY To demonstrate the clinical manifestation, diagnosis and treatment of myoclonus epilepsy with ragged-red-fibers ( MERRF) , a case of MERRF was presented with review of the literature. A 4-year-7-month-old girl was diagnosed with MERRF. She had tremor, fatigue and developmental delay for more than 2 years. Laboratory tests showed that the serum and urine lactic acid and pyruvic acid increased significantly. Electroencephalogram showed diffuse and focal spike slow wave and slow wave in right central and parietal regions. Electromyogram showed neurological damage. Gene mutational analysis showed mtDNA 8344 A>G mutation. The mutational rate was 78%. Mitochondrial disease MERRF syndrome was diagnosed. Cocktails therapy with vitamins B1, B6, B12, L-carnitine, and coenzyme Q10 was administra-ted to the patient. MERRF is a rare disease. The diagnosis can be made by gene mutational analysis. Cocktail therapy may slow down the deterioration of the disease. Gene therapy is still experimental.
