1.Application of Bedside Cranial Soft Channel Drainage in Intracranial Hematoma

Objective To investigate the application effect of drainage of intracranial hematoma in bedside skull soft channel.Methods In the hospital in October 2014 to 2015 years 5 months in the diagnosis and treatment of intracranial he-matoma in patients with selected 20 cases as the research object and the principles were randomly divided into 2 groups, and the observation group ( n=10) the application of bedside cranial awl soft channel drainage in the treatment of , control group (n=10) application of conventional drilling drainage treatment .Two groups patients therapeutic effect and complications in-cidence were compared .Results To observe the surgical treatment group the total effective rate was 90.0%, and complica-tion rate was 10.0%;Observation group , operation treatment , the total effective rate is 60.0%, and complication rate was 40.0%.Two groups of patients with surgical treatment of total efficiency and complication rates between the groups compared with statistical difference (P<0.05).Conclusion The application of bedside cone cranial soft passage drainage in intracra-nial hematoma is effective , can significantly improve the postoperative cognitive function and muscle strength , improve the a-bility to take care of themselves , and less complications .
2.Clinical study on endoscopic variceal ligation combined with endoscopic variceal sclerosis for treating patients with esophageal variceal bleeding

Objective To investigate the clinical efficacy and safety of endoscopic variceal ligation (EVL) combined with endoscopic variceal sclerosis (EVS) for treating patients with esophageal variceal bleeding (EVB).Methods Ninety patients with EVB were divided into observation group and control group by treatment methods with 45 patients each.The observation group was given EVL combined with EVS and the control group was given EVL.The elimination of varicosity after the first treatment,time and times required to eliminate,rehaemorrhagia,varicosity recurrence,complication and prognosis of the 2 groups were observed.Results The elimination rate of varicosity of observation group was 97.8%(44/45),the control group was 93.3%(42/45),there was no significant difference (P> 0.05).The elimination rote of varicosity after the first treatment of observation group was significantly higher than that of control group,the time and times required to eliminate were significantly lower than those of control group [71.1%(32/45) vs.48.9%(22/45),(17.24±5.65) d vs.(36.01 ± 11.81) d,(1.42 ±0.47) times vs.(1.87 ±0.61) times,P < 0.05 or <0.01].The rates of early-onset rehaemorrhagia,delayed rehaemorrhagia and varicosity recurrence of observation group were significantly lower than those of control group [4.4%(2/45) vs.17.8%(8/45),6.7%(3/45) vs.22.2% (10/45),2.2% (1/45) vs.22.2% (10/45),P <0.05].The rate of complication of observation group was significantly lower than that of control group [11.1% (5/45) vs.42.2% (19/45),P <0.01].Conclusion The EVL combined with EVS is an effective method for emergency hemostasis and preventing rehaemorrhagia in the patients with EVB.
3.Verification on hypothesis for optimum ratio in various active constituents of Chinese materia medica with desired effects

To propose the hypothesis of the requirement for optimum ratio of various active cons tituents of Chinese materia medica with desired effects, which suggests that both single drug and compound drug prescriptions have their own target effects corresponding to their main indications and their active constituents must be in optimum ratio This hypothesis was initially tested in an ischemic animal model The neuroprotection of Yushen Oral Liquor (YSOL, an oral liquor made of Chinese materia medica) on neuronal injury in CA1 region of rat hippocampus following transient forebrain ischemia was used as an index to evaluate the drug's effect By combination of HPLC and pharmacological method, the relationship of HPLC and pharmacodynamical results of each sample made of YSOL was studied to observe the chromatogram peaks representing the drug's neuro protective effects There are three peaks named as R1, R2, and R3 for YSOL, which closely correlate to the drug's neuronal protective effects and the relative weight but not the absolute quantity of those peaks plays a critical role in producing neuronal protective effects of YSOL on ischemia induced neuronal damage The results provide experimental data supporting our proposed hypothesis
4.The effects of intracellular calcium concentration and signal transducers and activators of transcription 3 on the invasion of Vibrio vulnificus clinical isolates B2 into dendritic cells

Objective To explore the effects of intracellular calcium concentration ([Ca2 + ]i )and signal transducers and activators of transcription 3 (STAT3 )signaling pathway on the invasion of Vibrio vuknificus (Vv )clinical isolates B2 into dendritic cells (DC).Methods The co-culture model of Vv B2 and DC 2.4 was constructed.Apoptosis and necrosis rates in different invasion phases were detected by flow cytometry.The calcium fluorescence probe Fluo-8-AM was used to detect the change of [Ca2 + ]i ,and Western blot and immunofluorescent techniques were used to explore the relative molecular expressions and intracellular distributions of STAT3 and phosphorylation STAT3 [p-STAT3 (705 )].Data were analyzed by t-test.Results After co-culture ofVv B2 and DC 2.4,the apoptosis rates of 1 ,2,3 and 4 h were (13.10±4.72)%,(30.10±3.52)%,(46.20±15 .61)% and(31 .00 ±19.10)%,respectively.The differences were statistical significant between Vv standard strain (1 .1758)and Vv B2 (t=4.30,22.33, 4.30 and 4.30,respectively;P =0.040,0.002,0.040 and 0.040,respectively).The necrosis rates of 1 , 2,3 and 4 h in co-culture of Vv B2 and DC 2.4 were(19.70 ±3.50 )%,(39.20±4.60 )%,(40.90 ± 13.80)% and (62.10 ± 8.20 )%,respectively.The differences were statistical significant between Vv 1 .1758 andVv B2 (t=9.93,14.09,4.30 and 14.09,respectively;P =0.010,0.005 ,0.049 and 0.005 , respectively).Compared to Vv 1 .1758,Vv B2 induced more apparent cell apoptosis and necrosis within the same time.By fluorescence microscope,intensity of calcium was enhanced with the rising of apoptosis. Although the total expression of STAT3 increased,the relative molecular expression of p-STAT3 (705 ) decreased.There was no significant difference in its distribution in cells compared to the blank group. Conclusion The rapid apoptosis and necrosis of DC during Vv B2 invasion into DC may be caused by increasing [Ca2 + ]i and inhibiting phosphorylation of p-STAT3 (705)simultaneously.
5.RET proto-oncogene mutation analysis in a pedigree with multiple endocrine neoplasia 2A

ObjectiveTo discuss the clinical diagnosis and treatment of multiple endocrine neoplasia ( MEN ) 2A, and report the mutation of the RET proto-oncogene in a pedigree of three patients with MEN 2A.MethodsBilateral adrenalectomy was performed on two of the three patients with hypertension and bilateral adrenal-conserving adrenal pheochromocytoma resection was performed on the other patient. All three patients were treated by total thyroidectomy and neck lymphadenectomy. Twelve family members were recruited to the study. Peripheral blood was collected and total genomic DNA was prepared for polymerase chain reaction (PCR). PCR products of exon 10 and exon 11 of the RET proto-oncogene were purified and a direct DNA sequence analysis was performed.ResultsThe pathological diagnosis of the specimens was bilateral adrenal pheochromocytoma and medullary thyroid carcinoma in all the three patients. There was no tumor recurrence or distant metastasis after 1.5 - 5 years of follow-up. A missense mutation of TGC (Cys)to CGC (Arg) at codon 634 in exon 11 of the RET proto-oncogene was detected in all three patients. Genetic screening identified two mutation carriers in the other members of this pedigree.ConclusionGenetic mutation screening and surgical intervention may be helpful to the members of high-risk families.
6.Chromatograph-digitized fingerprint spectrum of Liuwei Dihuang Dripping Pill

AIM:To establish some HPLC-digitized fingerprint spectrum(HPLC-DFPS)in Liuwei Dihuang Dripping Pill preparation and to apply it to identification of medicinal materials.METHODS:Based on loganin and paeonoside contents adopted as qualitative and quantitative factors,relative retention value as creteria,RP-HPLC was carried out on inertsil ODS:column,mobile phase consisted of methanol-water(0.2% formyl acid),detection wavelength was set at 240 nm.RESULTS:Using above optimum chromatograph conditions,HPLC-DFPS of relative medicinal materials and "Liwei Dihuang Dripping Pill" preparation were established.By comparison a group of characteristic peaks could be used to identify medicinal materials.CONCLUSION:This result indicates that it is practical to use HPLC-DFPS for identification of Liuwei Dihuang Dripping Pill.
7.Study on Detection Value of Platelet Activation Indexes and Platelet Parameters in Patients with Hepatitis B-induced Cirrhosis

Objective:To study the detection value of platelet activation indexes and platelet parameters in patients with hepatitis B-induced cirrhosis.Methods:40 patients with hepatitis B-induced cirrhosis in our hospital from November 2015 to August 2016 were selected as the observation group.40 healthy subjects went through physical examination during the same period were chosen as the control group.The platelet activation indexes and platelet parameters of subjects with same age in two groups were detected and compared.The detection results of the two items in patients with different stages of hepatitis B-induced cirrhosis were compared,too.Results:The PLT of observation group was lower than that of control group,while the other platelet parameters and platelet activation indexes were all higher than those of control group.The detection results of two items in patients with different stages of hepatitis B-induced cirrhosis were with significant differences,all P<0.05.Conclusion:The platelet activation indexes and platelet parameters in patients with hepatitis B-induced cirrhosis are obviously abnormally expressed.There are significant differences among the detection results in patients with different stages.Therefore,surveillance of those indexes in patients with hepatitis B-induced cirrhosis should be strengthened.
8.The effects of murine cytomegalovirus on the maturation, fertilization, cleavage and blastula formation of mouse oocytes in vitro.

To study the effects of mouse cytomegalovirus (MCMV) on the in vitro maturation, fertilization, cleavage and blastula formation of mouse oocytes, the immature oocytes were infected in vitro by MCMVs of different dosages (100 TCID(50), 10 TCID(50) and 1 TCID(50)). The oocytes were then observed for in vitro maturation, fertilization, cleavage and blastula formation and the ultrastructural changes after the culture with the viruses. Our results showed that no significant differences were found in IVM, IVF, cleavage and blastula formation among the groups treated with of virus of various dosages. And ultrastructural abnormality was observed in the oocytes treated by 100 TCID(50) of viruses. It is concluded that MCMV did not have any conspicuous effects on IVM, IVF, cleavage and blastula formation of murine immature oocytes.

9.Crystallography of ATP hydrolysis mechanism in rat brain kinesin.

Rat brain kinesin is a conventional kinesin that uses the energy from ATP hydrolysis to walk along the microtubule progressively. Studying how the chemical energy in ATP is utilized for mechanical movement is important to understand this moving function. The monomeric motor domain, rK354, was crystallized. An ATP analog, AMPPNP, was soaked in the active site. Comparing the complex structure of rK354 x AMPPNP and that of rK354ADP, a hypothesis is proposed that Glu237 in the Switch II region sensors the presence of gamma-phosphate and transfers the signal to the microtubule binding region.

10.Expression, purification and crystallization of rat brain kinesin.

Kinesin is a motor protein that uses the energy from ATP hydrolysis to move along the microtubule system. To investigate how the chemical energy stored in ATP is converted to mechanical movement, the corresponding N-terminal region of rat brain kinesin was expressed in BL21-Codon Plus (DE3)-RP competent cells. After SP-cation exchange chromatography and size exclusion chromatography, the protein yield reached 10 mg/L culture with the purity above 95%. The purified protein had ATPase activity and specifically reacted with the kinesin antibody in the Western blotting analysis. The purified kinesin was crystallized under the following condition: 1.7 mol/L (NH4)2SO4, 500 mmol/L NaCl, 20% glycerol. The kinesin crystal can diffract up to 2.0 angstroms resolution.
