1.Efficacy of deproteinised cancellous bone combined with BMP and fibrin in healing of rabbit segmental radial defect

[Objective]To repair segmental bone defect with the composite of deproteinised cancellous bone,bone morphogenetic protein(BMP) and fibrin.[Method] Bone defects were created in 36 New Zealand rabbits on the bilateral radii and treated with three kinds of implantations:A,composite of deproteinised cancellous bone,BMP and fibrin;B,of deproteinised cancellous bone;C,blank control.The defects were observed by entgenography,optical microscopy,and electron-microscopy at 2,4,8,12,16 weeks.[Result] The defects treated with A implantation regenerated much more new bone and bridged earlier than the defects treated with B implantation and got completely repaired 16 weeks after operation.The defects with B and C implantation couldn't get osseous tissue healing.[Conclusion]The composite of deproteinised cancellous bone,bone morphogenetic protein(BMP) and fibrin can be effectively used to repair segmental bone defect.
2.Antidotal effect of N-acetylcysteine on acute poisoning with sodium cyanide

AIM: To study the antidotal effect of N-acetylcysteine(NAC) on acute poisoning with sodium cyanide(NaCN).METHODS:After intraperitoneal(ip) injected of four sulphydryl compounds respectively,the mice which were acutely poisoned by NaCN were observed of the behavioral change,convulsion number and mortality rate(within 72 h).The median lethal dose(LD_(50)) of acutely poisoned mice was detected in NaCN group and NAC protection group.1 min after mice were poisoned by NaCN,the mice were divided into three groups:the first was ip normal saline(NS),the second was ip NAC in combination with sodium thiosulfate(Na_2S_2O_3),the third was ip sodium nitrite(NaNO_2) in combination with Na_2S_2O_3.Then the behavioral change,convulsion number and mortality rate(within 72 h)were observed,recorded,and compared the differences between the mice groups.RESULT: All of the four sulphydryl compounds had protecting effect on acutely NaCN poisoned mice,among them NAC had most prominent effect(P
3.Manufacture and evaluation of health education form for spinal tuberculosis patients

Objective To investigate the requirement of health education for patients with spinal tuberculosis,making clinical education form,and determine the evaluation system.Methods 134 cases of spinal tuberculosis patients were surveyed with self-designed questionnaires.The questionnaire included 5 aspects,using the Likert 4 scoring method.Delphi method was applied in 16 experts.Results The health demand of patients showed high positive correlation in domains of cognition and operation.There was a moderate positive correlation between affective domain and cognition,operation domain.Two rounds of expert consultation showed that the expert opinion coordination coefficient was 100%,the average of authority was above 0.8,the expert opinion coordination coefficients of all the indexes were between 0.2~0.6.Conclusions The health education form for spinal tuberculosis patients can help to understand the needs of patients,it is pertinent and get the recognition of experts.
4.Treatment of foot injury with free medial sural artery perforator muscle flap

This observation was to assess the treatment effects of free medial sural artery perforator muscle flap on foot reconstruction.Seven patients(6 men,and 1 woman;age 19 to 46 years,mean 33) with soft tissue defects on the foot underwent surgical procedures by using free medial sural artery perforator muscle flap.The coverage of the muscle flaps was performed by a meshed split-thickness skin graft.The donor site Was closed.Median follow-up Was 2.5 years(range 7 monks to 5.5 years).All the muscle flaps and skin grafts survived without major complications,and no morbidity was found at the donor sites.The muscle flaps seem to have advantage in blood supply,vascular anatomy and pedicle length,and may be helpful in the mpmr of soft tissue defects on the foot.
5.Fibrin glue/bone morphogenetic protein complex plus slow-release gentamicin for repairing infected bone defects in rabbits

BACKGROUND: Chronic osteomyelitis is difficult to manage clinically, and two or more operations were commonly needed. No satisfactory method for one-stage repair has been currently available.OBJECTIVE: To examine the possibility of using fibrin glue(FG) as the common carrier for both bone morphogenetic protein(BMP) and gentamicin for one-stage repair of infected bone defects.DESIGN: A completely randomized controlled experiment.SETTING: Center of Orthopaedic Surgery, Lanzhou General Hospital of Lanzhou Area Command of of Chinese PLA.MATERIALS: The experiment was conducted using 48 healthy adult Chinchilla rabbits of either sex on normal diet with body mass of 1.9 to 2.4 kg,provided by the Institute of Biological Products, Lanzhou, Gansu Province.INTERVENTIONS: Animal models of chronic osteomyelitis were established in 48 Chinchilla rabbits, and after thorough debriding of the wounds,an elongated semicircular bone defect was induced 1.5 cm to the medial proximal metaphysis in the tibias. Three treatments were subsequently given with FG, MP and gentamicin complex(Group A), FG/BMP complex(Group B), and nothing(control group, Group C), respectively.MAIN OUTCOME MEASURES: The general conditions of the rabbits were observed postoperatively. Bacterial culture of the bone specimen, bacterial counting, X-ray and histological examination were performed.RESULTS: Rabbits in Group A exhibited obviously better infection control and greater regenerated bone mass than those in Group B, and no significant difference was noted in infection control between Groups B and C, the latter group showing poor bone defect repair.CONCLUSIONS: FG, BMP and gentamicin complex helps to control infection and promotes bone formation, and can be used to repair bone defects caused by infection and open injury with severe contamination.
6.Icariin promotes osteogenic differentiation of bone marrow mesenchymal stem cells

BACKGROUND:Herba epimedi, a traditional Chinese medicine, has a long time in dealing with various orthopedic disorders. Icarinwithmany biological activites is one of the most important compositions of Herba epimedi. OBJECTIVE:Toinvestigate the effects of icarin on osteogenic differentiation of mesenchymal stem cels and the underlying mechanisms. METHODS:Bone marrow mesenchymal stem cels were treated using icarin with or without osteogenic mediumin vitro. Osteogenic differentiation markers, including runt-related transcription factor 2, osteocalcin and osterix, were detected by real time-qPCR. Alizarin red staining was used to measure calcium nodes generated by osteoblasts induced frombonemarrow mesenchymal stem cels. The proximal tibia bone structure of rats fed with icarin (2 mgperday) for 5 weeks was detected and analyzed by MicroCT. RESULTS AND CONCLUSION:Icarin was able to promote the expression of genes related to osteogenic differentiation in the absence or presence of osteogenic induction. Icarin could obviously increase the quantity of calcium nodes whenmesenchymal stem celswere cultured in the osteogenic medium. The animal experiment showed that icarin improved formation of trabecular bone.
7.Effect of Sodium ferulate on glucocorticoid-induced osteoporosis

Aim To study the influence of Sodium fer-ulate ( SF) on bone metabolism in glucocorticoid–in-duced osteoporosis rats. Methods Thirty cases of fe-male Wistar Rats(3-month-old) were divided into con-trol group, model group and SF group ( low-dose group, middle-dose group, high-dose group ) by ran-domized block design. Double fluorochrome labeling with calcein was performed before necropsy. The left tibia was taken for bone histomorphometry. Results In static parameters, the proximal tibia cancellous bone trabecular thickness, trabecular quantity and area ratio were significantly reduced in model group compared with control group;while compared with model group, those were increased in middle and high-dose SF group. Trabecular separation degree was increased in model group compared with control group, while it was decreased in middle and high-dose SF group compared with model group. In dynamic parameters, the calcula-tion parameters of cancellous bone mark perimeter rate and the bone formation rate were increased in model group compared with control group, in middle and high-dose SF group the bone formation rate was in-creased compared with model group. In bone cells, os-teoclast number per mm, osteoblast number per mm, percent osteoblast surface perimeter and percent osteo-clast surface perimeter were increased in model group compared with control group. In growth-plate, the thickness of growth-plate was increased in model group compared with control group. In bone cells and growth-plate there was no statistical significance between treat-ment group and model group. Conclusion This study demonstrates that SF can increase bone mass and im-prove bone structure,which may be related to the im-provement of bone formation. SF is effective for GIOP in rats.
8.Research advance in Enterobacteria ceaebacteria resistant to polymyxins

In order to deal with the increasing number of multidrug resistant Enterobacteriaceae , polymyxins were re-introduced into clinical practice .The resistance of polymyxins had gained global attention.In addition to chromosomal mediated drug resistance , a new plasmid-mediated colistin-resistance gene mcr-1 made transferation of polymyxins resistance more easily .Here, the mechanisms of chromosome mediated polymyxin resistance and plasmide-mediated drug resistance , including the distribution , prevalence, transfer mechanism, and genetic environments of mcr-1, current methods for polymyxins susceptibility testing methods and novel qualitative detection techniques were reviewed to provide information to cope with the growing problem of bacterial drug resistance .
9.Efficacy of ultrashort wave combined with Etroricoxib in treatment of periarthritis of shoulder

Objective To observe the effect and adverse effect of ultrashort wave combined with Etroricox-ib in treatment of periarthritis of shoulder. Methods 80 cases with periarthritis of shoulder from March 2015 to March 2016 were randomly assigned into treatment group and control group. Etroricoxib therapy was provided in control group while ultrashort wave plus Etroricoxib therapy in treatment group. VAS ,ROM and MBI were applied for the evaluation before and after the treatment. The differences of clinical cure rate ,recurrence rate and adverse reactions were observed between 2 groups. Results There were higher cure rates ,lower recurrence rates and less adverse reactions in 2 groups after the treatment. The shoulder joint pain ,rang of should motion and BI of both groups were improved with significant differences after the treatment ,but treatment group witnessed more improve-ment(P<0.05). Conclusions Ultrashort wave combined with Etroricoxib therapy can relieve shoulder pain ,and further improve the function of shoulder joint activity in the treatment of periarthritis of shoulder. It is recommended for clinical application.
10. Impact of PRDM1 gene inactivation on C-MYC regulation in diffuse large B-cell lymphoma

Objective:
To investigate the role of PRDM1 gene inactivaion in the regulation of C-MYC in diffuse large B-cell lymphoma (DLBCL), and to explore the correlation of its immunophenotype and prognosis.
Methods:
100 cases paraffin-embedded DLBCL tissues were collected from January 2009 to December 2015 at the First Affiliated Hospital of Xinjiang Medical University along with 20 cases of reactive proliferative lymph nodes as control. Immunohistochemical methods were used to detect the expression of CD20, CD10, MUM1, Ki-67, bcl-6, PRDM1/Blimp1, C-MYC and PAX5 protein. The tumors were classified into two subtypes according to Hans classification.The expression of PRDM1 and C-MYC gene in tumor group and control group was detected by reverse transcription PCR (RT-PCR) and the relationship between PRDM1 and C-MYC gene was analyzed.OCI-LY1 (GCB subtype) and OCI-LY3 (non-GCB subtype) cell lines were transfected with small interfering RNA by cationic liposome reagent transfection, and the expression of C-MYC in the transfected cell lines was detected by RT-PCR and Western blot. The Kaplan-Meier method was used to analyze the prognostic significance of PRDM1/Blimp1 and C-MYC at protein and mRNA levels.
Results:
There were 27 cases of GCB subtype and 73 cases of non-GCB subtype according to Hans classification. The positive expression of Blimp1 in DLBCL group and proliferative lymph nodes in control group was seen in 26(26.0%) and 20 cases(100%), respectively. There were 58 cases with high expression of PRDM1 at mRNA level, including 22 cases of GCB subtype and 36 cases non-GCB subtype, and the difference was statistically significant (
