1.Diffusion-Weighted MR Imaging of Kidney

Objective To explore the characteristics and applied value of diffusion-weighted MR imaging of kidney in healthy people.Methods Using Philips Gyroscan Intera 1.5T MR System,DWI of kidney was performed in 40 healthy adult volunteers,the ADCs of renal cortex and medulla were measured.Results The cortex and medulla of the normal kidney could be clearly showed by DWI.The ADCs of the cortex medulla and average were 3.08?0.52,2.74?0.68 and 2.91?0.58;2.79?0.51,2.53?0.65 and 2.63?0.62;2.64?0.43,2.32?0.47 and 2.48?0.44 when the b values were 300,500 and 800 s/mm2,respectively.The ADC was higher in the cortex than that in the medulla.Conclusion DWI can be used in kidney,it may be helpful for the diagnosis of renal diseases.
2.Primary Study of Chronic Kidney Disease with MR Diffusion-Weighted Imaging

Objective To study the applied value of MR diffusion-weighted imaging(DWI)and apparent diffusion coefficient(ADC)value in chronic kidney disease(CKD).Methods 10 healthy volunteers and 45 patients with different period of CKD underwent routine MRI and DWI.The DWI features were observed.The ADC values were acquired by using Philips soft package on the ADC images.Results Under different b value(300 s/mm2,500 s/mm2 and 800 s/mm2),the ADC value of CKD were 2.29?0.27,2.07?0.21 and 1.91?0.22,respectively,which were significantly lower than that in normal kidneys,and there was statistical difference between different group of CKD.Conclusion DWI may be a noninvasive method to evaluate the renal function.
3.Effect of gypenosides on hepatic function and fibrosis in rats with immune fibrosis

Objective:To study the effect of gypenosides (GPs) on the hepatic function and fibrosis progression in Wistar rats with immune fibrosis. Methods: The model of immune fibrosis was established by injection of human albumin. Rats in treated group were also administered with 0. 5 mg GPs during 30 d human albumin attack. The hepatic function of rats was e-valuated by determining the levels of ALT, TBIL and TBA in serum. The progression of hepatic fibrosis was evaluated by the levels of HA, PC Ⅲ and LN by radioimmunoassay (RIA) and confirmed by rats tissue pathology analysis. 4.The role of neutrophil collagenase in acute lung injury induced by endotoxin

Objective In the LPS-induced acute lung injury (ALI) there is massive accumulation of neutrophils in the lung. The effect of neutrophil collagenase on the degradation of lung matrix collagen ig still unknown. The aim of this study was to investigate the role of neutrophil collagenase in LPS-induced ALI.Methods Thirty SD rats of either sex weighing 190 ? 40 g were randomly divided into five groups of 6 animals : group 1 control and group 2-5 LPS. In LPS groups animals were anesthetized with intraperitoneal 3% pentobarbital 30 mg?kg-1. Right jugular vein was cannulated. LPS (O55: B5 Sigma USA) 5 mg?kg-1 was injected iv. The animals were sacrificed at 2 h (group 2), 4 h (group 3), 6 h( group 4) and 8 h( group 5) after iv LPS administration. Bronchoalveolar lavage was performed immediately with 3 ml of normal saline. The fluid washed out was collected and subjected to hypothermic centrifugation (3000 r?min-1, 10 min 4℃) . The supernatant was collected for determination of its protein content. Lung tissue was obtained for determination of myeloperoxidase (MPO) activity, expression of matrix metallo-proteinase-8 (MMP-8) , CrossLaps protein content (the degradation product of type Ⅰ collagen) and wet/dry lung weight ratio(W/D), and microscopic examination. Pulmonary permeability index was calculated (protein content in bronchoalveolar wash-out fluid / plasma protein concentration) .Results At 4, 6 and 8 h after iv LPS administration (in group 3-5) the MMP-8 expression, crosslaps protein content, MPO activity, W/D and lung permeability index were significantly increased as compared with those in control group ( P
5.Protective effect of genistein pretreatment against endotoxin-induced acute lung injury in rats

Objective To investigate the effects of genistein pretreatment on endotoxin-induced acute lung injury in rats and assess the possible mechanism. Methods Thirty-two male Wistar rats weighing 240-280 g were randomly divided into 4 groups ( n = 8, each group) : group Ⅰ control; group Ⅱ genistein; group ? LPS and group Ⅳ genistein pretreatment. The jugular vein was cannulated for administration of fluid and drug. The animals were anesthetized with intraperitoneal 3% pentobarbital 40 mg? kg-1 . In group Ⅰ and Ⅱ Ⅳ normal saline (NS) 1 ml?kg-1 was Ⅳ given 30min after IP NS 1 ml?kg-1 (Ⅰ ) or genistein 50 mg?kg-1 (Ⅱ ). In group ? and Ⅳ LPS 6 ml? kg-1 was Ⅳ given 30 min after IP NS 1 ml?kg-1 (?) or genistein 50 mg? kg -1 (Ⅳ). Four hours after.LPS injection, rats were sacrificed. The lungs were removed for evaluation of histological injury and determination of wet/dry lung weight (W/D) ratio, myeloperoxidase (MPO) activity, malondialdehyde (MDA) content, expression of TNF-f55 mRNA, HO-1 mRNA, TNF-f55, and HO-1. Bronchoalveolar lavage fluid (BALF) was collected for determination of PMN count, protein content, and MPO activity.Results LPS administration induced marked lung injury and significant increases in W/D ratio, MPO activity, and MDA content in the lung tissues, and PMN count, protein content, and MPO activity in BALF. All of these changes were significantly reduced by genistein pretreatment. Genistein also markedly suppressed LPS-induced expression of TNF-a mRNA and protein, and enhanced LPS-induced expression of HO-1 mRNA and protein. Conclusion Pretreatment with genistein has protective effect against endotoxin-induced acute lung injury. The underlying mechanism is via an inhibition of neutrophilic recruitment and activity, a down-regulation in TNF-f55 production, and a up-regulation in HO-1 expression.
6.The role of neutrophil collagenase in endotoxic acute lung injury.

The aim of this study was to determine the role of neutrophil collagenase in the pathogenesis of acute lung injury induced by endotoxin. 28 Sprague-Dawley were randomized into control group and LPS-enduced groups. Samples of left lung were obtained in 2 h (group L1), 6 h (group L2), 12 h (group L3) after intravenous LPS. Immunohistochemsitry was employed for detection of expression of neutrophil collagenase. Pathological scores, lung wet/dry weight ratio and the number of neutrophils were measured. The results showed that the concentration of neutrophil collagenase in LPS-enduced groups (group L1, L2, L3) were significantly higher than that of control group (P<0.01). Pathological scores, lung wet/dry weight ratio and the number of neutrophils in LPS-enduced groups (group L1, L2, L3) were also significantly higher than that of control group (P<0.01). Moreover, among group L1, L2 and L3, there were significant correlations in concentration of neutrophil collagenase and pathological scores, lung wet/dry weight ratio, the number of neutrophils (P<0.05). The present study showed that neutrophil collagenase play an important role in the pathogenesis and progress of endotoxic acute lung injury.

8.Electrochemiluminescence Performance of Poly(aniline_luminol) Composite Nanowires Synthesized by Chemical Oxidation

Poly ( aniline_luminol ) composite nanowires were synthesized by chemical oxidation using ammonium peroxydisulfate. In contrast to the maximum fluorescence emitting wavelength of luminol at 425 nm, the maximum fluorescence emitting wavelength of the polymeric luminol in the composite nanowires was red shifted to 465 nm obviously. The poly ( aniline_luminol) composite nanowires were modified on graphite electrode surface by drop coating, forming a stable poly ( aniline_luminol ) composite nanowires film. The composite nanowires film modified electrode presented favorable electrochemiluminescence ( ECL ) performances, and the ECL response could be enhanced by hydrogen peroxide. Under the optimized experimental conditions, the modified electrode provided a linear range of 5. 0×10-9-1. 0×10-5 mol/L for the detection of hydrogen peroxide with a detection limit of 2 ×10-9 mol/L.

Objective To investigate the number changes of the ribbon synapses(RS) in the inner hair cell of C57BL/6J mice with age .Methods The cochlear basilar membrane was obtained from C57BL/6J mice in 2 ,6 ,10 , 12 months old .RIBEYE on the presynaptic membrane and AMPA receptor on the postsynaptic membrane were double labeled by immunofluorescence histochemical technique .The stained sections were observed under a confocal laser-scanning microscope .Then the number of RS of the basilar membrane was calculated by three dimensional reconstruction images using 3DS max software .Results The numbers of RS reduced gradually from the cochlear a‐pex to base in the same developing stages .The numbers of RS in apex and middle turn of cochlea were gradually re‐duced with the age increasing .The number of RS in basilar turn reduced first and increased slowly then .Conclusion The decreases of the number of RS maybe the main pathological change at the early stage of presbycusis .In addi‐tion ,there may be a kind of compensatory mechanism by increaseing the number of RS to delay the hearing deficits .
10.Effects of acute normovolemic hemodilution on cardiac troponin I and myocardial enzyme

ve To investigate the effects of acute normovolemic hemodilution on cardiac troponin I (CTnl) and myocardial enzyme. Methods Twenty-nine ASA I-II patients undergoing hepatectomy or bilateral hip replacement were randomly divided into two groups: hemodilution group (H , n=15) and control group (C, n=14) . The patients were premedicated with diazepam and scopolamine. Left radial artery was cannulated for intra-arterial pressure monitoring and the letting of blood. Right internal jugular vein was cannulated for fluid administration and CVP monitoring. Anesthesia was induced with midazolam 0.15-0.30mg?kg-1 , fentanyl 4?g?kg-1 and vecuronium 0.08-0.1mg?kg-1 and maintained with infusion of midazolam (0.03-0.10 mg?kg-1?h-1), fentanyl (0.15-2.0?g?kg-1?h-1) and vecuronium (0.5-1.0?g?kg-1?min-1). After tracheal intubation the patients were mechanically ventilated with 100% O2 and PETCO2 was maintained at 35-45 mmHg. 15 min after induction of anesthesia hemodilution was started. Blood was withdrawn from radial artery and collected until Hct was reduced to 25%-28% . Ringer's lactate solution and hydroxyethyl starch (HES 6%) were simultaneously infused to maintain blood volume. Blood samples were taken before anesthesia, before blood transfusion, 4h after surgery and on the 1st postoperative day for determination of blood CTnI, CK and CK-MB activities. Results The demographic data including age and body weight were not significantly different between the two groups. Prior to blood transfusion Hct was reduced to 20.33% ?1.91 % in group H and 29.64%?1.78% in group C. CK increased significantly at 4h after operation and on the 1st postoperative day; CK-MB increased significantly on the 1st postoperative day. The difference between the two groups was not significant. CTnI showed little change during and after operation in both groups.Conclusions Our study suggests that hemodilution to 20% Hct does not result in myocardial injury in patients without preexisting cardiopulmonary disease as long as normal blood volume is maintained and patient suffers nohypoxia.
