1.The research on the effect of reading education in cultivation mode of nursing humanities

Objective To explore the application of reading education in cultivating nursing students' humanities in vocational college.Methods Nursing students were selected at random and divided into two groups,35 students in the experimental group and 50 in the control group.The students in the experimental group were given the 2-year related instruction through reading activities in the light of the principle enhance general humanities foundation in the first grade and strengthen humanistic features in the second grade.Nursing skill assessment and questionnaire oriented to humanities were used to test the educational effect.Results The result of nursing skill assessment in the experimental group was significantly better than that of the control group.The score of humanities' moral,law,culture and aesthetic dimension and the total score of the experimental group were significantly better than those of the control group.Conclusions Well targeted reading is effective in the cultivation of nursing students' humanities.Meanwhile reading education can serve as an important and beneficial way to improve the cultivation mode of vocational college's nursing humanities.
2.Diagnostic value of soluble B7-H4 in rheumatoid arthritis

Objective To investigate the diagnosis value of soluble B7-H4(sB7-H4) in rheumatoid arthritis (RA) .Methods By using the ELISA sandwich method ,the serum levels of soluble B7-H4 ,anti-cyclic citrullinated peptide(anti-CCP) antibody and glu-cose 6-phosphate isomerase(GPI) were detected in 67 cases of RA ,72 cases of osteoarthritis(OA) and 75 cases of healthy people . The immunoturbidimetric method was adopted to determine the serum rheumatoid factor (RF) .Then the diagnostic value of sB7-H4 in RA was compared and analyzed .Results The serum level of soluble B7-H4 in the RA group was significantly higher than that in the control group and the OA group with statistically significant difference between them (P<0 .01) .The sensitivity of sB7-H4 in diagnosing RA was higher than that of RF ,anti-CCP antibody and GPI ,but the specificity was lower than that of anti-CCP antibody and GPI .There was better correlation between sB7-H4 with RF and anti-CCP antibody .The combination detecting of sB7-H4 ,RF , anti-CCP antibody and GPI could increase the diagnostic rate of RA .Conclusion B7-H4 has better clinical application prospects in diagnosing RA .
3.Targeting macrophagic SHP2 for ameliorating osteoarthritis via TLR signaling.

Osteoarthritis (OA), in which M1 macrophage polarization in the synovium exacerbates disease progression, is a major cause of cartilage degeneration and functional disabilities. Therapeutic strategies of OA designed to interfere with the polarization of macrophages have rarely been reported. Here, we report that SHP099, as an allosteric inhibitor of src-homology 2-containing protein tyrosine phosphatase 2 (SHP2), attenuated osteoarthritis progression by inhibiting M1 macrophage polarization. We demonstrated that M1 macrophage polarization was accompanied by the overexpression of SHP2 in the synovial tissues of OA patients and OA model mice. Compared to wild-type (WT) mice, myeloid lineage conditional Shp2 knockout (cKO) mice showed decreased M1 macrophage polarization and attenuated severity of synovitis, an elevated expression of cartilage phenotype protein collagen II (COL2), and a decreased expression of cartilage degradation markers collagen X (COL10) and matrix metalloproteinase 3 (MMP3) in OA cartilage. Further mechanistic analysis showed thatSHP099 inhibited lipopolysaccharide (LPS)-induced Toll-like receptor (TLR) signaling mediated by nuclear factor kappa B (NF-κB) and PI3K-AKT signaling. Moreover, intra-articular injection of SHP099 also significantly attenuated OA progression, including joint synovitis and cartilage damage. These results indicated that allosteric inhibition of SHP2 might be a promising therapeutic strategy for the treatment of OA.
