1.EFFECT OF PLANTAGO ASIATICA L.SEED ON THE ANTIOXIDATION IN RATS

Objective: To investigate the effect of Plantago asiatica L. on antioxidation in rats. Methods: The antioxidative enzymes and LPO of serum, heart, and liver tissues were determined. Results: The activities of superoxide (SOD) in serum and heart were significantly lower than that of the control. The lipid peroxide (LPO) level in serum and heart was markedly higher than that of the control. Serum and liver catalase (CAT) activities in rats fed with high fat were decreased. Activities of liver glutathione peroxidase (GSH Px) in high fat rats were decreased significantly. The production of SOD activities in serum and liver and GSH Px in liver were increased significantly in rats maintained on Plantago asiatica L. supplemented diets, meanwhile serum and heart LPO were reduced. Conclusion: 15g/kg Plantago asiatica L. can increase the antioxidation against lipid peroxide in hyperlipidemic rats.
2.Molecular cloning, sequence characterization and mRNA tissue expression analysis of TDRP1 gene from the Banna minipig inbred line (BMI)

Objective To get TDRP1 gene of sterile and fertile boar of the Banna minipig inbred line (BMI), predict its function by bioinformatics analysis, and detect its expression patterns in the fertile boar.Methods Based on the NM_001198925 sequence, we designed specific primers and amplified BMI TDRP1 using RT-PCR method for sequen-cing and bioinformatics analysis.Meanwhile, the expression of TDRP1 in 17 organ tissues ( heart, liver, spleen, lung, kidney, thymus, lymph nodes, skin, duodenum, stomach, cerebrum, cerebellum, testis, epididymis, seminal vesicle, prostate, and bulbourethral gland) of fertile BMI boar and in the testis of sterile and fertile BMI boars was analyzed by semi-quantitative RT-PCR.Results The experiment obtained 680 bp cDNA sequence ( GenBank accession number:KJ186786) of BMI TDRP1, which encodes a protein of 186 amino acids with a predicted molecular weight (Mw) of 20.49 kDa and isoelectric point (pI) 5.86, and no signal peptide.It was a nuclear protein with a probability of 94.1%and had a leucine-rich nuclear export signals.Homology analysis of protein sequences revealed that BMI TDRP1 showed high identi-ty with that of humans, macaca mulatta, mouse and rat.The RT-PCR analysis showed that TDRP1 had a similar expression in the testes of sterile and fertile BMI boars.It was highly abundant in the seminal vesicle and prostate, moderately ex-pressed in cerebellum and testis and weakly expressed in cerebrum and kidney, while undetected in other 11 organ tissues. Conclusions We have cloned TDRP1 complete coding sequence, and found 2 SNPs,showing no difference in sequences and the testis mRNA expression levels between the fertile and sterile BMI boars.The multi-tissue transcription profile shows different expression levels in different organ tissues, being high in the seminal vesicle and prostate.The results of this study provide a foundation for further insight into the role of this gene in spermatogenesis.
3.Epidemiological feature of hepatitis B in Zaozhuang City, Shandong Province.

OBJECTIVETo study the prevalence and the epidemiological feature of hepatitis B in the population of Zaozhuang and the seroepidemiology of hepatitis B virus infection.
METHODSHBsAg, anti-HBs and anti-HBc were detected in 963 sera collected from 312 families by RIA, which were selected by multistage and stratified cluster sampling.
RESULTSThe results indicated that the standardized positive rates of HBsAg, anti-HBs and anti-HBc were 7.08%, 37.56% and 41.35%, respectively and the standardized prevalence rate of hepatitis B virus infection was 44.37%. The positive rate of HBsAg was higher in male subjects and in the urban area groups than those in female subjects (P<0.05) and in rural area groups (P<0.01), respectively. There was no significant difference in the positive rate of HBsAg among all age groups and occupation groups (P>0.05). The positive rates of anti-HBs and anti-HBc and infective rate of hepatitis B virus increased with age. The infective rate of hepatitis B virus was higher in male subjects and in rural area groups than those in female subjects (P<0.05) and in urban area groups (P<0.05), respectively.
CONCLUSIONSThe HBV infective rate of population was higher in Zaozhuang city; measures of prevention and control should be taken actively and effectively.

4.The use of the transabdominal ultrasound with different frequency scanning anterior lower segment uterine scar situation to guide motherhood

Objective To explore the use of the transabdominal ultrasound with different frequency scanning anterior lower segment uterine scar situation for maternal guidance .Methods Pregnant women had a history of cesarean section 323 cases, and were lower uterine segment transverse incision .Of which seven cases of cesarean section were 8 months to 12 months, 49 patients 13 months to 24 months, 89 patients 25 months to 36 months, and more than 169 cases of 37 months.High-frequency ultrasound was use for ab-dominal and dynamic monitoring of different-year anterior lower segment uterine scar thickness and healing .Pregnancy outcomes ob-served were compared with surgery .Results With the extension of cesarean time , grade I uterine scar was also increased to 25~36 months (2 years ) which was the most obvious , and grade II and III scars were reduced accordingly .When group 25~36 months was used as the control group , there was statistically significant difference relative to group 8 ~12 months and 13 ~24 months ( P <0.01), but was no significant difference relative to group >37 months ( P >0.05).Among 323 cases, seven cases had uterine rup-ture that was confirmed by surgery with a rupture rate of about 2.1%.The rupture rate was about 28.6%in group 8~12 months, a-bout 2%in group 13~24 months, about 1.0%in group 25~36 months, and about 1.2%in group >37 months.When group 25~36 months was used as the control group , there was statistically significant difference relative to groups 8~12 months and 13~24 months ( P <0.01 ) , and was no significant difference relative to group >37 months ( P >0.05 ) .Conclusions Abdominal ultra-sound with different frequency scanning can monitor the thickness and continuity of anterior lower segment uterine scar to estimate the scar healing situation , and guide uterine scar grading .Therefore, it is great significance in guiding clinical decision of motherhood ap-proach.
5.Ultrasound monitoring of cervical canal length combined with measurement of fibronectin in cervical secretion for predicting preterm delivery

The length of cervical canal was monitored by perineal ultrasonography in 377 pregnant women with 22 to 36 weeks of gestation,at the same time the contents of fibronectin (fFN) in cervical secretions were measured.The rate of cervical canal length shortening from the time of first visit to the appearance of preterm signs was calculated.When the cervical length shortening rate was 16% ~ 60%,fFN (+) was correlated with higher preterm birth rate (P <0.01),while when the shorten rate > 60% there was no significant difference in preterm delivery rate between fFN (+) and fFN (-) groups (P > 0.05).The results indicate that the monitoring of cervical canal length combined with fiberonectin measurement in cervical secretion may better predict pretenn birth.
6.Establishment and evaluation of rat obesity model induced by electrical damage of ventromedial hypothalamic nucleus and arcuate nucleus

Objective:To construct a rat model of hypothalamic obesity by two point electrical damage to the ventromedial hypothalamus and arcuate nucleus.Methods:Twenty adult male SD rats of SPF grade were randomly divided into experimental group and sham operation group.A 25GA (0.45 mm) solid iron needle was used, the needle was coated with an insulating layer, and the tip exposed a 0.5 mm conductive area.With reference to The Rat Brain in Stereotaxic Coordinates and using the stereotactic instrument (AP: -2.6 mm, ML: ± 0.6 mm, DV: -9.6 mm) as the coordinate, 1.5 mA current was continuously applied for 25 s, the ventromedial nucleus (VMH) and arcuate nucleus (ARC) of bilateral brain in SD rats was damaged.During the experiment, the body weight(BW), food intake(FI) and water intake(WI) of the two groups were recorded regularly.The rats were sacrificed on the 28th day after the operation, and the changes of periprenal fat mass and body length were measured.The changes of liver and adipose tissue were detected by HE staining method, leptin by ELISA, leptin receptor(LEPR) by Western blot.Results:(1) The body weight of rats in the experimental group ((427.5±17.7)g) and weight gain ((208.5±14.8)g) were significantly increased compared with the rats in the control group((349.2±17.7)g), ((136.2±21.4)g)on the 28th day after operation ( t=7.661, 6.806, both P<0.001). (2) The daily food intake of rats in the experimental group ((44.2±6.6)g) on the 28th day after surgery was significantly higher than that in the control group ((23.0±3.6)g) ( t=6.918, P<0.001). There was no significant difference of the daily drinking water of rats between experimental group((37.5±12.1)ml) and the control group ((35.0±11.8)ml) ( t=0.361, P=0.726). (3) Perikidney fat mass of experimental group rats ((13.4±2.7)g) significantly increased 28 days after operation compared with control group rats((6.3±0.9)g)( t=4.250, P<0.05). The naso-anal length of experimental group((21.8±0.4)cm) was significantly decreased compared with the control group ((23.4±0.2)cm) ( t=-6.788, P<0.01). The Lee index of the experimental group (348.9±8.5) was significantly higher than that of the control group(305.5±4.3)( t=7.898, P<0.01). (4) The serum leptin content ((8 324.10±159.00)μg/L) of the experimental group rats at 28 days after surgery was significantly higher than that of the control group((2 705.31±407.10)μg/L) ( t=25.712, P<0.001). The lateral hypothalamus area (LHA) LEPR protein expression (1.3±0.1) in the experimental group was significantly higher than that in the control group (0.9±0.1) ( t=4.932, P<0.01). Conclusion:Two-point electrical damage to bilateral VMH and ARC of rats can establish hypothalamic obese rat model.
