1.Protective Effect of Yulangsan Polysaccharide Against Hepatic Injury in Mice

Objective To investigate the protective effect of Yulangsan polysacharide ( YLSPS) and mechanism against ibuprofen-induced liver injury in mice. Methods The mice were randomly divided into the blank control(NC), the model control,YLSPS at 150 mg·kg-1 , 300 mg·kg-1 ,600 mg·kg-1 groups and biphenyldicarboxylate (150 mg·kg-1 BPDC) group. The mice were orally administered with corresponding agents once per day for consecutive 14 days, whereas the blank control group and model control group were orally administered with saline. Except the blank control group, all the other mice were orally administered IBU 200 mg·kg-1 body weight 2 h after last lavagedof medicimes. The mice were fasted and watered ad lib for 20 h after model establishment. Activities of ALT,AST and ALP,content of T-BiL,TNF-α,IL-6 in serum;activities of SOD,GSH-Px and content of MDA in liver tissue were detected. The morphological pathology test was used to examine degrees of hepatic injury. Results Compared with the model control, YLSPS could obviously reduce activities of ALT,AST and ALP,content of T-BiL, MDA,TNF-α and IL-6, and increase SOD,GSH-Px and CAT (P<0. 05), and then lessen the hepatic injury. Conclusion YLSPS showed potential protective effect against ibuprofen-induced liver injury in mice, the mechanism may be related to attenuating free radical injury and inhibiting lipid peroxidation and lowering release of inflammatory factors.
2.Effects of Early Rehabilitation and Hyperbaric Oxygen Treatments on Stroke Patients

ObjectiveTo observe effects of early rehabilitation and hyperbaric oxygen treatments on stroke patients.Methods80 patients were randomly divided into rehabilitation group (40 cases) and control group (40 cases). Patients in rehabilitation group were treated with rehabilitation and hyperbaric oxygen treatments; patients in control group were treated only with hyperbaric oxygen treatment. The activities of daily living (ADL) were assessed by modified Barthel index (MBI), and the cognition function was measured by mimi-mental state examination (MMSE).ResultsAfter treatment the scores of MMSE and MBI of two groups increased (P<0.05) and scores of rehabilitation group were higher than that of control group (P<0.05); the process of scores in rehabibilitation group were obviously superior to that in control group (P<0.001).ConclusionEarly rehabilitation and hyperbaric oxygen treatments can obviously improve cognition function and ADL of stroke patients.
3.Anti-infection effect of phage and its clinical application

In recent years, the problem of bacterial resistance has become more and more serious, which has brought troubles to global public health and medical care. The time and money required to develop new antibiotics is even greater than before. Bacteriophage is a kind of virus that can specifically infect bacteria, fungi, actinomycetes and other microorganisms. Relying on host bacteria to replicate in large numbers, rich species, low research and development cost, the value of anti-infection therapy is very considerable. It is a new generation of biological antimicrobial agents with great potential. This paper briefly describes the sterilization mechanism, progress of research on anti-infection aspect and clinical application of phage, in order to provide reference for phage anti-infection treatment and clinical application.
4.Cell-ELA-based determination of binding affinity of DNA aptamer against U87-EGFRvIII cell.

A15, a DNA aptamer with binding specificity for U87 glioma cells stably overexpressing the epidermal growth factor receptor variant III (U87-EGFRvIII), was generated by cell systematic evolution of ligands by exponential enrichment (cell-SELEX) using a random nucleotide library. Subsequently, we established a cell enzyme-linked assay (cell-ELA) to detect the affinity of A15 compared to an EGFR antibody. We used A15 as a detection probe and cultured U87-EGFRvIII cells as targets. Our data indicate that the equilibrium dissociation constants (K(d)) for A15 were below 100 nmol/L and had similar affinity compared to an EGFR antibody for U87-EGFRvIII. We demonstrated that the cell-ELA was a useful method to determine the equilibrium dissociation constants (K(d)) of aptamers generated by cell-SELEX.

5.Treatment of non-small cell lung cancer with EGFR-TKI

Epidermal growth factor receptor-tyrosine kinase inhibitors (EGFR-TKIs) are the key to the treatment of advanced non-small cell lung cancer (NSCLC),but there are various drug-resistant mutations in the latter stage.The first and second generation of EGFR-TKIs significantly prolong the survival of patients with EGFR-activated mutant tumors.The third generation of EGFR-TKIs effectively inhibit the progress of T790M mutant tumors.The fourth generation of EGFR-TKIs inhibit both L858R,T790M and L858R,T790M,C797S mutant tumors.Using circulating tumor DNA and exogenous RNA can effectively detect the mutation types of EGFR,and choose EGFR-TKIs therapy or combined chemotherapy according to the mutation types.
6.Application of aptamer in the study of cancer stem cells

Cancer stem cells have self-renewal and multilineage differentiation potentials,which play important roles in tumorigenesis,metastasis,recurrence and resistance to chemical therapy and radiotherapy,and so on.It is the hotspot of tumor research recently.Aptamer is a small molecular oligonucleotide with high targeting ability and affinity,which is a new type of nucleic acid in the diagnosis and treatment of cancer.Now more and more research is being done on the aptamer is applied in the diagnosis and treatment of cancer stem cells,which provides a new research tool for the diagnosis and treatment of cancer stem cells.
