1.Epidemiological characteristics related to the morbidity of carcinoma of large intestine

OBJECTIVE: To probe into the effects of living habit, diet, drug and hereditary factors related to daily life so as to reduce the incidence of carcinoma of large intestine through changing the life style.DATA SOURCES: Articles on carcinoma of large intestine published between January 1980 and December 2003 were retrieved in NCBI Entrez PubMed with the key words of "carcinoma of large intestine, epidemiology and prevention" and the language restricted to English. Meanwhile, articles on carcinoma of large intestine published between January 1994 and December 2003 were searched for in CNKI database with the Chinese key words of "carcinoma of large intestine, epidemiology and prevention" and the language restricted to Chinese.STUDY SELECTION: The related literature was selected by the primary tions, hormone level and genetic factors on the morbidity of carcinoma of excluding blind method was not required. Exclusion criteria: review articles and papers of meta-analysis and replicated studies.DATA EXTRACTION: Totally 84 related articles were collected, including 17 retrieved with retrospective approach. Fifty-eight articles met the inclusion criteria, and 26 papers were excluded. Among the excluded papers, 6 papers were about the basic biological and chemical research on carcinoma of large intestine, 12 were about different pathological types of carcinoma of large intestine and distribution of hospitalized cases, 4 were meta-analysis, and 4 were of popular science and delayed diagnosis due to physicians or patients themselves. The collected data showed that the morbidity and distribution of large intestine carcinoma were related to region,age, dietary factor, verminosis, heredity, hormone and chronic intestinal diseases and long-term stress.characteristics, trace element intake and living habits on the morbidity of responding preventive measures for reducing the incidence of large intestine carcinoma.CONCLUSION: With the improvement of living standard, the occurrence .and distribution of carcinoma of large intestine increase, presenting a tendency to occur in young people. Developing good life style and dietary habit and doing moderate exercise can prevent the occurrence of carcinoma of large intestine.
2.A Meta-analysis of the incidence of influenza reported during year 2005 to 2012 in China

Objective To investigate the epidemic patterns and the characteristics of influenza in Chi-na through a Meta-analysis based on the studies published in domestic literatures.Methods Related articles published during 2005 to 2012 were screened out from domestic databases and analyzed through a Meta-analysis with Review Manager 5.0 software.Results Twenty-two articles covering 957 901 patients with influenza-like-illness (ILI) and 148 233 pathogen samples were screened out according to the inclusion criteria.No significant difference with the ILI diagnosis rate was found between subjects at age 0-4 years and those at age 15-59 years. Higher ILI diagnosis rates were observed in those two groups as compared with subjects elder than 60 years old. Most of the pathogen samples were carried by subjects aged 25-59 years.More influenza virus strains were isola-ted in 2009 as compared with those of the seven other years (OR=2.25, 95%CI=1.27-3.70).There was sta-tistical difference between the numbers of influenza A H1N1 and seasonal influenza A strains (OR=2.25, 95%CI=1.30-3.91) .Significant difference was also observed between the numbers of influenza A and influenza B strains (OR=4.05, 95%CI=2.53-6.47).Conclusion There was significant difference with the diagnosis rate between subjects aged 0-4 years and those aged≥60 years.More attention should be paid to people at high risk of infection (0-4 years old and≥60 years old) and those at 25-29 years with high mobility and social inter-course for the timely prevention and control of pandemic influenza.The detection rate of influenza virus strains was increased during the outbreak of novel influenza A H1N1 infection in 2009.After that outbreak, the detec-tion rate of novel influenza A H1N1 strains was 2.25 times the rate of seasonal influenza strains.The detection rate of influenza A was 4.05 times the rate of influenza B virus strains.Therefore, it is necessary to strengthen the surveillance for influenza A virus and other epidemic influenza virus strains.
3.A feasibility study on calculating individualized scheme of dose of iodine contrast medium in enhanced abdominal CT imaging based on liver volume measurement

Objective:To explore the feasibility and value of individualized scheme of dose of iodine contrast medium in enhanced abdominal CT imaging based on liver volume (LV) measurement.Methods:The examination data and technical parameters of 64 patients, who underwent multi-phase dynamic enhanced abdominal CT imaging from February to September 2019 at First People's Hospital of Yunnan Province were retrospectively analyzed. The density of liver on CT pre-contrast images and post-contrast images, LV and average iodine content in the liver (AII) of all subjects were measured. The difference of liver density (ΔHU), total iodine dose of contrast agent injected (IM) and total intrahepatic iodine (TII) were calculated. Pearson correlation analysis was performed to evaluate the association between AII and ΔHU, as well as IM and TII. The difference between the injection volume of contrast agent predicted by the formula and the actual injection volume of contrast agent was tested by paired t test. Results:The ΔHU and AII showed highly positive correlation ( r=0.926, P<0.05). The regression equation was Y AII=0.034X ΔHU+0.296. The TII showed significantly positive correlation with the IM ( r=0.759, P<0.05), and the regression equation was Y IM=3.649X TII+16 486.754. With the liver enhancement ΔHU=50 HU as a reference, the individually dose of contrast medium injection could be deducted based on LV, with Y IM=7.225X LV+ 16 486.754. This formula was introduced into all the subjects to calculate the predicted value of contrast agent application, and the calculated injection volume of contrast agent was (64.27± 4.92) ml. Paired t test was conducted to compare the predicted injection volume of contrast agent with the actual injection volume of contrast agent, and the difference was statistically significant ( t= 6.009, P<0.05). Conclusion:The feasibility of calculating individualized scheme of contrast agent based on LV in enhanced abdominal imaging is verified. This method can be used to predict the enhancement level of liver and to reduce the dosage of iodine contrast agent.
4.Impact of co-cultured microglia and neural stem cells via transwell on differentiation from neural stem cells into dopaminergic neurons

Objective To investigate the influence of co-culture of microglia and neural stem cells (NSCs) on differentiation from NSCs into dopaminergic neurons in vitro.Methods (1) The microglia from neonatal SD rats was purified and identified.After the NSCs were isolated from 14-day pregnant SD rats,the cells were cultured and identified.(2) The identified NSCs were randomly divided into 2 groups:simple NSCs culture group and NSCs+microglia co-culture group.After the cells in both groups were cultured for 6 days,immunofluorescence assay and Westem blotting were performed to detect the protein expression of TH,DAT and Pitx3,the factors associated with the differentiation and maturity of dopaminergic neurons.(3) PCR was used to detect the gene transcription of TH,DA T and Pitx3 in the cells from the 2 groups and differences were compared between the 2 groups.Results (1)The staining of CD1 1b/c in the microglia mixed cultured in the neonatal SD rats was positive,with a purity of above 95%;the staining of NSCs identified via nestin was positive in the 14-day pregnant SD rats.(2) The immunofluorescence assay showed that the amounts of positive proteins of TH,DAT and Pitx3 in the NSCs+microglia co-culture group were significantly larger than in the simple NSCs culture group (P<0.05).The Western blotting showed that the protein expression levels ofTH,DAT and Pitx3 in the NSCs+microglia co-culture group were significantly higher than in the simple NSCs culture group (P<0.05).(3) The PCR detection showed that the gene transcription levels of TH,DA T and Pitx3 in the NSCs+microglia co-culture group were significantly higher than in the simple NSCs culture group (P<0.05).Conclusion Co-culture of NSCs and microglia via Transwell may promote differentiation from NSCs into dopaminergic neurons.
5.Correlation between Bone Density at Different Hip Positions with Muscle Parameters and Physical Performance in Middle-aged and Elderly People in Kunming Area

Objective To investigate the correlation between bone mineral density(BMD)at different hip positions with muscle parameters and physical performance in middle-aged and elderly people in Kunming area.Methods 531 middle-aged and elderly volunteers were recruited from the Radiology Department of the First People's Hospital of Yunnan Province from May 2021 to April 2022.All study subjects completed the five-times-sit-to-stand test(FTSST)and hip quantitative CT(QCT)examinations.Volunteers'total hip(TH),femoral neck(FN),and intertrochanteric(IT)BMD were measured by using QCT PRO workstation and using OsiriX software to measure the area and density of their gluteus maximus muscle,gluteus medius,and minimus muscle and midthigh muscle.Divide male and female volunteers into positive and negative groups respectively based on FTSST time≥12 seconds or<12 seconds,and analyze the differences in hip BMD between the groups;Also divide male and female volunteers into three groups(50~59 years old,60~69 years old,70 years old and above)at the age of 10,and analyze the correlation between hip BMD and muscle parameters in different age and gender stratification.Control for age and BMI,and then perform partial correlation analysis on the above indicators.Results The BMD of the hip in the female FTSST positive group was lower than that in the negative group(P<0.001),while there was no statistically significant difference between the male groups(P>0.05).After adjusting for age and BMI,among males,FN BMD was positively correlated with gluteus medius and minimus muscle density in the age groups of 50~59 and 60~69(P<0.05),while TH BMD,FN BMD,and IT BMD were strongly positively correlated with gluteus medius and minimus muscle density in the age group over 70(P<0.05).For females,the correlation between hip BMD and muscle density in the age groups of 50~59 and 60~69 was weak,while BMD in all parts of the hip was not correlated with muscle density in the age group over 70(P>0.05).There was a negative correlation between TH BMD,FN BMD,and gluteus medius and minimus muscle area in the age group of 50~59 years old for males(P<0.05),while there was a significant negative correlation between TH BMD,IT BMD,and gluteus maximus area in the age group of 70 years old and above for females(P<0.05).Conclusion The BMD of various parts of the hip in the female FTSST positive group is lower than that in the negative group.The density of the gluteus medius and minimus muscle can to some extent serve as a predictive indicator of femoral neck bone strength in middle-aged and elderly men in Kunming area.
6. Spectrum of somatic mutations and their prognostic significance in adult patients with B cell acute lymphoblastic leukemia

Objective:
To investigate the spectrum of gene mutations in adult patients with B-acute lymphoblastic leukemia (B-ALL), and to analyze the influences of different gene mutations on prognosis.
Methods:
DNA samples from 113 adult B-ALL patients who administered from June 2009 to September 2015 were collected. Target-specific next generation sequencing (NGS) approach was used to analyze the mutations of 112 genes (focused on the specific mutational hotspots) and all putative mutations were compared against multiple databases to calculate the frequency spectrum. The impact of gene mutation on the patients’ overall survival (OS) and recurrence free survival (RFS) was analyzed by the putative mutations through Kaplan-Meier, and Cox regression methods.
Results:
Of the 113 patients, 103 (92.0%) harbored at least one mutation and 29 (25.6%) harbored more than 3 genes mutation. The five most frequently mutated genes in B-ALL are SF1, FAT1, MPL, PTPN11 and NRAS. Gene mutations are different between Ph+ B-ALL and Ph- B-ALL patients.
