Purine drugs are for the treatment of autoimmune diseases,organ transplantation,acute lymphoblastic leukemia.The adverse reaction rate is 15% ~ 28%,which impacts on the clinical application in recent years.Studies have shown that inosine triphosphate pyrophosphatase (ITPA) are different between individuals,and there are adverse reactions in patients with defects of ITPA when purine drugs are used.This paper reviewes the ITPA pharmacogenetic research progresses.
2.Progress of cyclophosphamide pharmacology and therapy of maligant tumors in children

Cyclophosphamide is a widely used drug in clinic,which is a non-specific cell cycle nitrogen mustard cytotoxic drug.It is mainly used in the chemotherapy of malignant tumors and autoimmune diseases.Recent studies have found that its differences of metabolism in vivo are connected with the differences of individual genetic level.Some drugs can enhance the anti-tumor effect of cyclophosphamide.There age some progress in the treatment of acute leukemia and lymphadenoma in children.

Guidelines for the Prevention of Stroke in Women released by the American Heart Association/American Stroke Association on February 6,2014,and this makes us have further reflection on the risk factors for ischemic stroke and prevention in women.Compared with men,some of risk factors are unique for women or more common in the female population.The in-depth understanding of the risk factors is essential for the prevention of ischemic stroke in women.
4.The value of super-selective uterine arterial embolization in the treatment of massive postpartum hemorrhage

Objective To investigate the technical success rate of operation,safety,efficacy and clinical value of super-selec-tive uterine arterial embolization(SSUAE)in the treatment of massive postpartum hemorrhage.Methods The clinical data of 65 patients with massive postpartum hemorrhage who were not cured by medical treatment were retrospectively analyzed.Forty-one patients who underwent successful SSUAE were set as research team (group A)and 24 patients who underwent hysterectomy were set as control group (group B).The indications,success rate of hemostasis,bleeding volume and postoperative complications were compared.Results In group A,1 1 patients were in a critical condition and couldn′t tolerate the hysterectomy (26.8%),success rate of hemostasis was 95.1% (39/41 ),and 2 patients who with a poor outcome of embolization underwent hysterectomy after SSUAE (4.9%).In group B,the success rate of hemostasis was 95.8% (23/24),there was no statistical significance of hemostasis success rate between the two groups (P >0.05).There was significant statistical significance between the two groups in average operation time and bleeding volume in surgery,which were 33.8 min and 61.5 min,2 1 55.5 mL and 3 21 5.7 mL respectively(P <0.01).No postoperative complications occurred in group A,but 3 postoperative complications occurred in group B,which had signif-icant statistical significance (P <0.01).Conclusion SSUAE performed its excellent advantages,such as wide indications,minimal trauma,high success rate of hemostasis,low complications and reach the purpose of reserving uterus.
5.Assessment of Therapeutic Effects of Neovascular Glaucoma in Treatment Period of 28 Cases

To summarize the etiological factor and to assess the therapeutic effect for neovascular glaucoma(NVG).Methods A retrospective study of 28 cases (29 eyes) with NVG was performed. The mean age, proportion of sex and primary ocular disease, incidence of symptom, sign and postoperative complication were analyzed. Results 78 6% of the patients were over 40 years old. 62% of the patients were secondary to the ocular ischemic disease such as central retina vein occlusion and proliferative diabetic retinopathy. The clinical characters included intraocular pressure (IOP) increasing, pain, mixed conjunctival congestion and rubeosis iridis. The mean IOP reduction was 19.5 mmHg in 29 eyes after antiglaucomatous drags treatment and 19.13mmHg in 23 eyes after Hunan aqueous humor drainage (HAD) implantation. The common postoperative complications were flat anterior chamber and choroidal detachment. The success rate of HAD implantation was 87%.Conclusions NVG was commonly secondary to ocular ischemic disease. Although prevention was the principal, reducing IOP and ameliorating ischemic situation were as the same important.

7.Observation of the effect of Huoxue Tongmai decoction in the treatment of coronary heart disease with ;angina pectoris

Objective To observe the efficacy of Huoxue Tongmai decoction in the treatment of coronary heart disease with angina pectoris.Methods 60 coronary heart disease patients with angina pectoris were randomly divided into two groups,30 cases in each group.The observation group was treated with Huoxue Tongmai decoction concomitant with routine chemical medicine therapy,and the control group was treated with only routine chemical medicine therapy.The total effective rate,reduced angina occurrences after therapy and ECG were observed in both two groups.Results After treatment,the total effective rate of the observation group was 93.3%,which was signifi-cantly higher than that of the control group(χ2 =4.32,P <0.05).Compared with the two groups of ECG efficacy,the total effective rate of the observation group was 83.3%,which was significantly higher than 66.7% of the control group (χ2 =4.22,P <0.05).After one,three treatment courses,the number of angina attack in the observation group was significantly lower than that in the control group,and the differences were statistically significant(t =6.99, 10.82,all P <0.05).Conclusion Houxue Tongmai decoction combined with routine chemistry medicine is more effective than routine chemistry medicine in treating angina.
8.Computer-aided design of anatomic locking plate for the posterior malleolus

Objective To design an anatomic locking plate for the posterior malleolus using 3D reconstruction CT of the ankle joint and software Mimics.Methods The CT data of 20 normal ankles were imported into software Mimics in.dicom format for 3D reconstruction of the ankle joint.The angles of posteromedial and posterolateral varus,the twist height on the sagittal plane and the height of safe screw placement zone,the width of the posterior malleolus were measured for the posterior malleolus.After the anatomic locking plate was designed using the anatomic data measured and software Ungraphics NX5.0,it was 3D printed in a proportion of 1:1.Next,the model plate was tested and evaluated in 15 fresh cadaveric specimens of the ankle all the soft tissues of which were removed to fully expose the bony structure of the posterior malleolus.Results The anatomic measurements showed that the angles of postemmedial and posterolateral varus increased gradually towards the medial and lateral sides along the central axis of the posterior malleolus while the twist height reduced from the lateral to the medial side,and that the width of the posterior malleolus increased gradually from the superior to the inferior.It was found that after placement in the 15 fresh cadaveric specimens the anatomic model plate fit the posterior malleolus very well,fulfilling all the designed requirements for the anatomic locking plate for the posterior malleolus.Conclusion A novel anatomic locking plate for the posterior malleolu has been successfully designed and manufactured using 3D reconstruction CT of the ankle joint,software Mimics and 3D printing.
9.Negative immune regulatory molecule TIPE2 for treating SLE mice through regulating macrophage subtype

Objective To investigate the role of tumor necrosis factor (TNF)-alpha-induced protein 8-like 2 (TIPE2) for regulating the macrophage polarization in systemic lupus erythematosus and its curative effects on experimental SLE mice.Methods The mice were treated with activated lymphocytes derived DNA (ALD-DNA) for inducing mice model,randomly divided into AAV-scr control group and AAV-TIPE2 experimental group,and injected with AAV-TIPE2 or AAV-scr virus solution from the tail vein of mice.The expression of TIPE2 mRNA and protein in polarized macrophages,serum dsDNA antibody titer,urine protein and renal pathological index were detected.Results (1) The TIPE2 expression level of TIPE2 mRNA and protein in AAV-TIPE2-transfected cells was 13.5±1.6 times and 10.8±1.6 times of AAV-scr control group respectively.(2) M2 macrophage specific molecule MGL+ was 59.6% in AAV-TIPE2 group and MGL + cells in the AAV-scr group was 8.4%.M2/M1 odds ratio of AAV-TIPE2 experimental group to AAV-scr control group was 16.(3) The recombinant TIPE2 adenovirus related vector could stably expressed in transfected HEK-293.In vitro and in vivo experiments confirmed that AAV-TIPE2 was able to induce M2 polarization of macrophages in ALD-DNA-induced lupus mice.(4) The serum anti-dsDNA antibody,urinary protein and renal pathology in the AAV-TIPE2 group were significantly lower than those in the AAV-scr group(P<0.01).Conclusion TIPE2 alleviates the disease condition of ALD-DNA induced SLE mice through induction of macrophage polarization to M2 phenotype,which may be used as a promising therapeutic method for ALD-DNA induced SLE mice.
10.Effect of immunosuppression on the prevention of calcification in rat aortic valved homograft

Objective Calcification and graft failure may occur in aortic valved homografting as a result of immune rejection. This study was designed to examine the effects of immunosuppression in reducing immune rejection response and preventing calcification in rat aortic valved homograft(AVH). Methods Animals were randomly assigned into 3 test groups and 1 control group. SD-Wistar rats in the test groups received AVH of cryopreservation (group A), received AVH of cryopreservation followed by CsA treatment (group B), and received AVH preconditioned with monoclonal antibody against dendritic cells (DcmAb) diluted in 1:5 and cryopreserved then were treated with DcmAb postoperatively (group C), respectively. Rats in the control group(group D) were Wistar→Wistar.Rat AVH with myocardial cuff were transplanted into the abdominal aorta of rats. The rats were sacrificed in batchs at 2, 4, 8, 12 and 16 weeks postoperatively. Blood samples were obzained for detecting the expression of TCR-αβ, CD28. The AVH specimens were obtained for observing the change of endotheliocyte and smooth muscle cells with light and electron microscopes. At the same time, calcium contents of AVH tissues after transplantation were measured. Results As compared with the control group, the expression of TCR-αβ and CD28 in test groups was increased significantly at each time point(P<0.01) and reached peaks 2 to 4 weeks after operation, then gradually reduced and was closed to the level of controls at 12 weeks. Calcium contents of AVH tissues in the test groups was increased gradually since 4 weeks and reached the peak 12 to 16 weeks after operation. No significant difference in calcium level was found in the control group over 5 different periods (P>0.05). Calcium contents of AVH tissues at 4 and 8 weeks postoperatively were (2856±79)μg/g and (3587±168)μg/g in the groups with cryopreservation;(2518±73)μg/g,(3237±187)μg/g in the CsA treatment group;(2176±210)μg/g, (3089±176)μg/g in the DCmAb treatment group; (860±60) μg/g, (870±50) μg/gin the control group. Conclusion The immunosuppressive treatment had substantial effects on AVH calcification as a result of reduction in immune rejection response and delay in the development of calcification.
