1.Comparison of the effect of tigrine and clopidogrel in PCI in patients with STEMI

Objective To comparison of the effect of tigrine and clopidogrel in PCI in patients with STEMI .Methods 118 cases of routine PCI in patients with acute ST segment elevation myocardial infarction from March 2014 to May 2014 in our hospital were select, according to random number table method to all of the patients were randomly divided into clopidogrel group (group A) and tigrine group(group B), preoperative group A received 300 mg of aspirin combined with 600 mg of clopidogrel for treatment,group B received 300 mg aspirin combined with 180g of tigrolipine for preoperative treatment.Compared the two groups of patients with intraoperative infarct-related artery myocardial infarction thrombolytic test three blood flow and 1.5 hours after ST segment fall, postoperative heart B in the situation, the incidence of dyspnea, adverse cardiac events, bleeding events.Results The incidence of ST segment depression and the incidence of ST segment depression in group B were significantly higher than those in group A (t=12.234,χ2 =7.829,P<0.05).One month after surgery, B group of patients with LVEF value was significantly higher than the A group (t=5.393,P<0.05);there was no significant difference in LVDD between the two groups.The incidence of dyspnea and immediate TIMI grade 3 blood flow in group B were 24.6% and 88.5%, respectively, which were significantly higher than those in group A and 8.8% and 68.4% (χ2 =4.293,χ2 =4.392,P<0.05). The incidence of adverse cardiovascular events was 19.3% in group A, which was significantly higher than that in group B (χ2 =2.193,P<0.05). There was no significant difference in the incidence of bleeding between the two groups.Conclusion The use of STEMI patients underwent PCI surgery ticagrelor treatment, in case of the risk of bleeding can effectively improve the patient's myocardial reperfusion and prognosis does not increase , the increased incidence of immediate thrombolysis in myocardial infarction patients in the three grade .
2.Effects of chitosan on experimental rabbit atherosclerosis

Objective:To investigate the effects of chitosan on experimental rabbit atherosclerosis(AS).Methods:The models of rabbit AS were produced by a high fat diet.The rabbits were randomly assigned into normal diet(ND) group,high fat diet(HFD) group,chitosan and high fat diet(CHFD) group,chitosan and normal diet(CND) group.Fasting blood samples were collected for serum lipid assay on 0,4,8 weeks;the animals were killed on the 8th week to make aorta samples.AS plaque areas were estimated in the descending aorta dyed with oil red O and the ascending aorta dyed with H E.Results:TG,TC,LDL C in the CHFD group were lower than those in HFD group on 4 and 8 weeks;serum lipid concentration in ND group was similar with that in CND group;plaque areas in CHFD group were smaller than those in HFD group;intima thickness in CHFD group was thinner than that in HFD group.Conclusion:Chitosan reduces AS degree in rabbits of HFD group,which might be related to the decrease of serum lipid.
3.Effects of platelet-derived growth factor on vasoconstriction of the vascular vessels in rats

AIM: To investigate the effects of platelet-derived growth factor BB (PDGF-BB) on vasoconstriction of the vascular vessels. METHODS: The contractile effects of PDGF-BB at different concentrations on the rat thoracic aortic rings and the effects of verapimil, indomerthacin, phentolamine and propranolol on the response of the thoracic aortic rings were observed by using the technique of vascular perfusion in vitro, and norepinephrine was used to severe as control. RESULTS: PDGF-BB showed a remarkable contractile effect on the thoracic aorta rings of rats in a dose-dependent manner, more potent than norepinephrine at the same molar concentration. Verapimil and propranolol inhibited the contractile effect of PDGF-BB on the rat thoracic rings, but the effect of indomerthacin and phentolamine on the vasoconstriction of PDGF-BB was not observed. CONCLUSION: PDGF-BB significantly contracts the thoracic aorta rings of rats in dose-dependent and non-endothelium-dependent manners. Verapimil and propranolol inhibits the contractile effect of PDGF-BB on the rat thoracic rings.
4.Value of peripheral blood PCT, CRP, FIB and D-D levels for early diagnosing PTB complicated with bacterial pneumonia

Abstract: Objective To investigate the early diagnostic value of peripheral blood procalcitonin (PCT), C-reactive protein (CRP), fibrinogen (FIB) and D-dimer (D-D) levels in patients with pulmonary tuberculosis (PTB) complicated with bacterial pneumonia. Methods A total of 102 patients who admitted to Department of Tuberculosis of Affiliated Nantong Hospital of Shanghai University from Jan 2021 to May 2022 were enrolled in this study and divided into a group (52 cases) with pulmonary tuberculosis (PTB) patients and a group (50 cases) with PTB patients complicated with bacterial pneumonia. The levels of PCT, CRP, FIB and D-D in the peripheral blood were measured, the differences and correlations in all indicators were compared among two groups. The sensitivity and specificity of these indicators in the early diagnosis of PTB complicated with bacterial pneumonia were analyzed by receiver operating characteristic (ROC) curve. Results The levels of PCT, CRP, FIB and D-D in the peripheral blood from the PTB complicated with bacterial pneumonia group were 0.06 (0.04, 0.16) ng/mL, 38.00 (3.88, 96.10) mg/L, 4.51 (3.02, 6.07) g/L, and 0.59 (0.34, 1.88) mg/L, respectively, which were significantly higher than corresponding 0.04 (0.03, 0.04) ng/mL, 3.20 (0.84, 7.22) mg/L, 2.96 (2.48, 3.77) g/L, and 0.27 (0.17, 0.36) mg/L in the PTB group (Z=-4.784, -5.233, -3.853, -4.199, all P<0.001). Furthermore, the levels of CRP and FIB in the PTB complicated by bacterial pneumonia group were highly positively correlated (r=0.855, P<0.001). The area under the ROC curve (AUC) of PCT, CRP, FIB and D-D for early diagnosis of PTB complicated with bacterial pneumonia were 0.757, 0.794, 0.747 and 0.764, respectively. In addition, the AUC obtained by simultaneous measurement of PCT, CRP, FIB and D-D was as high as 0.916, and the sensitivity and specificity of diagnosing PTB complicated with bacterial pneumonia were increased to 85.7% and 96.9%, respectively, which were higher than those of individual indicators. Conclusions Levels of peripheral blood PCT, CRP, FIB, and D-D all show varying degrees of increase in patients with PTB complicated with bacterial pneumonia, and detecting the levels of all four markers, rather than any single marker, can assist in early monitoring whether the tuberculosis patients are complicated with bacterial pneumonia.
5.Dayuanyin Regulates TLR/MAPK/NF-κB Pathway for Preventing and Treating Acute Lung Injury Induced by H1N1 Infection

ObjectiveTo investigate the therapeutic effect of Dayuanyin on acute lung injury induced by H1N1 infection and decipher the potential mechanism. MethodThe constituents in Dayuanyin were analyzed by ultra-high performance liquid chromatography-quadrupole-exactive orbitrap mass spectrometry (UHPLC-Q-Exactive Orbitrap MS). Forty-eight female BALB/c mice were randomized into normal, model, oseltamivir (19.5 mg·kg-1), and low-, medium-, and high-dose (2.73, 5.46, 10.92 g·kg-1) Dayuanyin groups. The normal and model groups were administrated with deionized water by gavage, and the other groups were administrated with the corresponding drugs by gavage. On day 3 of drug administration, the normal group received nasal inhalation of normal saline, and the other groups were inoculated intranasally with A/RP/8/34 (H1N1) for the modeling of influenza virus infection. Mice were administrated with drugs continuously for 7 days and weighed daily. Sampling was performed 12 h after the last administration, and the lung tissue was weighed to calculate the lung index. Hematoxylin-eosin staining was performed to observe the pathological and morphological changes of the lung tissue and bronchi. The cytometric bead array (CBA) was used to measure the serum levels of interferon-gamma (IFN-γ), C-X-C motif ligand 1 (CXCL1), tumor necrosis factor-alpha (TNF-α), chemokine ligand 2 (CCL2), interleukin-12p70 (IL-12p70), chemokine ligand 5 (CCL5), interleukin-1β (IL-1β), chemokine (C-X-C motif) ligand 10 (CXCL10), granulocyte-macrophage colony-stimulating factor (GM-CSF), interleukin-10 (IL-10), interferon-beta (IFN-β), interferon-alpha (IFN-α), and interleukin-6 (IL-6). According to the results of mass spectrometry and network pharmacology, we analyzed the mechanism of Dayuanyin in treating acute lung injury caused by H1N1. The protein levels of extracellular signal-regulated kinase 1/2 (ERK1/2), p38 mitogen-activated protein kinase (p38 MAPK), nuclear factor-kappa B (NF-κB), and their phosphorylated forms were determined by Western blot. The mRNA levels of myeloid differentiation factor 88 (MyD88), Toll-like receptor 3 (TLR3), Toll-like receptor 7 (TLR7), and Toll-like receptor 8 (TLR8) in the lung tissue were measured by Real-time fluorescence quantitative polymerase chain reaction (Real-time PCR). ResultA total of 57 compounds, including paeoniflorin and baicalein, were detected in Dayuanyin. Compared with the normal group, the model group showed decreased body weight (P<0.01), lung edema and hemorrhage, increased lung index (P<0.01), and elevated levels of IFN-γ, IL-12p70, CCL5, IL-1β, CXCL10, GM-CSF, IFN-β, and IL-6 (P<0.01). Compared with the model group, Dayuanyin attenuated alveolar wall thickening, capillary congestion, and immune cell infiltration, reduced the alterations in body weight and lung index (P<0.01), and down-regulated the protein levels of IFN-γ, IL-12p70, CCL5, IL-1β, CXCL10, GM-CSF, IFN-β, and IL-6 (P<0.01). A total of 57 key genes were predicted by network pharmacological analysis, of which the MAPK signaling pathway was the main target signaling pathway. Compared with the normal group, the model group showed up-regulation in the protein levels of phosphorylation (p)-ERK1/2, p-p38 MAPK, and p-NF-κB (P<0.01) and the mRNA levels of TLR7, TLR8, MyD88, and TLR3 (P<0.05, P<0.01). Compared with the model group, Dayuanyin lowered the phosphorylation levels of ERK1/2, p38 MAPK, and NF-κB p65 in a dose-dependent manner (P<0.01) and down-regulated the mRNA levels of TLR3, TLR7, TLR8, and MyD88 (P<0.01). ConclusionDayuanyin can prevent and control H1N1 infection-induced acute lung injury by inhibiting the TLR/MAPK/NF-κB signaling pathway.
