Objective: To ascertain the factors and levels in the orthogonal design by drawing the extraction amount time curve of total alkaloid of Rhizoma Coptidis in different solvents. Methods:The extraction amount time curve of total alkaloid was plotted with the extraction amount of total alkaloid as a marker. The orthogonal design was used to select the optimum extraction process according to the contents of total alkaloid and berberine, and extract yield. Results:The extraction amound of total alkaloid related to volume and kinds of solvent and extraction time. The optimum exttraction process was as follows: adding 50% alcohol into meterials and refluxing and extracting for 3 times, 120 min every time. Conclusion:The investigation on the relation between total alkaloid extraction amount and time is of an instructive significance in orthogonal design. The optimum extraction process of Rhizoma Coptidis will provide an experimental basis for industrial production.

Objective: To ascertain the condition for extracting tetramethylpyrazine from Rhizoma Chuanxiong with alcohol. Methods: With the transferring rate of tetramethylpyrazine as index, the optimum extraction condition for the first time was selected by uniform test, and the times of extraction was studied. Results: Taking 80% alcohol as solvent, Rhizoma Chuanxiong was extracted two times with 8 and 6 times volume of alcohol for 2hr and 1.5hr, respectively. In this condition, tetramethylpyrazine could be effectively extracted. Conclusion: The experimental results provided the basis for the ascertainment of extraction process of Rhizoma Chuanxiong preparation.

Objective To evaluate the different effect of 50 g glucose challenge test (GCT) on screening the glucose intolerance at different gestational age. Methods Two thousand pregnant women were divided into 2 groups(1000 in each). Women in group A received 50 g GCT at 14 and 28 gestational weeks respectively and 75 g oral glucose tolerance test (OGTT) were performed in those cases with abnormal 50 g GCT. 50 g GCT was performed after fasting at 14 gestational weeks and repeated one hour after a meal at 28 gestational weeks. Women in group B were screened by 50 g GCT at 28 gestational weeks and followed by 75 g OGTT for those with abnormal 50 g GCT. The diagnostic effect of 50 g GCT for screening at different weeks of gestation with different ways was evaluated. Objective To investigate the clinical significance and safety of liquid-based cytology (thinprep cytologic test, TCT ) in pregnant women and cytological characteristics of the smears during pregnancy. Methods TCT were performed in 1951 pregnant women (group A), 3752 gynecological out-patients(group B)and 5069 routine screening women (group C ). The age of the women was ranged from 20 to 39 years. The cytological preparations were analyzed using TCT method following the Bathesda system (TBS) standard. Results (1) The incidence of abnormal epithelial patterns in groups A, B and C was 23.58%(460/1951), 19.22%(721/3752) and 13.40%(679/5069) respectively (P0.05). Conclusions TCT smear screening is necessary, safe and acceptable for routine prenatal tests. There is no difference in the cervical squamous intraepithelial lesions between pregnant and non-pregnant women.

Objective To observe the effects of phlorohlucinol on cervical dilatation. Methods Totaling 250 normal parturients were randomized into four study groups (50 in each group) and control group (n=50). Phloroglucinol was given in the study groups(group1,2,3 and 4)at the dose of 40 mg to the cervix and/or 160 mg and 200 mg intravenously during the latent phase. The control group (group1) received only atropine (0.5 mg) intravenously. Results The overall speed of cervical dilatation in the study group was (2.82?1.82) cm/h and (1.78?1.01) cm/h in the control group. Objective To observe the effects of diazepam on the cervix in relieving spasm, eliminating edema and improving dilation during the latent phase of labor. Methods One hundred normal parturients were randomly devided into study group ( n =50) and control group ( n =50). The study group was injected diazepam 10 mg in the cervix during the latent phase. The control group was given 10 mg of diazepam by intravenous injection. Results The dilating speed of cervix in study group was 2.81?1.92 cm/h. The mean time of the first stage of labor was 11.13?1.21 h, while the control group was 1.98?0.92 cm/h and 13.34?0.44 h respectively. Objective To investigate the characteristics of marital quality and social support status of the patients with breast cancer during the period of chemotherapy and the correlation of both factors.Methods Seventy-seven patients with breast cancer were divided into beginning-, middle-and final-staging groups of chemotherapy.And they were studied with Olson marital quality questionnaire and Social Support Rating Scale.Results The total scores of marital quality and other factors were the lowest in the mid-chemotherapy group.Among three groups,the mean of the objective support and availability were gradually increased with the chemotherapy going.The score of objective support was higher in the final than beginning chemotherapy groups (P =0.009).There was a positive correlation between marital satisfaction and the objective support (r =0.248, P =0.036), and between the total score of quality of marriage and subjective support (r =0.286, P = 0.015).Conclusion The marital quality of breast cancer patients receiving chemotherapy is poor,and subjective or and objective support might improve the patient's marital quality.

Objective To investigate the effects of angiotensin1-7 (Ang1-7) on renal tubulointerstitial fibrosis of diabetic rats.Methods Thirty-two male Wistar rats were randomly divided into four groups: normal control group,diabetic group,telmisartan group,Ang1-7-treated group.For 9 weeks after diabetes mellitus model established,24 h proteinuria,urine NAG/Cr,glucose,insulin,TG,TC,BUN,Scr,Na+ and K+ were assessed.Renal pathological changes were evaluated by PAS staining; Expression of TGF-β1,PPARγ and α-SMA mRNA was deteeted by real-time PCR; Protein levels of PPARγ,α-SMA and TGF-β1 were detected by Western blotting.Results (1)At the end of the ninth week,the blood pressure,proteinuria,renal weight/body weight in group DM were significantly higher than those in group NC (P＜0.05).(2)Renal interstitial fibrosis in group DM was obviously severe as compared to group NC (P＜0.05),but was improved in group TM and group T(P＜0.05).(3)TGF-β1 and α-SMA mRNA in group DM were significantly increased,and PPARγ mRNA was significantly decreased.Compared with group DM,TGF-β1 and α-SMA mRNA were significantly decreased,and PPARγ mRNA was significantly increased in group TM and group T,especially in group T.(4)TGF-β1 and α-SMA in group DM were significantly increased,and PPARγ decreased significantly.Compared with group DM,TGF-β1 and α-SMA decreased significantly,PPARγ increased significantly in group TM and group T,especially in group T.Conclusion Ang1-7 inhibits high glucose-induced α-SMA expression in vivo through up-regulating the PPAR expression and may inhibit renal tubulointerstitial fibrosis of diabetic rats.

The culture of umbilical cord-derived mesenchymal stem cells is extremely important for studies on umbilical cord mesenchymal stem cells. Optimization of cellculture technology is crucial for clinical application of mesenchymal stem cells and even celltherapy. Meanwhile, the labeling and tracer technique of umbilical cord-derived mesenchymal stem cells is a hotspot in stem celltransplantation. OBJECTIVE:To review the research and development of the cellmarkers and tracer methods of umbilical cord-derived mesenchymal stem cells. METHODS:A computer-based search of VIP, CNKI, Medline, Highwire and Foreign Journals Integration System databases was performed for articles concerning culture and labeling of umbilical cord-derived mesenchymal stem cells published from January 2001 to October 2013. The keywords were“stem cells, mesenchymal stem cells, umbilical cord-derived mesenchymalstem cells, cellculture, labeling methods”in Chinese and English, respectively. Final y, 35 articles were included in result analysis. RESULTS AND CONCLUSION:Umbilical cord-derived mesenchymal stem cells have not yet been widely used, mainly because of the immature isolation, culture and staining techniques of umbilical cord-derived mesenchymal stem cells. These techniques are worthy of further optimization studies. Although in recent years, cellmarkers and tracer technology of umbilical cord-derived mesenchymal stem cell s have made great progress, there are stil many problems need to be solved.

Objective To study the expression and significance of cyclin A1 in the skin of wild-type mice at RNA level.Methods Thirty 6-12-week old wild-type Kunming mice(15 male and 15 female)were included in this study.In situ hybridization was used to detect the expression of cyclin A1 mRNA in the skin of head and neck of the mice.The skin treated without probe was regarded as negative control and the testis of male mice was taken as positive contr01.Results The expression of cyclin A1 mRNA was found in sebaceous glands of 25 mice and in epidermis of 12 mice.Strong positive staining of sebaceous glands was seen in 50% and positive staining in 33.3% of sections,whereas strong positive and positive staining of epidermis was seen in 13.3% and 20% of sections.respectively.The positive rate of sebaceous glands was 83.3%,much higher than that of epidermis(33.3%).Conclusions There is a quite high expression of eyelin A1 mRNA in the skin sebaceous gland and epidermis of head and neck of wild-type mice.Especially a strong expression is in the sebaceous glands.It indicates that cyclin A1 mRNA may play a certain role in the physiological function of sebaceous glands and epidermis of the skin.
