1.Breast cancer metastasis suppressor gene 1 correlates with progression of cancer: a Meta-analysis

Objective To evaluate the association between the expression of breast cancer metastasis suppressor gene 1 (BRMS1) and the progression of cancer,and provide the basis for predicting the occurrence and metastasis of cancer.Methods Articles about BRMS1 with the clinical research literatures of cancer from PubMed,EMBase,China National Knowledge Infrastructure,Wanfang and VIP database were retrieved.The association between the expression of BRMS1 and tumorigenesis was evaluated by odds ratio (OR).The RevMan 5.3 software was used to complete the meta-analysis.Results A total of 27 articles were included,involving 2 282 cases in cancer group and 1 236 cases in the control group.The meta-analysis results showed that BRMS1 expression was significantly upregulated in cancer lesions compared with paired non-cancer (OR =0.10,95% CI:O.08-0.14,Z =15.37,P <0.000 O1).In the subgroup about the type of cancers,there was statistically significant difference in the pooled OR between breast cancer and the control group (OR =0.10,95% CI:0.05-0.20,Z =6.18,P < 0.000 01).Conclusion The level of BRMS1 is negatively correlated with tumorigenesis including breast cancer.Low or absent BRMS1 expression in the tumor tissue may be an adverse predictive factor of tumorigenesis and metastasis.
2.Analysis of genetic structure and gene flow of HLA class Ⅱ in 11 groups of the Chinese Nation

Objective: To ascertain the gene flow tracks and patterns and the origin of HLA class Ⅱ in the Chinese Nation.Methods : HLA classⅡ in 11 Chinese groups and 5 reference populations was studied by clustering analysis of genetic structure and distribution comparison of gene frequencies.Results and Conclusion: These analyses revealed that these genes decreased southwards and originated probably in Caucasoid or Negroids (DQA 1*0501, DQB1*0501, etc.),and those decreased northwards and originated probably in Southeast Asia (DQA1* 0601, DRB1*1202, etc.).It was found that these genes decreased both southwards and northwards (DQB1*0303, DRB1*0901,*1501,etc.) and originated probably in certain groups in mainland of Chinese Nation,and that HLA genes in different Chinese groups migrated from each other and amalgamated deeply,but Dais, Buyis etc were isolated from outside in some degree. Our results also supported that the Chinese Nation comprises two major populations of north and south.We proposed that the migration and mixing are the dominant driving force of the HLA gene flow while the genetic drift caused by isolation and the nature selection have an important influence on HLA genetic structure.
3.Mesenchymal stem cell transplantation for intracerebral hemorrhage

The onset of intracerebral hemorrhage is often sudden arid dangerous with high mortality. At present, it lacks truly effective theraputic measures and medications. Mesenchy-mal stem cell has multilineage differentiation potential induced trader a specific micro-environment, and it can be successfully induced and differentiated into osteoblasts, cartilage cells, myocardial cells, neurons, glial cells, and so on under the different micro-environment conditions. It has brought a new hope for the treatment of intracerebral hemorrhage.
4.SPIO and DAPI double labeled bone marrow mesenchymal stem cells of macaques:effects on cell viability and proliferation

BACKGROUND:Traditional cel transplantation tracer methods require histological analysis and identification in vitro, which limits the clinical application of stem cel transplantation. So it is urgent to establish an in vivo noninvasive and repeatable tracer method. OBJECTIVE:To observe the effect of SPIO and DAPI double labeling on survival and proliferation of bone marrow mesenchymal stem cel s from macaques. METHODS:Bone marrow mesenchymal stem cel s were derived from bone marrow aspirates of healthy macaques using whole bone marrow adherence method. Then, the cel s were identified using flow cytometry detection. Bone marrow mesenchymal stem cel s were labeled using SPIO and DAPI. Fluorescent microscope was used to detect DAPI positive rate, and Prussian blue staining and transmission electron microscope were employed to measure SPIO positive rate. MTT assay was used to detect cel viability and proliferation. RESULTS AND CONCLUSION:Bone marrow mesenchymal stem cel s were successful y isolated from healthy macaques using the whole bone marrow adherence method, and the cel purity was up to 95.1%. SPIO and DAPI were both successful to label the bone marrow mesenchymal stem cel s with a positive rate of 95%-98%, but had no influence on cel viability and proliferation.
5.Stem cell transplantation for inflammatory bowel diseases

BACKGROUND:Stem cell transplantation can, in theory, rebuild the immune system and regulate immunologic function. In addition, the transplanted stem cells may differentiate into tissues or cells we want. Therefore, this intervention may restore gastrointestinal inflammatory lesion and exert therapeutic effect on inflammatory bowel diseases (IBD). OBJECTIVE:To review the application of different kinds of stem cells in IBD treatment. METHODS:A computer-based search of CNKI, PubMed and FMJS was performed for articles concerning stem cel transplantation and IBD published after January 2002. The key words were stem cells, inflammatory bowel diseases, Crohn's diseases, ulcerative colitis, cell therapy in Chinese and English respectively. At last, 49 articles were included in result analysis. RESULTS AND CONCLUSION: Embryonic stem cells and adult stem cells both have certain therapeutic benefits in IBD. But which kind of stem cell is the safest and most efficacious cannot be confirmed, and at the same time the most effective and the optimal dose and way of transplantation of such cells are still unclear. All in all, there is still a lot of work to do to remove the obstacles.
6.Underlying mechanism of bone marrow mesenchymal stem cells in the repair of radiation-induced hematopoietic injury

BACKGROUND:Co-transplantation of bone marrow mesenchymal stem cells (BMSCs) and hematopoietic stem cells (HSCs) can improve the survival rate following radiation damage. OBJECTIVE:To review the mechanisms of BMSCs in hematopoietic reconstitution and immunomodulation after radiation damage. METHODS: Relevant articles included in the Web of science from 2005 to 2016 were retrieved, and the keywords were mesenchymal stem cells; radiation; radiation injury; hematopoietic stem cells; hematopoietic reconstitution; immunomodulatory in English. Then we sorted and analyzed the articles which were closely related to the theme. RESULTS AND CONCLUSION:BMSCs can repair broken DNA double bonds by autophagy and HR/NHEJ pathway; support and protect HSCs by intercellular interactions and paracrine action; and repair radiation-induced hematopoietic injury by inhibiting inflammation and immune responses to maintain HSC homeostasis. Therefore, BMSCs may function as an effective treatment for radiation-induced hematopoietic injury.
7.Effect of paclitaxel on telomerase activity, apoptosis and p53/bcl-2 gene protein expression of breast cancer MCF-7 cell line

Objective To investigate effect on telomerase activity, apoptosis and p53/bcl-2 gene expression of MCF-7 cells line induced by paclitaxel.Methods By techniques of cell culture in vitro, telomeric repeat amplification protocol with ELISA(TRAP-ELISA) and flow-cytometry (FCM), MCF-7 cell line was treated by paclitaxel in various concentration for 72h.Results Paclitaxel down-regulated telomerase activity of MCF-7 cell, induced apoptosis of the cell in a concentration-dependent manner, significantly decreased expression of bcl-2 gene protein and increased expression of p53 gene protein. There was a positive correlation between telomerase activity and apoptosis and the expression of p53/bcl-2 gene protein.ConclusionPaclitaxel could down-regulate telomerase activity,induce apoptosis, decrease expression of bcl-2 gene protein and increase expression of p53 gene protein, which may be one of important mechanisms of Paclitaxel′s anticancer action.
8.HLA-peptide tetramers and adoptive immunotherapy in prevention of cytomegalovirus disease

BACKGROUND: Antiviral drugs can reduce the incidence of early-onset cytomegalovirus(CMV)disease,but are associated with strong toxicity and the development of late-onset CMV disease.In order to prevent CMV disease better,cytotoxic T lymphocytes(CTL)may play a critical role in controlling CMV reactivation.Fluorescent HLA-peptide tetramers are used to monitor the recovery of CMV CTL in recipients of allogeneic transplants.OBJECTIVE: To explore the effect of HLA-peptide tetramers and adoptive immunotherapy in treating CMV disease.METHODS: A computer-based online search of Pubmed and Wanfang databases was performed for articles related to CTL detection,application of antiviral drugs and HLA-peptide tetramers,and adoptive immunotherapy with key words"HLA-peptide tetramers,cytomegalovirus,specific CTL,adoptive immunotherapy"in English and Chinese.Repetitive articles were excluded and 29 articles were included.RESULTS AND CONCLUSION: Adoptive immunotherapy with CMVs cytotoxic T cells as preemptive therapy is a very elegant strategy; however,generation of these cells is costly and time-consuming,and therefore the therapy is not available at every transplantation center.Magnetic selection of CMVs CD8+T cells from peripheral blood of CMV-seropositive donors by using HLA-peptide tetramers may be very hopeful,which simplifies adoptive immunotherapy.
9.Umbilical cord-derived mesenchymal stem cell culture:dyeing and tracer technique

The culture of umbilical cord-derived mesenchymal stem cells is extremely important for studies on umbilical cord mesenchymal stem cells. Optimization of cellculture technology is crucial for clinical application of mesenchymal stem cells and even celltherapy. Meanwhile, the labeling and tracer technique of umbilical cord-derived mesenchymal stem cells is a hotspot in stem celltransplantation. OBJECTIVE:To review the research and development of the cellmarkers and tracer methods of umbilical cord-derived mesenchymal stem cells. METHODS:A computer-based search of VIP, CNKI, Medline, Highwire and Foreign Journals Integration System databases was performed for articles concerning culture and labeling of umbilical cord-derived mesenchymal stem cells published from January 2001 to October 2013. The keywords were“stem cells, mesenchymal stem cells, umbilical cord-derived mesenchymalstem cells, cellculture, labeling methods”in Chinese and English, respectively. Final y, 35 articles were included in result analysis. RESULTS AND CONCLUSION:Umbilical cord-derived mesenchymal stem cells have not yet been widely used, mainly because of the immature isolation, culture and staining techniques of umbilical cord-derived mesenchymal stem cells. These techniques are worthy of further optimization studies. Although in recent years, cellmarkers and tracer technology of umbilical cord-derived mesenchymal stem cell s have made great progress, there are stil many problems need to be solved.
10.Serum uric acid level is positively related to albuminuria in type 2 diabetic patients

Objective To investigate association between serum uric acid (SUA),albuminuria and glomerular filtration rates (eGFR) in type 2 diabetic patients.Methods A total of 220 patients were enrolled in this cross-sectional study.According to urinary albumin excretion rates,patients were divided into 3 groups:normoalbuminuria (NAU) group,microalbuminuria (MAU) group,and macroalbumnuria group (MAAU).The first two groups were subdivided at SUA > 420 μmol/L (> 357 μmol/L,female) into normouricemia group and hyperuricemia group,at eGFR > 90 ml/min into high and low renal function groups.General information,blood biochemical results were collected to analyze the association between serum uric acid,eGFR,UAER and urine albumin quantification among different groups.Results The difference of SBP,duration of diabetes (DD),Scr,SUA and eGFR between every two groups were significant (P < 0.05).SBP,DD,Scr and SUA were highest in subjects with macroalbumnuria,second in microalbuminuria group,and lowest in normoalbuminuria group,while eGFR was lowest in macroalbumnuria group and highest in normoalbuminuria group.Prevalence of hyperuricemia in macroalbumnuria group (56.9%) and microalbuminuria group (51.2%) were also significantly higher than that in normoalbuminuria group (17.5%) (all P < 0.01).The difference of UAER in the subgroups of normouricemia and hyperuricemia was more significant in microalbuminuria group than in normoalbuminuria group.eGFR was significantly lower in hyperuricemia subgroups (P <0.01).Age and SUA were significantlg higher in subjects with low renal function compared with high eGFR (P < 0.05).Linear regression analysis indicated SUA was negatively correlated with eGFR after adjusted age,DD and UAER (β =-0.430,P < 0.01).Binary logistic regression analysis found that increased age,DD and SUA were risk factors of microalbuminuria β =1.092,95% CI(1.025,1.163),P < 0.01;β =1.005,95%CI(1.001,1.009),P < 0.05;β =1.407,95% CI(1.052,1.881),P < 0.05)] and SUA,age were risk factors of early renal function decline [β =1.015,95 % CI(1.00,1.023),P < 0.01;β =1.098,95% CI(1.006,1.199),P < 0.05].Conclusion SUA is independently associated with albumnuria and renal function decline in type 2 DM patients.
