After the first recognition occurs between the activated sperm and zona pellucida of the oocyte from the mammalians, ?-1,4-galactosyltransferase I (? 4GalT I) combines the N-GlcNAc terminals by O-ligands on the ZP3 of the zona pellucida, which plays a difunctional role in the fertilization. The G protein signal system on the sperm membrane then is consequently activated by ZP3 via ?4GalT I, contributing to the induction of acrosome reaction. It was proved that in the activation of the G protein system, both the BBXB and BBXXB motifs on the N terminal of the long ? 4GalT I are necessary.

Purpose To investigate the effects of chemotactic factor CCR4 on the abi1ity of pro1iferation,ce11 cyc1e,invasion,and mi-gration of human ga11b1adder cancer ce11. Methods Western b1ot was used to detect the expression 1eve1 of CCR4 in ga11b1adder carci-noma ce11s. Ga11b1adder carcinoma ce11s was infected by means of s1ow virus,the CCR4 gene si1encing was conducted using siRNA-CCR4 interference techno1ogy. Ga11b1adder carcinoma ce11s GBC-SD were divided into three groups( GBC-SD,GBC-SD/CCR4-RNAi and GBC-SD/contro1). CCL17,a 1igand of CCR4,was used to act on these three groups of ce11s. CCK8 method was used to detect the ce11 pro1iferation abi1ity of three groups. F1ow cytometry was used to test ce11 cyc1e. Tanswe11 assay was app1ied to detect ce11 migration and invasion abi1ity. Western b1ot was performed to detect the expression of its corresponding 1igands CCL17 and CCL22 proteins. Re-sults CCR4 gene si1ence did not inf1uence ce11 cyc1e and pro1iferation of ga11b1adder ce11 GBC-SD,but can significant1y inhibit GBC-SD ce11 invasion and movement abi1ity,CCR4 gene si1ence had no inf1uence on the expression of CCL17 and CCL22 gene in tumor ce11s. Conclusion Ga11b1adder carcinoma ce11s GBC-SD express chemokine receptor CCR4,chemokine receptor CCR4 can promote the invasion and metastasis of GBC-SD ce11s.
4. Comparison on the efficacy and safety of ultrasound-guided and X-ray guided for needle approach of percutaneous vertebroplasty

AIM: To compare ultrasound-guided vertebroplasty (percutaneous vertebroplasty, PVP) with X-ray guided vertebroplasty (PVP) to confirm the success rate of puncture point, X-ray radiation dose, operation time and postoperative outcome to explore the clinical application value of ultrasound-guided vertebroplasty. METHODS: Patients with thoracolumbar vertebral compression fractures treated by PVP in our hospital from November 2018 to October 2021 were divided into ultrasound-guided group and X-ray guided group. The success rate of puncture, the X-ray radiation dose, operation time and postoperative outcome were compared between the two groups. RESULTS: A total of 97 patients were included in this study, with an average follow-up time of (14.412±10.261) months. The success rate of one-time puncture was 60.4% in the ultrasound-guided group and 30.6% in the X-ray guided group (P<0.05). The X-ray radiation dose, fluoroscopy times and operation time in the ultrasound-guided group were significantly lower than those in the X-ray-guided group (P<0.05). The difference was statistically significant (P<0.05). The VAS of low back pain was significantly relieved in the two groups, but there was no significant difference between the two groups. There were no postoperative complications such as infection, puncture site hematoma and fracture nonunion between the two groups. CONCLUSION: Ultrasound guidance is a safe and effective method to determine the needle entry point of PVP. Skillful operation can improve the success rate of one-time puncture. Compared with traditional X-ray guidance, it can shorten the operation time, reduce the number of X-ray fluoroscopy and radiation during PVP operation.
5.Treating Displaced Patellar Fracture with Longitudinal Incision & Internal Fixation of the Sidelong Anterior Tension-band Wiring

Objective To explore the clinic effects of treating the displaced patellar fracture with a small longitudinal midline incision and internal fixation of the sidelong figure-of-eight anterior tension-band wiring. Methods 93 ea.ses of the displaced patellar fractures were internal fixated with the sidelong figure-of-eight anterior tension-band wiring(1.2 mm) simply, which made a longitudinal midline incision,it length can be made equal to the diam of patella approximately,that gives to enough expose the entire anterior surface of the patella and the patellar tendons. Results Follow-up for 6 to 30 months,93 cases were all obtained bony union. By Zhang Chun-cai score,there were 67 cases excellent,23 good and 3 fair,the excellent and good rate was 96.8%. Conclusion Treating patellar fracture with the fixation methods are very stabilization,and can functional exercise in early. It had similar results to reports using modified (with Kirschner wires) tension band techniques. The methods is simple and mini-trauma and invasive device minimally which results in fewer complications,especially as fractures traversed and comminuted with relative larger fragments are concerned.

ObjectiveTo evaluate the influence of electrotome on permanent and temporary cardiac pacemaker in laparoscopic cholecystectomy ( LC),and the application of cardiac pacemaker to the cases of cholecystolithiasis combined with bradyarrhythmia.MethodsClinical data of 215 patients with permanent or temporary cardiac pacemaker who underwent were studied for the preoperative and postoperative variation of pacemaker function,and for the influence of electricity coagulation during the operation on cardiac pacemaker function.ResultsLC was successfully completed in all 215 patients.The function of cardiac pacemaker was not obviously interfered during the operation,and the parameters of cardiac pacemaker did not remarkably change after the operation.ConclusionCardiac pacemaker is slightly interfered when electrotome and electrocoagulation were used in LC; LC is feasible and safe for patients with bradyarrhythmia by placement of cardiac pacemaker.
7.Anti-inflammatory Effects of Yuyang Capsule on Bacterial Dermatitis Model Rats and Its Effect on TLR/NF-κB Signaling Pathway

OBJECTIVE:To study the anti-inflammatory effects of Yuyang capsule on bacterial dermatitis model rats and its effect on TLR/NF-κ B pathway. METHODS:Minimum inhibition concentration (MIC)and minimal bactericidal concentration (MBC)of Yuyang capsule against Staphylococcus aureus were determined by microdilution test. Totally 50 SD rats were randomly divided into model group ,positive control group (amoxicillin and clavulanate potassium ,120 mg/kg as amoxicillin ),Yuyang capsules high-dose ,medium-dose and low-dose groups (according to MIC ),with 10 rats in each group. The model of bacterial dermatitis was established by using the burned skin of rats infected with S. aureus . 24 h after modeling ,administration groups were intragastrically given the corresponding drug ,and model group was intragastrically given the same amount of normal saline ,once a day,for consecutive 7 days. The skin healing rate was calculated on the 1st,3rd,5th and 7th day of administration ,and the scab formation,decrustation and healing were recorded. The contents of IL- 1β,IL-6,IL-10,TNF-α,hydroxyproline(HYP),collagen Ⅰ(Col Ⅰ)and Col Ⅲ in skin tissue were detected by ELISA. Morphology changes of skin tissues were observed by HE staining. The ultrastructure was observed by transmission electron microscope. Protein expression of TLR 2,TLR4 and p-NF-κB p65 were detected by Western blotting assay. RESULTS :MIC and MBC of Yuyang capsule were 25 mg/mL and 50 mg/mL,respectively. Dose of Yuyang capsules high-dose ,medium-dose and low-dose groups were set at 600,300,150 mg/kg. Compared with model group,scab appeared on the injured skin 3 days after administration in Yuyang capsule high-dose and medium-dose groups ,and decrustation appeared on the injured skin of part mice 5-7 days after administration ;the skin healing rate of the positive control group,Yuyang capsule high-dose and medium-dose groups were all significantly increased at each time point. The contents of IL- 1 β,IL-6,IL-10 and TNF-α,pathological score ,protein expression of TLR 2,TLR4 and p-NF-κB p65 were decreased significantly (P<0.05 or P<0.01);the pathological changes such as inflammatory cell infiltration in skin tissue were improved. The contents of HYP,Col Ⅰ and Col Ⅲ were increased significantly in positive control group and Yuyang capsule high-dose group (P<0.05 or P<0.01). There was no statistical significance in most above indexes in positive control group ,Yuyang capsule high-dose and medium-dose groups (P>0.05). CONCLUSIONS :Yuyang capsule can promote skin healing of bacterial dermatitis model rats and shows certain anti-inflammatory effects ;the mechanism may be related to inhibiting TLR/NF-κB signaling pathway and reducing inflammatory response.
